Abstract

Mangrove forests are one type of forest that grows in tropical and subtropical regions and plays a crucial role in maintaining ecosystem balance. One way to assess the health of mangrove communities is through regular monitoring and evaluation using modern technology such as digital image processing. The percentage of mangrove canopy cover can serve as a key indicator in evaluating the health and population density of mangrove communities. This study aims to implement the Otsu thresholding method in a digital image processing-based system capable of automatically determining the health status of mangrove communities based on the percentage of mangrove canopy cover. Mangrove canopy cover images were acquired using hemispherical photography techniques. Experimental results show that the system built using the Otsu thresholding method has an average Relative Absolute Error (RAE) of 0.034 and average Mean Error (ME) of 0.052, with an average processing time of 5.3 seconds. This indicates that the system can automatically determine the health status of mangrove communities in a relatively short time. It also suggests that the process of determining the health status of mangrove communities aligns with direct field observations.

Abstract

Wilayah pesisir sering kali menjadi sasaran bencana alam, yang dapat berdampak serius pada komunitas nelayan. Pengurangan ekosistem mangrove di pesisir dapat meningkatkan risiko banjir, erosi pantai, dan gelombang tinggi. Oleh karena itu, dalam program kegiatan Kosabangsa (Kolaborasi Sosial Membangun Masyarakat), pengabdian masyarakat ini bertujuan untuk membantu komunitas nelayan Sipatuo di Kelurahan Tahoa, Sulawesi Tenggara dalam menghadapi masalah bencana di wilayah pesisir yang merupakan tempat bermukim mereka. Kegiatan penanaman pohon mangrove dilakukan sebagai cara untuk melindungi pantai dan meningkatkan pemahaman komunitas tentang pentingnya pohon mangrove. Masyarakat dilibatkan dalam sosialisasi dan penanaman mangrove. Hasilnya sangat positif, dengan pemahaman masyarakat meningkat sekitar 18%, dan hampir semua orang ikut berpartisipasi. Ini menunjukkan bahwa kegiatan ini sangat efektif dalam memberikan pengetahuan kepada Masyarakat pesisir. Kegiatan pengabdian ini penting dalam upaya pelestarian lingkungan pesisir di wilayah Sulawesi Tenggara. Perlu diadakan penelitian  untuk melihat dampak jangka panjang dari upaya mitigasi bencana pesisir dan bagaimana pelestarian mangrove dapat membantu komunitas dalam menghadapi bencana pesisir.

Abstract

Pulau Simuang merupakan salah satu pulau kecil yang ada di guguan Kepulauan Tiworo dengan potensi mangrove yang cukup besar. Sayangnya, studi mengenai status kondisi komunitas dan stok karbon mangrove di Kepulauan Tiworo masih sangat terbatas. Penelitian ini bertujuan untuk menganalisis status kondisi komunitas mangrove, biomassa, stok karbon, dan penyerapan CO2 oleh vegetasi mangrove. Penelitian ini dilakukan di Pulau Simuang, Kepulauan Tiworo, dengan total empat stasiun penelitian. Analisis struktur komunitas mangrove menggunakan metode transek kuadrat. Garis transek dibentangkan tegak lurus dengan garis pantai. Disetiap transek diletakkan empat plot berukuran 100m2. Data yang dikoleksi berupa jenis, jumlah jenis, dan diameter batang (dbh). Analisis data dilakukan untuk menentukan kerapatan, status kondisi, biomassa, stok karbon, dan penyerapan CO2 oleh vegetasi mangrove. Hasil penelitian ini yaitu status kondisi komunitas mangrove di Pulau Simuang masih dalam kategori baik/padat (kerapatan berkisar antara 1825-2775 ind/ha2). Biomassa mangrove diperoleh berkisar antara 213,26 ton/ha2 sampai 506,24 ton/ha2. Stok karbon dan penyerapan CO2 di setiap stasiunberbanding lurus dengan data biomassanya. Stok karbon diperoleh berkisar antara 100,23 ton C/ha2 sampai 237,84 ton C/ha2. Selain itu, penyerapan CO2 oleh vegetasi mangrove berkisar antara 367,51 ton CO2/ha2 sampai 872,08 ton CO2/ha2. Meskipun kondisi mangrove masih dalam kategori baik/padat, masih terdapat sisa aktivitas penebangan mangrove di beberapa lokasi. Untuk itu diperlukan upaya perlindungan kawasan untuk menjaga fungsi ekosistem mangrove di kawasan tersebut.

