Abstract

Previous studies showed the potential of melatonin of green algae ( Ulva lactuca L ) to reduce the risk of coronary heart disease with the activity of antihiperlipidemia. Obesity and hiperlipidemia were the risk factor of degenerative diseases such as heart disease. This research aims was explore the effect of ethanol extract of green algae against weight, consumption of rat feed test white male rat were given high fat diet . Research was started with extraction green algae with ethanol 96 % to obtained concentrated extract. The extract was tested to wistar rats age of 2 months. Animals test divided into 6 group. Group I was control group, treated with high fat diet of lard 2 ml / 200g BW, of group II given high fat diet and simvastatin, group III, IV and V given high fat diet and green algae extract doses 50 mg/ Kg BW, 100 mg/Kg BW and 200 mg/ Kg BW. Group VI is the control group without diet fat high. Treatment was conducted over 28 days. Measuring normal of his weight every five day for 28 days and in measuring consumption feed with measure weight feed the rest of feed early 20 g. The difference of data weight rats per 5 days and consumption fodder for 28 days counted area under curve ( AUC ) from curves time versus weight. The measurement of triglycerides levels. The result showed the high fat diet with lard 2 ml / 200g BW for 28 days reduce weight and consumption feed mice in a significantly than control without high fat diet. The treatment with extract ethanol dose of 50 , 100 and 200 mg / Kg BW did not reduce weight, feed consumption, triglyceride levelsof  mice were given high fat diet.

Abstract

Penelitian sebelumnya membuktikan bahwa pemberian ekstrak pegagan (Centela asiatica) dapat mencegah peningkatan ekspresi protein caspase 3 dan menurunkan ekspresi Bcl-2 yang berkaitan dengan apoptosis di daerah CA1 dan CA2-CA3 sel pyramidal hippocampus  pada tikus model demensia.  Selain ekspresi protein tersebut, mekanisme apoptosis juga melibatkan protein lain yaitu caspase 8, yaitu protease sistein yang berperan sebagai inisiator apoptosis di jalur ekstrinsik. Tujuan penelitian ini adalah untuk mengetahui efek ekstrak pegagan dalam mencegah ekspresi gen caspase 8 di sel pyramidal hippocampus pada tikus model demensia yang diinduksi dengan trimetiltin (TMT). Sebanyak 30 ekor tikus Sprague Dawley, 3 bulan, jantan, berat 200-300 g dibagi menjadi 6 kelompok.  sebagai berikut: Kelompok Normal dan TMT diberi CMC-Na,  kelompok ekstrak diberi ekstrak dosis 100 (EP-100), 200 (EP-200)  dan 400 mg/kg BB (EP-400), sedangkan kelompok Sitikolin diberi sitikolin dosis 200 mg/kg B. Pemberian perlakuan selama 35 hari.  Injeksi TMT dilakukan pada hari ke-8 perlakuan dengan dosis 8 mg/kg BB secara intraperitoneal, kecuali pada kelompok normal. Pada hari ke 36 tikus dikorbankan, otak diambil, kemudian bagian hippocampus dipisahkan untuk pembuatan preparat imunohistokimia untuk pengamatan ekspresi gen caspase 8 di sel pyramidal daerah CA1 dan CA2CA3. Jumlah sel yang mengekspresikan gen caspase 8 pada sel pyramidal di daerah CA1 dan CA2CA3 dianalisis dengan uji anava satu jalur dilanjutkan dengan uji LSD dengan tingkat signikansi 0.05. Hasil penelitian menunjukkan bahwa pemberian TMT meningkatkan secara signifikan jumlah sel yang mengekspresikan caspase 8 di daerah CA1 namun tidak pada daerah CA2CA3. Pemberian ekstrak pegagan dosis 400 mg/kg BB menurunkan secara signifikan ekspresi caspase 8 namun tidak terjadi pada dosis 100 dan 200 mg/kg BB. Dengan demikian ekstrak pegagan berpotensi dikembangkan untuk  mencegah demensia.

