Abstract

This study to do examine the effectiveness of using heparin and hypotaurine to improve the in vitro fertilization rate. In first stage, oocytes were matured in the maturation medium supplemented with 0.3 mg/mL L-carnitine to improve nuclear maturation. In second stage, matured oocytes were fertilized with semen incubated in fertilization medium supplemented with 0.1 IU/mL heparin and 10 M hypotaurine. The final concentration of sperm used was 5x106 spermatozoa/mL, and its interaction with oocytes was carried out for 12 hours in 5% CO2 incubator at 38.5 C. The results showed that percentage of oocytes that reached metaphase II was significantly higher (P0.05) in media supplemented with 0.3 mg/mL L-carnitine compared to control (88.24.9% vs 79.85.1%). Furthermore, fertilization rate was characterized by formation of 2 PN was significantly higher (P0.05) in media supplemented with heparin and hypotaurine than control (82.62.0% vs 73.32.4%). In conclusion, L-carnitine supplementation at dose of 0.3 mg/mL on maturation media was effective in increasing percentage of matured oocytes. The combination of 0.1 IU/mL heparin and 10 M hypotaurine in fertilization medium was also successful in elevating the percentage of fertilized oocytes.

Abstract

This study aimed to determine the biopotency of pregnant mare serum gonadotropin (PMSG) in stimulating the formation of dominant follicles and the biopotency of human chorionic gonadotropin (hCG) in triggering an increase in the ovulation rate of the natural-estrus donor group and the prostaglandin F2 (PGF2) induced group using female Aceh cattle (n= 20) which were divided into two treatment groups: the group with natural estrus (A) and the group with PGF2 -induced estrus (B). Each donor group was superovulated using the injection of PMSGhCG (A1, B1) and only PMSG (A2, B2). The observations on total follicles and CL were carried out at D10, D14/D+0 and D21/D+7 using ultrasonography (USG). The results showed that PMSG gave a better superovulatory response to the development of dominant follicles (P0.01) at A1, A2, B1, and B2. The administration of hCG hormone 2000 IU i.m. was not able to increase the rate of ovulation. Total follicles after the addition of hCG were also not significantly different between A1:A2 and B1:B2. Statistically, the number of CL in all donor groups in D21 was not significantly different (P0.05). It concluded, superovulation using PMSG gave the same results to the donor cows with natural estrus and the donor cows with PGF2-induced estrus. The hCG hormone with a dose of 2000 IU injected i.m. was also not able to increase the ovulation rate of Aceh cattle superovulated with PMSG 2500 IU i.m. ____________________________________________________

Abstract

The objective of this study was to examine the effects of repeated freeze-thaw cycles and short-term storage of fecal extracts at ambient temperature on the stability of fecal glucocorticoid (fGCM) and estrogen metabolite (fEM) levels from crested macaques.In total 100 aliquots of fecal extracts from fecal samples collected from female crested macaques (Macaca nigra) living at the Tangkoko-Batuangus Nature Reserve, North Sulawesi were used. We performed two different experiments: (1) An experiment to investigate if levels of fGCM and fEM measured from fecal extracts that were exposed to two, four, six and eight repeated freeze-thaw cycles (test groups) differ to control samples (i.e. fecal extracts always stored frozen); (2) An experiment to evaluate whether storing fecal extracts at ambient temperature for two, four, six, and eight days (test groups)affects the levels of fGCM and fEM compared to the control group (i.e. fecal extracts frozen immediately).Results showed that hormone levels were significantly increased (P0.05) after four freeze-thaw cycles for fGCM and after eight freeze-thaw cycles for fEM. By contrast, there was no significant difference (P0.05) in levels of fGCM and fEM between the test groups and the control group in fecal extracts stored at ambient temperature. In conclusion, our data show that more than two and six repeated freeze-thaw cycles should be avoided when measuring fGCM and fEM in crested macaque fecal extracts, respectively. We also demonstrate that storing fecal extracts at ambient temperature is possible for at least 8 days without taking a risk of affecting the stability of fGCM and fEM levels.

