Abstract

Farming in Cibitung Tengah Village, Tenjolaya Subdistrict, Bogor District, West Jawa is characterized by vegetables and sheep farming. Several problems in farming are the use of chemical fertilizers, management of agriculture and domestic wastes properly; even some people disposing of garbage in the river and also, the institutional Farmer Group Association (Gapoktan) and Female Farmer Group (KWT) are not functioning optimally. The use of an integrated farming system can increase land and livestock productivities and reduce the volume of agriculture and domestic wastes all at once. This community empowerment activity aimed to increase the people's knowledge and skill of Cibitung Tengah Village in terms of processing agricultural and domestic wastes. Thus it will overcome the garbage and overuse of chemical fertilizer problems. The discussions and outreach activities were done by a door-to-door approach, especially for people who have not been actively involved in Gapoktan and KWT. This research was carried out with several educational activities such as counseling on community waste management systems, training on organic waste processing, Black Soldier Fly (BSF) training, processing organic waste with BSF technology, making silage as goat feed, and making branding of agricultural products. This empowerment program produced new products or processed products. The products will increase the income for the people of Cibitung Tengah, such as maggot processing products, CS-Fresh products, results from waste banks, and handicraft products from waste.

Abstract

This study aimed to isolate the endophytic fungi of soursop leaves that have the potential as anti-yeast. Soursop leaves were collected from 3 locations in West Java, Indonesia. The secondary metabolites produced by the endophytic fungi were extracted with ethyl acetate and tested for anti-yeast activity against Saccharomyces cerevisiae and Candida tropicalis. Profile of extracts contents characterization was carried out by using thin-layer chromatography (TLC). Twelve monoculture soursop leaves endophytic fungi isolates from Sukabumi (3 isolates), Cianjur (3 isolates), and Garut (6 isolates). It showed different abilities to inhibit C. tropicalis and S. cereviseae. The best extract is isolat from Cianjur (Sir-C1) and Sukabumi (Sir-S1). The stain patterns seen between the nine endophytic fungi extracts were similar. This indicates that the compounds in them may also be the same. But the extracts have a different absorbance for anti-yeast activities. This indicates that the total metabolites of each isolate are different. TLC results showed that ethyl acetate extraction produced almost the same metabolite components from nine endophytic fungi extracts, and all extracts had an anti-yeast activity with various absorbance. There are differences in the ability of 12 soursop leaf endophytic fungi to inhibit yeast model C. tropicalis and S. cerevisiae. Some extracts have similar TLC profiles, but their anti-yeast activity is not similar

Abstract

Belimbing wuluh merupakan tanaman jenis buah dan obat tradisional. Tanaman belimbing wuluh sudah sering dimanfaatkan masyarakat salah satunya untuk mengobati penyakit seperti batuk dan radang rektum. Tujuan penelitian ini adalah menguji senyawa fitokimia, menganalisis kandungan total fenolik dan flavonoid, serta aktivitas antioksidan dan antiinflamasi secara in vitro pada ekstrak etanol daun belimbing wuluh. Metode yang dilakukan pada penelitian ini adalah ekstraksi daun belimbing wuluh, skrining fitokimia, perhitungan total fenol dan flavonoid, uji aktivitas antioksidan, dan uji aktivitas antiinflamasi. Senyawa fitokimia yang terkandung pada ekstrak etanol daun belimbing wuluh adalah saponin, tanin, steroid, flavonoid, dan alkaloid. Kandungan total fenolik dan flavonoid ekstrak etanol daun belimbing wuluh secara berturut-turut sebesar 39,03 dan 97,28 µg QE/mg. Ekstrak etanol daun belimbing wuluh memiliki aktivitas antioksidan yang tergolong sangat kuat, sementara aktivitas antiinflamasinya terutama ditunjukkan pada konsentrasi ekstrak 200 µg/ ml, memiliki nilai persen inhibisi hemolisis yang paling tinggi. Kesimpulannya, ekstrak etanol daun belimbing wuluh dapat menjadi salah satu sumber antioksidan dan antiinflamasi alami.Ethanol Extracts of Averrhoa Bilimbi Leaf Demonstrated Antioxidative and Anti-inflammatory ActivityAbstractAverrhoa bilimbi, a fruit-bearing plant, has been traditionally used for medicinal purposes such as treatment of cough and rectal inflammation. In this current work, ethanol extract of Averrhoa bilimbi leaf was analyzed, with regard to phytochemical composition, i.e. total phenolic, and flavonoid, antioxidant activity, and in vitro antiinflammation activity. Extraction of carambola leaves, phytochemical screening, total phenolic and flavonoid contens, assay of antioxidant activity, and assay of anti-inflammatory activity were done in this research. As a result, the leaf extract positively contained some phytochemical compounds, i.e. saponin, tannin, steroid, flavonoid, and alkaloid. Furthermore, total phenolic and flavonoid of the leaf extract was found at 39.03 and 97.28 µg QE/mg extract, respectively. Additionally, antioxidant activity of the leaf extract was classified as very strong, while its anti-inflammatory feature at extract concentration of 200 µg/ml exhibited the highest inhibition of hemolysis. In summary, the ability of Averrhoa bilimbi leaf ethanol extracts to act as antioxidative and anti-inflammatory agents was determined and this may open the use for natural antioxidant and anti-inflammatory agents.