Abstract

Studi kerapatan dan perubahan tutupan mangrove menggunakan citra satelit sangat penting dilakukan untuk mengidentifikasi kondisi ekosistem mangrove secara eksisting. Tujuan dari penelitian ini adalah untuk menganalisis kerapatan dan perubahan tutupan mangrove menggunakan Citra Satelit Landsat 8 OLI dan Sentinel-2A Level 1C di Kecamatan Lasalepa, Kabupaten Muna. Pengamatan kerapatan mangrove secara insitu menggunakan plot pengamatan 100m2. Analisis perubahan tutupan mangrove menggunakan citra satelit multitemporal (2015-2020) dengan metode klasifikasi supervised classification. Terdapat lima jenis mangrove pada empat stasiun pengamatan. Pada Stasiun I-III dengan nilai kerapatan mangrove: 1.480 pohon/ha, 1.320 pohon/ha, dan 1.440 pohon/ha terkategori Baik (Sedang), sedangkan Stasiun IV dengan nilai kerapatan 2.120 pohon/ha terkategori Baik (Sangat Padat). Berdasarkan hasil analisis citra, perubahan tutupan lahan dalam rentang tahun 2015-2020 menunjukkan adanya peningkatan luasan.

Abstract

Soil bacteria could be the promising source of secondary metabolites with diverse pharmacological activities. Bacteria coded as P1 and M7 were isolated from soil obtained from Muna Island, Southeast Sulawesi. This research aimed to evaluate the antioxidant activity, cytotoxic property, and identify the chemical profile of the extract derived from soil bacteria P1 and M7. Based on 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, extract derived from P1 isolate was moderately active against ABTS free radical with IC50 of 126.22±10.82 µg/mL, and less active on DPPH (IC50: 721.95±16.59 µg/mL), while M7 extract was less active towards DPPH and ABTS. Interestingly, these extracts in the concentration of 100 µg/mL were cytotoxic to breast cancer cell (MCF-7), as tested by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. MCF-7 viability was decreased up to 69.83±1.46 % and 61.49±0.76 % after treated with P1 and M7 extract, respectively. The extracts were also induced membrane disruption and apoptosis of MCF-7 cells. The extracts contained phenol (9.52±0.22 and 10.41±0.09 mg EAG/g extract, for P1 and M7, respectively) and flavonoids (9.16±0.16 and 9.29±0.24 mg EQ/g extract, for P1 and M7, respectively). According to Gas Chromatography Mass Spectrometry (GC-MS) analysis, Pyrrolo[1,2-a] pyrazine-1,4-dione, hexahydro-3-(2-methylpropyl) that previously well known as antioxidant and cytotoxic compounds, were found in the M7 and P1 extract. This compound may play a vital role in the radical scavenging and cytotoxic properties of P1 and M7 extracts. The results of this study indicate that P1 and M7 extracts could be the potential source of antioxidant and cytotoxic compounds and need to be further developed for pharmaceutical use.

Abstract

The aim and objective of this activity is to assist the Mekar Jaya fish processing company in increasing the production volume and value of the company's finished products. Increasing production and increasing product value is carried out through fish ball and shredded meatball training, branding training, collective packaging and labeling of finished products. This will make it easier to increase the volume of Mekar Jaya fish processed by the group, as well as help increase business income and expand the product market. The name of this brand is taken from the local legend of Kolaka, namely Owete means fish, and Meambo means good. The goal is that in the future this product can be known by the people of Kolaka Regency and Southeast Sulawesi as a common component of regional cuisine. The results of laboratory analysis show that the nutritional value of processed fish balls and the product itself is quite significant. The goal is, by consuming these meatballs and floss, people's nutritional needs can be met. Apart from being an effort to increase the group's fish processing capabilities and increase the selling value of the group's processed products, this activity also increases the diversification of processed fish products in Kolaka Regency and increases the capacity of the MSME cluster in Tahoa Village. Kolaka. So that in the future, similar efforts need to be encouraged again in various locations in Kolaka Regency to expand the benefits of similar activities.

