Abstract

Vitamin C is a compound that cannot be formed by the body itself. Vitamin C is needed to regulate metabolic processes and normal body functions. Lack of vitamin C can cause a decrease in the immune system. During a pandemic, vitamin C is needed to improve the immune system to prevent and fight disease infections. The pandemic has caused an increase in the need and price of vitamin C supplements in the market. Fulfilling the body’s vitamin C needs can use alternatives such as Moringa leaves and green spinach. This study aims to determine the levels of vitamin C in Moringa leaves and green spinach with variations in temperature and concentration. Analysis of vitamin C was carried out using UV-Vis spectrophotometry at a wavelength of 516 nm. The results of the determination of vitamin C levels in Moringa leaves and green spinach obtained the highest levels of 24,9948% and 19,0205%, and the lowest levels of 2,2456% and 1,8710%. The results of the ANOVA test showed that there was a significant difference in the levels of vitamin C obtained at the given temperature and concentration variations.

Abstract

Rumput laut merupakan sumber alami senyawa bioaktif dengan potensi fotoprotektif, termasuk senyawa berbasis lipid termasuk lipid polar maupun non polar seperti asam lemak, karotenoid, sterol, steroid, dan triterpenoid. Kelompok senyawa ini dapat menyerap radiasi ultraviolet (UV), dan menetralkan radikal bebas, sehingga dapat melindungi kulit dari paparan sinar matahari dan mencegah terjadinya berbagai bentuk permasalahan kulit seperti penuaan dini (photoaging), hiperpegentasi, sunburn hingga kanker kulit. Tinjauan pustaka ini bertujuan untuk mengevaluasi potensi senyawa lipid dari berbagai spesies rumput laut dalam meningkatkan aktivitas Sun Protection Factor (SPF). Metode narrative review digunakan pada artikel review ini dengan menganalisis berbagai artikel penelitian yang memenuhi kriteria inklusi, yaitu diterbitkan antara tahun 2015 dan 2025, berasal dari jurnal terakreditasi SINTA atau terindeks Scopus, serta ditulis dalam bahasa Indonesia atau bahasa Inggris. Artikel yang tidak mencantumkan data uji SPF secara jelas, tidak meneliti senyawa lipid, atau hanya membahas metabolit lain seperti polisakarida dan protein dieksklusi dari tinjauan ini. Hasil review menunjukkan bahwa senyawa lipid seperti asam lemak (EPA, DHA), fucoxanthin, steroid, gliserol, dan terpenoid  dari rumput laut menunjukkan aktivitas fotoprotektif yang menjanjikan, terutama saat diekstraksi menggunakan pelarut non-polar seperti n-heksan. Artikel ini berfokus pada potensi lipid dari rumput laut sebagai bahan aktif dalam formulasi tabir surya alami.

Abstract

Sargassum ilicifolium merupakan salah satu jenis sargassum yang masih jarang diteliti, terutama data tentang aktivitas antioksan yang berfungsi sebagai agen fotoprotektor. Namun, Sargassum ilicifolium sudah terbukti memiliki aktivitas antioksidan, antijamur, antibakteri, antikanker, antitumor. Sargassum ilicifolium mengandung flavonoid, fenol, tanin, dan steroid. Terdapat pula kandungan kimia fucosantin, fucoidan, beta karoten dan protein yang memiliki aktivitas biologis penting, seperti antioksidan. Senyawa yang memiliki aktivitas sebagai antioksidan adalah senyawa fenol dan turunannya. Oleh karena itu, tujuan dari Literatur Review ini adalah untuk mengetahui metode yang paling optimal yang menghasilkan aktivitas antioksidan paling baik dan dapat berpotensi sebagai fotoprotektor pada Sargassum ilicifolium. Studi dilakukan melalui tinjauan pustaka dari database Google Scholar, PubMed, dan Elsevier. Dengan kriteria jurnal 10 tahun terakhir yaitu 2015-2025. Dalam penentuan antioksidan pada Sargassum ilicifolium metode yang paling banyak digunakan dan memiliki efektifitas dan efisiensi yang baik adalah DPPH, karena metode ini merupakan metode yang sederhana, mudah, cepat peka dan hanya menggunakan sampel dalam jumlah yang sedikit. Namun, kekurangan dari metode ini adalah jumlah pelarut pengencer yang dibutuhkan dalam pengujian cukup banyak. Faktor yang perlu diperhatikan untuk mempertahankan aktivitas antioksidan adalah proses ekstraksi dan kondisi penyimpanan sampel. Suhu yang rendah dan kondisi yang gelap akan mempertahankan aktivitas antioksidan dengan lebih baik. Selain memiliki aktivitas antioksidan, Sargassum ilicifolium juga memiliki potensi sebagai fotoprotektor.

