Abstract

The effect of Justicia gendarussa leaves extract has been investigated using oxonate-induced hyperuricemic male albino rats. In this experiment thirty five male albino rats were divided randomly into seven groups. There were three group doses variations of J. gendarussa extract that were 1.3 g/kg bw, 2.6 g/kg bw and 5.2 g/kg bw; two comparison groups that were allopurinol 0.68 g/kg bw and herbal “X” 0.85 g/kg bw; and two control groups that were induced control potassium oxonate 0.25 mg/kg bw and normal control CMC 0,5 %. Intraperitoneal administration of potassium oxonate 0.25 mg/kg bw was given one hour before administration of test drugs in eight day and plasma uric acid was measured in rats after two hours. Plasma uric acid was measured by spectrophotometric on 520 nm with enzymatic method. The results showed that all extracts could reduced uric acid level of rats. The decreasing potency of uric acid level was equal to doses increase, so the best result that can reduce uric acid level was J. gandarussa extract at a dose 5.2 g/ kg bw. This results indicated that J. gendarussa leaves extract may be effective for the prevention and the treatment of hyperuricemia.

Abstract

The community uses Tetracera indica (Christm. and Panz.) Merr extensively as a traditional medicine. This plant was the main source of raw materials for herbal medicinal products, so identification was needed to ensure its authenticity through pharmacognosy evaluation. The present study evaluated macros- and microscopic characteristics, performed phytochemical analysis, and performed total phenolic (TPC) and flavonoid content (TFC) analysis. The organoleptic and macroscopic studies were carried out through sensory organs. The microscopic characteristics of leaf powder and twigs identification using chloral hydrate and distilled water. Numerous chemical reagents, TLC methods and LC-MS/MS analysis were employed in the phytochemical study. Research employed DPPH and FRAP tests to assess the antioxidant activity. Furthermore, quercetin was used as a marker of antioxidant activity. The morphological marker of the leaf T. indica showed the surface of the leaves is rough bristles, shape elliptic-oblong, margin serratus and phyllotaxy alternate. The stem is lignosus, and the rough stem surface is slightly peeling. The lower leaf surface had paracytic stomata and uniseriate trichomes with tapering ends. Raphide crystals and cork cells were detected in the powder analysis of the twigs. The ethyl acetate extract's twig had the highest TPC and TFC values. Ethyl acetate extract from Tetracera indica twigs has potent antioxidant activity with IC50 = 99.84±0.011 µg/ml (DPPH) and 4296.67±0.024 mol/g (FRAP). The study showed that Tetracera indica contains possible active components that could be developed as novel antioxidants. The result of this study might provide early verification and identification to obtain quality medicinal raw materials.

Abstract

Saponin is a group of compounds contained in natural materials that have amphiphilic properties and can reduce surface tension. The reduction of surface tension caused by a soap compound (Latin = sapo) that can disrupt hydrogen bonds in water. Red frangipani plant (Plumeria rubra) is known to have saponin content. The research objectives were to identify the saponin content of red frangipani plant extract (Plumeria rubra) which could reduce the surface tension. Part of red frangipani plant (flowers, leaves and stems) was extracted using five kinds of solvents. Each of the extracts obtained was then tested for saponin content qualitatively. Extract from each part of the plant (flower, leaf, and stem) which has the highest foam was selected then tested surface tension using surface tensionmat equipment. The result of qualitative saponin test showed that flower, stem and flower extract of red frangipani with aqua demineralisata solvent had the highest saponin content compared to extract with other solvent. The content of saponins in plumeria rubra extract either from the leaves, stems or flowers could decrease the surface tension with the best results obtained from the flower extract with 8,61% of Critical Micelle Concentration (CMC).
