Abstract

Penyakit ginjal dapat disebabkan karena ada pembentukan batu yang terbentuk dari komplek kalsium sehingga dapat mengganggu fungsi ginjal. Biji buah kebiul (Caesalpinia bonduc (L.) Roxb) mengandung flavonoid yang dapat mencegah pembentukan endapanbatu di ginjal. Tujuan dari penelitian ini adalah untuk mengetahui efek ekstrak etanolbiji buah kebiul pada batu ginjal tikus Sprague Dawley (SD) yang diinduksi etilen glikol 0,75% dan amonium klorida 2%. Sebanyak 30 ekor tikus, dewasa, jantan, dibagi menjadi 6 kelompok yang mendapat perlakuan sebagai berikut: kontrol normal diberi akuades; kontrol sakit, diberi akuades; kontrol positif, diberi obat X dengan dosis 2,7 mL/kg BB, dan kelompok ekstrak diberi ekstrak biji kebiul dengan variasi dosis yaitu 250, 500, dan 1000 mg/kg BB. Induksi batu ginjal dilakukan dengan pemberian etilen glikol 0,75% dan amonium klorida 2% 2 jam setelah pemberian esktrak etanol biji buah kebiul dan obat X  selama 10 hari pada semua kelompok, kecuali kelompok normal. Efek terhadap batu ginjal ditentukan berdasarkan jumlah kalsium urin, kadar kreatinin dan ureum serum. Data jumlah kalsium yang diperoleh dianalisis secara statistik dengan uji Kruskall Wallis dilanjutkan dengan uji Mann Whitney, sedangkan data kreatinin dan ureum serum yang diperoleh dianalisis dengan one way ANOVA dilanjutkan dengan uji LSD. Signifikansi hasil dinyatakan dengan p<0,05. Jumlah kalsium urin, kadar kreatinin dan ureum serum kelompok ekstrak tidak berbeda bermakna (p>0,05) dibandingkan dengan kelompok kontrol sakit. Berdasarkan hasil penelitian dapat disimpulkan bahwa pemberian ekstrak etanol biji buah kebiul dosis 250, 500 dan 1000 mg/kg BB tidak memberikan efek pada batu ginjal tikus SD yang diinduksi etilen glikol dan amonium klorida.

Abstract

Obesity is an increase in body weight that exceeds normal and the risk of serious illness. Bambu Tali leaves (Gigantochloa apus (Schult.) Kurz) contain flavonoids, tannins, and saponins, which can inhibit the absorption of fatty acids in the digestive tract and excrete them through feces to lose weight. This study examines the tea activity of the Bambu Tali leaves in inhibiting weight gain, total cholesterol, and triglyceride levels of obese Wistar rats fed with a high-fat diet (HFD). A total of 30 Wistar rats were divided into 6 groups: normal control (standard feed), negative control (HFD), positive control (HFD + Orlistat), and 3 groups of tea doses of Bambu Tali leaves (HFD + 360, HFD + 720, HFD + 1440 mg/kg BW). HFD was given together with the Bambu Tali leaf tea for 35 days. On the 36th day, the blood sample was taken to measure cholesterol and triglyceride levels. The weighing was conducted weekly. The results showed that giving HFDincreased body weight significantly, total cholesterol, and triglyceride levels (p <0.05). Bambu Tali leaf tea at doses of 720 and 1440 mg/kg BW could significantly inhibit weight gain (p <0.05). Bambu Tali leaf tea with doses of 360, 720, and 1440 mg/kg BW can also inhibit the increase in total cholesterol levels (p<0.05). Meanwhile, the inhibition of a significant increase in triglyceride levels (p <0.05) only occurs at a dose of 1440 mg/kg BW. This study concludes that Bambu Tali leaf tea has antiobesity activity by inhibiting weight gain, total cholesterol levels, and plasma triglycerides of Wistar rats.

Abstract

Egg whites reportedly exhibit wound healing activities by forming new cell tissues and accelerating damaged cell tissue recovery. This study aimed to determine whether and how egg whites prepared in gel dosage forms healed incision wounds in mice (Mus musculus). The method began with formulating gels by mixing egg whites with Carbopol 940, methylparaben, propylparaben, propylene glycol, triethanolamine, and distilled water. Sixteen mice were divided evenly into four groups, and each was subjected to a 1 cm long midline dorsal incision. Group I was given 10% povidone-iodine ointment, group II was gel base, group III was 10% egg white gel, and group IV was 20% egg white gel. Based on the test parameters measured during macroscopic observation, namely wound length and healing time, the gel preparations were found to fulfill the physical properties requirements, and incision wounds treated with 10% and 20% egg white gels healed in 9 and 12.5 days, respectively. The statistical test revealed a significant difference in wound size on Day 10 and in healing time between wound treatments with 10% gel and base gel (p < 0.05). Therefore, it can be concluded that the 10% egg white gel is effective in healing cuts in mice as it speeds wound healing and decreases wound size.

