Abstract

Mapping expressed sequence tags (ESTs) to hexaploid wheat is aimed to reveal the structure and function of the hexaploid wheat genome. Sixty eight ESTs representing 26 genes were mapped into all seven homologous chromosome groups of wheat (Triticum aestivum L) using a polymerase chain reaction technique. The majority of the ESTs were mapped to homologous chromosome group 2, and the least were mapped to homologous chromosome group 6. Comparative analysisbetween the EST map from this study and the EST map based on RFLPs showed 14 genes that have been mapped by both approaches were mapped to the same arm of the same homologous chromosome, which indicated that using PCRbasedESTs was a reliable approach in mapping ESTs in hexaploid wheat.Key words: wheat, EST, PCR, homologous.

Prasetyo B, Chikmawati T, Walujo EB, Amzu E. 2018. Ethnoecology: The traditional landscape of Osing Tribe in Banyuwangi, Indonesia. Biodiversitas 19: 2003-2009. There is a concern that the decreasing area of Osing tribe in Banyuwangi may cause the decline of the farming culture that has been rooted in their daily lives. This research aimed to analyze the traditional knowledge of Osing tribe in the management and utilization of landscape units generated from farming activities and the benefit assessment of landscape units based on gender perception. The research used explorative method with emic and ethic approaches. The importance of each landscape element is assessed through MLA (Multidisciplinary Landscape Assessment) by scoring with Pebble Distribution Method (PDM) in Focus Group Discussion (FGD). The respondents consisted of females and males with three age categories: 11-17 years, 18-45 years, and ? 46 years. The results of PDM showed that the Osing tribes in the three villages of study sites considered their yards as the most important landscape unit in their lives (PDM = 42.82), followed by paddy field (PDM = 26.05), garden (PDM = 16.12), and finally field (PDM = 15.01). Based on gender perception, a house with a yard had higher importance (PDM = 45.09) in female’s perception than in male’s (PDM = 40.55). Three canopy strata or layerswere found in village landscapes, namely yard, paddy field, garden, and field.

Pratami, MP Chikmawati T, Rugayah. 2019. Short Communication: Further morphological evidence for separating Mukia Arn. from Cucumis L.. Biodiversitas 20: 211-217. Mukia Arn. is closely related to Cucumis L. based on molecular data, nevertheless, they have high morphological differences resulting in different opinion on taxonomical status of the two genera. Mukia Arn. has many similarities in pollen and leaf anatomical characters to Cucumis L., but both genera differ in seven seed characters, i.e color, shape, size, surface pattern, seed edge, transverse section at seed neck, and the markings of the inner seed coat surface. So, based on seed characteristics, Mukia Arn. is separated from Cucumis L.Keywords: Cucurbitaceae, leaf anatomy, Malesia

Abstract. Mahyuni R, Chikmawati T, Ariyanti NS. 2019. Short Communication: Two new species and new record of  Psydrax Gaertn. (Rubiaceae: Vanguerieae) in Borneo. Biodiversitas 20: 2011-2015. Psydrax sabahensis Mahyuni and P. wongii Mahyuni are two new species endemic to Borneo proposed in this present study with descriptions, illustrations and map of distribution provided. Psydrax sabahensis has blades ovate to elliptic, with angle 60º to the midrib, tertiary venation conspicuous on both side, petiole length 10-13 mm long, inflorescence sub-umbellate and pedicels 23-27 mm long. Psydrax wongii has blade elliptic, with angle 45º to midrib, apex acuminate to caudate, secondary veins 3-4 pairs, tertiary venation inconspicuous on upper side and slightly wrinkled on lower surface. The presence of P. maingayi (Hook.f.) Bridson in Borneo, previously thought to be a Malay Peninsula endemic species, is also confirmed as a new record.

