Abstract

Desa wisata merupakan kawasan yang mempunyai kearifan lokal yang berpotensi mempunyai daya tarik wisata. Meningkatnya peminat wisata desa dan melimpahnya desa wisata di Indonesia harus diimbangi dengan sumber daya manusia (SDM) yang memadai dalam mengelola dan mengembangkannya, terutama dalam hal kesehatan. Tujuan pengabdian masyarakat ini untuk memberikan edukasi dan pemahaman pada SDM dalam bentuk pelatihan dan penyuluhan kesehatan khususnya di desa wisata, desa kampung adat Segunung Wonosalam Jombang Jawa Timur. Metode yang diaplikasikan dalam pelaksanaan pengabdian masyarakat di desa wisata kampung adat Segunung ini ada dua langkah, yaitu penyuluhan dan pelatihan, dilaksanakan selama 3 hari. Hari pertama pelatihan dengan tema first aid, peserta diberikan pre-test dan post-test sebelum dan sesudah pelatihan. Hari kedua Penyuluhan kesehatan (tema: pengetahuan tentang tanaman obat, pengolahan bahan pangan, dan pola hidup sehat), peserta pada penyuluhan kesehatan diberikan pre-test dan post-test sebelum dan sesudah penyuluhan. Hari ketiga penyuluhan tentang NAPZA. Hasil pre-test dan post-test pada pelatihan first aid didapatkan peningkatan persentase jumlah peserta yang mendapatkan nilai ≥ 60 sebesar 56% dan pada penyuluhan kesehatan didapatkan peningkatan 47%. Penyuluhan dan pelatihan ini diharapkan dapat dipraktikkan dikemudian hari dan bermanfaat untuk kelangsungan objek wisata kampung adat Segunung.

Abstract

Background: Centella asiatica leaves ethanol extract (CALEE) has higher concentration compared to other structures within the plant. The extract contains alkaloids, saponins and flavonoids, which play an active role as antioxidant and antibacterial. Current study aimed to determine the effect of CALEE on Proteus mirabilis, Proteus vulgaris, and Yersinia enterocolitica, Gram-negative bacteria that cause diarrhea. Materials and methods: Simplicia of C. asiatica leaves was dissolved in 96% ethanol and macerated to get condensed extract, which then produced a concentration of 1 g/mL. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined to assess the effectiveness of CALEE on P. mirabilis, P. vulgaris, and Y. enterocolitica. MIC was determined through serial dilution test with Mueller Hinton broth media. After incubation, the bacteria were streaked on nutrient agar or McConkey agar to determine the MBC. Results: The MIC value of CALEE could not be determined since the color of CALEE was dark, hence the turbidity could not be compared. CALEE had the same MBC value (0.25 g/mL) in all bacteria species used in this study. Conclusion: CALEE is effective against Gram-negative bacteria, such as P. mirabilis, P. vulgaris, and Y. enterocolitica. Further research is needed, especially in vivo experiments and evaluation of the cytotoxicity effect of CALEE. Abstract

The antioxidant properties from medicinal plants have protective effects against oxidative stress-linked diseases. Their antioxidant capacities are related to phenolic compounds, including phenols and flavonoids. Clinacanthus nutans, a native to Asia, including Malaysia, Thailand, and Indonesia, are used for treating diabetes and other diseases, but the exploration of the plant in Indonesia is still limited. Therefore, this study aimed to reveal the phytochemical constituents, metabolic compounds, and antioxidant activity of C. nutans extracts. Dried leaf powder was extracted with 70% ethanol and aqueous, each extract was subjected to qualitative phytochemical screening. Subsequently, total phenolic and flavonoid content was calculated using Folin-Ciocalteu and AlCl3 colorimetric assay. The antioxidant activity was determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method. The profiles of phenolic and flavonoid compounds of 70% ethanolic extract were investigated by LC MS/MS. Phytochemical screening showed that the extracts contain phenolic, flavonoid, tannin, and saponin. The total phenolic content of the 70% ethanol and aqueous extract was 4.14±1.9% mgGAE/g and 3.89±3.2% mgGAE/g, respectively. Meanwhile, the total flavonoid content obtained was 0.19±1.0% mgQE/g and 0.03±0.5% mgQE/g for 70% ethanol and aqueous extracts, respectively. Both extracts showed high antioxidant activity, with DPPH IC50 30.57 ????g/mL for 70% ethanol and 37.24 ????g/mL for aqueous extract. LC-MS/MS analysis of 70% ethanol extract revealed 5 phenolic and 3 flavonoid compounds, where DL-glutamic monohydrate and Boesenbergin B were the most dominant based on the percentage of area under the curve. C. nutans originating from Indonesia can be a good source of natural antioxidants because 70% ethanol and aqueous extract show high phytochemical content and antioxidant activity.