Abstract

The present study examines anatomy of Gayo mare reproductive organs. This study used three sample of Gayo mare reproductive organs (n= 3) for observation of morphology and morphometric of the mare reproductive organs. The ovarium was fixed in 4% paraformaldehyde solution then followed by histological method and stained using hematoxylin and eosin (HE) and Massons trichome (MT). Three mares were observed for diameter and changes overview of uterus during estrous cycle in real time using ultrasound. The results showed that, in general, the anatomy of Gayo mares reproductive organs was similar to other mares, but smaller in morphometry. The total length of the Gayo mares reproductive tract from labia to apex cornua was 48.001.00 cm. Weight of Gayo mares left ovary was 19.077.70 g and the right was 24.430.83 g. Histologically, there was no difference between Gayo mares structure and other mares. In cortex uteri there were some follicles surrounded by capillary, various development stages of follicles, healthy follicles, atretic follicle, and corpus albican; while in medulla there were a lot of connective tissues. Ultrasound of the uterus showed the change in diameter during estrous cycle with the largest diameter of corpus uteri was 4.430.10 cm in horses with estrous cycle of 21 days and 6.300.93 cm in horses with 24 days estrous cycle. In conclusion, the morphometry of Gayo mare reproductive organs are smaller than the other horses and there are differences in diameter of the uterus during the estrous cycle due to the changes of endometrium thickness.

Abstract

This study aimed to evaluate the effect of Woton (Sterculia shillinglawii) leaves meal supplementation on the growth performance and digestibility of tilapia (Oreochromis niloticus) juveniles. The treatments consisted of Woton leaves meal at inclusion levels of 0% (A), 5% (B), 10% (C), 15% (D), and 20% (E), with each treatment replicated three times. The observed parameters included average daily growth (ADG), average body weight (ABW), growth rate (GR), specific growth rate (SGR), survival rate (SR), feed conversion ratio (FCR), and feed efficiency (FE), as well as protein digestibility and total digestibility. Analysis of variance showed no significant differences in ADG, ABW, GR, and SR among the treatments. However, Treatment B recorded a relatively higher SGR (2.73 ± 0.44% day-1) compared to the other treatments (P < 0.05). The analysis of digestibility revealed significant differences. Treatment C (15% inclusion level) resulted in the highest protein digestibility (99.39 ± 0.02%) and total digestibility (99.37 ± 0.01%), while a lower FCR (1.76 ± 0.24) and a higher FE (57.59 ± 7.85%) were found in Treatment B (P<0.05). This study highlighted that despite Woton leaves meal improving fish digestibility and feed efficiency, its supplementation in fish feed should be limited to 5-10% due to the adverse effects of its flavonoid compounds. Penelitian ini bertujuan untuk mengevaluasi pengaruh suplementasi tepung daun Woton (Sterculia shillinglawii) terhadap kinerja pertumbuhan dan kecernaan juvenil ikan nila (Oreochromis niloticus). Perlakuan terdiri atas tepung daun Woton pada tingkat inklusi 0% (A), 5% (B), 10% (C), 15% (D), dan 20% (E), dengan setiap perlakuan diulang tiga kali. Parameter yang diamati meliputi rata-rata pertumbuhan harian (RPH), rata-rata bobot tubuh (RBT), laju pertumbuhan (LP), laju pertumbuhan spesifik (LPS), tingkat kelangsungan hidup (TKH), rasio konversi pakan (RKP), dan efisiensi pakan (EP) serta kecernaan protein dan kecernaan total. Analisis varians tidak menunjukkan perbedaan yang signifikan pada RPH, RBT, LP, dan TKH di antara perlakuan. Namun, Perlakuan B menunjukkan LPS yang relatif lebih tinggi (2,73 ± 0,44% hari-1) dibanding dengan perlakuan lainnya (P<0,05). Analisis kecernaan mengungkapkan perbedaan yang signifikan. Perlakuan C (tingkat inklusi 15%) menghasilkan kecernaan protein tertinggi (99,39 ± 0,02%) dan kecernaan total tertinggi (99,37 ± 0,01%), sedangkan RKP yang lebih rendah (1,76 ± 0,24) dan EP yang lebih tinggi (57,59 ± 7,85%) ditemukan pada Perlakuan B (P<0,05). Penelitian ini menunjukkan bahwa meskipun tepung daun Woton meningkatkan kecernaan dan efisiensi pakan ikan, suplementasinya pada pakan ikan sebaiknya dibatasi hingga 5-10% karena dampak negatif senyawa flavonoid yang dikandungnya.

