Abstract

Desa Eko-Eduwisata Kemuning memiliki potensi produk lokal yang beragam sebagai penunjang sektor pariwisata. Salah satunya adalah UMKM Lumbung Rasa yang menghasilkan produk kripik, roti, dan herbal instan. Namun, usaha ini masih menghadapi kendala pada aspek produksi, manajemen, dan pemasaran. Kondisi tersebut menjadi tantangan dalam meningkatkan daya saing produk sekaligus mendukung hilirisasi produk lokal yang berkelanjutan. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk memperkuat kapasitas UMKM melalui pengembangan sistem produksi dan branding produk lokal sebagai pilar hilirisasi Desa Eko-Eduwisata Kemuning. Metode yang digunakan meliputi identifikasi kebutuhan mitra, penyuluhan, pelatihan, pendampingan, serta monitoring dan evaluasi. Program dilaksanakan melalui penerapan teknologi tepat guna produksi, edukasi manajemen mutu industri pangan yang mencakup penerapan keamanan pangan, pelabelan, dan penentuan tanggal kedaluwarsa, pelatihan manajemen digital, serta pendampingan branding dan pemasaran digital melalui media sosial. Hasil kegiatan menunjukkan adanya peningkatan pengetahuan dan keterampilan peserta dalam menerapkan prinsip manajemen mutu pangan, melakukan pencatatan keuangan secara digital, serta menyusun strategi branding yang lebih efektif untuk meningkatkan identitas dan daya saing produk lokal. Sinergi antara perguruan tinggi, pemerintah desa, dan pelaku UMKM juga memperkuat keberlanjutan program dalam mendukung pengembangan ekonomi berbasis potensi lokal. Kegiatan ini berkontribusi terhadap pencapaian Tujuan Pembangunan Berkelanjutan (SDGs), khususnya Tujuan 1 (Tanpa Kemiskinan), Tujuan 4 (Pendidikan Berkualitas), Tujuan 8 (Pekerjaan Layak dan Pertumbuhan Ekonomi), dan Tujuan 17 (Kemitraan untuk Mencapai Tujuan), melalui penguatan kapasitas masyarakat dan pengembangan UMKM yang berdaya saing.

Abstract

Empowerment of Jaya Farmer Group and MBS Farmers in Purworejo Sragen Through Complete Feed Preparation Training for the Village Development Program. The Village Empowerment Program implemented in Purworejo Village, Gemolong District, Sragen Regency, was motivated by the community's interest in raising dairy goats. The dairy goats that are quite developed include PE, Jawa Randu, and Sapera dairy goats. This Village Development Program is funded by the Ministry of Education, Culture, Research, and Technology. The development of dairy goats starts from the smallest unit, namely farmers, by forming a Livestock Farmers Group association. The Village Empowerment Program invites two Livestock Farmers Groups in Purworejo Village to become partners. The Livestock Farmers Groups targeted in the village empowerment activities are the Jaya Livestock Farmers Group and the Muda Bakti Sejahtera Livestock Farmers Group. The problems that arise in livestock farmers are related to milk production that is not yet optimal. Low milk production can be caused by the provision of feed that is not Accordance with the nutritional needs of livestock. The nutritional needs of livestock in each physiological phase are different. The solution provided through this Village Empowerment Program is the manufacture of complete feed for dairy goats. Complete feed is a fairly simple technology in livestock feed management. The ration for making complete feed is adjusted by looking at the availability of feed ingredients around Purworejo Village, namely cassava, bran, corn cobs, CGF, copra cake, and minerals. The Village Empowerment Program begins with socialization followed by workshops and mentoring. The conclusion after a series of programs have been completed is that farmers can understand the use and process of making complete feed technology which is assessed through the tests provided and their activeness during the mentoring session.

