Abstract

The objective of this study was to detect histological structure and histomorphometry of bali cattleand wagyu beef. The samples were collected from rib eye muscle of wagyu and bali cattle. The tissuesamples were fixed, dehydrated and embedded in paraffin and 4-5 mm sections, for histological examinationand stained with Haematoxilin-Eosin (HE). The light binocular microscope was used to identify the samples.The study showed that the beef histological structure consisted of: cells / muscle fibers, fatty and connectivetissues. The muscle fibers of wagyu were observed larger, regular, and longer compared to the Bali cattle.Additionally, intramuscular fat cells and connective tissue interlobulus were found less and thinner thanthe Bali cattle. In the area of muscle where the longitudinal cut was done, its observed that the presence ofI and A bands in Bali cattle beef were clearer compared to the wagyu. The diameter muscle fibers of balicattle beef (45.00 ±1.94 ?m), was smaller (P < 0.01) compared to the wagyu (75.00±1.82 ?m), as well as fatcell size of bali cattle beef (90.10 ±2.13 ?m) and wagyu beef (195.00±2.57?m). In conclusion, the structureof histology and histomorphometry of rib eye muscle in Bali cattle were different to the wagyu.

Abstract

The research objective want to track the expression of Toxoplasma gondii antigenes in the heart of the free-range chicken with immuno(cyto)histochemistry technique. In the present study were examined three methods to track antigen T.gondii, first method direct microscopic examination of the heart digests, the second method to detect antigen of T.gondii with immunocytochemistry technique of the chicken heart digests and the third immunohistochemistry examination of the heart free-range chicken. The number of material samples examined were 100 heart free-range chicken. Direct microscopic examination of the heart digests free-range chicken to track the bradyzoite form (inside cyst). Examination by immuno(cyto)histochemistry technique keep track T.gondii an antigen expression on cells and heart muscle tissue. The results showed that the direct microscopic examination on the heart tissue unable to detect cyst and antigen T.gondii. Immunohistochemical examination successfully detected the expression of antigenes T.gondii and was found 2% (2/100) positive. It can be concluded that T.gondii antigen expression in the heart of range chicken could be detected by immunohistochemistry.

Abstract

The purpose of this study is to make a monoclonal antibody against- Cysticercus cellulosae and itscharacterization. Samples antigen prepared from T. solium larvae (C. cellulosae) was then used to immunizeBalb/c. The immune response of mice assessed by ELISA test, then the lymphocytes of mice used for theproduction of monoclonal antibodies (MoAb). Origin lymphocytes of mice that produce antibodies againstC. cellulosae antigen, fused with myeloma cells (NS1). Results fusion of two cells produces hybrid cellscalled hybridomas; cells are then screened by ELISA test. Hybridoma cells that produce only MoAb, usedto produce large quantities in vitro. Characterization of MoAb against-C.cellulosae was tested by usingELISA and Western blotting. Mice were immunized with C.cellulosae antigen showed an immune responseproducing antibodies to C.cellulosae. Based on the results of fusion, produced a total of 51 hybridoma cellclones and after being screened, only three clones of hybridoma cells that produced MoAb against–C.cellulosae. MoAb produced, named after the hole where the growth of the ELISA micro plate, the BE6,BE7, and EE9. Characteristics of this MoAb capable of tracking cellulosae of fluid larvae and recognizeantigen protein bands with molecular weight 78kDa.

Abstract

Bali sejak 2008 mengalami kasus rabies dan saat ini menjadi daerah endemis rabies. Berbagai cara telah dilakukan untuk pengendaliannya namun kasusnya masih tetap ada. Diperlukan pendekatan one health yang berkolaborasi antara kesehatan manusia, hewan dan lingkungan. Tujuan dari penelitian ini untuk mengetahui tantangan program, peranan pemangku kepentingan dalam pengendalian rabies dan upaya pendekatan one health yang terintegrasi. Penelitian ini menggunakan pendekatan kualitatif untuk mengetahui tantangan program, peranan pemangku kepentingan dan pendekatan one health terintegrasi. Informan dipilih sesuai dengan kebutuhan peneliti. Hasil penelitian menunjukan tantangan dalam pelaksanaan program pada sistem pendataan masih kurang, vaksinasi anjing belum optimal, cara memelihara anjing yang masih diliarkan, program kontrol populasi, manajemen kasus gigitan anjing belum terpadu, edukasi masih rendah, partisipasi masyarakat yang rendah. Perlu peranan dari semua pemangku kepentingan untuk pengendalian rabies. Program pengendalian rabies dapat terlaksana dengan baik apabila mampu mengurangi kesenjangan yang terjadi. Masing-masing kelompok pemangku kepentingan berperan dalam program pengendalian rabies. Melalui pendekatan one health yang terintegrasi dilaksanakan kegiatan yang berkolaborasi dalam surveilan epidemiologi, manajemen kasus gigitan terpadu, manajemen hewan penular rabies dan faktor sosial-budaya-ekologi lingkungan.

