Abstract

Coronary heart disease is a serious health problem and it is known to be lethal. The disease can be triggered by the accumulation of cholesterol plaques in the walls of blood vessels This cholesterol formation is assisted by methylglutaril-CoA reductase (HMG-CoA reductase) enzyme. Red betel contains active compounds that have the potential to act as HMG-CoA reductase inhibitor. This study aimed to predict the potential of ethanol extract compounds, water fraction, ethyl acetate fraction, and red betel n-hexane fraction as HMG-CoA reductase inhibitors by in vitro method. The in vitro method which consisted of extraction by maceration, graded fractionation with n-hexane, ethyl acetate, and water, continued by thiobarbituric acid (TBA) method to see the potential antioxidant activity of the sample. The Result showed that the highest inhibition of HMG-CoA reductase is 94% was found in the concentration 200 ppm ethyl acetate fraction. Also antioxidant activity of red betel showed by the inhibition of malondialdehyde formation had the highest activity in the ethyl acetate fraction than other fractions used.

Abstract

Robusta coffee (Coffea canephora) is one type of plant that has long existed in Indonesia and has high economic value. Robusta coffee beans have the potential to be developed to reduce caffeine levels by treating them with lactic acid bacteria. This study aims to determine the metabolite profile of Robusta coffee from several samples treated with Lactobacilus plantarum bacteria. Samples were taken from the West Java location at the Patani Coffee Plantation. Sample extraction was carried out using the Ultrasonic Assisted Extraction (UAE) method with 96% ethanol. Metabolite content analysis was carried out using UPLC-QTOF-MS/MS with a C18 stationary phase column (Okta Decyl Silica), a mixture of formic acid/water 0.1/99.9 (v/v) and formic acid/acetonitrile 0.1/99.9 (v/v) mobile phases. The results of the analysis were interpreted using Masslynx software. The results showed that the caffeine content in coffee not fermented with Lactobacillus plantarum was 2.40%, coffee fermented with L. plantarum bacteria for 6 hours had an abundance% of 1.76%, and coffee fermented with L. plantarum bacteria for 24 hours had an abundance% of 2.38%. LC-MS/MS analysis identified compounds 35,26, and 25 in three Robusta coffee samples. These results were not significantly different from the fermentation of Lactobacillus plantarum in Robusta coffee beans

Abstract

Graphical Abstract    Highlight Research Feed formulations based on black soldier fly (BSF) larvae and tofu waste meet the quality requirements based on the Indonesian National Standard Feed formulations based on black soldier fly (BSF) larvae and tofu waste improve fish growth performance comparable to commercial feed Feed formulations based on black soldier fly (BSF) larvae and tofu waste did not increase the level of cholesterol content in fish flesh Fish flesh with black soldier fly (BSF) larvae and tofu waste diet have higher total fatty acid content compared to commercial feed     Abstract Despite growing interest in sustainable aquaculture, studies on the combined use of Black Soldier Fly Larvae (BSFL) and tofu waste as alternative protein sources in tilapia diets, particularly their effects on fatty acid profiles, remain limited. This study evaluates the effects of fish feed formulated with BSFL and tofu waste on the growth performance, proximate composition, and fatty acid profile of red tilapia (Oreochromis niloticus). Experimental diets included: commercial feed, dried BSFL, and a combination of BSFL:tofu waste. Fish were reared for eight weeks under controlled conditions. Treatment with BSFL:tofu waste exhibited superior growth performance, with the highest weight gain (102.88%) and length gain (33.75%). Proximate analysis revealed significant variations across treatments, with a combination of BSFL:tofu waste achieving the highest caloric value (110.71 cal/g) and fat content (3.75%) while maintaining protein levels comparable to commercial feed. Fatty acid analysis showed a combination of BSFL:tofu waste enhanced total fatty acid content (77.92%), predominantly polyunsaturated fatty acids (PUFAs), increasing from 37.34% in commercial feed to 47.04%. Essential fatty acids, including DHA and linoleic acid, were elevated in a combination of BSFL:tofu waste, indicating improved nutritional quality. Cholesterol levels remained unaffected among treatments. These findings suggest that combining BSFL and tofu waste can enhance fish feed quality, optimize growth performance, and improve the nutritional value of red tilapia. Future research should explore refining feed formulations to maximize nutrient stability and sustainability for broader aquaculture applications.

Abstract

Physalis angulata was known for its pharmacological potential, including antioxidant activity, yet the specific bioactive compounds responsible for enhancing endogenous antioxidant enzymes, such as superoxide dismutase 1 (SOD 1), remained unclear. This study aimed to identify and evaluate the active compounds of P. angulata as natural SOD 1 activators using an In silico molecular docking approach. Forty-nine ligands derived from the active constituents of P. angulata were screened based on Lipinski’s Rule of Five and ADMET properties. Molecular docking was performed using the SOD 1 protein (PDB ID: 5YTO), and docking validation yielded an RMSD value of 0.005 Å. Among the test ligands, withanolide exhibited the most favorable binding energy (−7.011 kcal/mol) and the lowest inhibition constant (7.0 pM), forming strong interactions with key catalytic residues of the enzyme. These findings indicated that withanolide had promising potential as a natural SOD 1 activator, providing a basis for future antioxidant drug discovery.
