Abstract

Stress condition has beeb reported to decrease intracellular antioxidant-superoxide dismutase(SOD) in the liver and kidneyof rats. This study was conducted to evaluate the antioxidant activies of ginger oleoresin on the profile of superoxide dismutase(SOD) in the kidney of rats under stress condition. The stress condition was achieved by five days of fasting together with swimming for 5 min/day. Ginger oleoresin was orally administrated in a dose of 60 mg/KgBW/day for seven days. Drinking water was provided ad libitum to all groups. The treatment of ginger oleoresin significantly decreased malondialdehyde (MDA) levels and increased SOD activity, as well as immunohistochemicall, increased the content of copper, zinc-SOD (Cu, Zn-SOD) in the kidney tissues compared to that of untreated group. The antioxidant content in ginger oleoresin such as shogaol, zingeron, and gingerol, etc. were shownto have activities in the kidney tissues of rats under stress condition that is increasunf the profile of SOD. Ginger oleoresin treatment in combination both before and after stress gave the best result. Key words : antioxidant, ginger oleoresin, kidney, stress, superoxide dismutase (SOD)

Abstract

Zingiber cassumunar Roxb known as Bangle, has antioxidative and antiinflammatory activities. It has been used for medicinal purposes traditionally. The aim of this study was to investigate whether administration of Z. cassumunar extracts orally may prevent acute liver damage induced by carbontetrachloride (CCl4) in rats. The study was carried out on 5 groups of male Sprague-Dawley rats (n=5). Group 1 was given ethyl acetate fraction of Bangle rhizome extract (30 mg/kg bw) (KS), group 2 was given dry juice of Bangle rhizome (30 mg/kg bw) (SK), group 3 was given curcuminoid (30 mg/kg bw) (KUR), group 4 as negative control (KN) given 5% Tween 80 solution (10 ml/kg bw) and Group 5 as control (K). Carbontetrachloride 0,1 ml/kg bw was given orally after 7 days to group KS, SK, KUR and KN. Rats were terminated 24 hours after CCl4-induction. Liver injury was evaluated by analyzed SGPT and SGOT activities from the serum and histopathologycal examination was conducted on the liver. The results clearly indicated that extracts of Z. cassumunar could reduced significantly the degree of liver damage induced by CCl4. It may be concluded that Z. cassumunar rhizome could be used as substance for hepatoprotector. Key words : Zingiber cassumunar, hepatoprotector, Liver damage.

Abstract

The present study was conducted to observe the role of antioxidant content of ginger oleoresin (Zingiber officinale) on inflammation in the kidney of rats under stress condition, as an anti inflammation. A total of sixty male Wistar rats were used for this study. They were divided into twelve groups; (1) control group, without treatment of both stress and oleoresin, (2) stress group, was treated by stress only, without oleoresin treatment, while (3) and (4) are groups that were treated by stress and then fed by standard feed for three and seven days, without oleoresin. Group (5) to group (12) were treated by stress and then followed by treatment of oleoresin for three and seven days. The doses of oleoresin were 20, 40, 60, and 80 mg/kgBW/day. Stress condition was done by 5 days fasting and swimming for 5 minutes/day, while drinking water was provided ad libitum. The highest number of inflammatory cells in the kidney of rats was observed in the stress group. The treatment of oleoresin after fasting stress showed decreasing of the number of inflammatory cells in the tissues. The decreasing rate was higher in the higher dose of oleoresin. The treatment groups that showed the number of inflammatory cells not significantly different from that of control group are treated groups receiving oleoresin 60 mg/kgBW/day for seven days, and 80 mg/kgBW/day for three and seven days, respectively. These results showed that ginger oleoresin has anti inflammatory effect in the kidney of rats kept under stress condition. Key words : Oleoresin, ginger, inflammation, kidney, rat, stress.

Abstract

The objectives of this experiment were to evaluate the effects of isoflavone-riched soybean flour with different treatment level of isoflavone on MDA level, SOD activity, and profile of Cu,Zn-superoxide dismutase (Cu,Zn-SOD) spermatogenic cells in the seminiferous tubules of male rat testes. Diet was given as isonitrogen and isocaloric with 10% of dietary protein. Twenty five males of Sprague Dawley rats were divided into five groups and treated with isoflavone-riched soybean flour by oral administration with different concentrations. The treatment was done for 2 months. The optimum concentration of isoflavone was 1.5 mg/day and resulted in : decreased level of MDA testes (2.29±0.05 nmol/g), mantained high SOD activity of rat testes (729.4±23.73 U/g), and increased content of Cu,Zn-SOD by immunohistochemical technique on spermatocytes and early spermatids. It was concluded that isoflavone-riched soybean flour diet (1.5 mg/day isoflavone) can prevent the formation of lipid peroxide and improve the antioxidant status of rat testes.