Abstract

Melasma is a skin disorder characterized by hyperpigmentation of the skin in areas exposed to sunlight and is included in the dominant skin disorder category in several countries. Meanwhile, data from the Food and Drug Administration shows that cosmetic products have progressed from 0.01% to 57.4% in terms of registered products over the last six years. Currently, melasma treatment and cosmetic applications as a whitening agent were carried out by regulating tyrosinase activity through the use of compounds such as kojic acid which poses a health risk. Therefore, it was necessary to have other alternatives that can regulate tyrosinase activity without harming health. This study aimed to determine inhibitory effects of wungu leaf (Graptophyllum pictum (L) Griff) bioactive component as an agent to inhibit tyrosinase activity through a molecular docking approach. A total of 28 wungu leaves bioactive components as a result of the analysis of Gas Chromatography-Mass Spectrometry (GC-MS) and Gas Chromatography-Flame Ionization Detector (GC-FID) were tested for their inhibitory effects on tyrosinase activity. Linolenic acid and phytol were ligands that have the best inhibition on tyrosinase enzyme activity with affinity energies of -5.8 kcal/mol and -5.4 kcal/mol, respectively, and inhibition constants of 55.718 M and 109.489 M.

Abstract

Cholesterol plaque buildup in artery walls occurs due to oxidation of Low-Density Lipoprotein (LDL) molecules by free radicals, which are a risk factor for coronary heart disease. Piper crocatum contains active compounds that can act as HMG-CoA reductase inhibitors, such as flavonoids, alkaloids, polyphenols, tannins, and essential oils. This study aimed to predict the potential of Piper crocatum extract and fraction compounds as HMG-CoA reductase inhibitors by investigating the ligand affinity to the HMG-CoA reductase enzyme. Ligand and receptor preparation was conducted using BIOVIA Discovery Studio Visualizer v16.1.0.15350 and AutoDock Tools v.1.5.6. Molecular docking used AutoDock Vina, while ligand visualization and receptor binding used PyMOL(TM) 1.7.4.5.Edu. The receptor used was HMG-CoA reductase (PDB code: 1HWK) with atorvastatin as a control ligand. Catechin, schisandrin B, and CHEMBL216163 had the highest inhibition with affinity energies of -7.9 kcal/mol, -8.2 kcal/mol, -8.3 kcal/mol, respectively. Amino acid residues that played a role in ligand and receptor interactions were Ser684, Asp690, Lys691, Lys692.

Abstract

Traditional medicines that can be used as α-glucosidase inhibitors are turmeric (Curcuma longa L.), black tea (Camellia sinensi L), and ginger (Zingiber officinale). It is necessary to research the use of a combination of tea, turmeric, and ginger in inhibiting the α -glucosidase enzyme. This study aims to determine the optimal combination of extracts of turmeric, black tea, and ginger in inhibiting the α-glucosidase enzyme. The research carried out was the measurement of IC50 values for the inhibition of α-glucosidase of each extract, and the inhibition of the enzyme α-glucosidase in combination. The results of the test of ethanol extract of turmeric, ginger and black tea water extract obtained the inhibition value of α-glucosidase with IC50 values of 9.48±0.05 g/mL, 66.64±0.44 g/mL and 9.52±0.25, respectively. F7 is a combination of turmeric, ginger and black tea which produces the highest α-glucosidase inhibition, which is 67.86±0.93%.