Abstract

Biologi mikroskopis mempelajari organisme pada level seluler dan menjadi bagian tak terpisahkan dari pengetahuan IPA modern. Di SMP Djama’atul Ichwan Surakarta, keterbatasan fasilitas membuat pembelajaran konsep-konsep biologi mikroskopis sulit dibarengi dengan pengalaman praktek di laboratorium sekolah. Mitra mengharapkan ada pendampingan untuk meningkatkan kompetensi praktek pada mata pelajaran biologi. Kegiatan pengabdian ini bertujuan meningkatkan kompetensi praktek dan wawasan umum siswa mengenai biologi mikroskopis yang tercakup dalam materi IPA di SMP. Kompetensi praktek biologi mikroskopis yang belum maksimal pada mitra berakar dari tidak adanya laboratorium IPA, media praktikum, dan mikroskop. Tim PKM UNS menghadirkan solusi berupa program pendampingan kepada siswa selama satu semester yaitu, 1) pengenalan mikroskop dan aplikasinya; 2) praktek membuat preparat mikroskopis sederhana sesuai materi SMP; 3) pengadaan mikroskop cahaya; modul praktikum; foldscope untuk mitra; 4) komunikasi selama semester berjalan dan umpan balik mengenai aktivitas praktikum dengan mikroskop di SMP. Keberhasilan kegiatan dapat dilihat dari indikator skor pre-/post-test, aktivitas praktikum, dan pengadaan mikroskop untuk SMP. Siswa menjadi paham prinsip kerja dan aplikasi mikroskop untuk visualisasi struktur seluler dan jaringan, hal ini tampak dari peningkatan skor post-test (>25% kenaikan skor) pada 72% siswa. Pada sesi praktikum, siswa mampu membuat dua preparat berbeda untuk diamati dengan mikroskop dan diidentifikasi bagian-bagiannya. Selanjutnya, siswa dapat melakukan praktek mandiri di SMP menggunakan mikroskop dari investasi kegiatan pengabdian ini Kata kunci: kompetensi praktik, biologi mikroskopis

Abstract

A mechanism controlling a growing cancer cells is by a programmed cell death (apoptosis). The wildtype-p53 enable to stop cleaves that follow DNA repair or cell death (apoptosis). The mutation of wt-p53 caused loosing its ability to inhibit cancer cells proliferation. Healing methods like surgery, radiation, immunotherapy and chemotherapy still have some weaknesses, and clinical medicine to cancer is also still has any dissatisfactory. Much of chemotherapy was not given optimal result yet, because no specific action to cancer cells only, but also to the normal cells. These problems encourage important effort to find specific and sensitive anticancer. Empirical evidence indicates that the crude extract of Pandanus conoideus Lamb var. yellow fruit has potential effect as an anticancer. Method of Freshney was used in growing T47D cell line, counting cells was done by direct counting, and apoptotic evaluation was done by TUNEL enzymatic labeling assay. The results of theresearch demonstrated that the LC50 of yellow fruit extract are 0.25 μL/mL. The percentage of apoptotic of 0.125 μL/mL,0.0625 μL/mL, and 0.03125 μL/mL are 34.38±2.26, 30.03±3.87 and 21.07±1.14 respectively.Key words: T47D, Apoptotic, p53, Pandanus conoideus Lamb. var. yellow fruit.

