Abstract

Control of plant diseases is usually carried out by the community using chemical pesticides. These chemical pesticides can damage the environment and human health. As an alternative, a technique for using fluorescent pseudomonad biological agents has been developed as an antagonistic, environmentally friendly microorganism to reduce the negative impact of using chemical pesticides. Community service has been carried out in Korong Kampung Guci Nagari Lubuk Pandan, Padang Pariaman regarding the application of compatible fluorescent pseudomonad biological agents. After the material was presented to the participants, it was continued with the application of compatible fluorescent pseudomonads to the tissue cultured rhizosphere of banana plants. All participants enthusiastically applied compatible fluorescent pseudomonads in the field.

Abstract

PPUPIK Mooghurt is a business unit that is part of the Entrepreneurship Unit of the Student Association majoring in Biology, FMIPA UNP. To improve the performance of the PPUPIK Mooghurt business unit, a more reliable marketing strategy is needed. The purpose of PKM PPUPIK Mooghurt: to find out the marketing capability of the KWU HMJ Biology FMIPA UNP unit team and offer solutions to their problems. The marketing ability of the PPUPIK Mooghurt team in general has not been satisfactory, so solutions are given to carry out training and socialize mooghurt products to the public. The results obtained were an increase in marketing capabilities for the PPUPIK Mooghurt team, and the product became better known in the community because teachers were also able to disseminate information to their students.

Abstract

Padi merupakan komoditas utama pertanian rakyat di Indonesia yang merupakan makanan pokok selain jagung dan sagu. Salah satu gizi yang terdapat pada beras adalah protein. Protein berguna untuk pertumbuhan, pemeliharaan sel dan pembentukan antibodi. Tujuan penelitian ini adalah mengetahui pengaruh lokasi tanam dan varietas terhadap kandungan protein beras padi sawah (Oryza sativa L.) varietas lokal Sumatera Barat. Penelitian menggunakan Rancangan Acak Kelompok Faktorial dengan menanam tujuh varietas padi lokal Sumatera Barat di empat lokasi berbeda di Sumatera Barat. Varietas yang digunakan adalah Ciredek, Anak Daro, Randah Putiah, Cantiak Manih, Mundam, Bakwan dan Sarai Sarumpun. Lokasi penanaman di Solok, Agam, Padang Pariaman dan Pesisir Selatan. Hasil penelitian diketahui bahwa interaksi antara lokasi tanam dengan varietas terhadap protein tidak berbeda nyata. Lokasi dan varietas berbeda nyata mempengaruhi kandungan protein beras padi sawah. Kandungan protein rata-rata beras di lokasi tanam yang tertinggi adalah Padang Pariaman sebesar 8,56%, dan terendah adalah Solok sebesar 5,99%. Varietas yang rata-rata kandungan protein tertinggi adalah Sarai Sarumpun sebesar 8,45%, dan terendah adalah Anak Daro sebesar 5,88%

Abstract

ABSTRAKPangan jajanan merupakan sarana bagi anak-anak dalam pemenuhan kebutuhan kecukupan gizi, tetapi juga berisiko menimbulkan keracunan jika tidak higienis. Menurut Undang–Undang RI nomor 18 tahun 2012 tentang pangan menyatakan bahwa keamanan pangan adalah usaha mencegah pangan dari kontaminasi biologis, kimia dan fisik. Laporan kasus kejadian luar biasa (KLB) yang dikeluarkan oleh BPOM tahun 2016 menyatakan bahwa 16,35% keracunan makanan di Indonesia berasal dari pangan jajanan di sekolah dan sebanyak 42,14%  berasal dari jajanan yang dibuat dari pangan rumah tangga. Tujuan penelitian ini adalah Untuk menguji kualitas  jajanan anak sekolah dasar secara mikrobiologi di Kecamatan Koto Tangah Padang. Penelitian ini bersifat deskriptif untuk mengidentifikasi mikroba patogen dalam jajanan yang di jual pada 19 Kantin Sekolah Dasar pada bulan Maret hingga April 2018. Total sampel yang dapat disampling sebanyak 49 sampel. Pengambilan sampel dilakukan pagi hari hingga menjelang jam istirahat siang. Sampel yang disampling dari kantin dibagi dalam tiga bentuk yaitu jajanan kering, jajanan basah dan minuman yang diberi es batu. Hasil penelitian dengan ditemukannya S.aureus pengkontaminasi pada 1 sampel (2,04%) dari jajanan kering dan E. coli sebanyak 3 sampel (6,12%) dari minuman yang diberi es batu, dan bakteri jenis lain sebanyak  91.82 %.Kata kunci:  Kualitas Mikrobiologi, Jajanan Anak Sekolah, Mikroba Patogen Pengkontaminan. ABSTRACTSchool Children Food (SCF) played an important role in providing nutrient intake of children nutritional  needs, in the other hand, it may has a risk of poisioning if unhygienic. According to the Law of Republic Indonesia number 18 of 2012 on food states that food security is an attempt to prevent food from biological, chemical and physical contamination. Report on Outbreaks issued by BPOM in 2016 states that 16.35% of food poisoning in Indonesia caused by contaminated SCF, and as much as 42.14% caused by contaminated of household SCF. The aim of this study is to test the microbiological quality of Elementay SCF in Sub-district Koto Tangah Padang. This descriptive research would like to identify pathogenic microbial in SCF which sold at 19 elementary school canteen during periode of time March to April 2018. The total samples are 49. Time of sampling conducted from morning until right before lunchtime. The samples categorized into three forms which are dried, wet and ice-cubed drinks. The result showed that one sample (2.04%) of dried SCF was contaminated by S. aureus, three samples (6.12%) of Iced-cube drinks was contaminated by E.coli and the rest (91.82%) was contaminated by other bacteria. Abstract

