Abstract

Hepatitis C Virus (HCV) is a world health problem. HCV infection is initiated by various structural and non-structural proteins. The HCV NS3 protein has an important function in viral replication. The N-terminal domain of NS3 acts as a protease to process most of the viral polypeptides. NS3 also acts as an RNA helicase and NTPase and triggers liver fibrosis which accelerates the development of liver disease. Thus, this study aims to provide information on potential new antiviral candidates against HCV that target the NS3 protein. This study was conducted in-silico with a ligand-based and structure-based pharmacophore model to the cavity of the active protein site generated after virtual screening and molecular docking. The results of this study showed that three compounds, namely stigmasterol, gamma-mangostin, and erycristagallin, were found as HCV antiviral candidates that target the NS3 protein with a lower binding affinity than the native ligand. The binding energy of each compound is -9.23 Kcal/mol, -8.58 Kcal/mol, and -8.17 Kcal/mol. Based on ADMET analysis, the three compounds have high absorption in the small intestine. The cytotoxicity analysis of stigmasterol compounds is not potentially mutagenic, and the LD50 value of stigmasterol is also lower than other compounds.

Abstract

Caffeine, a widely consumed stimulant, has demonstrated significant effects on cancer cell behavior, particularly in triple-negative breast cancer (TNBC) cells. This study investigates the impact of caffeine on MDA-MB-231 cells, focusing on cell morphology, viability, antioxidant gene expression, and cell migration. Caffeine at concentrations of 10 mM and above induced notable morphological changes, including cell rounding, detachment, and decreased cell density, indicative of cytotoxic effects and cellular stress responses such as apoptosis. Viability assays revealed a dose-dependent reduction in cell survival, with a substantial decline in total cell count at higher caffeine concentrations. The observed decrease in cell viability is associated with the downregulation of antioxidant genes SOD2 and GLO1, suggesting disrupted redox balance and impaired detoxification systems. A significant positive correlation was noted between SOD2 and GLO1 expression levels, indicating their interdependence in antioxidant defense mechanisms. Additionally, caffeine treatment impaired cell migration, as the wound healing assay shows, with higher concentrations significantly hindering wound closure. This effect on migration, relevant for metastasis, aligns with findings from previous studies on caffeine’s influence on cancer cell motility. The results suggest that 10 mM caffeine may serve as an optimal concentration for inducing cellular stress without immediate, widespread cell death, positioning it as a promising candidate for further therapeutic exploration.

Abstract

This research aims at finding out whether the implementation of guided questions technique can improve the students’ ability in writing descriptive text or not. The researcher applied a quasi-experimental research design which involved experimental class and control class. The population of this research was the second-grade students of SMP Negeri 1 Toribulu. The sample was VIII B as the control class and VIII C as the experimental class that consist of 20 students for each class. The researcher applied the purposive sampling technique. The instrument used to collect the data was test. The pre-test was administered to measure the students’ ability in writing descriptive text before the treatment and the post-test was to measure the students’ ability after the treatment. The data gathered from the test were analyzed statistically. The result of the data analysis shows that the hypothesis of the research was accepted regarding the analysis that the tcounted (2,076) was higher than the ttable (2,0042). The degree of freedom (df) of the table is Nx+Ny – 2 = 20+20-2 = 38 with the level of significance at 0.05. it means that guided questions technique can improve the students’ ability in writing descriptive text.

Abstract

Social stratification and legal awareness are topics of study, especially in the social sciences, which are very interesting to do. The paradigm that the higher the education will increse the more of law awareness, becomes a phenomenon that needs to be discussed. This study aims to; 1) analyze whether there is an effect of educational stratification on legal awareness, 2) analyze how to build legal awareness in the community. The study involved 30 respondents who were academics from University X, Bekasi. Education Straktas are classified into three; S1, S2 and S3. Indicators related to legal awareness are legal knowledge, legal understanding, legal attitudes and legal behavior. Analysis using statistical parametric. The results of the study show that education strata do not have an effect on legal awareness.

