Abstract

Dendrobium orchid is one of the ornamental plants with high economic value. Public interest in orchids, especially Dendrobium species is quite high and dominates the markets which make the seedling request also getting higher. The critical stage in determining the success of orchid seedling growth is also determined by acclimatization techniques. Differences in environmental conditions at the acclimatization stage can cause transpiration and interfere with nutrient absorption of plantlets. Chitosan as a natural organic material is a simple derivative of chitin which can function as a biofertilizer and bioimmuner. The purpose of this study was to determine the response of orchid seedling growth to the application of chitosan for the success of the acclimatization stage. The research method used a completely randomized design (CRD) with one factor, concentration of chitosan. The acclimatization stage using chitosan application with concentrations of 0 ppm, 2 ppm, 2.5 ppm, 3 ppm, 3.5 ppm, 4 ppm, 4.5 ppm and 5 ppm. The data obtained were then analyzed for variance with the F test at the 5% level. The treatment with significant effect was further tested with Duncan Multiple Range Test (DMRT) at 95% level. The results showed that the application of chitosan with a concentration of 3 ppm had a significant effect in increasing the parameters of plant growth percentage, plant height, number of leaves, leaf width, leaf length, root length, number of roots, and fresh weight of Dendrobium sonia orchid plants at the acclimatization stage.

Abstract

The aim of this study is to identify the arrowroot morphology characters and its distribution pattern in Jember District, East Java Province. Plant inventory was carried out in 6 sub-districts, i.e. Arjasa, Sumbersari, Silo, Tanggul, Semboro, and Wuluhan from July to October 2021 using survey method. The sampling technique was done by accidental sampling. The results of dendogram analysis based on morphological characters showed that Sumbersari, Wuluhan, and Arjasa were sub-district areas with a lot of arrowroots, if compared to other 3 sub-districts. Three main groups were formed based on the similarity of arrowroot morphological characters, accessions from geographically adjacent sub-district or village had greater morphological similarities. Leaf length has a close relationship with leaf width, leaf sheath length, and leaf petiole diameter. The distance between stem segments has a correlation with stem height. The longest fibrous roots and stem diameter contributed less to arrowroot diversity. The number of internodes positively correlated with tuber circumference, tuber length, and tuber weight. Arrowroot plant accessions grew in groups according to certain morphological characteristics, which was also showed a close relationship between these accessions. Abstract

The porang plant (Amorphophallus muelleri B.) is a tuber plant. Porang plant propagation still uses conventional methods by using seeds or frogs that experience dormancy and take a long time to make seeds. The tissue culture technology approach through organogenesis is a solution for the supply of large-scale porang seed material. The research aims to determine the effect of TDZ (Thidiazuron) on the formation of organogenesis. This study used porang leaf explants grown on MS media. The research design used a completely randomized design with one factor, namely TDZ hormone at concentrations of 0 mg/L, 1 mg/L, 2 mg/L, 3 mg/L, and 4 mg/L. The results showed that leaf explants on the addition of TDZ hormone could form organogenesis indirectly, but gave a response to form shoots multiplication. TDZ concentration of 2 mg/L gave the highest proportion value in the parameter of callus formation power of 95%, regeneration power of 88%, and  number of shoots of 10.2 shoots.
