Abstract

APLICATION OF WASHING MACHINE AND ROTARY DRYER FOR INCREASING ADDED VALUE VALUE OF EMPON-EMPON IN BANDAR VILLAGE, BANDAR DISTRICT, PACITAN. Bandar Village is located in Bandar District, Pacitan Regency. The economy of the people in Bandar Village is supported by traditional medicinal plants, especially empon-empon as the main commodity. Most of the farmers in Bandar Village are small farmers who depend on selling their crops to middlemen. The middlemen argue that prices at the farm level are low because the empon-empon is still dirty. The aim of this project is to carry out post-harvest processing activities by applying of washing machine and rotary dryer to increase the added value of the empon-empon products. Washing machine can clean it from dirt while rotary dryer can reduce the water content thereby increasing the yield of the active ingredients of empon-empon. Currently, both machines can function properly. This transfer of technology is very helpful for farmers in processing their harvested products on a large scale and they can sell them at higher prices. Kelompok Tani Suroloyo I as a partner actively participated in this project from post-harvest processing to marketing. This project is carried out on an ongoing basis in collaboration between Pusat Penelitian Lingkungan Hidup Universitas Sebelas Maret Team (PPLH UNS) and Kelompok Tani Suroloyo I.

Abstract

Dampak pandemi COVID-19 telah menurunkan pendapatan beberapa industri jamu di Kabupaten Sukoharjo. Industri jamu terkendala dalam proses produksi dan penjualan produknya. Cara pemasaran berubah, terutama saat diberlakukan pembatasan kegiatan masyarakat darurat (PPKM Darurat). Salah satu cara alternatif untuk meningkatkan kapasitas penjualan adalah melalui pemasaran digital online yang berfokus pada pemasaran digital melalui website yang digunakan sebagai e-commerce, media sosial, mesin pencari, dan membentuk tim reseller untuk menjual produknya. Di masa pandemi, industri jamu harus cepat beradaptasi seiring dengan PPKM Darurat, karena hal ini akan mengubah tren perilaku konsumen dalam membeli produk jamu. Salah satu industri jamu yang menjadi mitra proyek ini adalah Usaha Kecil Menengah (UKM) Ching Lung. Melalui transformasi digital ini dapat menjadi solusi atas permasalahan yang dihadapi oleh UKM Ching Lung saat ini. Selain itu, dengan meningkatkan kreativitas model penjualan produk, UKM dapat bertahan dengan melakukan diversifikasi produk yang sesuai dengan kebutuhan di masa pandemi COVID-19. Bekerjasama dengan UKM Jamu Ching Lung, dibuatlah website online yang dirancang dengan konsep toko online untuk meningkatkan penjualan jamu dan obat tradisional di Kabupaten Sukoharjo. Melalui kegiatan ini diharapkan dapat membantu UKM untuk memasarkan produknya agar dapat dikenal oleh masyarakat luas.

Abstract

The Bagas Waras Women's Farmers Group (KWT) is one of the women's groups that cultivates a lot of TOGA in Banyuanyar Village, Boyolali. Their seriousness in cultivating TOGA is proven by the establishment of Omah TOGA, which has become an educational tourism destination. KWT members also have high enthusiasm for developing TOGA's potential and have produced many processed food products from TOGA. KWT Bagas Waras needs innovations in TOGA products, namely traditional cosmetic products. The service team provides scientific insight through training on developing traditional cosmetic products, namely body scrubs made from various types of TOGA. This activity involves 40 KWT members who will participate in a series of stages: preparation, training, practice, and evaluation. This training activity can increase the scientific insight of KWT members and help them develop traditional body scrubs made from TOGA and good packaging methods. This training has gone well, with all participants very enthusiastically listening to material related to traditional cosmetics and demonstrating how to make body scrubs. The results obtained are traditional body scrubs with various variants originating from TOGA.