Abstract

Cysticercosis is a zoonotic parasitic disease which is still problem in Indonesia. The purpose of thisstudy was to investigate immune responses of mice that had been immunized using Taenia solium larval(Cysticercus cellulosae) antigens originated from infected pigs. Three kinds of the C.cellulosaeantigens,secretory and whole antigens were used to immunize three different groups Balb/c mice consisted of 15animals. The serum samples before and after immunization were tested with ELISA test. The resultsshowed that the third antigens induced highly significant titre (P<0,01)compared to unimmunized animals.However no significant different (P>0,05) were found when the third antigens were compared. It wasconcluded that immunization with the three kinds of C. cellulosae antigens can cause immunity in mice.

Abstract

Mouse has been an animal model that is very sensitive towards the Toxoplasma gondii infection. This research infects 30 mice with free-range chicken heart inoculate and the detection of T. gondiiby immunohistochemistry on the infected mice. The purpose of this study is to track T.gondii antigen expression by immune(cyto)histochemistry in mice. Observation over mice was conducted over 10 weeks. After 10 weeks, the mice are examined for its peritoneal fluid by microscopic and immunohistochemistry examination. Continued by the examination of internal organs to track the expression of T.gondii antigens by immunohistochemistry. The study result showed microscopic examinations on peritoneal fluid did not found any cyst or tachyzoites form of T.gondii. Examination of peritoneal fluid by immunohistochemistry was found to detect expressions of T.gondii antigen. Examinations of the mice’s internal organs detected expressions of T.gondii antigen only in the brain by immunohistochemistry examinations.

Abstract

Kualitas daging adalah ukuran yang penting dalam hal palatabilitas dan penerimaan kepada konsumen. Kandungan gizi yang tinggi dalam daging merupakan media yang sangat baik untuk pertumbuhan mikroorganisme sehingga menyebabkan daging mudah rusak dan busuk. Evaluasi ini bertujuan untuk mengetahui kualitas daging dan produk olahan daging dengan menggunakan sampel yang didapat dari pasar tradisional di Kota Denpasar seperti Pasar Batu Kandik, Pasar Poh Gading, Pasar Badung, dan Pasar Kumbasari. Pengujian ini dilakukan di Laboratorium Kesehatan Masyarakat Veteriner dan Epidemiologi Veteriner, Fakultas Kedokteran Hewan, Universitas Udayana. Pengujian dilakukan secara subjektif dan objektif. Uji subjektif daging dan olahan daging meliputi warna, aroma, konsistensi, tekstur, tenunan pengikat, serta kepualaman. Uji objektif daging meliputi uji pH, daya ikat air, kadar air, dan cemaran mikrob. Pengujian daging dan olahan daging menunjukkan bahwa warna, aroma, dan konsistensi dalam beberapa sampel dikategorikan normal sesuai dengan jenis daging dan produk daging, kecuali daging ikan dari Pasar Batu Kandik yang konsistensinya berair dan memiliki aroma amis. Tenunan pengikat dan kepualaman daging sapi dan babi masing-masing memiliki mutu II dan 10%, sedangkan daging ayam dan ikan memiliki mutu I dan 0%. Pada uji objektif, kadar pH daging dikategorikan normal berdasarkan jenisnya berkisar 5,4-6,4, sedangkan produk olahan daging seperti bakso berkisar 6-7 dan sosis 6-6,5; daya ikat air berkisar 64,43-83,75%; kadar air berkisar 71,7-81,9%, dan angka lempeng total bakteri (ALTB) 196 x 103-440 x 103 CFU/g. Namun, pertumbuhan Coliform melebihi batas normal dengan nilai 68-222 CFU/g. Hasil dari pengujian tersebut menunjukkan produk olahan daging memiliki kualitas baik. Seluruh daging dari Pasar Poh Gading dan Pasar Batu Kandik memiliki nilai organoleptik yang baik, kecuali daging ikan dari Pasar Batu Kandik. Seluruh daging memiliki nilai cemaran Coliform di atas batas normal kecuali daging ikan dari Pasar Poh Gading.
