Abstract

The present study examines anatomy of Gayo mare reproductive organs. This study used three sample of Gayo mare reproductive organs (n= 3) for observation of morphology and morphometric of the mare reproductive organs. The ovarium was fixed in 4% paraformaldehyde solution then followed by histological method and stained using hematoxylin and eosin (HE) and Masson’s trichome (MT). Three mares were observed for diameter and changes overview of uterus during estrous cycle in real time using ultrasound. The results showed that, in general, the anatomy of Gayo mare’s reproductive organs was similar to other mares, but smaller in morphometry. The total length of the Gayo mare’s reproductive tract from labia to apex cornua was 48.00±1.00 cm. Weight of Gayo mare’s left ovary was 19.07±7.70 g and the right was 24.43±0.83 g. Histologically, there was no difference between Gayo mare’s structure and other mares. In cortex uteri there were some follicles surrounded by capillary, various development stages of follicles, healthy follicles, atretic follicle, and corpus albican; while in medulla there were a lot of connective tissues. Ultrasound of the uterus showed the change in diameter during estrous cycle with the largest diameter of corpus uteri was 4.43±0.10 cm in horses with estrous cycle of 21 days and 6.30±0.93 cm in horses with 24 days estrous cycle. In conclusion, the morphometry of Gayo mare reproductive organs are smaller than the other horses and there are differences in diameter of the uterus during the estrous cycle due to the changes of endometrium thickness.

Abstract

This study aimed to determine the biopotency of pregnant mare serum gonadotropin (PMSG) in stimulating the formation of dominant follicles and the biopotency of human chorionic gonadotropin (hCG) in triggering an increase in the ovulation rate of the natural-estrus donor group and the prostaglandin F2α (PGF2α) induced group using female Aceh cattle (n= 20) which were divided into two treatment groups: the group with natural estrus (A) and the group with PGF2α -induced estrus (B). Each donor group was superovulated using the injection of PMSGhCG (A1, B1) and only PMSG (A2, B2). The observations on total follicles and CL were carried out at D10, D14/D+0 and D21/D+7 using ultrasonography (USG). The results showed that PMSG gave a better superovulatory response to the development of dominant follicles (P0.01) at A1, A2, B1, and B2. The administration of hCG hormone 2000 IU i.m. was not able to increase the rate of ovulation. Total follicles after the addition of hCG were also not significantly different between A1:A2 and B1:B2. Statistically, the number of CL in all donor groups in D21 was not significantly different (P0.05). It concluded, superovulation using PMSG gave the same results to the donor cows with natural estrus and the donor cows with PGF2α-induced estrus. The hCG hormone with a dose of 2000 IU injected i.m. was also not able to increase the ovulation rate of Aceh cattle superovulated with PMSG 2500 IU i.m. ____________________________________________________

Abstract

This study to do examine the effectiveness of using heparin and hypotaurine to improve the in vitro fertilization rate. In first stage, oocytes were matured in the maturation medium supplemented with 0.3 mg/mL L-carnitine to improve nuclear maturation. In second stage, matured oocytes were fertilized with semen incubated in fertilization medium supplemented with 0.1 IU/mL heparin and 10 µM hypotaurine. The final concentration of sperm used was 5x106 spermatozoa/mL, and its interaction with oocytes was carried out for 12 hours in 5% CO2 incubator at 38.5° C. The results showed that percentage of oocytes that reached metaphase II was significantly higher (P0.05) in media supplemented with 0.3 mg/mL L-carnitine compared to control (88.2±4.9% vs 79.8±5.1%). Furthermore, fertilization rate was characterized by formation of 2 PN was significantly higher (P0.05) in media supplemented with heparin and hypotaurine than control (82.6±2.0% vs 73.3±2.4%). In conclusion, L-carnitine supplementation at dose of 0.3 mg/mL on maturation media was effective in increasing percentage of matured oocytes. The combination of 0.1 IU/mL heparin and 10 µM hypotaurine in fertilization medium was also successful in elevating the percentage of fertilized oocytes.