Abstract

The aim of the study was to observe the ability of indigenous probiotic lactic acid bacteria (LAB) Lactobacillus plantarum 2C12 and Lactobacillus fermentum 2B4 as antidiarrheal agent in rats infected by Enteropathogenic Escherichia coli (EPEC), and also to observe their effect as immunomodulator (malonaldehyde level and proliferation of lymphocyte cell). A total of 90 male Sprague Dawley rats were used for this study and divided into 6 groups i.e.: (1) Negative Control (not infected with of EPEC), (2) LAB L. plantarum 2C12, (3) LAB L. fermentum 2B4, (4) LAB L. plantarum 2C12 + EPEC, (5) LAB L. fermentum 2B4 + EPEC, and (6) Positive Control (infected with EPEC). The treatment of LAB was undertaken from 1st-21st day, while infection of EPEC using 106 cfu/ml per day was undertaken during 8th -14th day. Groups administered with LAB L. plantarum 2C12 + EPEC, LAB L. fermentum 2B4 + EPEC, and positive control, showed decreased body weight during 12th-21st day. At the 21st day, positive control group underwent acute diarrhea (fecal water content was 68.2 % b/b). Statistical analysis with Duncan Test showed that the treatment given to six groups of rats gave significant effect (p<0.05) toward PER value, number of lymphocyte cells and malonaldehyde level in liver and kidney of the rats.

Abstract

Purple Dioscorea alata (DA) tuber has health benefits due to its bioactive anthocyanins, which belong to polyphenolic group. Tuber is commonly made into flour to optimize its uses, however, the anthocyanins undergo significant degradation during processing because of the endogenous polyphenol oxidase activities. This research investigated factors that retain anthocyanins and polyphenols in the purple DA flour as well as its antioxidant capacity. The types of treatments during milling process should be taken into account; for instance, soaking in citric acid and blanching in order to preserve the bioactive compounds. To examine the inhibitory effects of acidic soaking and steam blanching on polyphenol oxidase activities, these experiments used four levels of citric acid (0, 0.25, 0.5, and 1%) and two levels of steam blanching time course (5 and 10 minutes). It was found that steam blanching for 5 or 10 minutes could reduce the activity of polyphenol oxidase, and consequently, retard the oxidation process and retain the polyphenolic compounds. Soaking the purple DA slices into a 1% citric acid solution followed by steam blanching for 10 min resulted in the highest total anthocyanins (104.36 mg/100 g), polyphenols (198.52 mg equivalent gallic acid/100 g), with an antioxidant capacity of 1.300 mg trolox equivalent/100 g. This study showed that the retention of bioactive compounds of DA tuber through soaking the tuber slices in solution containing inexpensive chemicals like citric acid at low concentrations, combined with 10 minutes of steam blanching resulted in flour containing total anthocyanins and phenolic as high as 44.51% and 62.58% of fresh tuber, respectively.

Abstract

Bawang dayak (Eleutherine palmifolia) is an indigenous plant in Borneo traditionally used by Dayak tribes to treat any kind of degenerative deseases including diabetes mellitus. The purpose of this research was to measure antioxidant and antidiabetic capacities of water and ethanolic extracts of bawang dayak bulb. Parameters evaluated in this research were phytochemical screening, total phenolics, flavonoid content, DPPH free-radical scavenging activity, and alpha glucosidase inhibiting (AGI) activity. The result showed that the total phenolics and flavonoid content in bawang dayak ethanolic extract (217.71 mg GAE/g and 65.35 mg QE/g) were higher than that of the water extract (139.93 mg GAE/g and 16.95 mg QE/g). The ethanolic extract also had higher antioxidant and AGI activities (IC50 112 and 241 ppm) than that of the water extract (IC50 526 and 505 ppm). In addition, the IC50 values for AGI in bawang dayak ethanolic extract was lower than acarbose which is known as a commercial antidiabetic agent.

