Abstract

Diabetes melitus adalah penyakit metabolik yang ditandai dengan hiperglikemia dimana pankreas tidak memproduksi cukup insulin atau tubuh tidak dapat menggunakan insulin secara efektif. Daun yakon, kayu secang dan daun salam memiliki beberapa yang berpotensi sebagai antidiabetes, namun belum diketahui senyawa spesifik mana yang terlibat dalam mekanisme inhibitor SGLT-2. Penelitian ini bertujuan untuk menganalisis potensi senyawa bioaktif yang terkandung dalam Daun yakon, kayu secang dan daun salam dalam menghambat enzim SGLT-2 secara in silico. Data senyawa aktif didapatkan dari laman PubChem. Kristal struktur dari Structure of human SGLT2-MAP17 complex bound with empagliflozin didapatkan dari Protein Data Bank dengan PDB ID: 7VSI. Molecular docking menggunakan Molegro Virtual Docker dan simulasi molecular dynamic menggunakan YASARA Dynamics. Hasil molecular docking didapatkan 4 senyawa memiliki nilai rerank score lebih negative dari senyawa pembanding (empagliflozi). Hasil visualisasi menunjukan residu asam amino yang berperan penting ialah PHE 98 dan VAL 157. Hasil molecular dynamics menunjukan kompleks ligan memiliki kestabilan ikatan paling baik dan stabil dari ligan uji lainnya. Dihasilkan senyawa yang stabil, yaitu 3,4-Dicaffeoyquinic acid.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh latihan naik turun tangga dan latihan tanjakan terhadap peningkatan kecepatan lari 100 meter pada atlet U18 binaan club yang ada di kabupaten Pesawaran yaitu Atletik Teluk Pandan Club & Haidar Running Club. Metode penelitian menggunakan eksperimen dengan desain two group pretest-posttest. Sampel penelitian berjumlah 10 atlet sprint Putra dengan rata-rata Latihan diatas 1 tahun yang dibagi menjadi dua kelompok perlakuan. Kelompok pertama diberikan latihan naik turun tangga dan kelompok kedua diberikan latihan tanjakan. Instrumen yang digunakan adalah tes lari 100 meter. Perlakuan diberikan selama 6 minggu dengan frekuensi latihan tiga kali per minggu dengan tambahan program lainya sama. Analisis data menggunakan uji t. Hasil penelitian menunjukkan bahwa kedua metode latihan memberikan pengaruh yang signifikan terhadap peningkatan kecepatan lari 100 meter, namun latihan tanjakan menunjukkan peningkatan yang lebih besar dibandingkan latihan naik turun tangga. Disimpulkan bahwa latihan tanjakan lebih efektif dalam meningkatkan performa sprint 100 meter dan Latihan naik turun tangga juga dapat digunakan, guna meningkatkan kecepatan frekuensi ayunan kaki dan tangan serta power. Abstract This study aimed to determine the effect of stair up-and-down training and hill training on improving 100-meter sprint speed in U18 athletes fostered by clubs in Pesawaran Regency, namely Atletik Teluk Pandan Club and Haidar Running Club. The research method used was an experimental design with a two-group pretest–posttest design. The sample consisted of 10 male sprint athletes with an average training experience of more than one year, divided into two treatment groups. The first group was given stair up-and-down training, while the second group received hill training. The instrument used was a 100-meter sprint test. The treatment was conducted for 6 weeks with a training frequency of three times per week, while other additional training programs were kept the same. Data analysis used the t-test. The results showed that both training methods had a significant effect on improving 100-meter sprint speed. However, hill training showed a greater improvement compared to stair up-and-down training. It can be concluded that hill training is more effective in improving 100-meter sprint performance, while stair training can also be used to increase the frequency of arm and leg swing as well as power.

Abstract

Background: Diabetes mellitus (DM) is a chronic metabolic disease with a steadily increasing prevalence globally. Long-term use of synthetic antidiabetic drugs is often associated with side effects, making exploration of alternative therapies from natural ingredients important. Pluchea indica L. leaves have been reported as a traditional Indonesian plant empirically used to lower blood sugar levels. Objective: This study aims to explore the potential of active compounds from Pluchea indica L. leaves as candidate multi-target antidiabetic agents through an in silico approach. Methods: An in silico study was conducted using molecular docking with Molegro Virtual Docker (MVD) to evaluate the interaction of four active compounds from beluntas leaves [(+)-Lirioresinol B, β Stigmasterol, (+)-Pinoresinol, and Plucheoside A against diabetes target proteins DPP-4 (PDB: 4FFW), α-glucosidase (PDB: 3L4W), SGLT-2 (PDB: 7VSI), and PPAR-γ (PDB: 5UGM)]. Method validation was performed by re-docking the original ligands (RMSD < 2.0 Å). Lipinski's 5 Rule analysis and ADMET prediction were performed to evaluate the drug-likeness and pharmacokinetic profiles of the compounds. Results: Method validation showed RMSD of 0.31-1.71 Å for all target proteins. Docking results showed that (+)-Pinoresinol had the best affinity as a DPP-4 inhibitor (Rerank score -103.452), (+)-Lirioresinol B as an α-glucosidase inhibitor (-100.173), Plucheoside A as an SGLT-2 inhibitor (-126.555), and β Stigmasterol as a PPAR-γ agonist (-131.023). Lipinski's 5 Rules analysis showed that all compounds most often violated 1 criterion (PSA). ADMET predictions showed an acceptable pharmacokinetic profile with low toxicity. Conclusion: The active compounds from beluntas leaves show potential as multi-target antidiabetic agents with good binding affinity to diabetes target proteins. These findings are still predictive and require further validation through in vitro and in vivo studies to confirm their biological activity, selectivity, safety, and pharmacological relevance as effective antidiabetic candidates in the future.
