Abstract

Background: The diagnosis of Dengue Fever (DF) involves physical examination, laboratory tests, and confirmation primarily through thrombocytopenia and leukocyte abnormalities. Laboratory procedures play a crucial role; however, delays in complete blood count (CBC) analysis may influence hematological parameters, potentially affecting diagnostic accuracy. Objective: To determine the effect of 2-hour and 4-hour delays in CBC testing on leukocyte and platelet counts in patients with Dengue Fever. Methods: This experimental study used a posttest-only control group design with purposive sampling. Blood samples from DF patients were divided into three groups: immediate testing, 2-hour delay, and 4-hour delay. Data were analyzed using the Shapiro-Wilk test followed by Multivariate Analysis of Variance (MANOVA). Results: The mean leukocyte and platelet counts in the immediate testing group were 4.86×10³/μL and 61.55×10³/μL, respectively. In the 2-hour delay group, leukocyte counts slightly decreased to 4.79×10³/μL and platelet counts increased to 65.77×10³/μL. In the 4-hour delay group, leukocyte counts increased to 4.85×10³/μL while platelet counts decreased to 64×10³/μL. MANOVA results (Sig. = 1.000) indicated no significant differences among the groups. Conclusion: Delays of 2 and 4 hours in CBC testing did not significantly affect leukocyte and platelet counts in Dengue Fever patients. These findings suggest that short-term delays in sample analysis may not compromise hematological results in DF diagnosis.

Abstract

Lawar plek is a traditional Balinese dish made from raw meat, which poses a potential risk of contamination by pathogenic bacteria, including Enterotoxigenic Escherichia coli (ETEC). This study aimed to identify the presence of ETEC in lawar plek samples from Ketewel Village, Gianyar. This research uses molecular methods. The identification was conducted in two stages: initial isolation of suspected bacteria on Eosin Methylene Blue Agar (EMBA), followed by molecular confirmation using Polymerase Chain Reaction (PCR). The study results show that in EMBA culture, 4 out of 12 samples (33.33%) showed colonies presumptively identified as E. coli. However, PCR analysis revealed that none of these isolates carried the LT (heat-labile toxin) or ST (heat-stable toxin) genes characteristic of ETEC. The conclusion shows that neither the LT (heat-labile toxin) nor the ST (heat-stable toxin) virulence genes of Enterotoxigenic Escherichia coli were found in any of the samples tested as suspected Escherichia coli colonies on EMBA.

Abstract

The nutritional status of pregnant women can be determined by measuring the size of mid-upper arm circumference (MUAC) and body mass index (BMI). Women with excessive nutritional status have a high risk of various complications. This study aims to describe the nutritional status and blood glucose levels of pregnant women at the Ubud Health Center, Bali. This descriptive study was conducted on 27 pregnant women who had antenatal care at the Ubud Health Center. MUAC size was measured using a measuring tape, while BMI was measured by calculating body weight in kilograms divided by height in meters squared. Blood glucose levels were measured using the point of care testing (POCT) method. From 27 respondents, it was found that 23 respondents had normal MUAC, 3 respondents had less than normal, and 1 respondent was obese. Based on BMI, 14 respondents had normal BMI, 5 respondents were overweight, and 8 respondents were obese. For blood glucose levels, all respondents have normal levels. Respondents who have MUAC less than normal can be at risk of chronic energy deficiency, while respondents with the obese category can be at risk of GDM, hypertension, and cesarean section. Pregnant women with GDM can increase the risk of type 2 DM, cardiovascular disease and have the potential to give birth macrosomic babies. Pregnant women with GDM almost never give complaints, so screening is necessary. Screening can be done by measuring the nutritional status of pregnant women and blood glucose levels during antenatal care at health center.