Abstract

Praktik Destructive Fishing menggunakan bom ikan oleh beberapa nelayan masih marak dilakukan di kawasan perairan Kabupaten Buton Tengah. Oleh karena itu, beberapa ekosistem terumbu karang di perairan Buton Tengah telah mengalami kerusakan. Tujuan dari kegiatan pengabdian ini yaitu untuk melakukan transplantasi dan pelepasan media penempelan karang melalui pendekatan berbasis kelompok nelayan di Perairan Teluk Lianabanggai, Desa Lanto, Kabupaten Buton Tengah. Transplantasi menggunakan media kubus beton, dan media penempelan larva karang berupa bioreeftek. Selain itu, kegiatan ini juga bertujuan untuk mensosialisasikan bahaya dan dampak destructive fishing bagi keberlanjutan sumberdaya perikanan dan meningkatkan kapasitas mitra dalam upaya rehabilitasi terumbu karang. Pelaksanaan sosialisasi dan penguatan kapasitas mitra dilakukan secara langsung dengan metode ceramah. Pembuatan media transplantasi menggunakan media beton berbentuk kubus dan bioreeftek menggunakan batok kelapa. Pelepasan media dilakukan secara partisipatif, yaitu masyarakat kelompok nelayan terlibat secara aktif. Hasil kegiatan ini berupa peningkatan pengetahuan masyarakat terhadap bahaya dan dampak destructive fishing, peningkatan kapasitas masyarakat mengenai rehabilitasi terumbu karang. Masyarakat sangat antusias mengikuti rangkaian kegiatan ini mengingat kegiatan ini merupakan kegiatan yang pertama kali dilakukan di Desa Lanto, Kabupaten Buton Tengah. Diperlukan adanya upaya penelitian lebih lanjut mengenai status kondisi terumbu karang di perairan Selat Lianabanggai, Kabupaten Buton Tengah.

Abstract

Tsunami that swept the Tanjung Lesung coast in 2018 caused Anak Krakatau to collapse in Sunda Strait which hit the coastal areas of Banten and Lampung. Tanjung Lesung is one of the worst areas affected. Tsunami in Tanjung Lesung has taken lives, damaged infrastructure, and caused terrible natural damage. One of the damages affecting residents' lives in the Tanjung Lesung coastal community is exposure to the marine ecosystem. The main ecosystem that has the highest productivity level is the coral reef. Coral biota is the main benthic biota of reefs that are directly affected by earthquakes and tsunamis. However, until now there has been no data on the status of coral reefs after the 2018 Sunda Strait tsunami. Therefore, researchers feel the urgency of this research is high enough to determine the condition of coral reefs after the Sunda Strait tsunami in Tanjung Lesung. There are 3 research stations for data collection before the tsunami, and we conduct research in those 3 stations again after the tsunami. Then we added 3 research stations again, so there are 6 research stations. Observation of coral reefs uses the Line Intercept Transect (LIT) method. The line transect is made by stretching a roll meter with a scale parallel to the coastline along 25 meters with three replications with an interval of about 0-5 meters between replications so the total observed transect was 75 meters. The deterioration of coral reef conditions at three research stations (1-3) proved that the tsunami waves affected the damage to coral reefs. This has an impact on the diversity of coral reef species inhabitants that have decreased. Disturbance form coral-damaged structure and composition changes of the base substrate as habitat can have an impact on reef fish and coral reef inhabitants. So that this research is expected to be a reference for policymakers in determining rehabilitation steps for areas affected by the 2018 Sunda Strait tsunami.

Abstract

The global increase in multidrug-resistant (MDR) bacterial infection has rapidly gained concern globally. This study aimed to investigate antibacterial and antibiofilm potential of 25 soil actinomycete strains against MDR strains including Escherichia coli strain M4, Pseudomonas aeruginosa strain M19, Klebsiella pneumoniae strain M19, Bacillus subtilis strain M18, and Methicillin Resistant Staphylococcus aureus (MRSA). In this study, three actinomycete isolates encoded APM-7, APM-11, and APM-21 showed a strong and broad antibacterial spectrum. The minimum inhibitory concentration (MIC) of extracts derived from these isolates was ranged from 78 μg/ml to 10,000 μg/ml. In addition, The extracts also displayed significant biofilm inhibition values ranging from 6.06 to 72.4%. Based on the results, APM-21 extract had the best antibacterial and antibiofilm activities with the strongest values against MRSA. According to the nucleotide sequencing of the 16S rRNA gene, APM-7, APM-11, and APM-21 strains possessed similar identities with Streptomyces cyaneus, Streptomyces coerulescens, and Streptomyces panayensis, respectively. Based on Liquid Chromatography Tandem-Mass Spectrometry (LC-MS/MS) analysis, two antibacterial compounds, namely rancimanycin III, and enteromycin were detected in all those three extracts. Interestingly, APM-21 extract also contained two prominent antibacterial substances including paramagnetoquinone C, and caerulomycin I, suggesting their contribution to the most potential activities. Moreover, new insights were provided into a promising candidate for use in an active compound combating strategy to control MDR bacterial strain infection.