Abstract

Diabetes is metabolic disorders caused by oxidative stress that has always been linked to male reproductive dysfunction. Spirulina platensis has been used for therapeutic supplements for diabetes and repairing reproduction organs based on its antioxidant effects. The aim of this study was to examine the effect of Spirulina platensis suspension (SP) on quality and quantity of sperm also the expression of caspase-3 on testis in male Wistar rats induced by streptozotocin (STZ). This study used 30 adult male Wistar rats that divided into 6 groups of K1 (normal), K2 (single injection of STZ 45 mg/kg BW), K3 (normal rats treated with SP 36/kg BW/day) and treatment group K4, K5 and K6 were diabetic rats given with SP 36 mg/kgBW, 72 mg/kgBW and 144 mg/kg BW. The treatment was given orally for 28 days. Blood glucose level levels were measured using the biochemical analyzer. Quality and quantity of sperm were analyzed using Neubauer slide under the microscope. Apoptosis of Leydig cells was detected by immunohistochemical staining using Caspase-3 polyclonal antibodies. The result showed that there were significant differences between groups (p < 0.05) in final blood glucose level, quality, and quantity of spermatozoa and expression of caspase-3. Our findings clearly highlight the role of Spirulina platensis as an antidiabetes and protective agent for reproducton organs of STZ-induced in diabetic male Wistar rats that improves quality and quantity of sperm cells and reproductive organs.

Abstract

Carotenoid compound, include licopene and lutein, has ability to increaseimmune system. One of organs included in immune system is spleen. The objective of thestudy is to observe the effect of licopene to the histopathologic appearance of ratspleen. Twenty female Sprague Dawley rat, 1 month old, 100 grams of weight devideinto 4 groups. The first group got licopene dosage 3 ug/kg of body weight, the secondgroup got licopene dosage 6 ug/kg of body weight, the third group got corn oil (vehiclecontrol) and the fourth group got aquadest. The treatment were given during 17 days,orally. At the day 14, the first, second and third groups were injected by aloxansubcutaneuslly. Three days after injection of aloxan, all of rats were sacrified, spleenwere taken in order to histopathological examination. Histopathologic appearances ofspleen were analized descriptivelly. The result showed that licopene dosage 6 ug/kg ofbody weigh able to increase centrum germinativum formation of spleen white pulp morethan licopene dosage 3 ug/kg of body weight. Base on the result can be concluded thatlicopene administration can increase centrum germinativum formation of spleen whitepulp.

Abstract

Ganyong rhizome (Canna edulis Ker.) empirically use for treating gastritis (peptic ulcers). Ganyong rhizome contains carbohydrates and flavonoid that can reduce inflammation in the gastric tissues and againsts free radicals that are contributing to the pathogenesis of peptic ulcers. This research aims to know the gastroprotective effect of ganyong rhizome on white female rats, Wistar strain, which were given alcohol (ethanol 96%) by dose 1 ml/200gBB and measuring the total flavonoids content in ganyong rhizome. This research is Post Test Only Control Group Design, using 30 rats, weight 150-200 g, age 6-8 weeks, health condition. It is divided into six groups. Group I (normal) were fed and watered only, Group II (control) were given a CMC-Na 0,5%, Group III-V were given a suspension of ganyong rhizome starch (SGRS) with each doses 250, 500, and 1000 mg/kgBB, then Group VI were given a sucralfate dose 360 mg/kgBB. Rats were given oral treatment for 14 days. An hour after the treatment on 14th day, the whole groups unless the group I were given ethanol 96% 1 ml/200gBB orally. Twenty four hours later the rats were sacrificed, dissected and taken its gastric for further analyzed the number of ulcers as well as the description of histopathology of the gastric. The results showed that ganyong rhizome starch can decrease the index ulcer and increase the protection ratio value to the control group with results of doses SGRS 250, 500, 1000 mg/kgBB sequentially i.e., 12,5; 50; and 75%. The description of histopathology also showed significant improvement of gastric tissue at a dose of 1000 mg/kgBB. In addition, on TLC-Densitometri test also obtained the total flavonoid compound in ganyong rhizome is 112,49 ± 7,97 ppm. Conclusions of this research are that the ganyong rhizome positive contains flavonoid and it has gastroprotective effect by decreasing the number of index ulcer, increasing the protection ratio value, and improving the histopathology of gastric tissue which had given alcohol.