Abstract

Visfatin, a protein with a molecular weight of 52 kDa, has been demonstrated to increase in the bloodstream. Visfatin has been shown in several studies to be a potential marker of preeclampsia. The objective of this study was to describe visfatin levels in pregnant women in Gianyar, Bali. An observational design was employed in this study to describe visfatin levels in 41 pregnant women in Ubud, Gianyar regency, Bali. Visfatin levels were determined by examining EDTA blood samples using the Elisa method and reading them at 450 nm on a microplate reader. The average value of visfatin levels was 6.49 ng/ml, according to the results. The average visfatin level based on hypertension blood pressure respondents was 3.74 ng/ml. This result was lower than that of normal blood pressure respondents.  This study's decrease in visfatin levels may have a smaller effect on physiological insulin resistance. The average visfatin level in second trimester respondents was 7.9 ng/ml higher than 5.7 ng/ml in third trimester respondents. The increase in visfatin levels suggests that visfatin may play a role in the pathogenesis of preeclampsia. Visfatin has the potential to be used as a biomarker to detect preeclampsia earlier, allowing pregnant women with preeclampsia to receive appropriate treatment.

Abstract

SARS-CoV-2 is the causative agent of the disease known as COVID-19. COVID-19 is spreading very fast around the world. One of the immune responses that play a role in against SARS-CoV-2 infection is the production of antibodies, which is 3 weeks after infection. Where within 3 weeks after infection, antibodies will be produced against RBD and the S1 and S2 domains in glycoprotein S and nucleocapsid protein N. The ability of an antibody to inhibit viral infection is determined by its level or titer. This study aims to determine the description of antibody levels against SARS-CoV-2 after vaccination. This type of research is descriptive research. Measurement of antibody levels for SRBD SARS-CoV-2 was carried out using the CLIA method using the MAGLUMI tool. Of the 30 respondents, 23 people had received the third vaccine. The results of this study showed that the average level of SRBD antibodies against SARS-CoV-2 in respondents with 2 doses of vaccine (1.063,786 BAU/mL) was higher than in respondents with 3 doses of vaccine (535.651 BAU/mL). Vaccine intervals of more than 6 months (908.338 BAU/mL) have higher antibody levels than respondents with vaccine intervals of 1-6 months (228.006 BAU/mL). The conclusion of this study is the highest antibody titers are produced >6 months after vaccination, antibody titers are still detectable after 12 months of vaccination, and for further research, it can be measured antibody levels against SARS-CoV-2 from people who have got vaccination for a duration of 2 years or more.

Abstract

Cemcem (Spondias pinnata (L.f) Kurz) is a native Indonesian plant that has long been used as a traditional medicine. The ethanol extract of S. pinnata contains alkaloids, carbohydrates, flavonoids, triterpenoids, steroids, tannins, resins, and saponins. It has proven antibacterial activity against gingivitis and periodontitis bacteria, such as Streptococcus mutans. This study aims to develop a toothpaste formula with the active ingredient of S. pinnata leaf extract with various concentrations of 5, 15, and 30%; evaluate its properties; and measure its antibacterial effectiveness against Streptococcus mutans. The research method used a quasi-experimental method with a post-test-only control group design. The S. pinnata leaves were extracted with 96% ethanol solvent using the maceration technique. The properties of the toothpaste were evaluated based on homogeneity, spreadability, foam height, viscosity, and pH. The liking test was conducted to assess color, taste, scent, and texture based on the perception of 30 panelists. Antibacterial activity was tested using the well diffusion method. The test results confirm that all preparation formulas are homogeneous, the foam height is in the range of 4.33 - 21.33 cm, the spreadability is in the range of 3.44 - 4.53 cm2, the viscosity of toothpaste is in the range of 20767-67600 cps, and the pH value is in the range of 4.82 - 7.59. The results showed that toothpaste containing 5% of S. pinnata leaf extract met the physical and pH requirements and was most favored by panelists. This formula also showed antibacterial activity against S. mutans with an average inhibition zone diameter of 7.72 mm ± 0.439. Based on the study's results, it can be concluded that toothpaste containing 5% S. pinnata leaf extract effectively inhibits the growth of S. mutans bacteria by 54.33% compared to the positive control. Further research is suggested to optimize the concentration of S. pinnata leaf extract to maximize its antibacterial activity, without affecting its physical quality and organoleptic acceptance.