Oocyte competence is a primary factor for the success of embryo production in vitro. This research aims to determine the effect of supplementation glutathione in sheep oocytes’ maturation and fertilization rates in vitro. In the first research, maturation oocytes were conducted in a medium supplemented with GSH at graded doses for 24 hours. In the second research, 1.0 mM GSH (the best dose from research I) was supplemented in the maturation medium, fertilization medium, and their combination for 14 hours. The higher oocyte maturation rates were significant (P < 0.05) in the GSH supplementation of 1.0 mM (87.00%). Furthermore, supplementation of GSH on the maturation medium resulted in a higher normal fertilization rate (56.43%) than in other groups. It is concluded that GSH supplementation on the maturation medium is more effective in increasing the competence of sheep oocytes to be mature and results in improved fertilization rates.

Abstract

This study aims to find out the characteristic of gayo horse and to identify the population of gayo horse. Data collection was conducted through selection process over 100 local horse distributed in Central Aceh Sub-districts, Gayo Lues and Bener Meriah. The result was 30 gayo horses with varying age distribution. Detail observation was performed on six gayo horses of male (n=3) and female (n=3). The observation consisted of morphological observation and morphometry. Data collection of gayo horse population was based on annual report from local Animal Husbandry and Fishery Official over the last 5 years. Result showed that gayo horse has straight cranial shape similar to thoroughbred with smaller size, thick and stiff mane, and ears akin to donkey. Gayo horse body height ranges between 113-120 with body weight ranging around 215-280 kg. Gayo horse is agile despite living in mountain and able to carry heavy weight. There has been drastic decline of population in year 2010-2014. In conclusion, gayo horse is included as large pony category, and without any rescue effort, gayo horse is predicted to be extinct in 2037.

Abstract

Low-carbon aquaculture is increasingly recognized as a vital strategy to mitigate climate change, improve resource efficiency, and ensure sustainable food production. This systematic review, following the PRISMA 2020 protocol, analyzed 58 peer-reviewed studies selected from 312 published between 2020-2024, providing a transparent and replicable synthesis of the current evidence. Research is predominantly focused in Asia and Europe, regions where rapid aquaculture growth coincides with emerging climate policies. Key strategies for reducing environmental impacts include Integrated Multi-Trophic Aquaculture (IMTA), cultivation of low-trophic species such as seaweeds and filter feeders, and the restoration of coastal ecosystems, all of which enhance carbon sequestration, nutrient cycling, and ecosystem resilience. The review identifies the most commonly reported sustainability indicators as global warming potential (GWP), energy consumption, land use, nutrient loading, and eutrophication. Despite ongoing technological innovations, global aquaculture remains largely unsustainable with persistently low environmental performance. Barriers to improvement include incomplete carbon accounting, fragmented regulatory frameworks, and limited consumer awareness. Additionally, significant knowledge gaps in greenhouse gas emissions and carbon sequestration constrain robust life cycle assessments. Transitioning to low-carbon aquaculture requires not only technological innovation but also with stronger governance, ecosystem-based management, and integrated policy frameworks to enhance sustainability and climate resilience. This review underscores the urgency of coordinated action across research, industry, and policy sectors to optimize low-carbon strategies, address existing knowledge gaps, and support the development of sustainable aquaculture systems that are both environmentally responsible and economically viable.
