Abstract

Talasemia is a genetic disease that causes globin chain synthesis disorder, a major component of the hemoglobin molecule. With the synthesis of unbalanced globin chains will decrease the production of red blood cells. This study aims to determine the presence of mutations in exon 2 gene beta globin thalassemia patients using PCR method, where 5 samples in DNA isolation and continued PCR amplification, the amplification result of electrophoresis region 2 beta globin gene Region II is the result of amplification of primary 4 and primer primer reverse 5 with a target of 350 bp, on PCR there is amplification or doubling of the desired DNA sequence based on the primary selection for the reaction, the PCR product of each of these regions which is subsequently performed by SSCP. PCR-SSCP could analysis the mutation in exon 3 beta globin genes. In the PCR-SSCP result, there was no mutation in region 4 exon 3 in five samples but probably there was mutation in another regions.

Abstract

Biosciences and biotechnology have become the interesting topics since long to be investigated in order to understand science and technology, develop many tools and instrumentation, discovery of new drugs including natural base drugs and biopharmaceutical, contribute in energy sources, interact with the environment as well as to improve the quality of human being. The researches of the field have dramatically contributed for many other field and for better life. Uncounted numbers publications in these field will be continuously rising. To contribute in dissemination of research output which have been done by researcher around the world, the Current Research on Biosciences and Biotechnology so called CRBB is published in this year. In the beginning this journal will publish 2 issue in the year and will be increased in the future. All researchers in the field of bioscience and biotechnology are invited to submit the manuscripts to CRBB. The manuscript will be peer reviewed prior to publish.

Abstract

A flavonoid, luteolin, was isolated from the leaves of Syzygium myrtifolium . The compound was purified using vacuum liquid chromatography with the help of a specific guidance for the flavonoid isolation. The chemical structure of this compound was determined based on measurements of UV, IR and NMR spectroscopic data. The occurrence of luteolin in S. myrtifolium is firstly reported from this study.

Abstract

Ursodeoxycholic acid (UDCA), a secondary bile acid (BA), is an acidic steroid synthesized from cholesterol in hepatocytes. UDCA is widely used for the treatment of various diseases related to liver injury. The use of UDCA to treat non-liver diseases has also been developed recently, such as neurodegenerative diseases, cancer, and obesity. Due to the important role of UDCA on human health, numerous studies in understanding its chemical and pharmacological properties have been published. Literature sources were obtained from online databases such as Science Direct, Google Scholar, Scopus and PubMed using keywords relating to the purpose of study. Critical analysis and review were performed for all literature. UDCA is a steroid compound with pharmacological properties. Seventeen enzymes are involved in its biosynthesis, which has been proposed in four pathways: classic, alternative, the Yamazaki, and 25-hydroxylation pathways. UDCA can be isolated from bovine bile, bear bile or all Ursidae, human, rabbit, cow, rat, hamster, sheep, pig, and plant. UDCA has been used in the treatment of several diseases such as primary biliary cirrhosis, intrahepatic cholestasis of pregnancy, hepatolithiasis associated with Caroli syndrome gallstones, cystic fibrosis, hepatitis C virus, chronic heart failure, neurodegenerative diseases, and obesity, as well as in the prevention of cancer. UDCA has a wide range of pharmacological properties. Further investigations on its efficacy and safety on humans are required before it could be used for several indications. All genes which are responsible in UDCA biosynthesis have been elucidated. That said, further genetic engineering studies in order to find other prospective sources of UDCA could be a challenge for the future research.

