Abstract

Located around the Mangrove Production and Protection Forest in Tanjung Harapan Village, Kubu Raya Regency, the Bongkok River quality needs to be studied to determine the quality of the river. This study aims to determine the physicochemical quality of water and the community structure of phytoplankton in the Bongkok River during the dry and rainy seasons. The survey method was used at three stations and 9 observation points in both seasons. Phytoplankton types are identified up to the species level. The physicochemical factors observed included temperature, transparency, water flow, pH, salinity, dissolved oxygen, dissolved carbon dioxide, and levels of nitrate and phosphate. The community structure is seen from the abundance, diversity, uniformity, and dominance index values. From a physicochemical perspective, the Bongkok River is a brackish water type with relatively low levels of phosphate and nitrate nutrients and high dissolved CO2, but dissolved oxygen is still good enough to support aquatic life. There are 41 phytoplankton species from 6 classes, including Bacillariophyceae, Chlorophyceae, Cyanophyceae, Dinophyceae, Euglenaphyceae, and Xanthophyceae. The phytoplankton abundance in the dry season is higher than in the rainy season but is still at a low to moderate level. The phytoplankton diversity in the Bongkok River is in the medium category (2.46 – 2.67) but has a high evenness (0.72-0.89) and low dominance (0.11-0.18). Overall, the water conditions in the Bongkok River are sufficient to support the life of aquatic biota. However, the abundance of phytoplankton is still at a low to moderate level, especially during the rainy season.

Abstract

Identification of sequence polymorphism on the D-loop region of mtDNA has been done for various purposes, including health and medical treatment. In this research, single nucleotide polymorphisms were identified in the D-loop region of mtDNA of the Sundanese population in western Java. A total of 118 unrelated and healthy Sundanese probands were collected from closed-traditional kampung adat and open communities distributed in 14 cities and regencies in western Java. DNA amplification and direct sequencing of the D-loop region were proceeded using primers L15990 and H409. Multi-alignment was conducted not only intrapopulation but also with D-loop sequence data stored in GenBank for comparison. In this research, we categorized high-frequency SNPs as less effective for identification in population studies because of their presence in other populations outside Indonesia. Meanwhile, lower-frequency SNPs showed typical variants of Sundanese haplotypes. On the other hand, rare or low-frequency SNPs should be re-examined in larger size of samples to have a better understanding of risk factors for many diseases.

Abstract

Betawi identity was assessed based on the diversity of maternal lines using genetic markers in the D-loop region of mitochondrial DNA. The results of the cleavage by the restriction enzymes DdeI, HaeII, HaeIII, and HinfI showed five patterns of genetic variants which were later named Haplotype I, II, III, IV, and V. Haplotype I had the highest frequency of 75.6%, followed by Haplotype III 11.5%, and Haplotype II 10.3%. Haplotypes IV and V were only found in 1 individual each and were assumed to be individual mutations. The highest haplotype in the Betawi population showed the same restriction enzyme cleavage pattern with all comparison samples from the Sunda and Malay populations. Meanwhile, Haplotype II and III were strongly suspected as the result of the divergence of Haplotype I and can also be found in other ethnic groups in Indonesia. This finding confirms Jakarta as a location for ethnic mixing which then produces a new local culture as Betawi Culture.

Abstract

The Segedong River flows between the Tanjung Prapat Muda Mangrove Protected Forest in Kubu Raya Regency. Mangrove forests contribute nutrients to water. However, nutrients and fluctuative environmental conditions influence the existence of phytoplankton. People’s settlements and fish farming activities downstream of the Segedong River influence environmental conditions. Research on phytoplankton is needed to determine the quality of the Segedong River. The aim of this research is to determine the abundance and diversity of phytoplankton in the downstream Segedong River. Sampling was carried out in the dry and rainy seasons using a survey method at 3 stations. Water physicochemical factors are measured in situ and ex situ. The identified phytoplankton species are then calculated for their abundance, diversity index, evenness, and dominance. 59 phytoplankton species are categorized into 6 classes, including Bacillariophyceae (35 species), Dinophyceae (15 species), Chlorophyceae (4 species), Cyanophyceae (3 species), Chrysophyceae (1 species) and Xanthophyceae (1 species). The phytoplankton abundance obtained in both seasons at all stations ranged from 1,314-10,185 Ind.L-1 or categorized into oligotrophic to mesotrophic. The diversity index ranges from 1.99-2.88 in the medium category. The evenness index ranges from 0.53-0.80, which is in the medium to high category, while the dominance index is low <0.4.

Abstract

The era of globalization requires students as the nation's next generation to have 21st century skills. However, most students have not received or been trained in 21st century skills while in college. Learning with a constructivist approach with the case method model and team-based project is one strategy to solve this problem, but most of the lecturers in the PMIPA FKIP Untan department have not been able to distinguish and apply the two learning models. This is due to the low understanding of educators related to the case method and team-based project learning models. The aim of this workshop is to improve lecturers' understanding and skills in developing learning tools using the case method and team-based projects. This workshop begins with a presentation of material on the case method and team-based project learning tools using the lecture method and continues with a review of the RPS that has been prepared by the lecturer. All activities are carried out online. The results of the workshop were that 96.5% of the lecturers had understood the RPS using the case method and 93% of the team-based project. As many as 65.5% of lecturers stated that this activity was very helpful in preparing lesson plans with the case method and team-based projects. Meanwhile, 96% of participants wanted to do similar activities again. This indicates that this workshop has provided understanding and assisted participants in preparing lesson plans using the Case Method and Team Based Project. The results of this workshop have motivated lecturers to produce learning tools, especially better lesson plans.
