Abstract

Kualitas warna ikan klown hasil budidaya lebih rendah dibandingkan tangkapan alam, hal ini dipengaruhi oleh berbagai faktor, namun belum ada data ilmiah sebagai dasar untuk melakukan perbaikan. Tujuan penelitian adalah menganalisis dan mengevaluasi kualitas warna ikan klown (Amphiprion percula) hasil tangkapan alam dibandingkan dengan hasil budidaya. Kualitas warna diukur pada dua zona: zona-I kulit berwarna oranye antara insang dengan band warna putih pada tengah badan dan zona-II adalah bagian kulit warna oranye antara band putih tengah badan dengan band warna putih pada pangkal ekor, dengan parameter nilai L* (lightness), a* (redness), b* (yellowness), C (chroma), H (Hue). Analisis total karotenoid (TC) dilakukan pada kulit kedua zona, sirip pektoral, sirip dorsal, sirip kaudal, dan serum darah. Analisis kromatografi lapis tipis (KLT) dilakukan pada kulit dan sirip untuk mengonfirmasi jenis karotenoid dalam kulit dan sirip. Data kualitas warna dianalisis menggunakan t-test, hubungan kualitas warna dengan TC dianalisis dengan regresi sederhana, dan analisis deskriptif untuk hasil KLT. Hasil penelitian menunjukkan bahwa kualitas warna ikan klown tangkapan alam lebih baik dibanding budidaya, didukung oleh tingginya total karotenoid pada kulit zona-I, kulit zona-II, sirip pektoral, sirip dorsal, sirip kaudal, dan serum darah berturut-turut 51,64; 51,24; 136,40; 124,37; 194,18 mg/kg; dan 2,2 mg/mL; pada ikan hasil budidaya berurut-turut 2,5; 3,5; 8,45; 10,01; 23,43 mg/kg; dan 0,8 mg/mL. Hasil KLT menunjukkan bahwa jenis karotenoid pada kulit dan sirip ikan klown adalah astaxanthin, serta satu jenis karotenoid diduga zeaxanthin. Berdasarkan hasil penelitian, maka perlu ditambahkan pigmen karotenoid, dan prekursor pigmen lainnya melalui pakan untuk ikan klown budidaya.The color quality of cultured clownfish is not as good as the wild one. However, it’s influenced by several factors. However, but there is not enough scientific data to be used as the basis for improvement. The purpose of this study was to analyze and evaluate the color quality difference between cultured and wild clown fish Amphiprion percula. Color qualities were measured in two zones: an orange-colored at zone-I was measured between gills and white band at the center of the body and an orange-colored at zone-II was measured between the center of white band and white band near the caudal peduncle. Parameters analyzed were L* (lightness), a* (redness), b* (yellowness), C (Chroma), H (Hue). Total carotenoid (TC) was analyzed on both zones-I and II, pectoral-fins, dorsal-fin, caudal-fin, and blood serum. Thin layer chromatography (TLC) was used to analyze the type of carotenoids in the skin and fin tissues. Color quality data was analyzed by T-test. Simple linier regression and descriptive analyses were used to analyzed the other parameters. The results showed that the color quality of wild clown fish was better than that of the cultured clown fish, indicated by high TC content in skin of zone-I, skin of zone-II, pectoral-fin, dorsal-fin, caudal-fin, and blood serum (51.64, 51.24, 136.40, 124.37, 194.18 mg/kg, and 2.2 mg/mL, respectively); and in cultured fish 2.5, 3.5, 8.45, 10.01, 23.43 mg/kg, and 0.8 mg/mL, respectively. TLC test results showed that carotenoid type in skin and fin of clownfish were astaxanthin, and one type of carotenoid was suspected as zeaxanthin. Based on the results of the study, it is necessary to add carotenoid pigments, especially astaxanthin or other precursor the feed to improve the color quality of cultured clown fish.

