Abstract

Program pelatihan dan sosialisasi mengenai pemanfaatan SIJAKA (sirih merah, jahe merah, dan kayu manis) sebagai minuman fungsional antidiabetes bertujuan untuk meningkatkan pemahaman dan praktik masyarakat dalam mengelola diabetes secara alami. Desa Sukamaju yang terletak di Kecamatan Cigudeg, Kabupaten Bogor memiliki peluang untuk mengembangkan produksi minuman fungsional berbahan dasar SIJAKA, namun potensi ini belum dimanfaatkan secara optimal. Program ini dirancang untuk meningkatkan taraf kesehatan masyarakat serta mendorong pertumbuhan ekonomi lokal melalui pendirian fasilitas produksi minuman fungsional SIJAKA. Pelaksanaan kegiatan melibatkan kader Posyandu dan PKK melalui kegiatan penyuluhan dan pelatihan. Metode yang digunakan mencakup pengisian kuisioner pre-test dan post-test guna mengevaluasi tingkat pengetahuan peserta sebelum dan sesudah pelatihan. Hasil pelatihan menunjukkan adanya peningkatan pemahaman masyarakat terkait minuman fungsional SIJAKA, meskipun sebelumnya pengetahuan mereka masih terbatas. Pelatihan ini juga mengungkapkan bahwa 79,31% peserta mengenal sirih merah dan jahe merah sebagai herbal potensial, sedangkan 72,41% mengetahui manfaat kayu manis. Hasil ini menunjukkan adanya peningkatan kesadaran akan potensi herbal dalam mengelola diabetes. Selain itu, mayoritas peserta (92,11%) menunjukkan ketertarikan yang tinggi terhadap pelatihan dan ingin lebih memahami manfaat serta cara pembuatan SIJAKA. Program ini terbukti mampu meningkatkan antusiasme dan wawasan masyarakat terhadap SIJAKA, serta memiliki potensi untuk dijadikan sebagai contoh bagi desa-desa lainnya. Secara keseluruhan, program ini memberikan dua manfaat utama, yaitu peningkatan kualitas kesehatan masyarakat dan penguatan ekonomi lokal, meskipun masih terdapat kendala dalam mendorong masyarakat untuk memproduksi dan mengonsumsi SIJAKA secara mandiri.

Abstract

Physalis angulata was known for its pharmacological potential, including antioxidant activity, yet the specific bioactive compounds responsible for enhancing endogenous antioxidant enzymes, such as superoxide dismutase 1 (SOD 1), remained unclear. This study aimed to identify and evaluate the active compounds of P. angulata as natural SOD 1 activators using an In silico molecular docking approach. Forty-nine ligands derived from the active constituents of P. angulata were screened based on Lipinski’s Rule of Five and ADMET properties. Molecular docking was performed using the SOD 1 protein (PDB ID: 5YTO), and docking validation yielded an RMSD value of 0.005 Å. Among the test ligands, withanolide exhibited the most favorable binding energy (−7.011 kcal/mol) and the lowest inhibition constant (7.0 pM), forming strong interactions with key catalytic residues of the enzyme. These findings indicated that withanolide had promising potential as a natural SOD 1 activator, providing a basis for future antioxidant drug discovery.

Abstract

The rising demand for halal products, especially within the cosmetics industry, has increased the need for cosmetic products to comply with Islamic laws. One of the challenges in achieving halal certification is the determination of the source of the active ingredients used in cosmetic products, such as enzymes. According to Islamic laws, enzymes derived from pigs or other animals that are not slaughtered are not considered halal. To ensure that the enzymes used in cosmetics are derived from halal sources, it is necessary to use enzymes generated from microorganisms through fermentation, provided that the raw materials and growth medium comply with Islamic laws. Additionally, halal cosmetics must meet regional and international standards, including the Indonesian Ulema Council’s Fatwa and the Malaysian Cosmetic and Personal Care Standards. This research provides a comprehensive review of the role of enzymes in selecting halal compliance cosmetic ingredients, focusing on the source of enzymes as a means of assessing the halalness of cosmetic products. It is essential to utilize halal enzymes such as those derived from plants to provide Muslim consumers with assurance when using cosmetic products.