Abstract

The hydrogen sulfide is one of compound which very often found in the shrimp pond caused by anaerobic decomposition or as a natural condition of the sea water which have volcano activity. This research was obtaining information of the differences of genetic expression between black tiger shrimp which could resist to H2S and the one which could not survive in this H2S. This research also trying to obtain information the genetic variety which could resist to H2S. The genetic variety of black tiger shrimp which could resist to H2S has been analysis with allozyme electrophoresis technique, using specific tissue meat and buffer CAPM (Citric Acid Aminopolimorpholine) pH 6.From the three enzymes analyzed it could be detected that IDH enzyme (Isocitrate dehydrogenase) has locus polymorphic, whereas enzyme α-GDP (α-glycerophosphate dehydrogenase) and LDH (lactate dehydrogenase) with monomorphic locus. The average heterozygosity for the group which could resist to H2S is 0.070, whereas the group which could not survive in the H2S is 0.042. The control group has heterozygosity 0.041. The group which could resist to H2S with higher heterozygosity will have bigger chance to survive and have better adaptation ability in environmental changes. Genetic diversity of Ongole Grade, Aceh, and Sumbawa cattle based on polymorphism on ND-5 fragment mitochondrial DNA using PCR-RFLP technique. Biodiversitas 20: 783-788. Genetic diversity is the basis of livestock breeding because it can be used as an initial improvement in livestock quality through artificial selection. This study aims to determine polymorphism in ND-5 fragment of mitochondrial DNA in Ongole Grade, Aceh, and Sumbawa cattle and their genetic diversity. The total DNA from the blood of the local cattle was extracted using the Wizard genomic DNA purification system from Promega and amplified using the PCR technique. The PCR product was then digested with HindIII enzyme using the RFLP technique to detect polymorphism. The genetic diversity of the Ongole Grade, Aceh, and Sumbawa cattle was analyzed using the formula from Nei and its genetic relationship was evaluated with the 2.02i NTSYSpc version program. Our findings showed there were polymorphisms in the ND-5 fragment of mitochondrial DNA. Digestion with HindIII restriction enzyme produces two types of haplotypes. Haplotype B is a 453 bp-sized DNA fragment that is not truncated by the HindIII enzyme, and haplotype A is a DNA fragment cut by HindIII enzyme into two with fragments of 336 bp and 117 bp. Polymorphism was found in Ongole Grade cattle, but not in Sumbawa and Aceh cattle. Haplotypes diversity in ND-5 fragments of mitochondrial DNA of Ongole Grade was 0.6250 while Sumbawa and Aceh cattle displayed no diversity of haplotypes. The genetic relationship shows that Sumbawa cattle belonged to the same cluster with Ongole Grade but separated from Aceh cattle.

Primer design of the CO1 gene (Cytochrome Oxidase-1) for Sumatran elephant (Elephas maximus sumatranus) for rapid detection using real-time PCR method. Biodiversitas 25: 3840-3849. The population of Sumatran elephants is classified as endangered according to the IUCN due to conflicts between humans and elephants, resulting in many elephants being killed by humans to obtain ivory to be sold. The traded ivory is often processed into other ornaments. A rapid forensic examination with real-time PCR using the molecular approach of the CO1 gene (Cytochrome Oxidase-1) is necessary to determine the original species of the processed elephant ivory. This research aims to design and optimize a primer for identifying Sumatran elephants (Elephas maximus sumatranus). The CO1 gene primer for Sumatran elephants was designed using the Primer3Plus and IDT websites. The primers were used to amplify and identify the species of Sumatran elephants from blood, feces, and urine samples. Blood and feces samples were collected from Sumatran elephants at Taru Jurug Zoo Surakarta, while feces and urine samples were taken from Sumatran elephants at Ragunan Zoo Jakarta. DNA from blood and urine samples was extracted using the TIANamp Genomic DNA Kit, and DNA from feces samples was extracted using the TIANamp Stool DNA Kit. The purity and concentration of the obtained DNA extracts were measured. The designed primers were synthesized by Macrogen (Korea), and primer optimization was performed with a PCR gradient. The identification process of Sumatran elephant DNA extract samples was conducted by amplification using real-time PCR quantification with the standard curve method. The results of this research include the COI-270 primer with the sequences 3'-TTAGGTCAACCAGGCTCTCTTC-5' and 5'-AGGATATACGGTCCAACCAGTG-3', capable of amplifying the target 270 bp and having a single peak at the melt curve temperature of 79°C. The identification protocol with specific COI-270 primer can be used in forensic examinations to identify samples originating from Sumatran elephants.