Abstract

This experiment was conducted to determine the effect of dietary single cell protein as substitution for fish meal in fish diets. Five isonitrogenous (38% crude protein) and isocaloric (3,040 kcal digestible energy/kg diet) practical diets, namely diets A, B, C, D and E with different levels of single cell protein were fed to juvenile common carp (Cyprinus carpio). Twenty juveniles/aquarium with an initial weight of 1.20±0.11 g per individual were used in this experiment. Fish were cultivated in aquarium of 60 l capacity under standardized conditions at 26±10 C. The aquaria assigned to 5 treatments were arranged in triplicates. Diet A contained low dosage of single cell protein (0 g /100 g diet), while diets B (9.8 g /100 g diet), C (19.5 g /100 g diet), D (29.3 g /100 g diet), and diet E (39.5 g /100 g diet ), combined respectively with 39.5 g fish meal/100 g diet (diet A), 29.3 g fish meal/100 g (diet B), 19.5 g fish meal/100 g diet (diet C), 9.8 g fish meal/100 g diet (diet D), and 0 g fish meal/100 g diet (diet E). Fish were fed ad satiation for 45 days using these diets. The dietary with different level of single cell protein affected growth rate and feed efficiency. Fish fed on diet 39.5 g fish meal/100 g (diet A) produced the highest growth rate (1.15±0.267%) and feed efficiency (61.68±9.89%). Growth rate and feed efficiency decreased synchronously with increases in single cell protein in the fish diets. Lysine and tryptophan were higher in single cell protein than in fish meal; whereas arginine, histidine, isoleucine, leucine, methionine, phenylalainine, threonine and valine were lower in single cell protein. On the other hand, fish fed on experimental diets did not show any significance differences in the survival rate.

Abstract

This experiment was conducted to determine the dietary n-3 fatty acid requirement for reproduction of broodstock zebrafish, Brachydanio rerio. Three isonitrogenous (39% crude protein) and isocaloric (3,260 kcal digestible energy/kg diet) practical diets, namely diets A, B, and C, with different levels of fatty acids were fed to zebrafish broodstock. The broodstock were cultivated in aquaria. Diet A contained low dosage of n-3 fatty acids (0.4%) and 2% n-6 fatty acids, while diets B and C contained 2% n-6 fatty acids, combined respectively with 1% and 1.5% n-3 fatty acids. Fish were fed ad satiation for 60 days using these diets. During feeding periode, gonade maturation stages were examined. The n-3 and n-6 fatty acids affected fecundity, fertilization rate, and hatching rate. On the other hand, fish fed on diets A, B, or C did not show any significance differences in the gonade somatic index and total Survival Rate (SR) of larvae produced. Fish fed on diet B produced the highest fecundity (616 eggs/g of fish), fertilization rate (94.6%), and hatching rate (93.8%). The total lipid content of eggs were significant, ranging from B (29.7%), C (23.7%), and A (16.1%). At a dosage of 2% n-6 fatty acids, zebrafish require 1% of dietary n-3 fatty acids in the diet for reproduction. Excess dosage of n-3 fatty acid in the diet adversely affected fecundity, fertilization rate, and hatching rate.

Abstract

A triplicate experiment was conducted to evaluate cassava (Manihot esculenta) meal as alternative carbohydrate source for wheat flour in the diet of common carp (Cyprinus carpio L.). The diet contained 0%, 25%, 50%, 75%, and 100% of wheat flour were replaced by cassava meal in five treatment groups. Abstract

The synthetic steroid 17α-Methyltestosteron (MT) is commonly used as a feed additive to produce male population of Nile tilapia (Oreochromis niloticus). The use of synthetic testosterone hormone is not recommended in Indonesia. This study was conducted to evaluate the effect of natural testosterone hormone in bull testes meal (BTM) on the masculinization of Nile tilapia using validated aceto carmine squash method of gonads of the fish. Experimental design was utilized two factors experiments in completely randomized design. Fry kept in 40-L glass aquaria at a density of 40 fry/aquarium. Fry (7 dph) received the BTM  for 7 days (T1), 14 days (T2) and 21 days (T3) and doses 0% (D1), 3% (D2), 6% (D3), and 9% (D4).  When treatment was these results, indicated that significant (P≥0.05) masculinization occurred only in the group treated of BTM and no treated of BTM. In the group treated of BTM, doses and duration treatment is not significant. The percentage of male fish 83.3% (9%-7d, 9%-21d, 6%-21d: doses and duration, respectively), higher than all group. Survival rate of fry (95-99.5%) is not affected by treatment BTM (no significant P≥0.05). Fish growth was significantly affected by treatment BTM compare with no treated of BTM. The highest growth performance of fry were obtained with the 9% BTM.Key words: masculinization, nile tilapia, bull testes meal ABSTRAKSteroid sintetik 17α-Methyltestosteron (MT) umumnya digunakan sebagai aditif pakan untuk menghasilkan populasi ikan nila jantan (Oreochromis niloticus). Penggunaan hormon testosteron sintetis tidak dianjurkan di Indonesia. Penelitian ini dilakukan untuk mengevaluasi pengaruh hormon testosteron alami dalam makanan testis banteng (BTM) pada maskulinisasi ikan nila menggunakan metode divalidasi aceto carmine squash, dari gonad ikan. Desain eksperimental dimanfaatkan dua eksperimen faktor dalam desain benar-benar acak. Fry disimpan dalam 40-L akuarium kaca pada kepadatan 40 fry/akuarium. Fry (7 DPH) menerima BTM selama 7 hari (T1), 14 hari (T2) dan 21 hari (T3) dan dosis 0% (D1), 3% (D2), 6% (D3), dan 9% (D4). Ketika pengobatan hasil ini, menunjukkan signifikan (P ≥ 0,05) hanya terjadi maskulinisasi pada kelompok perlakuan dari BTM dan tidak diperlakukan BTM. Pada kelompok diobati BTM, dosis dan durasi pengobatan tidak signifikan. Persentase ikan jantan 83,3% (9%-7d, 9%-21d, 6%-21d: dosis dan durasi, masing-masing), lebih tinggi dari kelompok semua. Tingkat kelangsungan hidup benih (95-99,5%) tidak dipengaruhi oleh pengobatan BTM (tidak ada P yang signifikan ≥ 0,05). Pertumbuhan ikan secara signifikan dipengaruhi oleh BTM pengobatan dibandingkan dengan tidak diobati BTM. Abstract

