Abstract

Diabetes mellitus is a metabolic disorder characterized by hyperglycemia, which causes damage to tissues and organs through the process of oxidative stress. Malondialdehyde (MDA) as a biomarker of oxidative stress has a significant increase in hyperglycemia. Stem cell therapy can be an option for diabetes mellitus such as Wharton's jelly mesenchymal stem cells. This study aimed to determine the effect of MSC-WJ administration on serum MDA levels of hyperglycemic Wistar rats. This experimental study applied a post-test only design with a control group. The sample consisted of 21 male Wistar rats divided into 3 groups, including the negative control group (K-), namely the group that was only given a standard feed diet, the positive control group (K+), namely the group that was injected with alloxan with a dose of 0.2 ml intraperitoneally, and the treatment group (P), which was the alloxan-induced group, and was given MSC-WJ at a dose of 3 million cells intravenously in the rat tails. In all three groups, blood was taken from the retro orbital plexus to measure serum MDA levels with the TBARS test. The results of the analysis showed that the mean serum MDA levels of the K-, K+, and P groups were 1.83 nmol/ml, 2.97 nmol/ml, and 1.73 nmol/ml, respectively. The results of the comparison test showed a value of p = 0.000. It was concluded that there was a significant difference in serum MDA levels between the three groups, which indicated that MSC-WJ administration could reduce alloxan-induced hyperglycemia Wistar rats serum MDA levels.Keywords: hyperglycemia; malondialdehyde; Wharton's jelly mesenchymal stem cells ABSTRAK Diabetes mellitus adalah gangguan metabolik yang ditandai dengan hiperglikemia, yang menyebabkan kerusakan jaringan dan organ melalui proses stress oksidatif. Malondialdehid (MDA) sebagai biomarker stress oksidatif mengalami peningkatan signifikan pada hiperglikemia. Terapi stem cell dapat menjadi pilihan untuk diabetes mellitus seperti mesenchymal stem cell wharton’s jelly. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian MSC-WJ terhadap kadar MDA serum tikus wistar hiperglikemia. Penelitian eksperimental ini menerapkan rancangan post-test only with control group. Sampel adalah 21 ekor tikus wistar jantan yang dibagi menjadi 3 kelompok, di antaranya kelompok kontrol negatif (K-) yaitu kelompok yang hanya diberi diet pakan standar, kelompok kontrol positif (K+) yaitu kelompok yang diinjeksi aloksan dengan dosis 0,2 ml secara intraperitoneal, dan kelompok perlakuan (P) yaitu kelompok yang diinduksi aloksan, serta diberi MSC-WJ dengan dosis 3 juta sel secara intravena pada ekor tikus. Pada ketiga kelompok dilakukan pengambilan darah dari pleksus retro orbitalis untuk pengukuran kadar MDA serum dengan tes TBARS. Hasil analisis menunjukkan kadar rerata MDA serum kelompok K-, K+, dan P secara berturut-turut adalah 1,83 nmol/ml, 2,97 nmol/ml, dan 1,73 nmol/ml. Hasil uji perbandingan menunjukkan nilai p = 0,000. Disimpulkan bahwa terdapat perbedaan kadar MDA serum yang bermakna di antara ketiga kelompok, yang menunjukkan bahwa pemberian MSC-WJ dapat menurunkan kadar MDA serum tikus wistar hiperglikemia yang diinduksi aloksan.Kata kunci: hiperglikemia; malondialdehid; mesenchymal stem cell wharton’s jelly

Abstract

Atherosclerosis is the cardiovascular disease’s main pathological mechanism. Basil leaves are food companion that s widely consumed by Indonesians and contains phenolic and flavonoid bioactive compounds that can increase HDL levels. The purpose of this study was to see the effect of basil leaves extract (Ocimum basilicum L.) on HDL levels in atherosclerosis model rat. This study was conducted using the post test only control group design method on 29 rats. The rats were divided into 5 groups, K-, K+, P1, P2, and P3. K- is group of rats without treatment. K+ was treated with partial carotid ligation (PCL) and high fat diet (HFD), P1 was treated with PCL, HFD and given a dose of 100 mg/kgB basil leaves extract, P2 was treated with PCL, HFD and given a dose of 200 mg/kgBB basil leaves extract, P3 was treated with PCL, HFD and given simvastatin 1.5 mg/rat/day. HFD was given for 7 days, basil leaves extract and simvastatin given for 14 days using oral feeding tube. HDL levels tested with CHOD-PAP method using a microlab 300 spectrophotometer. The result from this study were analyzed by Saphiro-Wilk test, and Kruskal-Wallis test. The mean HDL levels obtained in the K- group were 52,65(49,70‒68.10) mg/dL, K+ 49,67±8,77 mg/dL, P1 52.70±1.58 mg/dL, P2 54.40±9.94 mg/dL, P3 58,05 ±5,34 mg/dL. The results showed there was a non-significant increase in HDL levels (p=0.377) in atherosclerosis model rat by giving basil leaves extract on both dose, and simvastatin.