Abstract

Oral drug absorption was highly influenced by dissolution rate, especially for poorly and insoluble drugs. Piroxicam is a nonsteroidal anti-inflammatory drug that is practically insoluble in water. The oral absorption rate of piroxicam is dependent on its dissolution rate in the GI tract. Polyvinyl Pyrrolidone (PVP) as surfactant can increase drug solubility by means of a micelle forming mechanism. The aim of the study was to know the influence of addition and variation of Polyvinyl Pyrrolidone (PVP) on Piroxicam absorption with everted intestinal sac method.  The  concentration of piroxicam solution was prepared by PVP in  1.0,2.0 and 3.0 % respectively. Crane and Wilson tube containing 75 ml of themucosal fluid was taken at 37 0 C in waterbath. Than, serosal solution of 1.5 mlwas added to intestinal sac by turned upside down and tied to a cannula, then put into the tube containing the mucosal fluid and constantly flowing oxygen gas.Serosal solution of 1 ml were taken every 15 minutes and then diluted with 2 ml of Ba (OH) 2 and 2 ml of ZnSO4 then centrifuge until 25 minute. The absorbant of supernatant was measured by UV spectrophotometer and data analyse wascalculated by one-way ANAVA.  PVP at 1%,2% and 3% increased piroxicamabsorption from the phosphate buffer pH 7.5 compared with negative control.According to P app , the values were 2.52 ± 0.43 cm/minute (negative control), 3.41 ± 2.17 cm/minute (1% PVP), 2.75± 1.14 cm/minute (2% PVP) and 4.77 ± 4.93 cm/minute (3% PVP) respectively. In conclusion. Lower doses of the surfactant (1%, 2%, and 3% PVP) significantly increased absorption of the drug by altering the membrane permeability.

Abstract

The schools have been allowed for doing face-to-face learning process with many requirements.Making own hand sanitizer will help schools to keep applying health protocol beside washing hands. Essential oils have been trusted for a long time give a calm effect and reduce stres. This activity have purpose to assistance hand sanitizer production with essential oils fragrance at SMP Muhammadiyah 1 Prambanan Yogyakarta. Knowledge have been transfered by team followed with demo and training to produce hand sanitizer with lemongrass fragrance at first activity. Knowledge score was determined from pre-test and post test answer score from multiple choice questions about hand sanitizer. Follow up became the next activity to ensure school can make their own hand sanitizer independently. The result showed knowledge score was increased from 52,11 to 92,11. School also have been made hand sanitizer independently with another essential oil varians. This products were limited used for school. Communication between team and school still continues.

Abstract

As one of the leading causes of death in worldwide, cervical cancer requires the effective therapies to reduce its mortality rate. One of the chemotherapy agents that frequently used in the treatment is cisplatin. However, due to drug resistance and its side effects, an agent that can be combined with cisplatin is needed. Parijoto fruit (Medinilla speciosa Reinw.ex.Bl) contains secondary metabolites compounds that have potential as anticancer. The study aims to determine the cytotoxic effect of methanol extract of Parijoto fruit calculated from the IC50 value and the synergicity of the combinational treatment with cisplatin evaluated from the Combination Index (CI) value and its cell viability by using MTT assay. Results showed that methanol extract of Parijoto fruit (MEP) performed cytotoxic effect on HeLa cell line with IC50 of 209.6 μg/mL while the value of IC50 of cisplatin against HeLa cells amounted to 12.8 μg/mL. The combination of 26.205 ppm (1/8 IC50) of MEP and 1.601 ppm (1/8 IC50) of Cisplatin performed synergistic effect on HeLa cell line with the CI value of 0.69. From the above results, it can be concluded that MEP is potential as co-chemotherapy agent based on the synergistic cytotoxicity effect with cisplatin.Keyword: cytotoxic, Medinilla speciosa, cisplatin, co-chemotherapy, MTT