Abstract

Introduction: Diabetes mellitus is a metabolic disease characterized by hyperglycemia. Various epidemiological studies have shown a trend of increasing incidence and prevalence of diabetes mellitus in various parts of the world. Therefore, diabetes mellitus is currently a global health threat. Dandang gendis (Clinacanthus nutans) is a widely used plant as a traditional herbal treatment in Indonesia, and it has been proven that the ethanol extract of dandang gendis leaves shows an antidiabetic effect. This research aims to determine the compatibility among the flavonoid compounds in C. nutans with Aldose reductase, glucokinase, and GSK3β target drugs for type 2 diabetes mellitus using in silico method.Methods: 45 compounds were obtained from multiple sources. The screening method used Lipinski’s rule of five and Pyrx until 8 compounds were selected. Avogadro, AutoDock 4.2, and Biovia Discovery Studio 2016 were used for molecular docking and visualization analysis.Results: Molecular docking results demonstrate that the ligand-protein interaction’s binding energy was -7.31 to 35.25 kcal/mol for 1AH3, -7.55 to 0.15 kcal/mol for 1V4S, and -7.99 to -2.85 kcal/mol for 3D0E.Conclusion: We can conclude that flavonoid compounds Apigenin, Vitexin, 3,3-di-O-Methylellagic Acid, and Clinacoside C show a high binding affinity with Aldose Reductase, Glucokinase, and GSK3β proteins and have the potential to be oral antidiabetic drug compounds for Diabetes Mellitus. However, its binding affinity has not been able to exceed that of the native ligand of the protein. Further research is needed to determine the significant efficacy and potential as an antidiabetic.

Abstract

Acne is a skin disease that is common in teenagers. In Indonesia, acne occurs around 85-100% of cases. According to the records of the Indonesian cosmetic dermatology study group, it was shown that 60% of acne sufferers in 2006, 80% in 2007 and 90% in 2009. A face with acne is an image that is not ideal for a teenager and creates dissatisfaction, causing a feeling of lack of self-confidence. One way that can be used to overcome the problem of acne is by giving a celery mask (Apium graveolens L.). Masks are cosmetic preparations for facial skin care which have the benefit of curing acne and acne scars. Celery (Apium graveolens L.) has been shown in several studies to have anti-inflammatory and antibacterial activity to prevent acne. This study aims to prove the effect of using a celery mask to reduce the number of acne lesions. The method chosen in this study was the Pretest-Posttest Control Group Design, by comparing the difference in reducing the number of acne lesions from the celery mask treatment with the paired T-test formula. The results obtained from giving celery masks to 10 young women aged 16-18 years with the classification of mild, moderate and severe acne for 2 weeks showed a significant decrease in the number of acne lesions (p = 0.00). So it can be concluded that giving celery masks can reduce the number of acne lesions in young women aged 16-18 years.

Abstract

Pseudomonas aeruginosa is the main cause of nosocomial infection which is responsible for 10% of hospital-acquired infection. Pseudomonas aeruginosa tends to mutate and displays potential for development of antibiotic resistance. Approximately, 10% of global bacterial isolates are found as Multidrug-resistant Pseudomonas aeruginosa. Pseudomonas aeruginosa have a quite tremendous severity index, especially on pneumonia and urinary tract infections, even sepsis, which 50% mortality rate. Pineapple (Ananas comosus L. Merr) has antimicrobial properties. The active antimicrobial compounds in Ananas comosus L. Merr include saponin and bromelain. This research aims to find the potency of antimicrobial effect of pineapple (Ananas comosus L. Merr) extract towards Multidrug-resistant Pseudomonas aeruginosa. Multidrug-resistant Pseudomonas aeruginosa specimen is obtained from patient's pus in orthopaedic department, Dr Soetomo Public Hospital, Surabaya. Multidrug-resistant Pseudomonas aeruginosa specimen is resistant to all antibiotic agents except cefoperazone-sulbactam. This research is conducted by measuring the Minimum Inhibitory Concentration (MIC) through dilution test with Mueller-Hinton broth medium. Pineapple extract (Ananas comosus L. Merr.) is dissolved in aquadest, then poured into test tube at varying concentrations (6 g/ml; 3 g/ml; 1.5 g/ml; 0.75 g/ml, 0.375 g/ml; and 0.1875 g/ml). After 24 hours' incubation, samples are plated onto nutrient agar plate, to determine the Minimum Bactericidal Concentration (MBC). The extract of pineapple (Ananas comosus L. Merr) has antimicrobial activities against Multidrug-resistant Pseudomonas aeruginosa. Minimum Inhibitory Concentration (MIC) could not be determined, because turbidity changes were not seen. The Minimum Bactericidal Concentration (MBC) of pineapple extract (Ananas comosus L. Merr) to Multidrug-resistant Pseudomonas aeruginosa is 0.75 g/ml. Further study of in vivo is needed.

