Abstract

Artemisia annua is a herbaceous plant that produces artemisinin as a malaria drug, haemorrhoids therapy, aromatherapy, antiviral, anticancer and antibacterial. Gloriosa superba is a plant that contains high colchicine compounds, especially on the seeds. Gloriosa superba extracts of tubers, stems, seeds, and leaves were used as biomutagen for many plants. Colchicine contains of these plants as antimitotic have been studied and proven by the mitotic index plants. Water extracts of Gloriosa superba seeds was used as a mutagen for Artemisia annua. The aim of this study was to determine the size and density of Artemisia annua stomata soaked in water extract of Gloriosa superba seeds as a mutagen. Extraction of Gloriosa superba seeds obtained naturally on Krakal Beach, Gunung Kidul by using a maceration method with water solvent (1:1). Artemisia annua sprouts were obtained from B2P2TOOT Tawangmangu. Variables treatment on sprouts using water extract concentration of Gloriosa superba seeds and soaking time of Artemisia annua sprouts. Measurements of stomatal length, width and density were conducted in epidermis of Artemisia annua leaf. Observation and measurements of the stomata were conducted by using a light microscope. The results showed that the length and width of stomata were 0.025 mm and 0.017 mm respectively. The stomatal density of the control leaf (174.69 amount/mm2) was lower than the other treated plants. Stomatal size and density has increased with the increasing concentration extracts on treated plants. Water extracts of Gloriosa superba seeds proved the effects on stomatal size and density of treated plants.

Abstract

Dendrobium sylvanum orchid is categorized as lowland habitus orchid and able to survive wihtout any shade. Orchid cultivation takes a very long time so it needs faster cultivation and producing large quantities. Therefore, it is necessary to do orchid culture by using tissue culture processes and then given gamma ray irradiation to bring up new characters in the orchid .The purpose of this research was to discover the impact of gamma ray irradiation in vitro to the diversity of D. sylvanum. The doses of gamma ray used in this research were 0 Gy, 15 Gy, 30 Gy, 45 Gy, and 60 Gy. The results obtained indicated that mutation mostly occurred on the treatment with 15 Gy (4.4 cm of plant height, 2.67 of leaves, 2.36 cm of leaf length, 0.49 cm of leaf width, and 5.33 of root strands) compared to the other doses and the control plants. The new finding in this research is the gamma ray dose that can optimally stimulate the mutation in D. sylvanum. This information is useful to generate the new variety in orchid cultivation in Indonesia. This research provides innovation in orchid cultivation and new variety that is possible to arise after the mutation.

Abstract

ABSTRACT   This research was conducted from March to May 2017in the Koto Masjid Village, Kampar, Riau and observation parameters has done in the Environmental Quality Laboratory of Aquaculture Faculty of Fisheries and Marine University of Riau. The aim of research was to knowing the layers on the profile of bottom soil of yellow podzolic red (YPR) pond and its characteristic with different age which catfish cultivated (Pangasius sp.) intensively. This research used eksperiment method and had direct observation with 1 factor, 4 treatments and 3 replications. The highest basal soil profile of the initial layer of flocculent layer (F) was P1 (0-5 years age) of 0.98 cm while in F final was P3 (age of 11-15 years). Furthermore, in the highest mixed sediment layer (S) layer, P3 is 3.98 cm, whereas in the final S layer is P2 (aged pond 6-10 years) that is 7.91 cm. Furthermore, the highest stable matures is stable sediment (M) is P4 (16-20 years age pond) which is 8.59 cm, while in the final M layer is P2 which is 11,50 cm. The next stage of the highest transitional layer (T) layer is P3 which is 10.54 cm, whereas in the final T layer it is P2 that is 17.16 cm. The measured soil quality parameters are still considered good. Soil sand fraction 74,16-89,66%, silt fraction 2,66-9,83%, clay fraction 7,66-16,00%. Ground BV 0.45-1.58 g / cm3, ground BJ 1,53-2,18 g / ml, soil porosity 14,75-66,26%, C organic soil 1,97-5,60% and pH 6,80-6,96.Keywords: Bottom Soil Pond Profile, Flocculent layer, YellowPodsolik Red, Pangasius sp.