Abstract

Meniran (Phyllanthus niruri L.) is a wild plant in the Euphorbiaceae family which grows in tropical climates. The plant is known to possess antioxidant activity and is rich in phenolic compounds. Since plants with high phenolic content and high antioxidant activity are known to exhibit high tyrosinase inhibitory activity, the purpose of this study is to determine the inhibitory activity of the tyrosinase enzyme by the methanol extract of meniran (P. niruri L.) and to isolate secondary metabolites from the extract and fractions which exhibit inhibitory activity against tyrosinase. The in vitro tyrosinase inhibitory activity assay was conducted using the 96-well microplate method with kojic acid as the positive control, while the separation and purification of compounds were carried out using chromatography techniques. The methanol extract and n-hexane fraction showed tyrosinase inhibitory activity with IC50 of 11.05±0.16 and 2.57±0.08 mg/ml, respectively. Three bioactive compounds were succesfully isolated from the n-hexane fraction. Based on the UV and NMR spectra, compounds 1, 2, and 3 were identified as phyllanthin, phyltetralin, and hypophyllanthin, respectively. Furthermore, phyllanthin (1) was shown to demonstrate inhibitory activity against tyrosinase with IC50 of 0.6322 mM. In conclusion, both meniran extract and phyllanthin (1) were found to exhibit inhibitory acitivity against the tyrosinase enzyme. In the future they can be candidates for the treatment against hyperpigmentation and also as an active ingredient in skincare formulations.

Abstract

Berdasarkan data WHO pada tahun 2019 lebih dari 463 juta orang di dunia menderita diabetes, dan cenderung jumlahnya semakin meningkat. Dengan bertambahnya penderita diabetes menyebabkan penggunaan obat antidiabetes meningkat. Berbagai macam obat diabetes telah digunakan untuk mengobati penyakit ini, diantaranya penggunaan obat herbal. Salah satu tumbuhan obat yang berkhasiat sebagai penurun kadar gula darah adalah bratawali (Tinospora crispa). Tujuan penelitian adalah menguji aktivitas antidiabetes produk obat herbal kategori jamu yang diproduksi oleh usaha mikro obat tradisional (UMOT) yang mengandung bratawali. Aktivitas antiabetes diuji dengan menggunakan metode toleransi glukosa dan efek pada mencit diabetes imbasan aloksan. Hasil pengujian kualitas menunjukkan ekstrak mengandung senyawa golongan alkaloid, flavonoid, dan steroid/triterpenoid. Pengujian aktivitas antidiabetes menunjukkan ekstrak dapat menghambat kenaikan kadar glukosa pada uji toleransi glukosa sebesar 18,29; 51,17 dan 75,35 % (p<0,05) dengan dosis berturut-turut 125, 250 dan 500 mg/kgBB. Sementara itu, pada mencit diabetes imbasan aloksan ekstrak mampu menurunkan kadar glukosa darah dengan penurunan yang terbesar 75,35% (p < 0,05) setelah pemberian dosis 500 mg/kgBB. Dari data yang diperoleh dapat disimpulkan bahwa ekstrak dari produk obat herbal yang mengandung bratawali sebagai komponen aktif mempunyai aktivitas antidiabetes. Senyawa aktif pada ekstrak bratawali yang bertanggung jawab untuk aktivitas tersebut masih perlu ditentukan.

Abstract

Kumis kucing mengandung metabolit sekunder sinensetin yang termasuk ke dalam golongan senyawa flavonoid. Sinensetin berpotensi sebagai agen antivirus dan imunomodulator.  Tujuan penelitian ini adalah standardisasi tanaman kumis kucing varietas putih dan upaya produksi senyawa sinensetin. Ekstraksi dilakukan dengan metoda maserasi. Tahap pemisahan lanjutan dilakukan dengan metoda ekstraksi cair-cair, kromatografi kolom dan kromatografi cair vakum. Hasil ekstraksi cair-cair terpilih tiga fraksi  yaitu fraksi air, etil asetat dan n-heksana. Sebanyak 2,08 gram fraksi etil asetat dilanjutkkan pada tahap pemisahan lanjutan menggunakan kromatografi cair vakum dengan fasa diam silika gel H60 dan fasa gerak n-heksana dan etil asetat. Hasil dari kromatografi cair vakum diperoleh sebanyak 11 subfraksi. Penggabungan dilakukan pada subfraksi 8-11 yang terdeteksi adanya senyawa sinensetin, selanjutnya terhadap subfraksi gabungan dilakukan pemisaahan lanjutan dengan kromatografi kolom. Hasil pemisahan dengan kromatografi kolom diperoleh subfraksi sebanyak 142 vial. Pada subfraksi hasil kromatografi kolom nomor 91-114 terdeteksi adanya isolat sinensetin. Kromatografi lapis tipis preparatif (KLTP) gabungan subfraksi kromatografi kolom 91-114 (SFK) menunjukkan adanya senyawa sinensetin. Berdasarkan hasil pemeriksaan kemurnian dengan menggunakan KLT 2 dimensi dan analisis profil spektrum UV isolat diduga senyawa sinensetin.