Nata de coco adalah sejenis makanan yang mengandung kadar serat tinggi, dan sudah dikenal luas oleh masyarakat. Di Indonesia, nata de coco sering disebut sari air kelapa atau sari kelapa. Nata de coco biasanya dikonsumsi berupa minuman segar yang ditambahkan ke dalam sirup, agar jeli, fruit coctail, dan lain-lain. Tujuan dari kegiatan pengabdian adalah dalam rangka memenuhi dan menggapai harapan dari panti asuhan Al Yusra Padang, yaitu memberikan pendidikan dan pelatihan-pelatihan pada anak panti. Pelatihan yang diberikan adalah pembuatan nata de coco, sejenis makanan yang mengandung kadar serat tinggi. Pelatihan pembuatan nata de coco dimulai dari pengenalan tentang bibit nata de coco, proses pembuatan nya, hingga pengemasan nata de coco menggunakan mesin pres gelas platik agar bisa di jual. Setelah mendapatkan pelatihan, peserta yang awalnya tidak mengetahui tentang nata de coco menjadi tahu. Peserta pelatihan baru mengetahui bahwa air kelapa merupakan bahan dasar pembuatan nata de coco. Setelah kegiatan berakhir, peserta pelatihan sangat antusias untuk mencoba membuat nata de coco secara mandiri. Kata kunci: Nata de coco, Serat, Air kelapa, Sari kelapa, Minuman segar ABSTRACT Nata de coco is a type of food that contains high fiber content, and is well known to the public. In Indonesia, nata de coco is often called coconut water extract or coconut juice. Nata de coco is usually consumed in the form of fresh drinks added to syrup, jelly, fruit coctails, and others. The purpose of this service activity is to fulfill and reach the expectations of the Al Yusra Padang orphanage, which is to provide education and training to orphanage children. The training was about making of nata de coco, a type of food that contains high fiber content. The training in making nata de coco starts with an introduction to nata de coco starter, the manufacturing process, to the packaging of nata de coco using a plastic glass pressing machine so that it can be sold. After receiving the training, participants who initially didn't know about nata de coco came to know about it. The training participants have just learned that coconut water is the basic ingredient for making nata de coco. After the activity ended, the training participants were very enthusiastic about trying to make nata de coco independently. Keyword: Nata de coco, Fiber, Coconut water, Coconut juice, Fresh drinks

Abstract

Jatropha curcas L. is one of the plants that can be developed as raw material of biodiesel. One of the efforts to increase the production of J. curcas is the propagation of stem cuttings horizontally. Efforts to stimulate root growth can be accomplished by adding Plant Growth Regulators (PGRs). PGRs can be obtained naturally or synthetically. Indole acetic acid (IAA) is one of PGRs, which is not only produced by plants but can also be produced by rhizobacteria. One such rhizobacteria is fluorescent pseudomonad. The objectives of the study were to know the response of growth of horizontal stem cuttings after being introduced with fluorescent pseudomonad. This research is an experimental research. Complete Randomized Design with 6 treatments and 4 replications. The treatments were soaking of J. curcas stem cuttings with fluorescent pseudomonad isolate suspension PfPj3 suspension concentration of 0% (without suspension), 5%, 10%, 15%, 20%, and 25%. Observation of the research is the beginning of the shoots, the number of shoots, and the length of the J. curcas stem shoot. The data obtained were analyzed using Anova and DNMRT significant level of 5%. The results showed that horizontal stem cuttings of J. curcas with fluorescent pseudomonad suspension influenced early shoot, shoot number, and shoot length. A 10% fluorescent pseudomonad concentration can cause shoots to appear early, while a concentration of 20% causes the largest number of shoots.