Abstract

Indonesia is a multi-cultural country and has a plurality in many dimensions. This pluralistic culture is a nation's wealth capital in understanding and interacting with other nations. Current developments have led us to wider interactions across national borders where the relationship involves intercultural aspects. However, building and fostering ethnocultural empathy is an accumulative and continuous process and requires consistency over a long period of time. The most important first process is that we need to analyze the state of cultural empathy in society. This exploration, of course, requires a valid and reliable measuring instrument. As a challenge, we don't have limited instruments to explore this yet. In developing the instrument, the vast territory of Indonesia requires us to research in several periods to get maximum results. In this study, the Scale of Ethnocultural Empathy will be adapted and developed by Wang et al. and tested its validity and reliability in the central region of Indonesia. The number of items tested was 59 on 399 people, consisting of 18 males and 381 females. As for the distribution of the tribes living on Kalimantan, Bali, and Sulawesi, 24% came from the Javanese, and 44.86% of the total population came from the Makasarese, Bugis, Balinese, Ambonese, Minahasa, Dayak, and Kalimantan Malays. The study results showed that the scale of cultural empathy (The Scale of Ethnocultural Empathy / SEE) as a result of adaptation and development of 59 items was declared valid as many as 55 items and 4 items dropped. The results of the study also showed that who selected the scale of cultural empathy (The Scale of Ethnocultural Empathy / SEE) as many as 55 valid items, 58 items that met the item discriminatory value with riX 0.30 in the item discrepancy test stage, thus remaining 55 items based on the validity and item discrepancy. The results of the study also show that the scale of cultural empathy (The Scale ofEthnoculturall Empathy / SEE) as many as 55 items that meet the value of item discriminating power and validity was tested for reliability and obtained a Cronbach's Alpha score of 0.797 so that the instrument with 55 items can be accepted as a scale with reliability. Very high.

Abstract

Tribulus terrestris merupakan salah satu tanaman obat yang mempunyai beragam manfaat antara lain sebagai diuretik (peluruh kencing), meningkatkan kadar hormon testoteron, memulihkan vitalitas dan menambah kebugaran. Hambatan dari budidaya tanaman ini secara konvensional adalah relatif rendahnya persentase daya kecambah, bentuk bijinya yang kecil dan budidaya yang tergantung pada musim tertentu. Perbanyakan tanaman dengan menggunakan biji secara in vitro merupakan solusi yang dapat digunakan untuk memperbanyak tanaman ini. Penelitian bertujuan untuk mengetahui pertumbuhan in vitro T. terrestris dengan perlakuan IBA dan BAP. Penelitian menggunakan Rancangan Acak Lengkap (RAL) yang disusun secara faktorial. Faktor pertama adalah konsentrasi IBA dan faktor kedua adalah konsentrasi BAP. Analisis data dilakukan dengan uji DMRT dan deskriptif. Hasil penelitian menunjukkan adanya interaksi pemberian IBA 1 ppm + BAP 1 ppm menghasilkan tinggi tunas tertinggi dan jumlah akar terbanyak, interaksi pemberian IBA 1 ppm + BAP 1 ppm dan IBA 1,5 ppm + BAP 2 ppm menginduksi saat muncul akar tercepat. Pemberian IBA secara tunggal tidak mempengaruhi pertumbuhan eksplan T. terrestris, pemberian IBA 0,5 ppm dikombinasikan dengan BAP 2 ppm dapat menginduksi panjang akar terpanjang. Pemberian BAP 1 ppm dapat menginduksi saat muncul tunas tercepat, saat muncul daun tercepat dan jumlah daun terbanyak.

Abstract

Cekaman air merupakan salah satu faktor pembatas pertumbuhan dan produksi tanaman termasuk cabai (Capsicum annuum L.). Produktivitas tanaman merupakan karakter yang kompleks sehingga hubungan antarkarakter perlu diketahui untuk mendapatkan kriteria seleksi yang tepat untuk perbaikan tanaman. Penelitian ini bertujuan mengetahui kriteria seleksi terhadap produksi tanaman cabai toleran kekeringan pada fase pembungaan. Penelitian disusun menggunakan Rancangan Acak Lengkap dengan tiga taraf cekaman air, yaitu 100% kapasitas lapang (kontrol), 50% kapasitas lapang (medium stress), dan 25% kapasitas lapang (extreme stress). Hasil analisis korelasi menunjukkan bahwa berbagai karakter yang diamati memperlihatkan perbedaan asosiasi dengan level cekaman kekeringan. Karakter panjang akar tidak dapat digunakan sebagai kriteria seleksi tanaman cabai yang toleran terhadap kekeringan karena panjang akar tidak berkorelasi secara signifikan dengan bobot buah per tanaman pada 50% dan 25% kapasitas lapang. Berdasarkan korelasi dan analisis lintas pada kondisi normal (100% kapasitas lapang) dan kondisi tercekam (50% kapasitas lapang), karakter jumlah buah dan persentase fruit set berkorelasi positif dan berpengaruh langsung terhadap bobot buah per tanaman sehingga dapat digunakan sebagai kriteria seleksi untuk perbaikan tanaman cabai yang toleran kekeringan.KeywordsCekaman air; Cekaman kekeringan; Kriteria seleksi; Cabai; Capsicum annuum LAbstractWater stress is one of limiting factors for plant growth and production, including chili (Capsicum annuum L.). Plant productivity is a complex character so the relationship between characters needs to be known to get the right selection criteria for crop improvement. This study aimed to determine the selection criteria for the production of drought-tolerant chili pepper at the flowering phase. The research arranged under completely randomized design with three levels of water stress, namely 100% water field capacity (control), 50% water field capacity (medium stress) and 25% water field capacity (extreme stress). The results of the correlation analysis showed that the various characters observed exhibited differences associations with drought stress levels. Root length cannot be used for selection criterium of drought tolerant on chili because it was no positive significantly correlated to fruit weight per plant at 50% and 25% water field capacity. Based on correlation and path analysis in normal and drought conditions, the number of fruits and percentage of fruit set can be used as selection criteria for genetic improvement of drought-tolerant on chili plants because these characters were significantly positively correlated and had a direct effect on fruit weight of the plant.

