Abstract

Genetic relatedness among duku, kokosan, and pisitan from Indonesia were investigated using random amplified polymorphic DNA (RAPD) markers. Eleven primers (OPA-01, OPA-02, OPA-10, OPB-07, OPB-11, OPB-12, OPB-15, OPT-16, OPU-14, OPU-19, and OPU-20) were used for amplification and yielded a total of 174 DNA bands, of which 167 were polymorphic. Primer OPA-10, OPB-11, OPB-12, OPB-15, and OPU-19 produced all of the polymorphic DNA bands. The size of the amplified DNA fragments ranged from 41-1546 bp. The dendrogram separated into two clusters at a genetic similarity coefficient of 0.76. The cluster 1 consisted of subclusters duku and several pisitan (pisitan OKI, pisitan Sleman, pisitan Hatu, pisitan Punggur, and pisitan Tanjung), and cluster 2 consisted of subclusters kokosan and pisitan. In the kokosan subclusters, including duku Drendan. Dendrogram supported the determination of taxonomic status of duku, kokosan, and pisitan as one species, namely Lansium domesticum Corr. and its divided into two groups, namely L. domesticum ’duku group’ and L. domesticum ’pisitan-kokosan group’. Thus, RAPD analysis was useful tool for determining the genetic variation and the genetics relatedness among duku, kokosan, and pisitan in Indonesia.Key words: duku, kokosan, pisitan/langsat, genetic relatedness, RAPD

Abstract

Lansium domesticum Corr. with vernacular name in Indonesian duku has been reported containingtherapeutic bioactive compounds, and some of these compounds shown to be potent antitumor, anticancer,antimalaria, antimelanogenesis, antibacteria, and antimutagenic activities. This plant is commonly known asduku, kokosan and langsat by the local community in Indonesia. The morphological appearance of all varieties isnearly the same, and identifi cation of the varieties is very diffi cult for growers. Variation of DNA sequences ofthe ITS (Internal transcribed spacer) region can be used as a molecular character to determine the phylogeneticrelationship of different varieties of L. domesticum. The aims of this study were to determine taxonomy status ofduku, kokosan, and langsat, also phylogenetic relationship among varieties of L. domesticum based on ITS rDNAsequencing. DNA was isolated from leaves of plant and then amplifi ed using F1 and R1 primers. Nucleotidesequences were identifi ed using Sequence Scanner Software Programm version 1.0, nucleotide sequences from18S, ITS1, 5.8S, ITS2 and 26S region, that has been mergered using EditSeq and SegMan in software Suite forSequence Analysis DNASTAR Lasergene DM version 3.0.25. The results of study showed that DNA fragmentsranging in size from 782-810 bp. Different pattern of DNA fragments indicated polymorphism among duku,kokosan, and langsat. Based on the results of the ITS rDNA sequencing and phylogenetic tree analysis. Itwas determined that Lansium and Aglaia are a separated genus with the similarity index value of 0.98. Duku,kokosan and langsat were divided into two cluster, namely cluster kokosan-langsat and cluster duku with thesimilarity index value of 0.996. Keywords : Phylogenetic relationship, ITS region, L. domesticum, duku, kokosan, langsat