Abstract

The purpose of this study was to determine the potency and optimum pH of AAS thermophilic bacteria isolates in producing bioethanol. This research was carried out from September 2021 to December 2021, at the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Padang State University. This research is an experimental study consisting of 6 treatments and 3 replications with variations in incubation pH of 6.5, 7, 7.5, 8, 8.5, and 9. The data obtained were analyzed by means of variance (ANOVA) with Duncan's further test. New Multiple Range Test (DNMRT). The results showed that the isolates of thermophilic bacteria SSA had the potential to produce bioethanol with isolates SSA 8 being the producer of bioethanol with the highest concentration of 1.001%, and variations in incubation pH affected the concentration of bioethanol produced with the optimum pH of thermophilic bacteria in producing bioethanol, namely pH 8 which produced the highest concentration of bioethanol is 1.001%.Tujuan dari penelitian ini adalah untuk mengetahui potensi serta pH optimum isolat bakteri termofilik SSA dalam menghasilkan bioetanol. Penelitian ini dilaksanakan pada bulan September 2021 hingga bulan Desember 2021, di Laboratorium Mikrobiologi  Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Negeri Padang. Penelitian ini merupakan penelitian eksperimen yang terdiri dari 6 perlakuan dan 3 ulangan dengan variasi pH inkubasi 6,5, 7, 7,5, 8, 8,5, dan 9. Data yang diperoleh dianalisis dengan sidik ragam (ANOVA) dengan uji lanjut Duncan’s New Multiple Range Test (DNMRT). Hasil penelitian menunjukkan bahwa isolat bakteri termofilik SSA memiliki potensi dalam menghasilkan bioetanol dengan isolat SSA 8 merupakan penghasil bioetanol dengan kadar tertinggi yaitu 1.001%, dan  variasi pH inkubasi berpengaruh terhadap konsentrasi bioetanol yang dihasilkan dengan pH optimum bakteri termofilik dalam menghasilkan bioetanol yaitu pH 8 yang menghasilkan konsentrasi bioetanol tertinggi yaitu 1.001%.

Abstract

Mudiak Sapan hot spring, South Solok Regency, West Sumatra is one of the hot springs where there are various species of thermophilic bacteria that produce xylanase enzymes. The xylanase enzyme is a group of enzymes that has the ability to hydrolyze xylan (hemicellulose) or polymers into xylose and xylo-oligosaccharides. The purpose of this study was to test the compatibility of bacterial isolates to become a xylanase-producing bacterial consortium. This research is a descriptive research that informs the compatibility of thermophilic bacterial isolates from the Mudiak Sapan hot springs. The method used in this study was disk diffusion, based on the research, it was found that 5 combinations of compatible xylanase enzyme-producing isolates were MS18 x MSS11; MS18 x MSS15; MSS18 x MS16 ; MSS11 x MS16 ; MSS15 x MS16 and 1 combination of incompatible isolates namely MSS11 x MSS15 which had an inhibition zone of 0.645 cm.

Abstract

Endophytic bacteria have been extensively studied as a source of new antimicrobial compounds. Research into new antimicrobials is important as an alternative to the treatment of resistant microbes. One potential source of antimicrobial compounds is the Taxus (Taxus sumatrana). This study aimed to investigate the antimicrobial activity produced by the endophytic bacteria of the Taxus. Endophytic bacteria were isolated from the roots of Taxsus plants growing in Mt. Singalang, X Koto District, West Sumatra. Bacteria were isolated by inoculating pieces of Taxus root tissue into a PDA medium. Endophytic bacteria were identified macroscopically and microscopically. Antimicrobial activity was tested with the test microbe using the spot test diffusion method. As a result of the research, 23 isolates of endophytic Taxus bacteria were obtained, of which 21 isolates were gram-positive and 2 isolates were gram-negative bacteria. The results of the antimicrobial activity test yielded only 2 isolates of endophytic bacteria with antibacterial activity.

Abstract

Advances in the field of biotechnology have led to the use of enzymes in industry increasingly. The high public awareness of environmental pollution makes enzyme technology as an alternative to replace various chemical processes. Enzymes can be used in various industrial sectors such as textiles, food, detergents, paper and cosmetics, as well as biofuels. One of the enzymes that is often used is the enzyme xylanase. Thermophilic microorganisms that can produce thermostable enzymes are thermophilic bacteria. One of the important factors affecting the success of the manufacturing process of the production of the xylanase enzyme is the concentration of the starter. The production of the thermophilic bacterial xylanase enzyme SSA 4 produced the highest specific enzyme activity at a concentration of 4% which was 0.137 U/mg. This shows that low starter concentrations can overwhelm microbial growth well. The increase in the concentration of the starter causes the enzyme activity to decrease, this happens.