This study was conducted to analyze the production of freshwater Spirulina sp. cultured with photoperiod manipulation treatment. In this study, photoperiod manipulation treatment performed on cultured spirulina using fiber tanks (100 L). Spirulina was grown with different photoperiod (bright/T and dark/G) that are six hours per day (6T-18G), 12 hours per day (12T-12G), 18 hours per day (18T-6G), and 24 hours per day (24L-0G). The parameters were observed include dry biomass, population density (N), specific growth rate (SGR), doubling time (G), proximate analysis, and water quality. The results of this study showed that the optimum population density was achieved on day-3 days of cultured, and manipulation photoperiod showed no significant effect to the dry biomass harvest and population density, but significantly affect the specific growth rate and doubling time. Treatment of lighting 12, 18 and 24 hours per day to produce the maximum specific growth rate (0,345 to 0,366 per day) and a maximum doubling time (1,89 to 2,01 days) were not significantly different, whereas the old treatment six hours per day lighting showed the lowest maximum growth rate (0,323 per day) and highest doubling time (2,15 days). At treatment of lighting 12 hours per day, relatively higher protein content (39,73%) than others. In conclusion, the lighting 12 hours per day resulted in optimum production efficiency than other treatments.Keywords: Spirulina sp., photoperiod, density, biomass, growth, nutrition value. ABSTRAKPenelitian ini dilakukan untuk menganalisis produksi spirulina Spirulina sp. air tawar yang dikultur dengan manipulasi fotoperiode.  Dalam penelitian ini, spirulina dikultur dalam wadah fiber 100 L dengan perlakuan fotoperiode (terang/T dan gelap/G) berbeda, yaitu enam jam per hari (6T-18G), 12 jam per hari (12T-12G), 18 jam per hari (18T-6G), dan 24 jam per hari (24T-0G). Parameter yang diamati meliputi biomassa kering, kepadatan populasi (N), laju pertumbuhan spesifik (SGR), waktu penggandaan (G), dan analisis proksimat sprirulina, serta kualitas air. Hasil penelitian menunjukkan bahwa kepadatan populasi optimum dicapai pada hari ke-3 umur kultur dan manipulasi fotoperiode tidak berpengaruh nyata terhadap hasil biomassa kering dan kepadatan populasi, namun secara nyata mempengaruhi laju pertumbuhan dan waktu penggandaan. Perlakuan pencahayaan 12, 18 dan 24 jam per hari menghasilkan laju pertumbuhan spesifik maksimum (0,345 sampai dengan 0,366 per hari) dan waktu penggandaan maksimum (1,89 sampai dengan 2,01 hari) yang tidak berbeda nyata, sedangkan perlakuan pencahayaan enam jam per hari menunjukkan laju pertumbuhan maksimum terendah (0.323 per hari) dan waktu penggandaan tertinggi (2,15 hari). Pada perlakuan pencahayaan 12 jam per hari, kandungan protein relatif lebih tinggi (39,73%) dari yang lain. Secara umum dapat disimpulkan bahwa  pencahayaan 12 jam per hari menghasilkan efisiensi produksi yang lebih baik daripada perlakuan lainnya.Key word: Spirulina sp., fotoperiode, kepadatan, biomassa, pertumbuhan, kandungan nutrisi.