Abstract

Duku, kokosan dan langsat merupakan buah-buahan yang populer dan memiliki nilai ekonomi yang penting. Secara morfologi duku, kokosan dan langsat  hampir mirip satu sama lain sehingga menyulitkan dalam penentuan pada tingkat infraspesies bagi para pemulia tanaman. Berbagai pendekatan untuk penentuan morfologi ini telah banyak dilakukan. Tujuan penelitian ini menentukan status taksonomi duku, kokosan dan langsat pada ketegori infraspesies berdasarkan karakteritik makro dan mikromorfologi. Berbagai variasi duku, langsat dan kokosan dikoleksi dari beberapa pusat pertanaman di Indonesia, meliputi wilayah Sumatera, Jawa dan Kalimantan. Preparasi herbarium tumbuhan meliputi koleksi sampel, persiapan, penempelan dan identifikasi. Analisis morfologi menggunakan metode pengamatan dan deskripsi, pengukuran karakter morfologi dilakukan baik pada organ generatif dan vegetatif meliputi bunga, batang, biji dan buah, dan semua sampel didokumentasikan dengan menggunakan foto. Hasil penelitian ini menunjukkan ditemukan 29 variasi dari duku, kokosan, dan langsat yang terdiri dari 19 duku, 2 kokosan dan 8 langsat dari semua wilayah. Hasil karakterisasi makro dan mikromorfologi menunjukkan duku, kokosan, dan langsat dari berbagai daerah di Indonesia dapat dinyatakan duku, kokosan, dan langsat merupakan marga Lansium, untuk kategori jenis adalah L. domesticum Correa dan pada kategori infraspesies dapat dibagi menjadi dua group yaitu group duku dan group kokosan-langsat.

Abstract

Every tarsier species performs different vocalization behaviour. Cephalopachus bancanus as one of the tarsier species listed as vulnerable in the IUCN red list has limited and different information about their vocalization. This research was designed to explore the species vocalization in the vicinity of Petaling Village, District of Bangka, Bangka Island, Indonesia. Tarsier vocalization inside temporary enclosures was recorded using a handy recorder and analysed using bioacoustics software Audacity 2.3.3 and Raven Pro 1.6.1. We described seven vocalization types with different functions and spectrogram patterns. One type of vocalization, squeak, is produced only by the infant. Two types of vocalizations (whistle and cheeps) were produced by the infant and adult, and four vocalization types were performed by adults. Those types of vocalizations can be heard within human hearing. Some types of vocalizations have peak frequencies at the ultrasonic level, i.e.: agonistic scream, alarm call, distress call, and hysteresis.

Abstract

Analysis and identification of land use in coastal areas to study the coastal environment Sembilang National Park in South Sumatra using remote sensing system (Geoinformation System) has been conducted from June until November 2014. The objective to study the changes that occur in the coastal environment as well as to identify and estimate the area use change existing mangrove areas in Region I of the Section of Sembilang National Park Management, South Sumatra. Methods for the analysis and identification based on Landsat image data 8 in 2013 using ENVI 4.3 and Arc GIS 9.3 software as well as the conversion of ground check. The results showed the use of land in the mangrove areas in the region I of the Section of Sembilang National Park Management classified into 5 classes, namely mangrove, water area, mangrove shrubs, mainland mud, and ponds areas. The area of each class were observed in sequence as follows 8921.19 ha (41.69%) is a mangrove area; 3934.3 ha (23.28%) is water areas; 3.608,52ha (17.41%) is an area of mangrove shrubs; 1.649,11ha (9.05%) is mainland mud and settlement area; and 1.561,04ha (8.57%) is the ponds areas. The percentage of the class obtained as a result of the conversion of ground check is based on the land use map data with the total area observed.

Abstract

Indonesia is estimated to have 157 species of bamboo which is more than 10% of the world's bamboo species. Almost 50% of bamboo growth in Indonesia is endemic bamboo and more than 50% of bamboo species life in Indonesia have been utilized by the society conventionally. Further, the bamboo utilization is still having high potential to be developed by increasing the industrial development. This research aimed to investigate the diversity of the bamboo species in South Sumatera, particularly in Musi Rawas district. The result showed that in Musi Rawas district of South Sumatera province there are 10 species of bamboo located at 11 locations in protected forest area of 883.60 ha and production forest area of 177,976.26 ha. The large number of bamboo potentials viewed from various aspects and the many types of bamboo scattered in this district can be a supporting data in doing bamboo conservation efforts, especially in Musi Rawas district of South Sumatra province.