Abstract

Parijoto (Medinilla speciosa, Reinw. ex. Bl.), a tropical plant native to Southeast Asia, contains flavonoids, tannins, and saponins, which have the potential as an anticancer. Gold nanoparticle-based drug formulations are applied to increase the anticancer effectiveness of herbal medicines. The compounds in the stalk of parijoto have the potential to be bioreductor in the biosynthesis of gold nanoparticles. This study aims to determine the physicochemical characterization, cytotoxic activity, and expression of protein caspase-9 after treatment with nanogold parijoto (AuNPs-PR) on HeLa cell. The nanogold biosynthesis process was done by reacting 1 mM HAuCl4 with parijoto aqueous extract (EP). Physicochemical characterization measure of particle size, Polydisperse Index (PdI), and zeta potential of AuNPs-PR was carried out using a particle size analyzer. The cytotoxic effect and viability cell of AuNPs- PR were carried out using the MTT assay. The expression of caspase-9 was observed by immunocytochemistry assay. Physicochemical characterization of AuNPs-PR shows that the particle size value is 160.8 nm with PdI and zeta potential values of 0.430 and -4.56 mV respectively. In the MTT assay, both AuNPs-PR and EP demonstrated a reduction in the viability of Hela cells after 24 h in a dose-dependent manner, yielding IC50 values of 3.28 μg/mL and 19.22 μg/mL, respectively. AuNPs-PR and EP showed low cytotoxic activity against Vero normal cells, with IC50 values of over 500 μM. Further, the immunocytochemistry assay indicated that there was upregulation of caspase-9 by their expression. These results indicate that AuNPs-PR could effectively induce apoptosis in HeLa cells by upregulating caspase-9.Keywords: caspase-9, HeLa cells, MTT, nanogold, parijoto.

Abstract

Cisplatin (Cisp) is a non-specific chemotherapeutic agent for breast cancer. Hesperetin (HST), a flavanone found in various citrus fruits, exhibits bioactive properties, functioning as an antioxidant, anti-inflammatory, and anticancer agent. The objective of this research was to investigate the potential of HST as a co-chemotherapeutic agent in conjunction with Cisp, specifically focusing on its cytotoxic effects against 4T1 triple-negative breast cancer cells and senescence modulatory effect on Vero normal kidney cells. The cytotoxic effect and viability cell of HST were evaluated through 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium (MTT) assay. In addition, the effect of cellular senescence inhibition on the Vero cell line was measured using senescence-associated β-galactosidase (SA-β-gal) staining. In the MTT assay, both HST and cisplatin demonstrated a reduction in the viability of 4T1 cells in a dose-dependent manner, yielding IC50 values of 498 μM and 2 μM, respectively. The co-treatment of HST and cisplatin showed an increase in sensitivity of the 4T1 cells with a combination index of <1. HST showed low cytotoxic activity against Vero cells, with IC50 values of over 500 μM. HST decreased cellular senescence induced by cisplatin exposure on Vero cells. These results indicated that HST in co-treatment with cisplatin decreased 4T1 cell viability synergistically. HST independently reduces the cellular senescence of normal cells. Consequently, HST holds promise for potential development as a co-treatment agent in combination with cisplatin for breast cancer cells, and it may also serve as an alternative for counteracting senescence in healthy tissues.Keywords: cytotoxic, senescence, hesperetin, cisplatin, breast cancer.

Abstract

Bay leaves are used as food flavoring and also have medicinal properties. They may have cytotoxic effects derived from natural ingredients. The low efficacy of the therapy with an adequate dose preparation of the plant extract is due to its low solubility and oral bioavailability that is less than the maximum. Hence, this study aimed to improve the solubility and oral bioavailability of the extract mainly for the chloroform extract of leaves that are not soluble in water by preparing a self-nanoemulsifying drug delivery system (SNEDDS). Then, the potential cytotoxic effects of the SNEDDS of bay leaves were determined by calculating the value of IC50 on the T47D cell line. The cytotoxic effect of the SNEDDS of bay leaves was determined using an MTT assay, and the findings were read using an ELISA reader. Data analysis is calculated via linear regression methods by using Microsoft Excel software. The results showed that the SNEDDS of bay leaves performed cytotoxic effects on the T47D cell line with IC50 138 μg/mL. The results showed that the optimal composition formula SNEDDS, namely, Tween 20:PG:oleic acid = 2.25:2.25:0.5 in 5 mL SNEDDS preparation, which had a value of transmittance of 83.81% with emulsification time was less than 5 min; the average droplet size was 165.5 nm, and the zeta potential was −0.4 mV. The data analysis showed that the cytotoxicity effect of the SNEDDS of bay leaves is included in the moderate cytotoxic category.Keywords: Bay leaf, optimization, nanoemulsi, cytotoxicity.
