Abstract

BACKGROUND: Obesity is linked to chronic low-grade inflammation and increased oxidative stress that may disrupt hypothalamic metabolic homeostasis. Excess nutrient intake activates inflammatory signaling and promotes glial reactivity, contributing to neuroinflammatory remodeling. Superoxide dismutase (SOD) is a key enzymatic antioxidant marker reflecting hypothalamic defense capacity. However, whether natural antioxidants can simultaneously restore SOD activity and attenuate obesity-induced glial reactivity remains unclear. Among many natural antioxidant, Moringa oleifera is rich in bioactive compounds with antioxidant properties that may attenuate neuroinflammation. Therefore, this study was conducted to evaluated the effect of M. oleifera fruit extract (MOFE) on hypothalamic total SOD activity and histopathological features of glial reactivity in obese Wistar rats.METHODS: Male Wistar rats were divided into four groups: normal control, obese control, and two treatment groups. For obese control and treatment groups, obesity was induced for 8 weeks using a high-fat/high-sucrose diet. Fresh M. oleifera fruit pods were macerated to produce MOFE. Following the obesity-induction, the two treatment groups were treated with 500 and 1,000 mg/kgBW/day MOFE for 4 weeks. A hydroxylamine-based assay was employed to measure total SOD activity of hypothalamic tissue homogenate. Meanwhile, hematoxylin-eosin (HE) was used to stain hypothalamic sections for glial reactivity scoring.RESULTS: After the obesity induction, the obesity-induced rats showed elevated Lee Index, but after the treatment with 500 and 1,000 mg/kgBW/day MOFE, the Lee Index decreased for 26.61±3.53% and 26.32±0.93%, respectively, which is greater compared to obese control. MOFE administration was also able to improve hypothalamic SOD activity (64.75±1.29 U/mL and 65.78±0.74 U/mL). MOFE groups also exhibited predominantly milder histopathological changes and glial reactivity than obese controls.CONCLUSION: MOFE administration lowers Lee Index, improves hypothalamic SOD activity, reduces glial reactivity and improves neuroinflammation changes in obese rats, suggesting that MOFE might be potential agent for obesity-related oxidative-inflammatory brain injury. Abstract

Penyuluhan kesehatan dengan pendekatan dokter keluarga pada kasus demam berdarah dengue (DBD) adalah solusi bagi penatalaksanaan kasus kedokteran yang berfokus pada pemberian layanan kesehatan primer yang komprehensif, berkesinambungan, dan berpusat pada individu, keluarga, serta komunitas. Terdapat peningkatan kasus DBD dari tahun 2023 ke 2024 sehingga peran kedokteran keluarga penting untuk menyelesaikan masalah ini, dimulai dari screening hingga edukasi kepada keluarga untuk mengurangi presentase angka kejadian terutama di wilayah UPDT Pengasinan, Depok. Penyuluhan kesehatan diperlukan untuk mengidentifikasi daftar masalah klinis pada pasien dan keluarga serta factor-faktor yang mempengaruhi masalah demam berdarah dengue, mengubah pengetahuan dan perilaku dalam pencegahan demam berdarah dengue yang berikatan terhadap kesehatan pasien serta keluarga. Keterlibatan partisipasi keluarga dalam mengatasi masalah kesehatan pasien dengan pendekatan kedokteran keluarga sangat diperlukan. Kegiatan pengabdian kepada masyarakat ini dilakukan di UPTD Pengasinan Depok dengan pendekatan kedokteran keluarga. Kegiatan penyuluhan kesehatan pada kasus demam berdarah dengue di UPTD Pengasinan Depok dengan pendekatan dokter keluarga di dapatkan bahwa dari hasil evaluasi dan penatalaksanaan terdapat perbaikan terhadap pengetahuan dan sikap yang menjadi baik, salah satunya yaitu perilaku untuk melakukan 3M dan mencoba membuka diri untuk berkegiatan sosial di lingkungan rumah. Pengetahuan dan perilaku tersebut sangat mempengaruhi pasien dan keluarga karena merasakan kualitas hidupnya yang membaik dan nyaman saat berada di lingkungan rumah.