Abstract

The objective of this study was to examine the effects of repeated freeze-thaw cycles and short-term storage of fecal extracts at ambient temperature on the stability of fecal glucocorticoid (fGCM) and estrogen metabolite (fEM) levels from crested macaques.In total 100 aliquots of fecal extracts from fecal samples collected from female crested macaques (Macaca nigra) living at the Tangkoko-Batuangus Nature Reserve, North Sulawesi were used. We performed two different experiments: (1) An experiment to investigate if levels of fGCM and fEM measured from fecal extracts that were exposed to two, four, six and eight repeated freeze-thaw cycles (test groups) differ to control samples (i.e. fecal extracts always stored frozen); (2) An experiment to evaluate whether storing fecal extracts at ambient temperature for two, four, six, and eight days (test groups)affects the levels of fGCM and fEM compared to the control group (i.e. fecal extracts frozen immediately).Results showed that hormone levels were significantly increased (P0.05) after four freeze-thaw cycles for fGCM and after eight freeze-thaw cycles for fEM. By contrast, there was no significant difference (P0.05) in levels of fGCM and fEM between the test groups and the control group in fecal extracts stored at ambient temperature. In conclusion, our data show that more than two and six repeated freeze-thaw cycles should be avoided when measuring fGCM and fEM in crested macaque fecal extracts, respectively. We also demonstrate that storing fecal extracts at ambient temperature is possible for at least 8 days without taking a risk of affecting the stability of fGCM and fEM levels.

Abstract

This study aimed to obtain the ultrasound image of preovulatory follicle growth in gayo mares to predict the ovulation time. This study used three Gayo mares and one gayo stallion for teaser aged 5-10 years and weighed 200-250 kg. Preovulatory follicle growth was observed using transrectal ultrasonography which was performed when the dominant follicle size was ≥ 3.5 cm. Observations were made for 3 consecutive estrous periods on gayo mares at relatively the same time. The absence of previous follicular appearance and the presence of hypoechoic area in the same ovary indicate that ovulation has occurred. The result revealed that the diameter of preovulatory follicle of gayo mares was 4.02±0.32 to 5.05±0.04 cm. There was no significant follicle growth in the last 48 hours before ovulation and the preovulatory follicle shape altered from spherical to pearshape. Many small follicles (2 cm in diameter) were observed in peripheral area of the ovary indicated the recruitment of follicles which form the first follicular wave. To conclude, the alteration in the size and pearshape of preovulatory follicle and the recruitment of small follicles can be used as a marker to determine the time of ovulation on gayo mares.

Abstract

This study aimed to evaluate the effect of breeds and bulls ages on semen volume, sperm concentration, total number sperm, progressive motility, and sperm abnormalities of semen bull. The data used in the form of semen quality data from the Singosari National Artificial Insemination Center from the period 2021 to 2022. The bulls used each returned 4 bali, simmental and limousin. With age categories 3-4 years, 5-6 years, 7-8 years and 9-10 years. The research design used was a completely randomized factorial design. Data sperm quality were analyzed by general linear model method followed by LS mean. Sperm quality studied showed an interaction (P<0.05) between breed and age on the volume semen, sperm concenration, total number sperm,  progressive motility and sperm abnormalities. Bali cattle have a longer semen production period than simmental and limousine cattle. Bali cattle can be used until the age of 9-10 years, while simmental and limousine cattle can only be used until the age of 7-8 years.

Abstract

Pasak bumi is popular as an aphrodisiac. Traditionally, pasak bumi infusion is used to increase libido and duration of erection. The research was focused on: Activity level of FSH cells after three days of pasak bumi treatment. The research was carried out by evaluation using immunohistochemistry method via antibody anti FSH staining. The result of the research showed that a dosage of 18 mg/200 g body weight (bw) orally of pasak bumi maintained certain endoceine positive cells e.g. FSH production cells and were found stabile in the third days. Pasak bumi serves a stabilizer for intracellular levels of FSH in the anterior hipophysis/pituatary.