Abstract

Monoclonal antibodies (mAbs) have become a key component in modern biotechnology and healthcare, playing essential roles in the treatment of diseases such as cancer, viral and bacterial infections, asthma, and in diagnostic applications. However, conventional mAb production using mammalian cell systems is costly, time-consuming, and presents scalability limitations. To address these challenges, current technological advances have led to the development of alternative platforms using plant-based expression systems. This review aims to explore the potential and progress in producing monoclonal antibodies through the Nicotiana benthamiana plant-based expression system, which offers several advantages including rapid production, cost efficiency, greater production capacity, and reduced risk of contamination. The method employed in this review is a literature study conducted through database searches (Google Scholar, PubMed, Science Direct, and others) focusing on peer-reviewed articles from 2015 to 2024. The articles were selected and then analyzed thematically was performed to extract relevant data. The results indicate that Nicotiana benthamiana can efficiently produce functional mAbs with glycosylation profiles comparable to mammalian systems. In addition, plant antibodies have also been successfully applied in various therapeutic and diagnostic studies. Further research is needed to ensure the safety and effectiveness of Nicotiana benthamiana plant-based mAb, as well as efforts to develop production scale for wider medical applications.

Abstract

Cancer is a disease characterized by uncontrolled abnormal cell growth, originating from cancer stem cells. These cells form a side population with stemness properties similar to normal stem cells, have high tumorigenicity, and contribute to the development of cancer. Among all cases, breast cancer is one of the causes of cancer death in women worldwide, estimated to reach 28% of new cancers. The application of one of the genetic engineering techniques in the form of CRISPR / Cas9 which can be used as an alternative choice in optimizing breast cancer therapy. This literature review aims to determine CRISPR-Cas 9 Genetic Engineering as Breast Cancer Therapy. This journal review method is through searching for articles from databases such as PubMed, ScienceDirect, and Google Scholar using relevant keywords, namely breast cancer therapy, Genetic Engineering, CRISPR / Cas9 Technique. The results obtained show something promising in the use of one of the genetic engineering technologies in the form of the CRISPR / Cas9 technique which is able to weaken or suppress genetic activity related to the continuity of growth and development of breast cancer cells. In conclusion, CRISPR/Cas9 technology shows promising potential in breast cancer therapy with its ability to inhibit the expression of certain genes that contribute to the growth, invasion, and metastasis of cancer cells.

Abstract

Indonesia merupakan negara dengan biodiversitas tinggi termasuk biota lautnya. Spons laut merupakan sumber alami yang dapat ditemukan dalam jumlah melimpah di perairan Indonesia dan mengandung berbagai komponen bioaktif termasuk collagen yang bermanfaat untuk kesehatan kulit. Pada penelitian ini, dilakukan suatu studi efek ekstrak collagen dari spons laut Stylissa flabelliformis terhadap profil kulit mencit (Mus musculus L.). Spons laut tersebut diambil dari perairan Lombok Utara Indonesia kemudian ekstraksi collagen dilakukan dengan menggunakan Tris-HCl Buffer diikuti dengan pengendapan collagen menggunakan asam asetat. Selanjutnya dilakukan karakterisasi ekstrak collagen dengan menggunakan FTIR-Spectroscopy. Studi efek ekstrak collagen terhadap profil mencit dilakukan selama 14 hari. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak collagen spons laut Stylissa flabelliformis secara topikal ini dapat meningkatkan kadar minyak (sebum) dan kelembapan kulit (moisture), meskipun tidak signifikan. Sedangkan elastisitas kulit dan kandungan collagen kulit tidak terpengaruh. Dapat disimpulkan bahwa penggunaan ekstrak collagen ini dapat memperbaiki profil kelembapan kulit, sehingga dapat dikembangkan sebagai pelembap alami.