Abstract. Miftahudin, Hasibuan RS, Chikmawati T. 2019. Antioxidant activity of ethanolic extract of three Selaginella species from Java Island, Indonesia. Biodiversitas 20: 3715-3722. Three Selaginella species, S. ornata, S. plana, and S. willdenowii, from Java Island, Indonesia, have been known to have antioxidant properties; however, in vivo antioxidant activities of these species have not been reported. This research aimed to evaluate the in vivo antioxidant activity of ethanolic extract of three Selaginella species. The 70% ethanol extract of three Selaginella species at four different doses was administered to mice one day before being treated with oxidative stress. The liver tissue of mice treated with or without oxidative stress was analyzed their lipid peroxidation by measuring MDA concentration and Superoxide Dismutase (SOD) activities. The results showed that there were variations in antioxidant activity among the three Selaginella species. In general, the dose of 0.3 g extract kg-1 BW has been able to reduce lipid peroxidation and increase SOD activity. The administration of S. ornata extract to the mice at 1.2 g extract kg-1 BW reduced the MDA concentration to the lowest level, but the same dose of two other Selaginella extracts caused toxic effects in mice. The antioxidant activities of S. ornata and S. plana were better than that of S. willdenowii extract, and among those species, S. ornata has the best antioxidant activity.

Abstract. Nikmah IA, Rugayah R, Chikmawati T. 2020. Leaf anatomical variation in Desmos Lour. and Dasymaschalon (Hook. f. & Thomson) Dalla Torre & Harms species (Annonaceae). Biodiversitas 21: 3317-3330. The relationships between Desmos and Dasymaschalon are debated for along time. Those two genera have high morphological similarities, especially in their generative character (moniliform monocarps). Therefore, sterile specimens of Desmos are difficult to be distinguished from Dasymaschalon. Leaf anatomy in paradermal section of 20 taxa (12 species of Desmos, eight species of Dasymaschalon) have been carried out. The data were used to support the interspecific and intergeneric delimitation of Desmos and Dasymaschalon. Desmos and Dasymaschalon are two distinct genera mainly distinguished based on the anticlinal wall undulation of epidermal cells and supported by variation of the crystal type, and size. The anticlinal wall undulation of Desmos is almost straight to slightly wavy, and never sinuous, meanwhile, Dasymaschalon varies from almost straight to deeply sinusoid. The crystals of Desmos consist of rhombohedric, druse type A, and druse type B crystals, whereas Dasymaschalon has prism, druse type A, druse type B, and drue type C crystals.

Abstract. Mildawati, Sobir, Sulistijorini, Chikmawati T. 2020. The diversity of pteridophytes in Siberut National Park, Mentawai Islands, West Sumatra, Indonesia. Biodiversitas 21: 3200-3208. The diversity of Pteridophytes in the Siberut National Park (Siberut NP) Mentawai Islands, West Sumatra, is still very well preserved, but the information is limited. This research aimed to study the divesrity of Pteridophyte species in the Siberut NP in order to provide its taxonomic information using morphological data. The study consisted of exploration to collect plant specimens directly in the field, observation and identification of herbarium specimens in Herbarium Bogoriense and verification using online websites from various sources. A total of 69 species of Pteridophytes classified into 36 genera, and 20 families were recorded in the Siberut NP with Polypodiaceae has the most species diversity with 12 species of 6 genera. As many as 14 species are new records for Siberut Island, i.e. Selaginella involvens (Sw.) Spring (Selaginellaceae), Asplenium simile Blume (Aspleniaceae), Diplazium montanum V.A.V.R (Athyriaceae), Stenochlaena palustris (Burm) Bedd. (Gleichnaceae), Davallia solida (Forst.) Sw. (Davalliaceae), Callistopteris apiifolia (C.Presl) Copel., Crepidomanes humile Bosch (Hymenophyllaceae), Lindsaea carvifolia Kramer, Lindsaea doryphora Kramer, Lindsaea napaea Aldrew. (Lindsaeaceae), Nephrolepis biserrata (Sw.) Schott (Nephrolepidaceae), Ceratopteris thalictroides (L.) Brongn., Haplopteris ensiformis (Sw.) E.H.Crane and Syngramma alismifolia (Pr.) J.Sm. (Pteridaceae). The habitats of Pteridophytes found were mostly terrestrial and epiphytic (34 species each), while aquatic habitat was only found to one species. The results of this study will serve as baseline information of the plant diversity in the Siberut NP that can be used as the reference for future studies on Pteridophytes and to develop their conservation strategy in the region.