Abstract

Standar kualitas semen yang berperan penting dalam keberhasilan inseminasi buatan (IB) sebagai upaya peningkatan populasi dan mutu ternak sapi di Indonesia terbukti tidak seutuhnya mampu menentukan tingkat fertilitas sapi pejantan di Indonesia. Studi terbaru membuktikan bahwa meskipun sudah memenuhi standar kualitas semen yang sudah ditetapkan, namun tiga dari enam ekor sapi pejantan di Indonesia masih tergolong fertilitas rendah (<60%). Potamine-1 (PRM1) merupakan molekul pada inti spermatozoa yang telah terbukti memiliki hubungan erat dengan fungsi fertilitas dan kualitas spermatozoa pada berbagai sapi pejantan di Indonesia. Hal tersebut mendasari usulan pengembangan uji berbasis molekuler PRM1 sebagai penentu tingkat fertilitas yang diharapkan dapat digunakan untuk proses seleksi dan kebijakan pengafkiran sapi pejantan unggul di Indonesia. Melalui policy brief ini diharapkan usulan pengembangan uji tersebut dapat lebih tersampaikan dengan segala solusi yang diberikan, termasuk terkait pendekatan uji berbasis molekuler yang paling tepat digunakan, kajian-kajian lanjutan, dan berbagai harapan dukungan dari segala pihak demi terwujudnya pengembangan uji PRM1.

Abstract

Whiteleg shrimp (Litopenaeus vannamei) is a major aquaculture commodity that continues to be developed through intensive culture systems. This study aims to evaluate the growth patterns and survival rates of L. vannamei reared in an intensive aquaculture system at the Busmetik pond of the Sorong Marine and Fisheries Polytechnic during the 2021–2024 period. Observed parameters included average daily growth (ADG), average body weight (ABW), survival rate (SR), feed management practices, and water quality indicators. Data were analyzed using ANOVA and Kruskal-Wallis tests to identify interannual differences. Results revealed a consistent increase in SR, from 78.05% in 2021 to 91.7% in 2024, along with a steady improvement in shrimp length and weight, although the differences were not statistically significant (p > 0.05). Factors such as feed quality, feeding frequency, water pH and dissolved oxygen (DO), as well as disease control measures, were shown to influence culture performance. These findings suggest that integrated and adaptive management practices play a critical role in enhancing the growth efficiency and survival of L. vannamei in intensive aquaculture systems. Keywords: Litopenaeus vannamei, intensive culture, growth performance, survival rate, water quality

Abstract

This study aims to observe the changes in the characteristics of vaginal mucus to diagnose early pregnancy in dairy cattle. The characteristics of vaginal mucus include pH value, consistency, volume, and Vaginal Electrical Resistance (VER) value to diagnose early pregnancy using a Draminski Estrous Detector. The sample used that collected in this research was 52 dairy cattle. The research method consists of taking samples and observing the characteristics of the mucus. Data analysis includes normality test, T-test and Pearson correlation. The results showed that the mucus volume of pregnant cattle was less (1.280.356 mL) than that of non-pregnant cattle (2.210.437 mL), the pH value in pregnant cattle tended to be acidic, while non-pregnant cattle were classified as neutral, and there were differences in the consistency of mucus in pregnant cattle (4.00.0), while non-pregnant cows tended to be liquid (2.60.204). The volume and consistency of vaginal mucus in pregnant and non-pregnant cattle were significantly different (P0.05). The VER value of pregnant cattle was higher (36563.2 ohms) than non-pregnant cattle (19055.3 ohms). The VER value was highly correlated (r= 0.819) with the pregnancy status of dairy cattle. Thus, VER can be used as an accurate indicator to determine the status of early pregnancy in dairy cattle.