Abstract

Abstract. Early marriage is a social and public helath issue that affects reproductive health, education, psychological well-being, and socioeconomic status. From a public helath perspective, early marriage is associated with an increased risk of adolescent pregnancy and may contribute to various complications during pregnancy and childbirth, as well as impaired child growth. Therefore, educating adolescents about these risks is an imprtant promotive and preventive strategy. This community service activity aimed to improved adolescent’s knowledge of stunting prevention efforts before entering marriage and pregnancy. The activity was conducted in Pangkalan Jati Village, Depok, targeting adolescents involved in youth organizations and mosque-based youth groups. The activities included problem identification, a pretest, health counseling and education, the distribution booklets and leaflets, interactive discussion, and a posttest. In addition to the educational intervention, hemoglobin level screening was conducted to provide an initial assessment of the adolescent’s health status. The activity was attendedd by 20 male and female adolescents. The results demonstrated an 20% improvement in participants understanding of the risks associated with early marriage. Community-based education that actively enggages adolescents can serve as a promotive and preventive approach to strengthening adolescent health literacy and supporting sustainable stunting prevention efforts.

Abstract

Targeting the precursors of trimethylamine-N-oxide (TMAO), such as choline, betaine, and enzymes produced by the FMO3 gene, through natural compounds may reduce risk factors such as cardiovascular and metabolic diseases. Protein precursors involved in high TMAO level production include cntA, CUTC, and FMO3. Manalagi apple (Malus sylvestris) is rich in bioactive compounds, including pectin, phloretin, and chlorogenic acid, with potential health benefits, and would affect the decreasing production of TMAO by inhibiting some protein precursors. This research aims to investigate the potential of three small molecules in it as a dietary source candidate and drug candidate that could mitigate the risk of TMAO-related diseases based on their interaction and binding affinity through an in-silico study. The methods include the prediction of protein and small-molecule structures, including FTIR, molecular docking, and analysis of small-molecule bioavailability. Pectin is considered the top candidate compared to phloretin and chlorogenic acid based on its high binding affinity to interact with CUTC, cntA, and FMO3 (-10.2, -8.6, and -9.1 kcal/mol, respectively). Phloretin is a promising compound for drug development based on its binding site to interact with more residues and bonds, its bioavailability with a drug likeness score of 0.55, and high gastrointestinal absorption.

Abstract

Background: Repeated heating degrades cooking oil quality, increasing peroxide values above national safety standards (SNI 01-3741-2002). Household waste cooking oil typically reaches 20-40 meq/kg, potentially triggering systemic oxidative stress and organ toxicity.Objectives: To analyze the mechanism of waste household oil as a trigger of oxidative damage leading to DNA oxidation.Methods: This prospective, controlled, true experimental in vivo time-course study initially allocated 32 male Swiss Webster mice into four parallel cohorts (n=8 per group), with 28 completing the final analysis (n=7 per group). The mice were orally treated with used cooking oil at a dosage of 10 μl/g body weight. The animals were distributed into four experimental cohorts. Group A (control) received fresh oil (peroxide value 8-10 meq/kg) for eight weeks, while Groups B, C, and D received waste oil (peroxide value 20-40 meq/kg) for 8, 12, and 16 weeks, respectively. Following the treatment, assays evaluated serum MDA and SOD, hepatic 8-OHdG via HPLC, TNF-α and IL-6 expression via IHC, and liver histopathology (congestion, steatosis, and necrosis) via H&E staining. Data were analyzed using the Kruskal-Wallis and Wilcoxon-Mann-Whitney post-hoc tests.Results: Used cooking oil administered over a 16-week period led to an increase of harmful effects. These included oxidative stress, primarily through lipid membrane peroxidation, evidenced by significant increases in both serum MDA and activated SOD p<0.05. The oil also induced chronic inflammation, marked by elevated expression of TNF-α and IL-6 in the cytoplasm and sinusoids. Furthermore, it resulted in severe liver structure changes (including congestion, steatosis, and necrosis) and a time-dependent upward trend in 8-OHdG, reaching statistical significance at 16 weeks compared to controls (p<0.05).Conclusion: These findings suggest that prolonged consumption of household used cooking oil promotes progressive hepatic lipid peroxidation and structural damage, with oxidative DNA damage accumulating significantly after extended exposure (16 weeks).