Abstract

Spirulina is blue green algae that have high nutrient content like protein and vitamin so itcan beused as source ofsingle cell protein (SCP). This research was conductedto know minimum doses of Spirulina in different protein feed on growth performance of tilapia. Experimental diets devided by two different protein level (25% of protein and 28% of protein) and three levels supplementation of Spirulina 0% (25A), protein level 25% supplementedSpirulina 3% (25B), protein level 25% supplementedSpirulina 6% (25C), protein level 28% supplementedSpirulina 0% (28A), protein level 28% suplemented Spirulina 3% (28B), and protein level 28% supplementedSpirulina 6% (28C). Testeds feedwere tilapia with average initial weight 17,24±0,29 g and density five fish/aquarium. Tilapia rearedin aquarium sized 50×40×35 cm3 filledwith 30 cm waterand provided altrecirculation system. Fish were cultured for 40 days and fed at satiation with three times feeding frequency. All data analyzed statistically by one-way ANOVA.Result demonstrated that various dosesof Spirulina in different protein level of feed influencedspecific growth rate, feeding efficiency, protein deposition, and fat deposition. Abstract

This study was aimed to determine the immersion dose of recombinant giant grouper growth hormone (rElGH) to increase growth of eel juvenile (glass eel). After shock salinity treatment (NaCl 3% for 2 min), glass eel were immersed in water containing 0.9% NaCl, 0.01% bovine serum albumin, and different of dose of rElGH (0, 0.12, 1.2, 12 and 120 mg/L). Glass eel were fed on blood worm for the first month, and commercial diet for the second month of rearing. Fish rearing was performed in 60 L glass aquarium at density of 150 fish per aquarium. The results showed that higher in growth body weight was obtained in immersion dose of 12 mg/L, by increment of about 37.4% higher compared to that of control. The immersion dose of 12 mg/L also increased specific growth rate by 29.3% higher (p<0.05) compared to control. Furthermore, at that immersion treatment, feed conversion ratio of blood worm and commercial diet decreased by 33.7% and 25.6% compared to control, respectively. Protein (7.15±0.08%) and lipid (9.95±0.10%) retentions in 12 mg/L rElGH-treated fish were higher (p<0.05) than those of control (protein 6.17±0.07%; lipid 5.73±0.06%). Lower in protein content (12.73%), while higher content in lipid (8.35%) and crude carbohydrate (3.22%) were found in 12 mg/L rElGH-treated fish compared to those of control (13.24%, 5.90%, and 1.76%, respectively). Thus, in general, rElGH immersion dose of 12 mg/L could generate high performances of eel juvenile, and application of rElGH can be useful to increase aquaculture eel production.Keywords: immersion dose, growth hormone, growth, glass eel

Abstract

This study was conducted to determine the effects of inorganic selenium (sodium selenite) and organic selenium (selenomethionine) supplementation on growth and viability of juvenile humpback grouper (Cromileptes altivelis). The experiment was designed as a completely randomized design with eight treatments and three replications. The treatment being tested was source and dosages of selenium ie., inorganic selenium (sodium selenite) supplementation with dosages of 0.5, 1, 2, and 4 mg Se/kg diet and organic selenium (selenomethionine) with dosages of 1, 2, and 4 mg Se/kg diet. Another treatment was unsupplemented selenium. Juveniles humpback grouper at an initial average length of 6,39±0,41 cm and body weight of 4,49±0,65 g were reared in 90×40×35 cm3 aquaria and fed artificial diet (pellet) two times daily (08.00 and 16.00) at satiation. Fishes were reared for 40 days at a stocking density of 12 fish per aquarium on sea water with salinity of 30–31 ppt and temperature of 28–29 °C. The results of this study showed that the source of selenium supplementation (inorganic or organic) affected growth performance, glutathione peroxidase (GPx) enzyme activity, and blood profiles of the experimental fish. Generally, it was found that selenomethionine supplementation resulted in better fish performance than sodium selenite. In sodium selenite supplementation, survival declined with the increased dosages of Se in the diet, and the supplementation at dosage of 0,5 mg Se/kg diet showed a toxic effects. Histopatological test showed that there were damage in livers, kidneys, and intestines of fish supplemented with sodium selenite from 0.5 to 4 mg Se/kg diet. On the contrary, supplementation of selenomethionine up to 4 mg Se/kg did not show any signs of toxicity and the survival was on 86.11 to 97.22%. Feed efficiency, protein retention, lipid retention, and Se retention indicated that a dosage of 4 mg Se/kg selenomethionine supplementation was the best dosage.Keywords: selenium, viability, growth, Cromileptes altivelis, humpback grouper