Abstract

The neutrophil lymphocyte ratio (NLR) and platelet lymphocyte ratio (PLR) tests are simple tests can be done before a COVID-19 patient hospitalized. This examination is useful for determining the diagnosis and prognostic of the patient. The aim of the study was to determine the correlation between NLR and PLR with the severity of COVID-19. This research was an analytical study with a cross-sectional approach using secondary data from 289 medical records of patients with confirmed COVID-19 for the period June-August 2021. The total sampling technique was used and processed with the chi-square test. Most of the research subjects were in the range of 26-45 years (28.4%). More than half of the subjects were female (61.6%). There were 99 patients (34.3%) with severe conditions. Less than half of the patients had abnormal leukocyte and platelet examination results, namely 64 (22.1%) and 56 (19.4%). Patients with abnormal NLR and PLR examinations were 135 (46.7%) and 123 (42.6%). There was a significant correlation between NLR and the severity of COVID-19 inpatients (p<0.001, OR=9.452). There is a significant correlation between PLR and the severity of COVID-19 inpatients (p<0.001, OR=7.268). This study concludes a significant correlation between NLR and PLR with the severity of COVID-19.

Abstract

Latar Belakang: Di layanan primer, OA sering ditangani dengan OAINS, namun efek samping gastrointestinal seperti dispepsia kerap menurunkan kualitas hidup. Objektif: Menilai hubungan pemakaian OAINS dan durasinya dengan derajat keparahan dispepsia pada pasien OA di wilayah kerja Puskesmas Kuranji, Kota Padang. Metode: Studi potong lintang menggunakan data primer melalui wawancara terstruktur terhadap 101 pasien OA yang sedang menjalani terapi OAINS. Analisis menggunakan uji chi square. Hasil: Mayoritas responden berusia >60 tahun 54,5 persen, perempuan 79,2 persen. Natrium Diklofenak merupakan OAINS tersering 84,2 persen, dengan durasi penggunaan >3 bulan 56,4 persen. Terdapat hubungan pemakaian OAINS dengan dispepsia p=0,000, dan hubungan durasi penggunaan OAINS dengan derajat keparahan dispepsia p=0,001. Kesimpulan: Pada pasien OA di layanan primer, pemakaian dan durasi OAINS berkaitan dengan dispepsia dan tingkat keparahannya. Temuan ini menegaskan pentingnya skrining gejala cerna, edukasi pasien, serta peninjauan regimen analgesik yang lebih aman. Pertimbangkan gastroproteksi dan pemantauan berkala pada kelompok berisiko tinggi secara klinis.

Abstract

Penelitian ini bertujuan untuk mengetahui efek ekstrak daun kemangi (Ocimum basilicum) terhadap kadar LDL pada tikus (Rattus Norvegicus) model aterosklerosis. Desain yang digunakan adalah post-test only control group design dengan 30 ekor tikus jantan yang dibagi menjadi lima kelompok: kontrol negatif (K-), kontrol positif (K+), kontrol statin (KS), perlakuan I (P1; 100 mg/kgBB ekstrak daun kemangi), dan perlakuan II (P2; 200 mg/kgBB). Induksi aterosklerosis dilakukan melalui ligasi parsial arteri karotis dan pemberian diet tinggi lemak selama 7 hari. Pemberian simvastatin dan ekstrak kemangi dilakukan secara oral selama 7 hari. Kadar LDL diukur menggunakan metode CHOD-PAP dengan alat Microlab 300. Hasil analisis statistik menunjukkan perbedaan bermakna kadar LDL antara kelompok K-, KS, dan P2 dibandingkan dengan kelompok K+ (p<0,05). Rerata kadar LDL tertinggi ditemukan pada kelompok K+ (64,56 mg/dL) dan terendah pada kelompok KS (31,62 mg/dL). Dapat disimpulkan bahwa ekstrak daun kemangi, khususnya dosis 200 mg/kgBB, berpotensi menurunkan kadar LDL pada tikus model aterosklerosis.