Abstract

Tempe is a potential source of protein for 250 million people in Indonesia. Indonesian soybean demand reached 2.2 million tons per year, and 1.3 million tons of which are used for tempe production. Approximately 70% of the soybeans are imported from different countries. To improve national food security, it is important to use local soybean as a raw material. Therefore, this study used Grobogan local soybean as raw material. The purpose of this study was to evaluate the impact of the consumption of tempe (from local soybean Grobogan) in the long term, ie over a 90-day subchronic test using rats as a model. Parameters measured were serum biochemical profile, hematology, malonaldehyde (MDA), and superoxide dismutase (SOD) in liver and kidney organs of rats. This research was conducted using three groups of rats were fed tempe, boiled soybean, and casein as a source of protein in the ration. Data feed convertion efficiency showed tempe of Grobogan soybean had a better quality protein than that of boiled soybean and casein. As compared to the groups of rats fed boiled soybean and casein, the tempe group had higher HDL cholesterol, but not significantly different in hematological parameters, MDA and SOD in liver and kidney organs. This suggests consumption of tempe made from Grobogan soybean in the long term does not have a negative impact on health.

Abstract

Penelitian ini bertujuan untuk menganalisis potensi antioksidan kulit pisang kepok dan kulit pisang uli pada hati tikus hiperkolesterolemia. Kulit pisang kepok dan uli diekstraksi dengan metode maserasi menggunakan pelarut air, etanol 70% dan etanol 96%. Ekstrak diuji fitokimia secara kualitatif. Ekstrak diuji aktivitas antioksidanya menggunakan metode DPPH. Ekstrak dengan nilai IC50 terendah dipilih untuk uji in vivo. Desain penelitian pada uji in vivo meliputi pencegahan dan pengobatan. Uji in vivo dilakukan dengan memberikan varian ekstrak kepada kelompok tikus percobaan. Parameter yang diamati adalah aktivitas SOD dan kadar MDA organ hati. Ekstrak kulit pisang kepok memiliki rendemen 17,97% (pelarut air), 17,18% (pelarut etanol 70%) dan 15,02% (pelarut etanol 96%). Ekstrak kulit pisang uli memiliki rendemen 33,04% (pelarut air), 27,2% (pelarut etanol 70%) dan 34,42% (pelarut etanol 96%). Ekstrak kulit pisang kepok dan uli pada berbagai pelarut memiliki karakteristik warna coklat. Semua ekstrak mengandung senyawa flavonoid. Saponin terdapat pada semua ekstrak kecuali ekstrak kulit pisang uli dengan pelarut etanol 96%. Triterpenoid hanya terkandung pada ekstrak kulit pisang kepok dengan pelarut etanol. Ekstrak kulit pisang kepok dengan pelarut etanol 70% memiliki nilai IC50 terendah (P<0.01) dibandingkan dengan jenis ekstrak lainnya. Kelompok pencegahan dan kelompok pengobatan yang dicekok ekstrak etanol 70% kulit pisang kepok memiliki aktivitas SOD secara nyata lebih tinggi dan kadar MDA lebih rendah dibandingkan kelompok yang tidak dicekok ekstrak (P<0,05). Ekstrak kulit pisang kepok dengan pelarut etanol 70% menunjukkan aktivitas antioksidan yang paling baik. Ekstrak etanol 70% kulit pisang kepok menaikkan aktivitas superoksida dismutase dan menurunkan kadar malondialdehid organ hati tikus percobaan model hiperkolesterolemia baik sebagai pencegahan maupun pengobatan.

Abstract

Perhaps, the Javan Rhino (Rhmocerossondaicus) is the most endangered species among large animals in the world. Only two population of ~t are known in the world: one in Indonesia and another in l'ietnam. None exists in the zoos. The Javan Rh~no'sc aptivityis an alternative conse~vatione ffort to ensure their existence In the world. But it is imperative to give a correct immobilization, stable preparation such as in the natural forest. and also disease monitoring. Medication and preventive medicine for the Javan Rhino are vely important. Some diseases which have been encountered in the zoos are a.0.: helminthiasis. endocarditls, coliform d~arrheah. epat~tsb iliari.abscesses and slun lacerations can result in septicaemia. The sources of diseases In captivity can be some kinds of foods and water, animal keepers, equipment, contaminated place, rats and mice, birds and insects.
