Abstract

Propolis, a resinous compound honeybees produce, demonstrates an extensive spectrum of powerful biological properties. However, the anti-cancer activity of propolis derived from Geniotrigona thoracica Sumatrans has yet to be reported. Thus, we sought to investigate the cytotoxicity of aqueous propolis extracts from Geniotrigona thoracica Sumatrans against Colo-201 colon cancer cell line and senescence Colo-201 colon cancer cell line induced by low-dose doxorubicin. This study was conducted at the Parasitology Laboratory of Human, Safety, and Environment, Universitas Padjadjaran Bandung from January to May 2024. This study assessed cell viability using the WST-1 test. Non-induced Colo-201 cells were treated with an aqueous extract of propolis (AEP) 100 ppm, or 5-fluorouracil (5-FU) 5 mg/ml as the positive control or water as a vehicle on untreated control. Colo-201 senescence was induced by doxorubicin 0.1 µM for three days. Doxorubicin-induced Colo-201 senescence was then treated with AEP 100 ppm, with 5-FU 5 mg/ml as the positive control, or with the combination of AEP 100 ppm and 5-FU 5 mg/ml, or water as a vehicle on untreated control. The data were analyzed using SPSS version 25.0, a one-way ANOVA, and Tukey’s post hoc test. The results showed that AEP has cancer-killing effects on Colo-201 cells and Colo-201 senescent cells induced by low-dose doxorubicin. AEP-treated Colo-201 cells and Colo-201 senescent cells induced by low-dose doxorubicin viability were significantly reduced to 37.15% and 13.72%, respectively, although slightly higher than those of the 5-FU-treated one at this concentration. There was also a decrease in the cancer-killing effect of 5-FU from 88.55% in non-induced Colo-201 cells to 41.5% in the doxorubicin-induced Colo-201 senescence model. In conclusion, aqueous extract of propolis from Geniotrigona thoracica Sumatrans showed cancer-killing-effects both on the Colo-201 colon cancer cell line and senescence Colo-201 colon cancer cell line induced by low-dose doxorubicin.

Abstract

Background: Coronavirus disease 2019 (COVID-19) is still becoming a problem in several countries with the emergence of new variants with the latest variant named Omicron. Studies show that patients with COVID-19 tend to have Acute Respiratory Distress Syndrome (ARDS) because of high pro-inflammatory cytokines and chemokines levels. Since its role as an antiviral agent, anti-cytokine, immunomodulator, antioxidant and has potential effect in maintaining body functions, vitamin C can be used as one of therapy. There is still arguable about giving treatment of COVID-19 patients with Intravenous Vitamin C (IVC). Previous research has not been discussed related to accelerating recovery and decreasing mortality. Purpose: To identify the impact of IVC intervention given to COVID-19 patients admitted in hospital by reviewing clinical outcomes particularly on complexity or mortality. Review: Literature information was sourced from PubMed, ScienceDirect, and Google Scholar databases through keywords to retrieve inclusion criteria published during 2019-2021. Result: We found that IVC influenced inflammatory biomarkers such as CRP, d-dimer, ferritin, and several inflammatory cytokines, mortality, length of stay, oxygenation, organ and immune system function but showed no significant difference between the control group. Conclusion: IVC treatment might be an essential adjuvant therapy in the aggravation of mild, severe, and critical COVID-19 patients, but did not understate complexity or mortality. Hence, further research and evaluation in clinical trials are needed.

Abstract

Gastric Ulcer is a common side effect of Non-Steroid Anti Inflammatory Drugs (NSAIDs) use such as Aspirin. Probiotic has many benefits especially for the alimentary system, but the effect on gastric ulcer have yet to be explored. In this research we aimed to find the probiotic effect on histological structure changes during aspirin induced gastric ulcer healing process in animal model. This was simple random sampling animal experimental laboratory, that divided Wistar Rats into two main groups. Both groups were induced with Aspirin (300mg/Kg BW) per oral, once daily for three consecutive days,and subsequently the test group were given probiotic suspension (cfu >108/gr) per oral, once daily, for 14 days. Each subgroups were sacrified serially at day 0, 3, 7, 14. The gaster were collected and processed for histology examination. Better histological structures were shown in the test group at day 14. MANOVA result showed probiotic's effect in enhancing histological structure changes during gastric ulcer healing process. Better histological structure changes were observed in mucus thickness; reepitelization, gastric glands formation, and angiogenesis process of the test group. As a conclusion, probiotic enhanced mucus thickness, reepitelization, glands remodelling, and angiogenesis in Aspirin induced gastric ulcer healing process in animal model. Key words: probiotic, gastric ulcer, histological structure, aspirin, healing process

