Abstract

Faloak (Sterculia quadrifida R.Br) was used empirically as a traditional medicinal plant. The information of the active compounds contained in the faloak bark was not specifically published. This study aims to determine the active compounds contained in the fraction of ethyl acetate of faloak bark which has cytotoxic effect on breast cancer cell type T47D. Extraction method use maceration, isolation compound using gradient isolation method, structural elucidation using information from IR, H-NMR, C-NMR and LC-MS. Cytotoxic activity test on breast cancer cell line T47D using MTT method. The result show the isolate compound was 2-iminoethyl 2- (1-hydroxypentan-2-yl) phenyl) acetate with IC50 value on T47D breast cancer cell 7, 12 μg/mL and with selectivity index value was 47, 53.Keywords: Sterculia quadrifida R.Br, 2-iminoethyl 2-(2-(1-hydroxypentan-2-yl) phenyl) acetate, T47D.

Abstract

Zingiber purpureum Roxb. was a plant that has the potential to be developed as a chemotherapy agent. Bioassay guided fractination aims to examine the anticancer activity of n-hexane, ethyl acetate, and methanol fractions from ethanolic extract of bangle plant rhizome to colon cancer cell type WiDr and Vero cells. The apoptotic test and modulating cell cycle were observed by flow cytometry method. Isolation process of the most active fraction. The pure isolates obtained were identified by structures with structural elucidation using ultraviolet, 1D-NMR, 2D-NMR and liquid chromatography-mass spectrometer. Obtained 2,3-dihydro-5-hydroxy-7-methoxy-2-phenylchromen-4-one compound of ethyl acetate fraction number 21. With a value of CC50 to a WiDr cell of 11.24 ± 3.44 μg/mL and 342.22 ± 4.98 μg/mL to Vero cells.

Abstract

Objective: Indonesia is known with its natural resources, one of them is abundant herbs. The herbs are basic ingredients of herbal medicine which is called jamu. Jamu started to develop since hundred years ago and passed through generation which is trusted to cure many diseases without giving any side effect. Lately, there are many cases of combining jamu with some chemical substances. The most common chemical substances used in jamu are acetaminophen and phenylbutazone. Methods: This research develops method to detect acetaminophen and phenylbutazone by using spektrofotometric UV-Vis which is combined with chemometrics. The most common chemometrics multivariate calibration used is Partial Least Square (PLS) and Principal Component Analysis (PCA). From this method, the data analysis used is R Studio software. This research generates a regression equation. Results: Based on this research, the regression equation indicates that the result is significance which can be seen from the value of P < 0.05. Besides that, the value of correlation coefficient of acetaminophen as 0.9889, and phenylbutazone as 0,9728. Conclusion:  This method can be considered as qualified and meet the requirement.

Abstract

Faloak bark extract has antibacterial, antioxidant and anticancer potential but no optimal solvent has found to extract the active compounds of faloak bark. Therefore, it is necessary to optimize the solvent against the faloak bark. This study aims to determine the optimum solvent of methanol, ethyl acetate, petroleum ether, and water in order to produce an optimal response value in the extract, total phenolic, and inhibitory results in antifungal tests. Extracts obtained through reflux extraction for 2 hours and extract yield calculated. The extract results tested to determine the total phenolic content and inhibition of antifungal test. The determination of the optimum solvent composition carried out using Rstudio v.3.2.2 software with ANOVA parametric test or Kruskal-Wallis non-parametric test and using Design Expert v.11 software with Simplex Lattice Design (SLD) method. The results showed that the solvent composition of faloak bark extract (Sterculia quadrifida R.Br) in giving the yield of extract, total phenolic, and optimal antifungal inhibition was methanol:water 0,438:0,562.