Abstract

Diabetic patients are susceptible to severe wound conditions due to high blood sugar levels and bacterial infections. This study aims to determine the formulation and effectiveness of the hydrogel plaster combination of laying hen eggshells with button mushrooms in curing diabetic wounds. The research methods consisted of making the hydrogel, coagulation test, and antibacterial tests. Hydrogels were made with 5%, 10%, and 20% eggshell concentrations. The coagulation activity test used rabbit blood, which was added with glucose. Antibacterial test was performed using Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). This research shows that the coagulation activity of F2 and F3 formulations significantly differed from the control, with an average blood clotting time of 64.67 minutes and 63.00 minutes. The antibacterial test results produced a clear zone for S. aureus and E. coli with diameters of 10.27 mm and 9.18 mm for F2, 6.90 mm, and 11.66 mm for F3. This research concludes that hydrogel formulation 3, with 20% eggshell and 6.4% button mushroom, had the best blood clotting effect and antibacterial activity.

Abstract

Phytochemical screening to conduct a preliminary evaluation of the chemical content of black tea (Camellia sinensis), Turmeric (Curcuma longa), and Ginger (Zingiber officinale). In addition, the content of secondary metabolites in black tea, turmeric and ginger can be used as an indication of the quality of these plants. the need for a combination of black tea extract act, turmeric, and ginger from various people so that it can be liked by the public. This study aims to test the phytochemical extracts of black tea, turmeric, and ginger as well as organoleptic tests on the most optimum combination of -glucosidase enzyme inhibitory activity. Phytochemical screening showed that black tea and ginger extract contains alkaloids, flavonoids, tannins, saponins, terpenoids, quinones, glycosides, and phenolics, while turmeric extract contains alkaloids, flavonoids, tannins, terpenoids, quinones, glycosides, and phenolics. The organoleptic test of the F7 combination consisting of a combination of black tea extract, turmeric, and ginger showed the highest level of preference by P3 and P2 on color, aroma, consistency, and aftertaste, while taste was at P3.

Abstract

Heart disease has become the leading cause of death in the world in the last 15 years. This disease has several primary risk factors, one of which is hypercholesterolemia. Cholesterol biosynthesis in the liver is influenced by the activity of the rate-limiting enzyme, HMG-CoA reductase. Traditional herbal medicine provides alternative treatment for hypercholesterolemia. Both pulai and papaya leaves were reported as HMG-CoA reductase inhibitors. There has been very limited study however, on the efficacy of a mixture of aqueous extracts of the two in inhibiting HMG-CoA reductase. This study aims to determine the activity of a mixture of water extracts of both pulai and papaya leaves. The activity of the enzyme HMG-CoA reductase was measured by Abcam® kit No. Ab204701 by spectrophotometric method at 334 nm. The results showed that the three extracts of the combination of pulai and papaya leaves had four times better inhibitory action than the single extract (p0.05). Abstract

Javanese goat and Garut sheep hair contain α-keratin, a protein that can be broken by hydrolysis to produce simpler amino acids. Feather waste generates millions of tons of α-keratin biomass originating from animal slaughterhouses, thereby raising health concerns. The utilization of acid hydrolysis is considered to be more cost-effective compared to enzymatic hydrolysis, and it provides a broader range of amino acid cleavage sites compared to enzymes, which exhibit specific cleavage. This study aimed to isolate amino acids from Javanese goat and Garut sheep hair through acid hydrolysis. The methods included hair sample preparation, acid hydrolysis used 6 M HCL at 110°C, reflux isolation, amino acid separation based on isoelectric pH 4.9 –5.4, functional groups analysis using FTIR, and analysis of amino acid content by HPLC methods. The results showed that the yield produced after isolation on Javanese goat hair samples was 0.92% and Garut sheep hair 0.32%, respectively. The FTIR spectrum showed amino acid functional groups in both samples, including carboxyl (COOH), amine (C-N primer), (C-S disulfide), and amide I (-CONH2). Successful breakdown of α-keratin proteins into simpler amino acids was achieved for Javanese goat and Garut sheep hair. Amino acid analysis of Javanese goat hair isolates revealed the presence of aspartic acid, threonine, serine, glutamate, proline, glycine, alanine, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, histidine, lysine, and arginine amino acids, respectively. The highest content was isoleucine at 0.60% w/w. In conclusion, the isolated amino acids from Javanese goat hair can be used as a halal supplement that serves as nutrition in the body.