Abstract. Miftahudin, Putri RE, Chikmawati T. 2020. Vegetative morphophysiological responses of four rice cultivars to drought stress. Biodiversitas 21: 3727-3734. Each rice genotype develops certain morphophysiological responses to drought stress. The study aimed to analyze the morphophysiological responses of vegetative aspect of four rice cultivars to drought stress. A 10% Polyethylene glycol-6000 was added to a Yoshida nutrient solution medium as a drought stress stimulant for four rice cultivars, i.e., IR64, Hawara Bunar, Situbagendit, and Inpago 10. Fourteen-days-old rice seedlings were grown on the media with and without drought stress treatment for 9 days, and morphophysiological characters of vegetative aspects were observed. Drought stress inhibited the shoot growth of cv. Hawara Bunar, but increased shoot growth of cv. Inpago 10. The physiological responses in the form of leaf relative water content, proline, malondialdehyde (MDA), and total chlorophyll contents in cv. Hawara Bunar was inversely proportional to those of cv. IR64 showed an inferior response to drought stress. The rice cv. Hawara Bunar might develop better response mechanisms to drought than that of cv. IR64. The physiological responses of cvs. Situbagendit and Inpago 10 were in between the other two cultivars. We conclude that the variation of morphophysiological responses to drought stress among rice cultivars is an indicator of tolerance capability to drought that could be used as early-growth selection criteria in rice breeding programs for drought tolerance.

Abstract. Damayanto IPGP, Rustiami H, Miftahudin, Chikmawati T. 2020. A Synopsis of Bambusoideae (Poaceae) in Lombok, Indonesia. Biodiversitas 21: 4489-4500. There is little documentation of bamboo on Lombok. Non-native Indonesian bamboos have been reported occurring in Lombok. As such, suspicious bamboo specimens and information in those reports need to be examined and possibly re-determined. This study aimed to provide updated taxonomical information of bamboo species in Lombok and to investigate the accuracy of previous studies regarding the existence of several non-native Indonesian bamboo species on the island (i.e. they do not have original distribution in Indonesia). Data from fieldwork were combined with specimens in Herbarium Bogoriense to create a synopsis of bamboo in Lombok and to develop an identification key to the species. Eleven species of bamboos in Lombok were reported from this study, namely Bambusa glaucophylla, B. maculata, B. multiplex, B. spinosa, B. vulgaris, Dendrocalamus asper, Gigantochloa apus, G. atter, Schizostachyum brachycladum, S. lima, and Thyrsostachys siamensis. Several doubtful species (B. albustiata, B. ventricosa, Guadua angustifolia, S. jaculans, Pseudosasa japonica) were presented to clarify the existence of a non-native Indonesian bamboo in Lombok. Moreover, there is not enough evidence to state Dinochloa sp. occurred in Lombok. The results of this study can be used by stakeholders for bamboo identification and as the baseline taxonomic account for future studies and conservation of bamboo species in Lombok.

Abstract. Makmur K, Chikmawati T, Sobir. 2020. Genetic variability of lemon basil (Ocimum × africanum Lour.) from Indonesia based on morphological characters and ISSR markers. Biodiversitas 21: 5948-5953. Lemon basil (Ocimum × africanum Lour.) or kemangi is a highly beneficial plant, yet still underutilized. Therefore, providing necessary information on genetic diversity of this species is essential for further utilization. The aim of this study was to elucidate genetic diversity of 33 accessions of O. × africanum collected from four Islands of Indonesia along with three accessions of Ocimum basilicum L. Morphological observation was conducted on 37 morphological characters following the International Union for the Protection of New Varieties of Plants descriptor guidelines; subsequently, 13 ISSR primers were employed in molecular analysis. Both morphological and molecular data were analyzed based on simple matching similarity index using UPGMA method. Morphologically, O. × africanum and O. basilicum were clearly separated at the similarity index of 0.52, and among two species were divided into two groups according to two either character. ISSR analysis using 13 ISSR primers produced 111 DNA bands, and 108 of them (97.29%) were polymorphic. Cluster analysis based on ISSR data could not explicitly separate O. × africanum and O. basilicum accessions. Besides, Ocimum accessions collected from the same area did not always cluster into one group.