Abstract

ABSTRACTThis experiment was conducted to evaluate the use of by product of corn ethanol industry, DDGS (distiller dried grain with soluble) and hominy as feed raw material on digestibility and the growth performance of humpback grouper Cromileptes altivelis juvenile. Three experimental diets containing isoprotein (48%) but different in DDGS contain were used. Diet A containing 0% of DDGS and hominy while diet B and C containing 10.13% (6.13% of DDGS and 4% of hominy) and 19.05% (15.05% of DDGS and 4% of hominy). Reduce of fish meal level in the diet was happen as a consequence of balancing amino acid. For digestibility experiment and Cr2O3 at level of 0.05% was used as a tracer. The feces were daily collected 30‒60 minutes after feeding for 14 days. Dry matter and protein were used as digestibility parameters. Completely randomized design with three treatments and two replicates was used in this experiment. Humpback grouper with an average weight of 11‒14 g were reared at floating net cages (FNG) of 3×2×1.5 m3. The stocking density for each cage was 167 fish/FNG and reared for six months. For digestibility measurement 30 fish were held in cylindrical fiber tank filled with 800 L of sea water. Fish were fed three times daily at satiation level. For maintaining net in a clean condition net was changed in every ten days. Feed consumption, digestibility of protein and total, growth performance indicator i,e specific growth rate, feed efficiency protein and lipid retention were used as evaluating parameters. The results showed that the fish fed with diet containing DDGS (6.13%) dan hominy (4%), at the level of 10.13% (diet B), shows similar growth performance indicators with the fish fed diet A (0% of DDGS and hominy) and the growth performance indicator was higher when being compared to those fish fed diet C (containing 19.05% of DDGS (15.05%) and hominy (4%)) (p<0.05). More over the inclusion of DDGS and hominy at the level of 10,13% give no negatif impact to feed palatability (shown by total feed consumption value) and digestibility of protein. The increasing level of DDGS and hominy more than 10% reduced palatability and protein digestibility. Based on this research, DDGS and hominy can be used up to 10% in the feed formulation of humpback grouperKeywords: digestibility, growth performance, DDGS, hominy, humpback grouper, Cromileptes altivelisABSTRAKPenelitian ini dilakukan untuk mengevaluasi pemanfaatan limbah jagung dari industri etanol dalam bentuk distillate dried grain with soluble (DDGS) dan homini sebagai bahan baku pakan terhadap kinerja pertumbuhan ikan kerapu bebek Cromiliptes altivelis. Ikan yang digunakan dalam penelitian ini adalah juvenil ikan kerapu bebek berukuran 11‒14 g. Ikan dipelihara dalam karamba jaring apung yang berukuran 3×1,5×2 m3 dengan kepadatan 167 ekor/jaring. Ikan diberi pakan sampai kenyang sebanyak dua kali sehari. Untuk menjaga agar jaring tetap bersih maka dilakukan pergantian jaring setiap satu sampai dua minggu sekali. Ikan dipelihara selama lima bulan dan setiap bulan dilakukan sampling. Tiga macam pakan digunakan dalam penelitian ini dengan kadar protein 47% dan rasio energi protein 10 kkal/g protein. Pakan A adalah pakan yang tidak mengandung DDGS dan homini, pakan B mengandung DDGS 6,13% dan 4% (total 10,13%), dan pakan C mengandung DDGS 15,05% dan Homini 4% (total 19,5%). Untuk mengevaluasi ketercernaan nutrien maka kromium oksida (Cr2O3) digunakan sebagai penelusur. Penelitian ketercernaan dilakukan di laboratorium dengan menggunakan fiber dengan volume 800 L. Rancangan acak lengkap dengan tiga perlakuan dan dua ulangan digunakan dalam penelitian ini. Hasil penelitian menunjukkan bahwa nilai ketercernaan nutrien tidak berbeda sampai total kadar DDGS dan homini 10,13% (pakan B) dan peningkatan DDGS serta homini sampai 19,05% (pakan C) menurunkan nilai ketercernaan. Ikan yang diberi pakan dengan DDGS sebesar 10,13% (pakan B) memiliki nilai efisiensi pakan, sintasan, laju pertumbuhan spesifik, konsumsi pakan dan retensi protein yang sama dengan pakan tanpa DDGS (pakan A), akan tetapi nilai-nilai tersebut lebih tinggi jika dibandingkan dengan ikan yang diberi pakan B (p<0,05). Retensi lemak ikan yang diberi pakan B memiliki nilai yang lebih tinggi jika dibandingkan dengan ikan yang diberi pakan A dan C. Dari penelitian ini dapat disimpulkan bahwa DDGS dan homini dapat digunakan sebagai bahan baku pakan ikan kerapu bebek sampai level 10,13% (DDGS 6,13% dan homini 4%).Kata kunci: pertumbuhan, ketercernaan, DDGS, homini, ikan kerapu bebek, Cromileptes altivelis