Abstract

ABSTRAK Kasus demam berdarah yang tinggi mengharuskan masyarakat untuk berusaha mengendalikan laju penyebaran virus dengue dengan menghindari atau mengurangi kontak dan gigitan dengan nyamuk. Dalam usaha mencari bahan alam yang mempunyai efek repelan, telah dilakukan uji efek repelan kombinasi minyak atsiri rimpang temulawak dan rimpang jahe terhadap nyamuk Aedes aegypti. Rimpang temulawak dan rimpang jahe diisolasi minyak atsirinya dengan menggunakan metode destilasi uap dan air. Bahan uji dibuat dalam berbagai konsentrasi 2% v/b, 5% v/b, 10% v/b, 15% v/b, 20% v/b, dan 25% v/b. Kontrol positif digunakan adalah repelan DEET 12,5% dan kontrol negatif digunakan basis unguentum leniens. Metode yang digunakan adalah metode “Comparative Efficacy of Insect Repellent against Mosquito Bites” yang telah dimodifikasi. Efek repelan dianalisis dari waktu penolakan yaitu waktu pertama kali nyamuk menggigit. Hasil percobaan diperoleh bahwa semakin besar konsentrasi minyak atsiri dalam repelan maka semakin besar waktu penolakan nyamuk Aedes aegypti. Repelan dengan konsentrasi 25% v/b (konsentrasi terbesar) merupakan sediaan yang paling baik dan memenuhi persyaratan sebagai repelan karena mempunyai waktu penolakan yang paling lama. Kata kunci : minyak atsiri, Aedes aegypti, unguentum leniens, repelan ABSTRACT The highes of the dengue fever cases make the society have to control the spread of the dengue virus with avoid and decrease contact and mosquito bite. In find the natural resource that has repellant effect, have been done repellant research effect with volatile oil combines from Curcuma xanthorrhiza rhizome and Zingiber officinale rhizome to Aedes aegypti. Volatile oil of Curcuma xanthorrhiza rhizome and Zingiber officinale rhizome were isolated using steam and water destilation method. The test composition made with different concentration, that are 2% v/b, 5% v/b, 10% v/b, 15% v/b, 20% v/b, and 25% v/b. The positive control is repellant DEET 12,5% and the negative control is unguentum leniens base. The method of this test is “Comparative Efficacy of Insect Repellent against Mosquito Bites” which modified. Repellant effect analyzed from refusal time, that is mosquito’s bite in the first time. The research result is higher concentration of volatile oil followed by increasing of refusal time on Aedes aegypti. Repelant with 25% v/b concentration (the biggest concentration) were the best preparation and good in standard as repellent. It caused that concentration had longest refusal time. Abstract

Sweet potato (Ipomoea batatas L. Lam) is a plant source of carbohydrates with chemical content of catechins, quercetin, asam klorogenat, anthocyanins, beta-carotene. These phytochemical compounds have been shown to have antioxidant activity that is closely related to tyrosinase through the oxidative stress pathway. The purpose of this study was to develop sweet potato plants as anti-tyrosinase candidates for cosmetic preparations by molecular docking of these 4 compounds to tyrosinase protein (pdb:2Y9X) with kojic acid control. The research was done in 4 stages : (1) Preparation of proteins (receptors) and ligands with Biovia Discovery Studio (2) Optimization of geometry with Avogadro and Chem 3D (3) Validation of docking and molecular docking methods for proteins (2Y9X) with Autodock-4 (5) and (4) ADMET study with pkCSM and SwissADME websites. The test results obtained the highest affinity for 3 active compounds, namely beta-carotene with (ΔG) -6.05 kcal/mol (IC) 36.72 M and quercetin has (ΔG) -5.87 kcal/mol (IC) 45.30 M . While the control kojic acid has a lower affinity of (ΔG) -5.27 kcal/mol (IC) 380.37 M. ADMET predictions provide information that the four active compounds Ipomoea batatas L. Lam meet the requirements to be developed as natural tyrosinase inhibitors based on lipophilicity (logP), topology polar surface area (TPSA), skin permeability, skin sensitivity, logKP, CYP1A2 inhibitor, lipinski rule. and bioavailability. The molecular docking study and ADMET prediction indicate that the active compound of sweet potato (Ipomoea batatas L. Lam) has the potential to be developed as a natural cosmetic ingredient.