Abstract

The study purple sweet potato is known to contain flavonoids, a class of natural polyphenols with the capability to mitigate skin pigmentation. The research aims to assess the antioxidant and tyrosinase inhibitory activities of purple sweet potato extracts obtained through hexane, ethyl acetate, ethanol, and water solvents. The results of phytochemical identification show that the extract contains various secondary metabolites which have the potential to act as antioxidants. The DPPH method IC50 values of 6948.12ppm for n-hexane, 3015.19ppm for ethyl acetate, 128.05ppm for ethanol, and 791.77ppm for water extract. The MDA inhibitor test IC50 values of 2067.02ppm for n-hexane, 1968.13ppm for ethyl acetate, 116.14ppm for ethanol, and 921.14ppm for water extract. In the tyrosinase inhibitor assay, IC50 values were 1328.29ppm for n-hexane, 1245.13ppm for ethyl acetate, 217.35ppm for ethanol, and 391.21ppm for water extract. Tuckey test statistics, ethanol extract was not significantly different from the positive control in the DPPH test, MDA inhibitor and tyrosinase test. These findings suggest that purple sweet potato extracts, particularly the ethanol extract, hold promise as natural ingredients with antioxidant and tyrosinase inhibitory properties, making them potential candidates for safe and effective skin brightening formulations.

Abstract

Tamanu or nyamplung oil, derived from Calophyllum inophyllum L., is commonly used in traditional medicine and occasionally used as a moisturizer in skincare cosmetics. In silico predictions suggested that tamanu oil contained compounds capable of inhibiting tyrosinase activity. This study aimed to measure the surface tension of tamanu oil and evaluate its tyrosinase inhibitory activity in vitro. The surface tension of tamanu oil was measured using the du-Nouy ring method with a tensiometer, while tyrosinase inhibitory activity was assessed by spectrophotometry using a microplate reader. The study revealed that the surface tension of tamanu oil was measured at 41.83±0.76 mN/m, and the tyrosinase inhibitory activity of tamanu oil was determined to be 83.75±0.41%. Notably, the inhibitory activity of tamanu oil was comparable to that of the positive control, kojic acid, which exhibited a tyrosinase inhibition of 84.59±2.04%. We concluded that tamanu oil has potential as a natural surfactant raw material and may serve as an effective tyrosinase inhibitor.   Keywords: Calophyllum inophyllum L., nyamplung, surface tension, tamanu oil, tyrosinase inhibitor

Abstract

Green spinach (Amaranthus tricolor L.) is a plant that contains various vitamins, such as vitamins A, B, and C, as well as protein and fiber. Green spinach contains high iron. Besides that, spinach also contains secondary metabolites such as flavonoids, antioxidants, and phenolics, which are beneficial for degradative diseases. This study aimed to evaluate the impact of plant height, iron content, total phenolic content, and antioxidant activity in spinach plants using a 70% ethanol solvent. The samples were extracted through a maceration method in a microwave for 3 minutes, followed by filtration to obtain the test sample filtrate. Iron content was determined using the ICP-OES method, total phenolic content (TPC) was measured with the Folin-Ciocalteu method, and antioxidant activity was assessed using the DPPH method. The findings revealed that treatment 2 (2 parts maggot fertilizer to 8 parts soil) had the highest iron content and antioxidant activity, whereas treatment 1 (1 part maggot fertilizer to 9 parts soil) showed the greatest plant growth and phenolic content.