Abstract

Research in the molecular field requires DNA with a high degree of purity. Local rice varieties of South Sumatra have different leaf texture so that in isolation process to obtain DNA with high purity level required the right method. This study aims to compare the quality and quantity of purity of local rice insulation of local rice varieties of South Sumatra with different methods of CTAB and Wizard Genomic DNA Purification System Kit from Promega. The research was conducted from August 2015 to December 2015 at the Laboratory of Microbiology and Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. The method used is DNA isolation method Wizard Genomic DNA Purification System Kit from promega and CTAB. Based on research that has been done result of DNA visualization showed that DNA isolate CTAB method still have smears while DNA isolate method of Wizard Genomic DNA Purification System Kit from promega not. The average DNA isolate concentration of Wizard Genomic DNA Purification System Kit method is A260 / 280 = 1.853 μg / ml. The average DNA isolate concentration of CTAB method is A260 / 280 = 1,705 μg / ml. Isolation of Rice DNA of local variety of South Sumatera using DNA method of Genomic DNA Purification System Kit from promega has higher quality and quantity compared to CTAB method.

Abstract

Research on the Morphological Diversity of Lansium domesticum Corr in South Sumatra aims to examine the morphological diversity of L. domesticum in South Sumatra. The research was conducted in April 2017 to June 2017. This research was conducted by means of exploration carried out in seven districts covering, East Ogan Komering Ulu District, South Ogan Komering Ulu District, Ogan Komering Ulu District, Musirawas District, Ogan Komering Ilir District, Muara Enim District and Musi Banyuasin District. Sampling was done using purposive sampling method. Observation of morphological diversity was done by characterization of morphology and analyzed by the descriptive method. The result of this research shows that in South Sumatera, there were two variants of L. domesticum namely L. domesticum variant duku and L. domesticum variant langsat. Morphological characters that distinguish between duku and langsat variants were found under canopy conditions, the presence of hair on top and bottom surfaces of leaves, fruit shape, fruit skin thickness, fruit taste, and the presence of sap on ripe fruit.

Abstract

The study aims to determine the diversity of vegetation in the Green Open Space of the Gasing Industrial Area. The study was conducted from April to July 2017. The method of analysis used quadratic method with 2 plots at different location with three repetitions, quantitative data analysis was done on density value, dominance, frequency, important value and diversity index of each plant species. The results showed that vegetation consists of 16 tribes and 19 species, dominated by Pteridophyta and Spermatophyta. Value of vegetation diversity (2,24) and uniformity value 0,30. The higher number of the species, the higher diversity index of the species. Such characteristics are caused by environmental influences such as organic matter content and low pH and factors derived from human intervention. Keywords: Vegetation, Diversity, industrial area

Abstract

There has been research on the Bioacumulation of Cadmium in the Leachate, Sediment, Sepat fish and watercress within water around the final solid waste disposal (FWD) of Sukawinatan Palembang using Atomic Absorption Spectrophotometric (SSA) technique. This study aims to determine the distribution of cadmium contained in water ecosystem within water around the final solid waste disposal (FWD) of Sukawinatan, as well as bioconcentration factor (BCF) of the biotic compartement to the abiotic compartement . The way is done by wetdestruction sample. The resultant destruction solution was analyzed at a wavelength of 228.8 nm. The results showed that the concentration of cadmium in leachate Inlet 0,0099 mg/L until the Sedapat River 0,0010 mg/L, Sediment Inlet 0,0427 mg/Kg until Sedapat River Sediment 0,0051 mg/Kg, Sepat fish reservoir 0,0099 mg/Kg and Sepat fish Sedapat River 0,0096 mg/Kg along watercress reservoir 0,0042 mg/Kg over Sedapar river 0,0027 mg/Kg. So that distribution of cadmium for water and sediment compartment getting further from leachate inlet to Sedapat river is decreasing. The value of cadmium consentration in leachate is still below the threshold conformable in regulation of environment and forestry ministers no.59 of 2016 is 0,1 mg/L. Distribution on the fish and watercress compartment also descreased to the Sedapat river. Consentration value on the fish and watercress also stil below the quality standard in this SNI 7378;2009 for fish 0,1 mg/Kg and watercress 0,2 mg/Kg. While the value of BCF fish and BCF watercress <100 is still in low category.