Abstract

Daun jarak pagar (Jatropha curcas L.) mengandung beberapa senyawa aktif, seperti alkaloid, saponin, tannin, terpenoid, steroid, glikosida, senyawa fenol, dan flavonoid yang diduga dapat menurunkan kadar trigliserida dalam serum darah, sehingga dapat mencegah keadaan hipertrigliseridemia. Penelitian  ini bertujuan untuk mengetahui aktivitas penurunan kadar trigliserida serum setelah pemberian ekstrak etanol daun jarak pagar (EEDJP) variasi dosis pada tikus hipertensi yang diinduksi dengan NaCl dan diberi pakan lemak tinggi. Penelitian eksperimental dengan pre-posttest control group design terhadap tikus jantan galur Wistar. Tikus dibagi menjadi 7 kelompok, yaitu  kelompok normal, kontrol (diinduksi NaCl dan diberi pakan  tinggi lemak), Captopril, Simvastatin, EEDJP dosis 1,8 g/KgBB, 2,7 g/KgBB, 4,05 g/KgBB. Analisis kadar trigliserida serum dengan metode GPO- PAP. Data dianalisis   dengan uji One-Sample Kolmogorov-Smirnov Test, Homogenity of Variances, One-Way ANOVA, Kruskal-Wallis, dan Mann-Whitney. Induksi NaCl dan pemberian pakan lemak tinggi dapat meningkatkan kadar trigliserida serum yang signifikan (p<0,050). Pemberian EEDJP dapat menurunkan kadar trigliserida serum yang signifikan (p<0,050) pada semua variasi dosis. Kesimpulannya EEDJP dapat menurunkan kadar trigliserida secara signifikan pada dosis 1,8 g/KgBB, 2,7 g/KgBB, 4,05 g/KgBB.

Abstract

The rare sulphated polysaccharides of green algae have been explored of several activities such as antioxidant, anti-inflammation, anti-bacterial, and anti-virus. Ulva lactuca L is one of the majoring algae species in Indonesia that promosing to be explored as natural anti-inflammation in the inflammation disorders treatment. The purpose of this study was to conduct an in-silico test of anti- inflammatory potency of sulphated polysaccharide chemical constituent of green algae (Ulva lactuca L) against the cyclooxygenase-2 enzyme. The methods used were the preparation of a protein structure database cyclooxygenase-2 enzyme (4COX), protein preparation using the Biovia Discovery Studio application, molecular docking simulation of sulphated polysaccharide compounds on proteins using the Autodock 4.0 application and visualization using Ligpot+ v2.2. The results of docking sulphated polysaccharide compounds with the cyclooxygenase-2 enzyme, showed a best binding affinity energy of Gluconic acid ulvan -7.62 kcal/mol similar to the control drug sodium diclofenac (-7.81 kcal/mol), followed by Iduronic acid Ulvan -7.57 kcal/mol, Fucoidan (-6.11 kcal/mol), Alpha Carrageenan (-6.93 kcal/mol), and Lambda Carrageenan (-5.38 kcal/mol). In the conclusion based on the molecular docking result, the sulphated polysaccharide compounds in Ulva lactuca L are potential to be developed as natural antiinflammatory agent by in vitro and in vivo investigation

Abstract

Binahong leaves are plants that have great potential as antihyperglycemics, and have chemical content including triterpenoids, flavonoids, and saponins. However, the development of binahong leaves in pharmaceutical preparations is still very lacking. Objectives of this study are knowing the transdermal patch formulation of binahong leaf extract (BLE) produces good physical properties and the development of BLE transdermal patches with different extract concentrations may blood glucose lower (BGL) levels in glucose-induced mice. This study uses a single-layer patch because it has the advantage of being able to penetrate faster and is suitable for compounds that have solubility in polar solvents. The research began with the extraction of binahong leaves, then formulated a transdermal BLE patch using a formula that includes HPMC (polymer), glycerol (plasticizer), oleic acid (enhancer) and tween 80 (enhancer). Diabetic rats were divided into 4 groups of 6 rats, negative control group (glucose + placebo), positive control group (glucose + insulin), FI group (glucose+ transdermal patch BLE 30%), and FII group (glucose + transdermal patch BLE 47%). BGL measurements are carried out on the 0th, 3rd, 5th, and 7th days. The physical characterization carried out includes organoleptic, weight uniformity, thickness, pH, folding resistance, and moisture content, obtained FI and FII results that meet the requirements set out by each test standard. BGL (mg/dL) from day 1 to day 7 decreased in each group, positive control (68.8±3.76), negative control (112.5±6.89), FI (81.0±4.04), and FII (72.33±5.12). Based on the statistical results of the T-test on the physical evaluation of BLE transdermal patch preparations of FI and FII there were significant differences in weight and thickness uniformity, but there were no significant differences in pH, folding power, and humidity tests. The administration of a 47% BLE transdermal patch provides a faster but insignificant BGL reduction effect compared to the 30% BLE FI transdermal patch.

