Abstract

The use of natural ingredients in dental care, such as manginang which has become part of the Minang community culture, is considered an alternative dental care that must be created to overcome the negative impacts of dental care made from artificial materials. The core of this study is to change the manginang tradition towards logical verification through screening of bioactive compounds of gambir leaves (Uncaria gambir Roxb.) as antibiofilms against Streptococcus mutans. This study was conducted using an in silico strategy approach with the help of computer programs such as MOE v.09 and Chimera 1.13.1, as well as the Way2Drug site, protein information bank, and protox_ii. This study began with the collection of ligand information from the writing, followed by transformation into 2D and 3D designs. Other antibiofilm actions were decided using the QSAR procedure through the website (http://www.way2drug.com/PASSOnline/predict.php). The selected antibiofilm compounds were then followed by observing their affinity for the receptor (pdb id. 3AIB) using an atomic docking strategy. Finally, the ligand damage profile was observed to anticipate. The results of this study indicate that of the 41 identified gambir leaf compounds, there are 20 compounds that have the potential to have antibiofilm effects with quinnic acid having the highest score (Pa 0.703), while the compound with the strongest affinity for the receptor is chlorogenic acid with an affinity score (Gbinding) of -16.03 kcal/mol and an average of low toxicity (level IV-VI). However, of the 20 bioactive compounds of gambir leaves, only 5 compounds are predicted to have the same potential as the -maltose control.

Abstract

Ovariectomy is a commonly performed medical procedure to investigate the effects of estrogen loss on the immune system. Secretory immunoglobulin A (s-IgA) is an antibody that is important in mucosal defense system. Probiotics, such as Lactobacillus, have been known to affect the local immune system. This study aims to evaluate the effect of administering Lactobacillus probiotics on s-IgA levels in Wistar rats undergoing ovariectomy. Female Wistar rats (n=30) were randomly divided into two groups, namely the treatment group (receiving Lactobacillus probiotics) and the control group (receiving placebo). After undergoing an ovariectomy, treatment was given for 4 weeks. After the treatment period, s-IgA levels were evaluated using the ELISA technique in mouse saliva samples. The results showed that the treatment group receiving Lactobacillus probiotics had significantly higher s-IgA levels than the control group (p 0.05). Administration of Lactobacillus probiotics can increase s-IgA levels in Wistar rats undergoing ovariectomy. This shows the potential of probiotics in enhancing mucosal immune responses in conditions associated with estrogen deficiency

Abstract

Bladder stones represent a significant and increasingly common health issue in captive green iguanas (Iguana iguana), often resulting from improper husbandry practices. This case report examines the diversity and characteristics of bladder stones in five green iguanas (three females, two males) presenting with abdominal enlargement and decreased appetite. Surgical removal (cystotomy) was performed, followed by detailed morphometric and chemical analyses of the extracted stones. The stones varied in size (15.3655.01 g), shape, and color (white, cream, and dark brown), yet chemical composition was consistently dominated by calcium oxalate (5260%), tricalcium phosphate (2328%), and uric acid (1012%). Morphometric analysis showed low variation (50%) across measured parameters, indicating homogeneity among the samples. Notably, lighter iguanas tended to harbor heavier stones, suggesting a correlation with disease progression rather than sex or stone color. These findings reinforce the role of dietary imbalances, dehydration, and the anatomical peculiarities of the iguana urinary system in stone formation. Early detection, appropriate diet, and optimal hydration are critical to prevent urolithiasis. This report contributes to the limited data on iguana urolithiasis in Indonesia and highlights the need for broader studies to explore preventive strategies.

Abstract

Pathogenic Escherichia coli is a type of bacteria that has the ability to cause disease in hosts, including humans and animals, although in general E. coli acts as a normal flora in the digestive tract. This study aimed to detect the presence of pathogenic E. coli in the intestines of ducks sold at Lambaro Market, Aceh Besar. E. coli is a normal flora bacteria in the digestive tract of animals and humans, but some strains are pathogenic and can cause zoonotic diseases. The presence of pathogenic E. coli in poultry products has the potential to be a source of infection for humans through consumption of contaminated food. This study used 15 duck intestine samples obtained from cecum, to identify the presence of pathogenic E.coli. The results showed that 13 samples produced clear colonies on Sorbitol MacConky Agar (SMAC), indicating the inability of bacteria to ferment sorbitol, which correlated to the presence of pathogenic E. coli. In contrast, two samples showed pink colonies indicating the presence of nonpathogenic E. coli. The conclusion of this study was the cecum of ducks sold at Lambaro Market, Aceh Besar positive for pathogenic E. coli.

Abstract

Bulbus oculi prolapse is a condition characterized by the protrusion of the eyeball from the eye socket, which can be caused by various factors such as trauma, elevated intraocular pressure, and abnormalities in the eye muscles. This case study aimed to examine the treatment of bulbus oculi prolapse in domestic cats. A female calico cat named Mici presented with a chronic left eye injury that had become infected, leading to inflammation and persistent ocular discharge. The cats body weight was 0.7 kg, and its temperature was 38.2C. Surgical treatment was performed using an enucleation technique. The procedure involved an incision along the mid-anterior eyelid, extending to the lower eyelid. Dissection was carried out to expose the bulbus oculi, which was then ligated and excised. The eyelid margins were sutured using simple interrupted sutures. Postoperatively, the cat was administered antibiotics and antihistamines.

