Abstract

Clarkeinda trachodes is an interesting agaric and has been acknowledged as one of the poisonous mushrooms worldwide. This species has mentioned to be found in tropical regions of Asia, including Indonesia. However, no clear record has been found regarding the primary information and herbarium collection of this species in Indonesia. During our regular mushroom foraging at IPB University campus forest Indonesia, the basidiomata of Clarkeinda was encountered. The current study aimed to confirm the taxonomical position of our specimens based on morphological and molecular evidence. Fresh basidomata was observed on the macro- and micromorphological features. The molecular analysis and phylogenetic tree construction were done based on ITS 1/2 sequence. The combination of morphological and molecular evidence confirmed our specimen as C. trachodes. Morphologically, Clarkeinda trachodes BO24637 can be determined by a large basidiocarp of Agaricales, prominent pellicle on disc pileus, notable ring, reddish brown context and stipe when injured, and truncate apex of basidiospores. The BLAST result showed that our specimen has a high similarity to C. trachodes as the top hits. In addition, the phylogenetic tree constructed by ITS sequence committed specimen BO24637 as C. trachodes with 100% BS value. The current study resolves a clear and accessible document of C. trachodes in Indonesia. In addition, we provided the morphological and molecular information as well as herbarium collection of C. trachodes in Indonesia, which can be used for future study of this species.

Abstract

Abstract

Macrofungi are a nutraceutical food containing high protein, low fat, and are rich in minerals that play an important role in animal physiology, particularly as food supplements because of their distinctive aroma. This research aimed to provide information on the macroscopic and microscopic characters of several macrofungi from the Agaricomycetes consumed by animals, and to describe their antibacterial potentials. This research employed opportunistic sampling, mushroom identification and description, proximate analysis, and antibacterial assays. A total of seven mushrooms of the Agaricomycetes were described, namely Lentinus sajor-caju, Gymnopilus sp., Marasmius sp., Phallus indusiatus, Panus lecomtei, Termitomyces sp., and Schizophyllum commune. Lentinus sajor-caju had carbohydrate, fiber, protein, fat, ash, and moisture contents of 72.02%, 30.03%, 11.63%, 1.37%, 4.26 %, and 10.74%, respectively. The antibacterial assay of L. sajor-caju indicated that both 0.8 mg/ml and 1 mg/ml concentrations showed no significant difference in activity. However, both exhibited potent inhibitory effects against Candida albicans. Lentinus sajor-caju extract inhibited Enteropathogenic Escherichia coli (EPEC) at a concentration of 5 mg/ml. In comparison, a dose of 0.8 mg/ml effectively suppressed the growth of Methicillin-resistant Staphylococcus aureus (MRSA). This current research provided a basic framework for future mycophagy analysis and bioprospection in Indonesia.

Abstract

HIGHLIGHTS- Glowing mushrooms were found at night in the forest of IPB University and showed a bright green light in the dark.- Examination of their morphological features and phylogeny material confirmed they are the species Mycena chlorophos.- This is the first confirmed record of Mycena chlorophos in Indonesia, extending its known distribution worldwide. ABSTRACTA bioluminescent fungus produce light as a result of a chemical reaction within its body. The majority of fungi exhibiting bioluminescence are classified within the Mycenaceae which comprises a significant proportion of the known glowing fungi. One of those species which well noted worldwide is Mycena chlorophos. However, no clear record of M. chlorophos has been discovered in Indonesia. While engaging in our routine late-night mushroom foraging in IPB University Campus Forest (IPBUCF), some glowing basidiomata were encountered. This study sought to determine the taxonomic classification of the collected specimens through comprehensive morphological and molecular examinations. The fresh fruiting bodies were underwent thorough examination based on macroscopic and microscopic characteristics. Phylogenetic analysis of the rDNA-ITS region was conducted using ITS 1/2 primers, and the integration of morphological and molecular data verified that our specimens belong to Mycena chlorophos. By morphology, our specimen poses convex pileus with shallow depression at centre, viscid, shiny, adnexed lamellae with series of lamellae, ellipsoid and smooth basidiospores. The pileus and hymenophore glow vividly with a bright greenish hue in the darkness. The BLAST analysis indicated a 98% similarity between our specimen and Mycena chlorophos from China, ranking it among the top matches. Furthermore, the phylogenetic tree positioned our specimens within the M. chlorophos clade with a 100% bootstrap support value. Our specimen was in sub clade with materials from Malaysia. The DNA sequence of our scpecimen has been registered to Gen Bank and can be used for future reference of bioluminescent fungi wordlwide. This research updates information regarding the distribution and characteristics of  M. chlorophos in Indonesia. The discovery of this species in the IPBUCF adds to the known geographical range of M. chlorophos near the anthropogenic activity in Indonesia. In addition, our result also contributes to the growing documentation of bioluminescent fungi in Indonesia, a region with high but underexplored fungal biodiversity.