Abstract

Penelitian ini bertujuan untuk mengetahui kemampuan mahasiswa dalam menyelesaian soal Higher Order Thinking Skills (HOTS) menurut taksonomi Bloom hasil revisi yaitu C4, C5, dan C6. Pengujian dilakukan pada 22 orang mahasiswa di Program Studi Pendidikan Fisika Universitas Papua Semester Genap 2016-2017 yang mengontrak mata kulah Laboratorium Sekolah. Instrumen yang digunakan dalam penelitian ini adalah soal pada materi tekanan yang terdiri dari 25 soal pilihan ganda. Penelitian ini merupakan penelitian deskriptif dengan menggunakan pendekatan kuantitatif melalui pemodelan Racsh. Hasil penelitian menunjukkan bahwa rata-rata nilai mahasiswa dalam menyelesaikan soal C4 yaitu 53,18, soal C5 yaitu 52,27 sedangkan soal C6 yaitu 68,83 dengan Rerata 57,27 ± SD 16,70 berada pada kategori baik. Pemodelan Rasch menunjukkan bahwa soal nomor 14 dan nomor 7 yaitu soal C4 (analisis) memiliki tingkat kesukaran paling tinggi untuk diselesaikan mahasiswa. Soal  nomor 5 dan nomor 6 yaitu C6 (mencipta) memiliki tingkat kesulitan paling rendah. Hasil penelitian menunjukkan bahwa kemampuan mahasiswa dalam menyelesaikan soal tingkat tinggi masih rendah terutama pada soal C4 dan C5.Kata-kata kunci: Higher Order Thinking Skill (HOTS),  menganalisis, mengevaluasi, dan mencipta

Abstract

Sambiloto (Andrographis paniculata) has been known empirically as a plant that has benefits as a traditional antidiabetic Javanese medicine to treat diabetes mellitus (DM). The main compound contained in sambiloto is andrographolide which has a hypoglycemia effect. Andrographolide has low solubility in water and poor bioavailability due to biotransformation by P-glycoprotein in the intestinal tract which causes andrographolide to metabolize more quickly in the duodenum and jejunum. To overcome this extract ethanol of sambiloto (EES) formulated to patch transdermal. Enhancers can affect the characteristics of transdermal patch preparations which affect the pharmacological activity of transdermal patch preparations. The critical point of transdermal patch preparation is the selection of enhancers which is one of the compounds to increase the penetration of active substances in the skin by disrupting the permeability of the stratum corneum. The aim of this research is to determine the effect of variations in the combination of oleic acid and tween 80 as an enhancer on the physical characteristics of EES patches.The methods of extraction sambiloto use soxhetation with ethanol 96% as solvent. The formulation of EES patch transdermal using solvent casting technique with various concentration enhancer tween-80 and oleic acid. This research was conducted by making 3 formulas, where F1  (1:1), F2 (1:3) and F3 (1:3). The evaluations include weight uniformity, thickness, water content, pH and folding endurance of the patch. The result of visual appearance the patch EES has a dark green with aroma specific of EES. The characteristics of EES transdermal patches were affected by the ratio of tween-80 and oleic acid as the enhancers. The combination of oleic acid and tween 80 as a permeation enhancer has an influence on physical characteristics (weight and thickness) of EES patches.

Abstract

Background: One of the main risk factors for cardiovascular diseases such as coronary atherosclerotic heart disease (CAHD) is dyslipidaemia or high levels of low-density lipoprotein (LDL) and triglycerides (TG) and low levels of high-density lipoprotein (HDL). Hypertension is also a cause of cardiovascular disease. One potential plant to lower LDL levels and blood pressure is Jatropha curcas, which is known to contain saponins, polyphenols, and flavonoids. Objective: The purpose of this study was to determine the effect of the ethanol extract of Jatropha curcas leaves (EEJCL) on blood pressure, LDL levels, and HDL levels in hypertensive rats given a high-fat diet. Methods: This study is an experimental study with a pretest-posttest control group design on male Wistar strain rats. Rats were divided into seven groups, namely the normal group, control group (induced with NaCl and given a high-fat diet), Captopril group, Simvastatin group, and EEJCL groups given doses of 1.8, 2.7, and 4.05 g/kg BW. The data obtained were analysed using the One-Sample Kolmogorov-Smirnov Test, Homogeneity of Variance, One-Way ANOVA, and Tukey Test. Results: The results showed that the administration of EEJCL could significantly lower LDL levels and blood pressure and increase HDL levels (p < 0.05) at doses of 1.8, 2.7, and 4.05 g/kg BW, and the dose of 4.05 g/KgBW was the most optimal dose. Conclusion: EEJCL has a potential for development in the treatment of hypertension and dyslipidaemia.