Abstract

Soil is growing media for plant growth. It preserves water and nutrients for plants. The physical  changes in soil affect the life of roots and plant performances. The goal of  research  was to  change physical condition in soil through mulching for increasing pepper plant production. Land for pepper cultivation was mulched using rice straws and plastic sheet (PE). Physical condition such as soil suhue and humidity and content of organic matters in the soil were measured or examined. Also, production components such as plant height and stalk diameter were measured as well as fruit production. The result showed that mulching increased significantly soil suhue and humidity as well as content of organic matters in the soil. Mulching increased plat height, diameter of stalk, and fruit production, especially in using  rice straws Therefore, mulching have  a role in increasing growth and production pepper fruit  through modification of physical condition in soil.Keywords :  mulching, growth, production, pepper

Abstract

Gloriosa superba merupakan herba merambat yang mempunyai nama lokal Kembang Sungsang. G. superba berkembangbiak secara liar di daerah Pantai Krakal, Gunung Kidul, Yogyakarta. Herba ini bermanfaat sebagai pengobatan tradisional dan pemuliaan tanaman. Populasi tanaman ini sangat melimpah di sekitar Pantai Krakal dan tidak diketahui manfaatnya oleh masyarakat sekitar. Tujuan penelitian ini adalah mengetahui morfologi secara umum G. superba dan mengetahui kadar kolkisin yang terkandung dalam biji G. superba. Pengambilan contoh G. superba dilaksanakan bulan November 2016 di Pantai Krakal dengan mengambil seluruh bagian tanaman untuk pengamatan morfologi tanaman. Analisis kadar kolkisin yang terkandung dalam biji G. superba menggunakan metode KLT-Densitometri. Hasil penelitian ini menyebutkan bahwa morfologi tanaman ini terdiri dari bunga yang berwarna merah dan kuning, daun berwarna hijau meruncing, biji yang dibungkus oleh buah kapsul, umbi berwarna putih dan batang yang merambat panjang hingga ±5 m. Kadar kolkisin yang terkandung dalam ekstrak air biji G. superba sebesar 12,84 µg/µl (± 2,88).

Abstract

Problems encountered in the development of herbal medicine industry is that most of the raw materials (80%) came from the forest or natural habitats and the rest (20%) of the results of traditional cultivation. The research is focused to get the propagation techniques of temulawak by in vitro using various combinations of IBA and BAP concentration, each consisting of 0, 1, 2, 3, 4 ppm. The purpose of this research is to gain a plant propagation technique of temulawak through the use of IBA and BAP concentration so as to provide a quality seeds. The results showed that the highest shoots obtained at 1 ppm IBA and 3 ppm BAP treatment with a buds height of 15.9 cm. Treatment of 0 ppm IBA and 4 ppm BAP produces the highest buds number by 4 buds. While the highest number of leaves obtained in 2 ppm IBA and 3 ppm BAP treatment with total of 18 leafs. The highest length of leaf obtained in the treatment of 3 ppm IBA and 2 ppm BAP with a length of 14.6 cm. Variable highest number of roots was obtained at treatment of 4 ppm IBA and 1 ppm BAP with root number exceeds 30 pieces with very many hair roots and the highest root length was obtained at treatment of 2 ppm IBA and 0 ppm BAP with a length of 15.6 cm. Based on these results it can be concluded that the IBA and BAP treatment at various concentrations can affect the propagation and growth of temulawak explants.