Abstract

The digital era has brought significant changes to the field of education, making technology an essential part of the teaching and learning process in elementary schools. This study aims to evaluate and improve classroom management in a digital learning context at Rosela Indah Public Elementary School. Using a descriptive qualitative method, data were collected through interviews with the fourth-grade homeroom teacher, classroom observations, and analysis of school documents. Data analysis involved reduction, presentation, and drawing conclusions. The results showed that the integration of digital technology enhances learning effectiveness and student engagement. Teachers play a key role in creating enjoyable and meaningful learning experiences through the use of self- developed learning applications. Digital learning also facilitates easier access to information, boosts students’ motivation, and encourages better interaction and collaboration. However, several challenges were identified, including increased workload for teachers and parents, inconsistent content quality, and risks related to students’ concentration and health. Effective strategies include the use of learning management applications and a point-based reward and punishment system. Achieving success in digital learning requires collaboration among teachers, students, parents, and sufficient technological support. This study recommends improving teachers’ digital competencies, developing a curriculum that supports digital approaches, involving parents, and conducting regular evaluations.

Abstract

Penelitian ini bertujuan untuk mengembangkan bahan ajar IPS berbasis kearifan lokal untuk SD atau MI. Objek penelitian ini adalah SD atau MI Penelitian menggunakan metode kualitatif dengan pendekatan fenomenologi. Hasil penelitian menunjukkan bahwa pengembangan bahan ajar IPS berbasis kearifan lokal untuk siswa SD/MI dapat memberikan kontribusi positif terhadap proses pembelajaran. Bahan ajar yang dikembangkan memuat nilai-nilai budaya dan tradisi lokal yang dekat dengan kehidupan siswa, sehingga membuat pembelajaran lebih kontekstual dan mudah dipahami. Selain itu, penggunaan bahan ajar ini juga mampu meningkatkan minat dan keterlibatan siswa dalam pembelajaran serta menanamkan nilai-nilai karakter seperti gotong royong, menghargai budaya, dan cinta terhadap lingkungan sekitar. Guru pun memberikan respons positif karena materi menjadi lebih relevan dan mudah disampaikan di kelas.

Abstract

Acne vulgaris is a common dermatological disorder that significantly impacts quality of life, yet its complex pathogenesis remains incompletely understood, contributing to the variability in clinical presentation and treatment response. This review summarizes recent advances in omics-based research on acne vulgaris and explores how these findings support the development of targeted therapy. A systematic literature search was conducted in PubMed using the keywords “acne genomic,” “acne transcriptomic,” “acne proteomic,” and “acne metabolomic.” Original research articles published in English, available in full text, and published between 2015 and 2025 were included. After screening for relevance and removing duplicates, 17 studies met the inclusion criteria. Additional relevant articles were also referenced to complement the discussion. The selected studies show that large-scale molecular analysis provides a more comprehensive understanding of the molecular mechanisms underlying acne vulgaris. These findings enable the identification of novel biomarkers, better insight into pathological pathways, and the development of more targeted therapeutic strategies. Further studies are needed to validate these findings and translate them into improved strategies for the diagnosis, prevention, and treatment of acne vulgaris.

Abstract

This paper explores the essential roles of teachers, students, and learning materials as the three foundational pillars of effective learning. The objective of this study is to address the imbalance often found in educational practices that emphasize only one or two elements, neglecting the importance of their interconnectedness. Using a qualitative descriptive method and literature review approach, this paper analyzes how these three components function individually and collectively within contemporary educational settings. Teachers are examined as facilitators and agents of pedagogical design, students as active participants and co-constructors of knowledge, and learning materials as dynamic tools that support cognitive and skill development. The findings reveal that meaningful learning experiences emerge when there is a harmonious integration of teacher guidance, student engagement, and relevant instructional content. The study also highlights how the neglect of one pillar can hinder the learning process and reduce educational effectiveness. In conclusion, this paper emphasizes the need for a balanced, holistic approach in educational planning and instructional practice, ensuring that teachers, students, and materials work in synergy to enhance learning outcomes.