Abstract

Mucosa surface is the body's initial barrier against pathogens, rely on secretory Immunoglobulin A (s-IgA) for localized immunity. This study investigated the impact of Etlingera elatior extract on s-IgA levels and mucosal fluid secretion in mice challenged with Staphylococcus aureus. The research method used an experimental design with seven treatment groups, i.e negative control (P1), positive control (P2), and five groups with E. elatior extract administration at concentrations of 40% to 80% (P3-P7). Data were analyzed using ANOVA and Tukey's subset test to determine differences between groups. The results showed an increase in s-IgA levels as the extract concentration increased, with the highest level at P7 (1842.25 ng/mL). Conversely, mucosal fluid secretion decreased, with the lowest level at P7 (21.5 L). Statistical analysis showed significant differences between treatment groups (p0.05). The increase in s-IgA levels and normalization of mucosal fluid secretion are thought to be caused by bioactive content such as flavonoids and alkaloids which have immunostimulant and anti-inflammatory properties. The study revealed that E. elatior extract is effective in increasing s-IgA levels while controlling mucosal fluid secretion in mice infected with Staphylococcus aureus. These findings suggest the potential of E. elatior extract as a natural potential immunostimulant to support the treatment of mucosal infections.

Abstract

The TP53 gene encodes the p53 protein, often referred to as the guardian of the genome due to its critical role in maintaining genomic stability and preventing tumorigenesis. Under normal conditions, p53 expression is tightly regulated by MDM2 and MDMX, which promote its degradation through ubiquitination. In response to endogenous or exogenous stress, this ubiquitination process is inhibited, leading to the stabilization of p53. Once stabilized, p53 forms a tetrameric complex in the nucleus and binds to DNA. It then activates the transcription of genes involved in cell cycle arrest, DNA repair, apoptosis, and senescence, aiming either to repair DNA damage or eliminate cells when the damage is irreparable. Nearly half of all cancer cases involve mutations in the TP53 gene. These mutations may include missense, nonsense, inframe, and splice-site mutations. Mutations in TP53 result in the production of mutant p53 (mutp53) proteins. These mutations can lead to a loss of tumor suppressor function or confer gain-of-function properties that promote tumor progression. Given its central role in cancer development, TP53 is considered a promising therapeutic target. Potential strategies include reactivating suppressed p53, restoring the function of mutp53, or inducing its degradation.

Abstract

Corneal ulceration is a frequent ophthalmic disorder in domestic cats that can cause ocular discomfort, visual impairment, and serious complications if left untreated. Conventional medical management often provides limited support for epithelial regeneration, leading to the need for alternative therapeutic options. This case study reports the clinical outcome of using autologous serum eye drops (ASEDs) in the treatment of corneal ulceration in a domestic cat. A cat presented with severe epiphora, conjunctival hyperemia, and a centrally located corneal ulcer confirmed by fluorescein staining. Autologous serum was prepared by weekly collection of 6 mL of blood from the dorsalis antebrachii vein, followed by centrifugation and aliquoting into sterile eye drop bottles stored at 4 C. The serum was applied topically twice daily, accompanied by supportive therapy consisting of vitamin A supplementation, multivitamin injections, and an Elizabethan collar to prevent self-trauma. Clinical scoring (03) was used to monitor epiphora, conjunctival hyperemia, and corneal opacity over the treatment period. Marked improvement was observed within 14 days: epiphora and conjunctival hyperemia resolved completely, while corneal opacity improved from moderate to mild, with fluorescein staining becoming negative, indicating complete epithelial closure. No adverse effects were recorded during therapy. This case highlights the potential of autologous serum eye drops as a safe and effective adjunct treatment for feline corneal ulceration, warranting further investigation in larger cohorts to establish standardized protocols for veterinary ophthalmology

Abstract

Scabies is an infectious disease which is caused by Sarcoptes scabiei mites and considered as an emerging/re-emerging parasitic disease. Recent study using the Polymerase Chain Reaction (PCR) method has a higher sensitivity for detecting mites than using the skin scraping method. This study aims to detect the Second Internal Transcribed Spacer (ITS-2) encoding gene of S. scabiei. ITS-2 is a highly conserved area and always used to identify and differentiate host species infected by S. scabiei. S. scabiei isolates were obtained from skin scrapings from stray cat suspected of being infected with Scabies from Pasar Induk Bondowoso, Pasar Tamanan and Pasar Pujer. Detection on the presence of S. scabiei was carried out microscopically and then confirmed by molecular examination using the PCR method and visualized by electrophoresis on 2% agarose gel. The Polymerase Chain Reaction (PCR) results showed DNA bands at 304 bp Based on the result it can be concluded that samples are positively infected by Scabies can be detected by the ITS-2 encoding gene of Sarcoptes scabiei.