Abstract

Cataract is one of the eye pathology that can cause blindness. Oxidative stresshas been identified as an initiating factor of cataract. The objective research was tostudy the protective effect of ethanol extract Centela asiatica toward cataract formationon Wistar rat induced by sodium selenite. Cataract was induced in 9 days old Wistarrat, which 5 groups of 7 each. Group I got injection aquadest and the test groups gotinjection CMC-na (group II), vitamin E dose 378 IU/kg bw (group III), ethanol extractGotu cola herb dose 100 mg/kg bw (group IV), and 200 mg/kg bw (group V), 400mg/kgbw, subcutaneously, respectively, for 3 days. After 4 hours, all rats got injection asingle dose 50 ug/kg of BW sodium selenite intraperitoneally. Then eyes were observedin all groups on postnatal day 16, when the eyes of the rat first opened. Then percentageof cataract incidence were calculated. Finally, rats were executed and eyes were takenfor lens histophatological analyses with hematoxiline eosin staining. Based onmacroscopic and microscopic analyses it can be concluded that ethano extract ofCentella asiatica had protective effect toward cataract formation on Wistar rat inducedby sodium selenite. The least cataract incidence was showed at dose 200 mg/kg bw.

Abstract

Previous studies have shown that the formulations of clove bud essential oil (CBEO) with a concentration of 5% in O/W cream and 2.5% in W/O cream have anti-inflammatory properties. The formulations are developed by adding enhancers, namely oleic acid and propylene glycol. The optimum compositions of oleic acid and propylene glycol are 50%:50% in W/O cream and 30%:70% in O/W cream. This study aimed to determine the effects of different compositions of creams on their anti-inflammatory activities. The creams were evaluated for their anti-inflammatory properties using mice with croton oil-induced inflammatory. The mice were sacrificed, and the back skin was removed to make histopathologic preparations at the end of the three-day inflammatory induction to obtain data of epidermal thickness and the number of cells with COX-2 expression. The results showed that the addition of oleic acid and propylene glycol as enhancers increased the anti-inflammatory activity of both O/W and W/O creams. Since the O/W cream decreased the thickness of the epidermis and the number of cells with COX-2 expression more than the W/O cream, the formulation of CBEO in O/W cream is concluded as a better anti-inflammatory than that of W/O cream.

Abstract

Liver is one of the most important organ for drug metabolism in the body. The damage to the liver can be marked by increased activity of the enzyme alanine aminotransferase (ALT) in the blood. Lycopene is an antioxidant that has the ability to eliminate free radicals and detoxifies electrophilic compounds that damage can be prevented. The purpose of this research is to know whether lycopene can inhibit the increase alanine aminotransferase activity of SD (Sprague Dawley) rats induced by parasetamol. This experiment using 20 male rats Sprague Dawley. They were divided into 4 groups. Group I is a healthy control (baseline) was only given aquadest, group II were given paracetamol as pain control, group III and IV are treatment group were each given lycopene with a dosage 15 µg/kg BW and 30 µg/kg BW for 21 days. On 19th day group II, as well as all treatment groups induced paracetamol. The blood sampling was performed on orbital sinus on 19th day (before induced by paracetamol) and on 21th day or two days after induced paracetamol to know the difference alanine aminotransferase activity. The data were analyzed using non-parametric statistical methods with Kruskal-Wallis test followed by Mann-Whitney test with 95% confidence level. The result has showed that the lycopene doses 15 µg/kgBW had alanine aminotransferase activity of 130.10 ± 11.83 U/I and lycopene doses 30 µg/kgBW had alanine aminotransferase activity of 106.46 ± 2.36 U/I. If compared with paracetamol control had alanine aminotransferase activity of 92.53 ± 2.57 U/I, should take a summary that the lycopene doses 15 ìg/kg BW and 30 ìg/kg BW couldn’t decrease alanine aminotransferase activity on SD (Sprague Dawley) male rats.

Abstract

Curcuma xanthorrhiza Roxb is one of the plants containing curcumin. Curcumin has anti-amyloidogenic,antioxidant, and anti-inflammatory activity that can prevent dementia. The aim of the research was to know theeffect of Curcuma xanthorrhiza Roxb rhizome ethanol extract (CREE) to spatial memory on dementia model ratinduced by trimethyltin (TMT). This research use 30 rats, divided into 6 groups, each group consisted of 5 rats. Allgroupswere given trimethyltin intraperitoneal lyexcep tgroup I.Group I (healthy controls) and group II (TMT/illness control) were given a solution of 1% CMC-Na orally. Group III (drug control) were given citicoline 200mg/kgBW orally. Group IV, V, and VI were given CREE orally dose 50 mg/kgBW, 100 mg/kgBW, and 150mg/kgBW respectively. Data of spatial memory were obtained by Morris water maze test consisted of acquisitiontrial, and a probe trial. Data from the test then statistically analyzed by one-way repeated measure ANOVA.Morris water maze test showed that there were significant differences (p<0,05) between TMT group comparedwith CREE dose 50 mg/kgBW, 100 mg/kgBW, and 150 mg/kgBW group. But there was no significant difference(p>0,05) between CREE dose100 mg/kgBW group anddrug control group.Based on the research results can beconcluded that ethanol extract Curcuma xanthorrhiza Roxb rhizome at dose of 100 mg/kgBW can improvespatial memory on dementia model rat induced by trimethyltin.