Abstract

Orthosiphon stamineus has long been used in traditional medicine in East India, Indo China, Southeast Asia, and tropical Australia, where this plant is usually found. Based on the color of the flowers and petals, Orthosiphon stamineus is classified into two varieties: white flowers (white varieties) and purple flowers (purple varieties). Orthosiphon stamineus has traditionally been used to treat hypertension, diabetes, bladder and kidney disorders, gallstones, gout, and rheumatism. The leaves of Orthosiphon stamineus were introduced to Europe and Japan as tea for health. The main compounds possessed by Orthosiphon stamineus are rosmarinic acid, eupatorium, and sinensetin. In addition, several studies have isolated the plant of Orthosiphon stamineus. This journal review aims to review studies related to the content of secondary metabolites, traditional uses, pharmacological activities, and levels of flavonoids contained in plants.

Abstract

Sappan wood extract (Caesalpinia sappan) provides better antioxidant properties than ascorbic acid. Empirically, Sappan wood is often used as traditional medicine. To support the utilization of Sappan wood, the isolation and identification of active antioxidant compounds from the methanol extract of Sappan wood had been conducted from this study. Extraction was performed with maceration and evaporation methods. The spots of active antioxidant compounds in TLC-DPPH observations were isolated using liquid vacuum chromatography to obtain two pure compounds. The results of the LC-MS/MS identification of the two compounds, respectively, exhibited peak fragmentation of molecular ions at m/z 271.09, 257.15, 167.03, 151.06, 123.12, and 104.06 at the retention time of 6.35 minutes which were translated as Alpinetine and peak fragmentation of molecular ion at m/z 287.09, 177.06, 164.05, 123.04 and 110.04 at a retention time of 5.75. minutes which were translated as 3-Deoxysappanone B. The antioxidant activity quantitatively showed that Alpinetine and 3-Deoxysappanone B were capable of scavenging DPPH radicals with IC50 values ​​of 20.11 M and 15.28 M, respectively.

Abstract

The cat's whiskers plant ( Orthosiphon aristatus (Blume) Miq) that grows in Indonesia has white flowers, purple flowers, and white flowers with purple patterns. The quality of the secondary metabolite content of each cat's whiskers plant is different because it depends on environmental influences such as rainfall or soil mineral content. Plant tissue culture methods such as callus culture or cell culture are used to overcome these obstacles because these methods do not depend on the environment and can produce secondary metabolites such as those produced by the original plant. Cat's whiskers contain the main secondary metabolite compounds: rosmarinic acid, eupatorin, and sinensetin. Rosmarinic acid belongs to the phenolic group, while sinensetin and eupatorin are flavonoids. This study aimed to determine the total levels of polyphenols and flavonoids in purple cat whiskers and determine the ratio of secondary metabolites of cat whiskers callus with wild-type plants—determination of polyphenol content using Folin Ciocalteau reagent. Determining flavonoid levels is based on colorimetry using AlCl 3 reagent, specific for flavone and flavonol compounds. The levels of polyphenols contained in the ethanolic extract of callus and cat whiskers of the purple variety were 16.056 ± 0.204 mgQE/g and 30.780 ± 0.425 mgQE/g, while the polyphenol content of the callus ethyl acetate extract and the purple variety of cat whiskers leaves was 15,489 ± 0.425 mgQE/g and 13,262 ± 0.044 mgQE/g. The levels of flavonoids contained in the ethanol extract of the cat whiskers leaf were 1.39±0.03 mgQE/g and callus 0.16±0.02 mgQE/g in the ethyl acetate extract of the leaves of the cat whiskers 2.16±0.012 mgQE/g and callus 2.24±0.010 mgQE/g. The polyphenols and flavonoids in the callus ethyl acetate extract were more significant than the ethyl acetate extract of the purple variety ( wild type ) cat whiskers leaf.