Abstract

Skin protects the body from various trauma, such as high temperature trauma known ascombustio. Pineapple (Ananas comosus) contains lots of substances such as bromelain,antioxidants, and zinc shown to accelerated healing process in previous experiments. Aim ofthis research was to analize the effect of topical pinneaple juice application in combustiohealing process. The method was true experimental laboratory. Wistar rats’ back skin wereinduced by 100 o C iron plat untill 2 nd degree combustio occured, then divided into two groups(n=20). The treatment group were given topical application of pinneaple juice, then the healingprocess were observed macroscopically and microscopically on day 0, 3, 7, and 14. The resultshowed better wound healing in treatment group on day 14. MANOVA test showed topicalapplication of pinneaple juice accelerated healing process (p=0.001). Treatment group showedbetter histological structure changes, as seen from better granulation tissue formed (p=0.003)and reepithelization (p=0,000), but not from angiogenesis (p=0.733). Conclusion, topicalapplication of pinneaple (Ananas comosus) juice accelerated healing process in 2 nd grade ofcombustio animal model measured by granulation maturation process and reepithelizationprocess, but has no effect on angiogenesis.Keywords : Ananas comosus juice, 2 nd degree combustio, granulation tissue, reepitelisation,angiogenesis.

Abstract

Background: Alpha-tocopheryl acetate (Vitamin E) and metformin hydrochloride have been tested as anti-aging compounds at various concentrations. This study aimed to identify the most effective concentrations of alpha-tocopheryl acetate and metformin hydrochloride in promoting the viability of human dermal fibroblasts (HDFs), a primary cell type in skin aging research.Methods: HDFs were isolated using a mechanical isolation method and cultured under standard conditions. Cells were treated with varying concentrations of alpha-tocopheryl acetate and metformin hydrochloride as independents agents. After 48 hours of incubation, cell viability was measured using the MTT assay.Results: Alpha-tocopheryl acetate had the highest HDF cell viability (107%) at a concentration of 50 μM. Metformin hydrochloride had the maximum HDF cell viability (158%) at 5 μM. However, the viability response varied across different concentrations for both agents, indicating that optimal dosing was essential for maximizing their effectiveness.Conclusions: Alpha-tocopheryl acetate at 50 μM and metformin hydrochloride at 50 μM yield the highest viability of HDFs in vitro. These findings suggest potential roles for both agents in anti-aging skin therapies. Further research is recommended to explore their mechanisms of action and to optimize dosing strategies for clinical applications.

Abstract

The cytoskeleton plays an important role in forming the cell framework and dynamics. The part that plays a role in supporting the spatial and mechanical functions of the cell is located in the cytoskeleton. Therefore, strategies to maintain the integrity and dynamics of the cytoskeleton have potential as therapies for age-related disorders. One thing that can be used to delay aging is metformin. Through AMPK activation, it can influence the cytoskeleton; namely, it can remodel the dynamics of the cytoskeleton by influencing structural changes in the actin cytoskeleton, which in turn will reduce the permeability of the filtration barrier in diabetic conditions and can increase insulin sensitivity, especially in old age, thereby delaying aging. In this review, we review the current understanding of the role played by the cytoskeleton in aging and review the opportunities and challenges for transitioning basic research into intervention development. Methods: Several major electronic databases, including PubMed, Scopus, and Cohcrane were used to select articles between April 2013 and April 2023. From the 8 existing literature, we present the activity of metformin as an aging delay. Cytoskeleton can change with age so it can affect the dynamics and structure of the cell. Metformin shows significant benefits, especially in delaying aging, through activation of the AMPK pathway so that it can increase the activity of mitochondria in producing ATP, which can affect the structure of the cytoskeleton. With this article on changes in the cytoskeleton with aging, it is hoped that insight into future research directions can be achieved.