Abstract

Purple leaf (Graptophyllum pictum L. Griff) is a plant that has antibacterial, antioxidant, anti-inflammatory and antidiarrheal benefits. Flavonoids, alkaloids, steroids, saponins, and tannins are some of the secondary metabolites found in the leaves that have antibacterial properties. This study aims to determine the content of phytochemical compounds in purple leaf twig extract. The method used is phytochemical screening to identify flavonoid, alkaloid, saponin, tannin, steroid and terpenoid compounds. The results of phytochemical screening showed that the purple leaf twig extract contained flavonoid, saponin and terpenoid compounds. However, it did not show any steroid compounds. The conclusion is that the identification of secondary metabolite compounds in purple leaf twig extract contains flavonoid, saponin and terpenoid compounds. In contrast to the content of secondary metabolite compounds in G. pictum twigs is different from that in its leaves.

Abstract

Turbinaria sp. is valuable source of various metabolites that exhibit pharmacological and biological activities. The diversity of these metabolites can be influenced by several factors, such as environmental conditions and the choice of solvent during the extraction process. This study aims to investigate the effect of different solvents and methods on the metabolite profiles of Turbinaria sp. extract Turbinaria sp. was extracted using maceration and soxhlet method with n-hexane and n-hexane:methanol as solvents. Phytochemical analysis was performed using tube tests. Meanwhile the determination of total phenols and total flavonoids was performed by spectrophotometry assay. The results has shown that Turbinaria sp. contain flavonoids, alkaloids, terpenoids, phenols, and tannins, with the highest total phenol and total flavonoid content found in the extracts using n-hexane as a solvent and soxhletation methods.

Abstract

UV radiation from sunlight can induce the formation of free radicals, which can disrupt cellular homeostasis or DNA by triggering inflammatory signal transduction. The occurrence of inflammation in the skin can be aggravated by the presence of the bacterium Staphylococcus aureus. Previous studies have shown that the marine sponge Stylissa sp. contains an abundance of fatty acids and lipids compared to other species. Although Stylissa sp. has great potential for health applications, research on the bioactivity of lipid and fatty acid compounds from this sponge remains limited. Therefore, this study aims to explore the lipid content of Stylissa sp. as an alternative source of antibacterial agents against pathogens responsible for photoaging. The metabolite profiling of the extract was conducted using GC-MS, while the antibacterial activity was performed using the Kirby-Bauer method. Based on GC-MS profiling results, six compounds were identified in the lipid extract of Stylissa sp. namely palmitic acid, butyl glycol acetate, n-eicosylcyclohexane, isopropyl laurate, oleic acid, and 1-tetradecanol. Antibacterial evaluation of Stylissa sp. lipid extract at concentrations of 25%, 75%, and 100% showed antibacterial activity against Staphylococcus aureus, with inhibition zone diameters of 1.83, 2.17, and 2.92 mm, respectively. The results in this study have shown that the lipid extract of Stylissa sp. contains lipid compounds with potential anti-bacterial activity towards S. aureus. Future research to isolate unsaturated fatty acid compounds from the lipid extract of Stylissa sp. to achieve higher antibacterial activity is recommended.

Abstract

Sargassum sp. is an abundant source of bioactive non-polar compounds, however their anti-bacterial potential remains understudied. This study aimed to compare the anti-bacterial activity between the methanol extract and non-polar fraction of Sargassum sp. Extraction was performed using sonication with methanol, followed by partition with petroleum ether and fractionation on silica gel G60 using n-hexane:ethyl acetate (9:1). FTIR analysis characterized the functional groups, while anti-bacterial evaluation was performed using the disc diffusion method against Staphylococcus aureus. Data were analyzed statistically using one-way ANOVA. FTIR spectrum of the methanol extract showed absorption bands corresponding to non-polar compounds. In the non-polar fraction, the bands were more dominant, suggesting a higher content of non-polar compounds. Anti-bacterial evaluation of Sargassum sp., methanol extract and non-polar fraction exhibited inhibitory effects against S. aureus. The non-polar fraction produced a larger inhibition zone compared to the methanol extract, indicating stronger anti-bacterial activity and significant difference between the two samples (p<0.05). The results of this study showed that the non-polar fraction of Sargassum sp. methanol extract has higher anti-bacterial activity compared to the extract. Further studies are recommended to elucidate the individual bioactive compounds and identify their anti-bacterial mechanisms.