Abstract

Cancer degenerative disease is a serious problem in Indonesia, caused by free radicals, and can be prevented by antioxidant compounds. Superoxide dismutase (SOD) is the strongest enzyme that acts as a protective organism against oxidative stress. The active compound of red betel can be studied in silico as an activator of the SOD enzyme. Molecular docking visualization, physicochemistry, toxicity of red betel active compounds were studied and continued with molecular docking. The studied parameters were Gibbs free energy (ΔG) and receptor-ligand chemical bonding analysis. The test results show that the active compounds of red betel 6-Amino-2-(3-aminopropylamino)-9-(cyclohexylmethyl)- 2,3,4,5,6,7-hexahydro-1H-purin-8-one was well-absorbed dan safe for consumption with Gibbs free energy value of -9.1490 kcal/mol. Hydrogen and hydrophobic interactions of these compounds were similar to the control ligand, namely β-amyrin and Trehalose. This study concluded that the active compound of red betel 6-Amino-2-(3-aminopropylamino)-9-(cyclohexylmethyl)- 2,3,4,5,6,7-hexahydro-1H-purin-8-one has the best potential to increase the work of Cu/Zn-SOD enzymes. This compound also meets the requirements of Lipinski regulations so that it can be well absorbed orally by the body and is not carcinogenic

Abstract

Endophytic fungi have been reported to promote plant growth and resistance to biotic and abiotic stresses. This study aimed to determine the effect of endophytic fungi application isolated from the sengon (Falcataria moluccana) seeds on the growth of sengon. The seed-borne endophytic fungi were applied when the session was three weeks old by sprinkling each suspension of endophytic fungi (103 to 106 spores/ml) on the soil around the plant roots. The experimental parameters evaluated were height, diameter, and total wet and dry weight. This study showed that endophytic fungi treatment has no significant effect on the stem diameter, total wet weight, and water content; but significantly affected the height and total dry weight. Cladophialophora sp. and Ascotricha sp. fungi increased the height growth of sengon, while Aspergillus sp. fungi increased the total dry weight against the control. Ochroconis sp. treatment was not significantly different from the control. The shoot-root ratio values range from 1.13-1.43, indicating that the seeds already have roots supporting plant growth. The Dickson quality index range from 0.65-1.03 suggested that the seedlings have good biomass distribution. Abstract

Inulin is a polysaccharide with linear fructan chain structure. Food industries widely use inulin as a low calorie sugar substitutions. Yacón (Smallanthus sonchifolius) tuber that are easily grown and contain higher inulin can be explored as local inulin sources. This research aimed to characterize the chemical properties of inulin isolated from yacón tuber. The methods of this study include proximate analysis of yacón tuber, inulin isolation, and characterization of purified inulin using FTIR. The results showed that yacón consisted of 91.23 water (analyzed as moisture), 0.12 proteins, 0.58 fats, 0.52 crude fibers, and 7.34% carbohydrates. The yield of inulin extracted from yacón tuber was 4.86% whereas its purity and actual content of inulin were 44.23 and 2.15%, respectively. The Osazon test revealed that the isolated inulin has similar crystalline with that from chicory. Based on characterization using the FTIR spectrophotometer, the isolated inulin had functional groups of C-O, C-H, CH2, O-H, and C=O. The spectrum of the isolated inulin has similarities with chicory inulin. Therefore, yacón tuber could be considered as a potential local inulin source in Indonesia.

Abstract

Obat adalah bahan yang digunakan untuk pencegahan, pemulihan, maupun peningkatan kesehatan manusia. Obat flu dan batuk dalam sediaan sirup harus memenuhi persyaratan kualitas obat sebelum diedarkan ke pasaran. Stabilitas farmasi merupakan salah satu kriteria yang amat penting untuk suatu hasil produk obat yang baik. Ketidakstabilan produk obat dapat mengakibatkan penurunan kualitas dengan hilangnya khasiat obat dan menjadi toksik atau beracun. Beberapa zat berkhasiat yang terdapat dalam obat flu dan batuk sirup adalah parasetamol dan amonium klorida. Parasetamol digunakan sebagai analgesik (pereda nyeri) dan antipiretik (penurun demam). Amonium klorida sebagai obat batuk bekerja dengan cara mengiritasi mucosa bronchial (mengencerkan dahak) sehingga mudah dikeluarkan. Jalur utama degradasi parasetamol yang menyebabkannya tidak stabil adalah peristiwa hidrolisis membentuk 4-aminofenol yang bersifat toksik bagi tubuh. Kadar parasetamol dan batas ketidakmurnian 4-aminofenol ditentukan dengan HPLC, dan kadar amonium klorida   ditentukan dengan potensiometer. Kadar parasetamol dalam sampel obat flu dan batuk sirup adalah 102.06%, kadar 4-aminofenol dalam sampel obat batuk sirup adalah 0%, dan kadar amonium klorida dalam sampel obat batuk sirup adalah 108.56%. Berdasarkan hasil tersebut, sampel obat batuk sirup memenuhi persyaratan yang telah ditetapkan Farmakope Indonesia V tahun 2014 dan United States Pharmacopeia (USP) 41 tahun 2018.