Abstract

Pemberdayaan kader kesehatan dalam program GERMAS melalui Posbindu di Desa Kentong, Kecamatan Glagah, Kabupaten Lamongan bertujuan untuk meningkatkan pengetahuan tentang pemanfaatan tanaman obat yang berkhasiat sebagai anti hipertensi. Tingginya prevalensi pasien hipertensi di daerah tersebut, diduga akibat dari kepatuhan berobat yang rendah, pengetahuan perilaku gaya hidup dan diet yang sehat masih belum optimal, serta minimnya informasi dan pengetahuan tentang pemanfaatan tanaman obat pada pasien hipertensi. Solusi yang ditawarkan untuk menyelesaikan permasalahan tersebut adalah memberikan penyuluhan tentang materi pentingnya mengelola hipertensi dengan baik, pengobatan rutin, perilaku gaya hidup serta diet sehat bagi pasien hipertensi,  sosialisasi tanaman obat berkhasiat sebagai antihipertensi dan demonstrasi pelatihan pembuatan sediaan dari tanaman obat. Metode yang digunakan melalui survei, kuesioner dan penyuluhan penyampaian materi, serta demonstrasi langsung di depan peserta. Ditemukan pada kuesioner pretes bahwa kader belum paham tentang ragam tanaman obat dan pengolahan yang tepat, setelah dilakukan penyuluhan dan demonstrasi terjadi peningkatan pengetahuan dengan para kader mampu menjawab pos tes dengan benar, peserta aktif berdiskusi secara oral, hal tersebut sebagai indikator bahwa peserta antusias dan dapat tercipta kader kesehatan yang mandiri.

Abstract

Background: Indonesia as an agricultural country has more than 33 million workers in the agriculture. Pesticides have an important role in agriculture because they function as powerful plant pest exterminators. However, its use is considered to cause various toxic effects, one of which is on the nervous system. This study aimed to evaluate the impact of pesticide exposure on cognitive impairment by assessing the butyrylcholinesterase biomarker in the blood of rats as experimental animals.Methods: This was a true experimental study conducted during September–November 2022 at the Pharmacology and Biochemistry Laboratory, Faculty of Medicine, University of Jember with a post-test only approach using Wistar rats which were divided into 1 control group and 4 treatment groups treated with chlorpyrifos 5 mg/kg orally. Serum butyrylcholinesterase enzyme levels were measured using the kinetic photometric method and then analyzed with a regression test in IBM SPSS® Statistics software.Results: The average value of butyrylcholinesterase was 920.23±145.27 (Control group); 904.48±91.6 (7 days after chlorpyrifos administration); 889.81±95.2 (14 days after chlorpyrifos administration); 457.84±69.5 (28 days after chlorpyrifos administration); 575.82±194.25 (56 days after chlorpyrifos administration). The results of the One-Way ANOVA test showed significant differences between groups as indicated by a significance test of 0.024 (p <0.05).Conclusions: There is a relationship between the duration of exposure to chlorpyrifos and butyrylcholinesterase levels in Wistar rats. This fact shows that the longer an organism is exposed to low doses of chlorpyrifos pesticides, the higher the level of neurological system toxicity that occurs in that organism.

Abstract

Diabetes Melitus adalah penyakit yang ditandai dengan hiperglikemia karena sekresi insulin yang sedikit, aksi insulin kurang poten, atau disebabkan keduanya. Daun dandang gendis (Clinacanthus nutans)diketahui mengandung antioksidan yang dilaporkan mampu menurunkan kadar glukosa darah. Penelitian ini untuk mengetahui pengaruh pemberian ekstrak daun dandang gendis terhadap kadar glukosa darah pada tikus Wistar model diabetes. Tikus sebanyak 30 ekoryang sesuai kriteria dibagi dalam 5 kelompok yang kemudian diinduksi hiperglikemia menggunakan streptozotocin 50 mg/kgBB. Setelah 2 hari pasca induksi, tikus diberi ekstrak daun dandang gendis sesuai dosis yaitu K1 (75 mg/kgBB), K2 (150 mg/kgBB), dan K3 (300 mg/kgBB) lalu dibandingkan dengan K0 (CMC-Na sebagai kontrol) dan K4 (metformin sebagai obat standar) selama 14 hari. Penurunan kadar glukosa darah yang paling signifikan terjadi pada kelompok K1 dengan pemberian ekstrak dandang gendis 75 mg/kgBB dibandingkan dengan kelompok K0, K2, K3, dan K4.

Abstract

Penelitian ini untuk mengetahui efek terapi ekstrak kulit manggis (Garcinia mangostana) terhadap sepsis yang diinduksi Shigella dysenteriae. Penelitian ini merupakan penelitian eksperimental, menggunakan 30 mencit dibagi menjadi 5 kelompok. Masing-masing mencit diinjeksi S. dysenteriae secara intraperitoneal. Perlakuan hari pertama post injeksi kontrol positif diberi Azithromycin 246 mg/kgBB, kontrol negatif diberi akuades, kelompok perlakuan diberi ekstra kulit manggis dengan dosis sebagai berikut: K1= 100 mg/kgBB, K2= 200 mg/kgBB, dan K3= 400 mg/kgBB. Perlakuan hari kedua post injeksi sesuai masa inkubasi bakteri skor MSS tiap mencit dihitung. Terdapat korelasi antara pemberian ekstrak kulit manggis dengan skor MSS mencit (p<0.05), tidak terdapat perbedaan signifikan antara pemberian ekstrak 200 mg/KgBB dan 400 mg/kgBB dengan Azithromycin.
