Abstract

Introduction: IL-6 acts as a biomarker of the inflammatory process. In elderly, especially women, the demand for tooth extraction treatment is quite high. A failure in the wound healing process during the menopausal period can prolong the inflammation process, thereby increasing the probability of infection, including alveolar osteitis. The menopausal process affects estrogen levels in the blood, which leads to reduced collagen formation as a key component of tissue regeneration. IL-6 levels can also increase during inflammation, especially in the chronic phase following tooth extraction. The study aims to analyze the differences in inflammatory IL-6 levels between menopausal and non-menopausal rats. Methods: This study applied an in vivo experimental design. A total of 30 Rattus novergicus were divided into two groups: a control group and a treatment group. The control group consisted of rats in which no oophorectomy was performed (non-menopausal), while the treatment group underwent ovariectomy to induce menopause. One after the procedure, tooth extraction were performed, and observations were conducted on days 3, 7, and 14 post-extractions. IL-6 levels were measured using ELISA from blood serum samples. The data were analyzed using One-Way ANOVA.  Results: The average IL-6 levels (in ng/L) in the control (non-menopausal) group on days 3, 7, and 14 were 0.087, 0.318, and 0.247, respectively. The average IL-6 levels in the treatment (menopausal) group were 0.191, 0.452, and 0.318, respectively. The One-Way ANOVA test showed no significant difference between the groups (p=0.799 > 0.05).  Conclusion: The trend of higher IL-6 levels in the menopausal group suggests a potential impact of hormonal changes on post-extraction inflammation. Although no significant difference was found, the elevated IL-6 levels in menopausal rats indicate a potential effect of estrogen deficiency on the inflammatory response following tooth extraction.

Abstract

Latar belakang: Madu Apis dorsata memiliki aktivitas untuk menghambat pembentukan biofilm Candida albicans . Madu mengandung fenolik, steroid, tannin dan flavonoid. Dimana flavonoid merupakan salah satu kelas dari keluarga fenolik (setengah dari jumlah total senyawa fenolik) berfungsi untuk menghambat pembentukan biofilm Candida albicans . Tujuan: penelitian ini bertujuan untuk mengetahui aktivitas antibiofilm madu Apis dorsata terhadap biofilm Candida albicans . Metode: Sampel yang digunakan berupa madu Apis dorsata yang diperoleh dari Gunung Mutis Nusa Tenggara Timur. Sampel madu dilarutkan dalam media RPMI 1640 dengan konsentrasi larutan konsentrasi 50%, 25%, 12,5%, 6,25%, dan 3,125%. Uji aktivitas antibiofilm madu terhadap biofilm Candida albicans  dilakukan dengan metode Mikrotiter plate menggunakan ELISA reader.  Hasil: Hasil penelitian menunjukkan bahwa semua konsentrasi madu memiliki aktivitas menghambat pembentukan biofilm Candida albicans  dengan penghambatan terbesar konsentrasi madu 50% sebesar 69,78%. Penelitian ini menunjukkan bahwa madu Apis dorsata dari Gunung Mutis NTT secara kuat menghambat pembentukan biofilm Candida albicans . Efektivitas ini bersifat dosis-dependen, di mana konsentrasi madu 50% memberikan tingkat penghambatan tertinggi sebesar 69,78%. Kesimpulan: Dengan demikian, madu ini patut dipertimbangkan sebagai sumber agen antibiofilm alami yang potensial, berkat kandungan senyawa fenolik dan flavonoidnya.

Abstract

Candida albicans infection is a serious health concern, particularly in immunocompromised individuals, due to its ability to form biofilms that are resistant to conventional antifungal therapies. Curcuma xanthorrhiza Roxb. (temulawak) possesses antifungal properties; however, its effectiveness is limited by poor solubility and bioavailability. Nanoemulsion formulation offers a promising strategy to enhance the penetration of active compounds. This study aimed to develop and evaluate the effectiveness of temulawak nanoemulsion as an antibiofilm agent against multidrug-resistant (MDR) C. albicans. Using a post-test control group only design, the antibiofilm assay was conducted via the microtiter plate method and analyzed based on Optical Density (OD) measurements. The results showed that temulawak nanoemulsion significantly reduced biofilm formation, with an 89% reduction in OD at the 100% concentration. These findings support the potential use of temulawak nanoemulsion as an alternative therapeutic strategy for Candida albicans infections, particularly those involving drug-resistant biofilms.

Abstract

Escherichia coli is one of the gram-negative bacteria that produces the Extended Spectrum Beta-Lactamase (ESBL) enzyme and has a role in resistance to the beta-lactam group. The presence of E. coli bacteria that produce ESBL in food, especially in fish such as milkfish (Chanos chanos) poses a potential source of the spread of antimicrobial resistance (AMR) in humans. This study aims to determine the prevalence of ESBL-producing E. coli in the intestinal flora of milkfish sold in traditional markets in Surabaya City. This study used a descriptive observational method with a cross-sectional approach. There were 50 samples of milkfish intestines taken from several stalls in traditional markets randomly. Identification of E. coli bacteria by planting in MacConkey media with 4 µL/ml cefotaxime followed by the Double Disk Synergy Test (DDST). Of the 50 milkfish intestine samples used, 34 (68%) samples showed positive results for ESBL-producing E. coli. The high prevalence may indicate that the potential for the spread of antimicrobial resistance through the food chain, food hygiene management and controlling the use of antibiotics in fish farming need serious attention.
