Abstract

Paederus dermatitis is an acute skin irritation caused by skin contact with toxins in the rove beetle (Paederus sp). Pederin poison was previously thought derived from the rove beetle's body. Based on other studies, poison pederin obtain from the symbiotic rove beetles and Pseudomonas aeruginosa which is able to secrete the matrix metalloprotease (MMP). Doxycycline is an antibiotic act as an inhibitor of MMP. This study aimed to verify the effect of Doxycycline gel to the number of macrophages in healing process Paederus dermatitis due to poison of rove beetle (Paederus sp.). The study design was a quasy experimental design using 40 mice (Mus musculus), were divided into 4 groups. Normal control group, negative control group, P1 group was given doxycycline 1%, and P2 group was given doxycycline 5%. Experimental animals were terminated on day 1, 3, 5, 7, and 15. The result of One Way Annova test on the number of macrophages on day 1, 3 and 5 showed a significance of 0.00 (p<0.05). Abstract

Cardiovascular disease is a major health problem in developed and developing countries and still be the first rank causing death in the world, including Indonesia. Most of cardiovascular disease caused by atherosclerosis. Macrophage apoptosis can reduce the size of atherogenic lesions and the progression of atherosclerotic plaque formation. Proapopstosis effects on macrophages owned by flavonoids. This study aims to know the potential of flavonoids found in Citrus grandis extract which works by suppressing the phosphorylation of AKT Ser473 so that the formation of foam cells (foam call) can be minimized. The research sample is 38 male Wistar rats weighing 90-150 grams. Samples were divided into 5 groups. All groups except normal control group were induced atherosclerosis by utilizing the shear stress mechanism at the branch of the abdominal aorta. Shear stress created by injecting adrenaline i.v. followed by hyperlipidemic diet (egg yolk) for 3 weeks. Then, during the next two weeks, the positive control group treated with simvastatin, while the treatment group were treated with extracts of C. grandis (ECG) known to contains flavonoids. Macroscopic showed that ECG could reduce atherosclerotic lesions. of citrus grandis. The sample data captured in the form of mouse blood and abdominal aortic tissues. Based on the results of the TLC screening, Ethanolic Extrxct Meanwhile, the group given simvastatin as a positive control also showed fewer atherosclerotic lesions. ECG proven to lower cholesterol and triglyceride levels of rats that had been induced atherosclerosis. Award ECG doses of 500 and 1000 mg/kg showed a significant decrease in cholesterol (p <0.01) compared to the negative control group. ECG is proven to reduce atherosclerotic lesions, so that ECG can be used as an adjuvant for simvastatin to achieve maximal therapeutic effect against atherosclerosis.

Abstract

Physalia utriculus is one of the invertebrate marine biota that is often found in Indonesia. Some symptoms of venoming due to jellyfish stings cause pain, itching, and hemolysis. In Indonesia, 13 cases of jellyfish stings were reported in 2005-2009 with three people dying in Java, Bali, and Bangka. Cocoa beans (Theobroma cacao L.) contain fat, carbohydrates, proteins, and polyphenol compounds that are useful as antioxidants. Polyphenols in the form of epicathechins, catechins, and procyanidins serve to provide protection to hemolysis. The purpose of this study was to determine the potential of ethanol extract of cacao (Theobroma cacao L.) in inhibiting the damage of erythrocyte induced by Physalia utriculus in vitro. This study used 28 samples of erythrocytes divided into seven groups, namely the normal control group, negative controls, and treatment with cocoa ethanol extract 0.2%, 0.1%, 0.04%, and 0.02%. Each subsequent group induced venom Physalia utriculus. The results showed that the average speed of erythrocyte lysis in the treatment group by giving cocoa ethanol extract 0.2%, 0.1%, 0.04%, and 0.02% respectively (seconds ± standard deviation) was 858,25 ± 94,44; 1.000,5 ± 159,93; 678,5 ± 19,71; and 1.006 ± 159,50. The mean speed of erythrocyte lysis in the negative control group was 1,025 ± 164.63 and the positive control group with the administration of N-Acetylcystein can last up to one hour after administration of venoms. Test for normality and homogeneity shows that data is normally distributed and homogeneous. One Way Annova analysis shows the significance value of p <0.05, then a post hoc analysis test was performed with the Bonferoni method to find out the differences in significance in each group. In this study it can be concluded that the administration of cocoa ethanol extract has no potential to inhibit erythrocyte damage that has been venomed by Physalia utriculus in vitro.