Biodiversity and conservation status of flora and fauna in Magetan, East Java, Indonesia: A focus on rare and protected species of mammals, birds and plants. Biodiversitas 26: 1606-1617. Indonesia is recognized for its high level of species endemicity across its islands, but this biodiversity is under significant threat due to high loss rates. Magetan District, East Java Province, Indonesia comprises 8% of protected areas, located adjacent to Mount Lawu, which harbor substantial biodiversity. This study aims to assess the diversity, distribution and protection status of wildlife in Magetan, based on the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) and the Minister of Environment and Forestry Regulation of the Republic of Indonesia, especially for mammals, birds and plants. The research utilized a combination of methods, including the collection of biodiversity data from 2013 to 2022, interviews with relevant stakeholders, and field observations. The findings identified three mammalian species and one plant listed under CITES Appendix I. Additionally, 19 wild animal species and one plant species were found to be rare and protected under the government regulation. Among these, five species belong to the mammal group, while 14 species are avifauna group. The highest concentrations of these protected species were observed in the Panekan and Plaosan Sub-districts, highlighting the importance of these two areas for conservation. Conservation strategies are required to protect these animals and plants by avoiding deforestation and habitat restoration. Other conservation strategies are by enforcing laws, addressing human-wildlife conflicts, raising awareness, and conducting regular monitoring. The study provides critical insights for conservation policy development and biodiversity documentation, specifically for Magetan District.

Assessment of biodiversity and conservation status of flora and fauna in Madiun City, East Java, Indonesia. Biodiversitas 26: 3965-3972. Madiun City in East Java, Indonesia is a small municipality that has evolved into an urban center due to its strategic position as the hub of socio-economic activities, including trade, industry, and healthcare. Urbanization in the town poses a threat to biodiversity. This is due to changes in land use and cover, natural exploitation, introduction of exotic species, air and water pollution, and climate change. This study aims to explore biodiversity in Madiun City to inform the development of strategies and policies for natural resource management. The observations revealed that Madiun City is home to 122 tree species, 76 species of ornamental plants, and 143 species of animals, including mammals, birds, reptiles, amphibians, fish, and insects. Eight plant species and five animal species are listed in the IUCN Red List. Additionally, there are eight plants and five animals listed under CITES, as well as six animals listed as protected under the Indonesian Ministry of Environment and Forests' P106/2018. Leucopsar rothschildi, Padda oryzivora, and Pavo muticus are birds listed under all the three protection instruments, suggesting their conservation importance. Green open spaces with aviaries are crucial for conserving biodiversity, as they offer a range of ecological, aesthetic, social, and economic benefits. Community participation with the local government is vital to conservation programs in urban areas. Active community involvement in conservation programs and socialization can significantly support the success of biodiversity conservation.

Abstract

Background: Sexual behavior among teenagers is worrying. It is influenced of permissive attitude shift, causing violation of norms, ethics in society. One factor is the opening of information about sex freely circulating in the community. In Surakarta adolescent at high school never did kiss the lips of 10.53%, a kiss in 5.6%, 4.23% masturbation and sexual intercourse 3.09%. This study aimed to determine the association of self-esteem, self-efficacy, peers, parents, with premarital sexual behavior in adolescent high school in Surakarta.Subjects and Method: This was an observational analytic study with cross sectional design. A total sample of 113 respondents was taken by using proportional random sampling. The population was in ten high schools of 1663 subjects in Surakarta. Data analysis used linear regression.Results: There is a relationship of self-esteem with premarital sexual behavior (b1= -0.202; 95% CI= -0.39 to -0.01; p= 0.036) were significantly higher self-esteem then reduce premarital sexual behavior. There is a relationship of self-efficacy with premarital sexual behavior (b2= -0.168; 95% CI= -0.33 to -0.01; p= 0.041) were significantly higher self-efficacy students then reduce premarital sexual behavior. There are peer relationships with premarital sexual behavior (b3= -0.271; 95% CI= -0.51 to -0.04; p= 0.024) were significantly better peer support and will reduce premarital sexual behavior. There is a parental relationship with premarital sexual behavior (b4= -0.236; 95% CI= -0.44 to -0.03; p= 0.025), the better the more parental supervision reduce premarital sexual behavior. There is a relationship of self-esteem, self-efficacy, peers, parents with premarital sexual behavior (F = 10.053; 95% CI= 24.16 to 43.61; p <0.001; R 2 = 24.4%)Conclusion: Self-esteem, self-efficacy, peers, and parents are associated with premarital sexual behavior in adolescents of 24.4%.
