Abstract

Three sesquiterpenoids, guaianediol (1), alismol (2), and spathulenol (3), were isolated from the n-hexane and ethyl acetate extracts of the stem bark of Dysoxylum amooroides. The three compounds were found in D. amooroides species for the first time. The structures of the isolated compounds were identified and established based on an extensive spectroscopic analysis involving HR-TOF-ESI-MS, IR, and NMR data, as well as a comparison with the previously reported works of literature. Compounds 1-3 were further assessed for cytotoxic effects against MCF-7 breast cancer cells. Guaianediol (1) showed inactive activity with IC50 > 100 µM, alismol (2) showed weak activity with IC50 value of 82.1 µM and spathulenol (3) showed considerable activity with an IC50 value of 15.2 µM. A brief structure-activity relationship and comparison with the previous works were also discussed to understand better the role of guaiane- and aromadendrane-type sesquiterpenoids in the biological activity perspective.

Abstract

Abstract. Curcumin is a natural compound from Curcuma longa with known antibacterial, antioxidant, and anti-inflammatory properties. However, its therapeutic use is limited due to poor water solubility and low bioavailability. One strategy to improve its biological performance is by conjugation with amino acids. This study aimed to synthesize curcumin conjugates with arginine, alanine, and leucine using Steglich esterification with DMAP and DCC. Conjugation with arginine and alanine was unsuccessful, likely due to solubility issues, steric hindrance or instability of functional groups. In contrast, the curcumin–leucine conjugate was successfully synthesized in dioxane, yielding a crude product with a 2,4% yield. High-resolution mass spectrometry (HRMS) confirmed the product with a [M–H]+ peak at m/z 580,2551, consistent with the theoretical mass of curcumin–leucine–Boc. Antibacterial activity against Escherichia coli was tested using the microdilution method, but no inhibitory effect was observed at test concentrations ≤1500 ppm. Abstrak. Kurkumin merupakan senyawa aktif alami dari Curcuma longa yang dikenal memiliki aktivitas biologis seperti antibakteri, antioksidan, dan antiinflamasi. Namun, penggunaannya secara terapeutik masih terbatas karena kelarutannya yang rendah dalam air dan bioavailabilitas yang buruk. Salah satu pendekatan untuk meningkatkan efektivitas biologisnya adalah melalui konjugasi dengan asam amino. Dalam penelitian ini, dilakukan upaya sintesis konjugat antara kurkumin dengan tiga jenis asam amino, yaitu arginin, alanin, dan leusin, menggunakan metode esterifikasi Steglich dengan katalis DMAP dan aktivator DCC. Konjugasi kurkumin–arginin dan kurkumin–alanin tidak berhasil dilakukan, diduga akibat faktor ketidaklarutan reaktan, hambatan sterik atau ketidakstabilan gugus fungsional dalam kondisi reaksi. Sementara itu, sintesis konjugat kurkumin–leusin berhasil dilakukan dalam pelarut dioksana, dan menghasilkan senyawa dalam bentuk krud dengan rendemen sebesar 2,4%. Hasil karakterisasi menggunakan spektrometer massa resolusi tinggi (HRMS) menunjukkan puncak molekul [M-H]+ pada m/z 580,2551, sesuai dengan massa teoritis konjugat kurkumin–leusin–Boc. Uji aktivitas antibakteri terhadap Escherichia coli dilakukan dengan metode mikrodilusi, namun tidak menunjukkan adanya penghambatan pada konsentrasi uji ≤1500 ppm.

Abstract

ABSTRACT. Triterpenoids are a large group of secondary metabolites commonly found in plants, exhibiting high diversity in both structural features and biological activities. Meliaceae family is knows as a rich source of the triterpenoid compounds. As the most extensive genus within the Meliaceae family, Aglaia is known to contain many bioactive triterpenoid compounds, including cytotoxic triterpenoids. Among the various types of triterpenoids, dammarane is frequently found in Aglaia and has demonstrated potential cytotoxic activity. This purpose of the research was to isolate and structure determination of four dammarane triterpenoids from the methanol extract of Aglaia eximia stem bark. All four compounds were successfully isolated and identified as, dammar-24-en-3a,20-diol (1), 20S,24S-epoxy-dammar-3a,25-diol (2), (E)-dammar-23-en-3a,20,25-triol (3), and (E)-25-hydroperoxydammar-23-en-3a,20-diol (4), respectively. The compounds were analyzed using a combination of spectroscopic techniques, including HRMS (high-resolution mass spectroscopy), FTIR (fourier transform infrared spectroscopy), and NMR (nuclear magnetic resonance, one and two dimensional). Cytotoxicity assays using the MTT method were applied to compounds 1-4. All isolated compounds (1-4) generated moderate cytotoxic activity against P388 murine leukemia cells with IC50 9.09, 11.03, 5.89, and 5.74 μg/mL, respectively. Preliminary structure-activity relationship (SAR) analysis suggested that the presence of hydroxyl and hydroperoxyl groups at C-25 increases cytotoxicity, whereas the cyclization in the side chain to form an epoxide ring decreases cytotoxicity. Abstract

Xylapeptide A, derived from the fungus Xylaria sp. x Sophora tonkinensis, exhibits potent and selective antimicrobial properties. Our research group has successfully synthesized xylapeptide A. In our recent work, two xylapeptide A analogues (An1 and An2) were synthesized using a combination of solid- and solution-phase synthesis methods. The linear precursors of An1 and An2 were synthesized on 2-CTC resin with the Fmoc strategy. The coupling reagents HBTU/HOBt and HATU/HOAt were employed. Subsequently, the linear precursor was cleaved from the resin using either 20% TFA or a TFE mixture, and then cyclized in solution phase with HBTU. The synthesized products were purified using semi-preparative RP-HPLC, giving the percent yields 16% for An1 and 12% for An2. Both compounds were then characterized by HR-ToF-MS, 1H- and 13C-NMR. The synthesized xylapeptide A and its analogues were evaluated against Bacillus cereus, Bacillus subtilis, Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, Klebsiella pneumoniae, and Candida albicans. The result showed that An2, possessing arginine residue, exhibited higher activity compared to xylapeptide A and An1. This research suggests that xylapeptide A analogues hold great promise as novel antimicrobial agents.

Abstract

This study departs from the urgency of early childhood religious education as the foundation for shaping religious and moderate character. The lack of Qur’an learning facilities and qualified teachers in Karang Mulya Village has hindered children aged 6–10 years in mastering Qur’anic literacy skills, even though this stage is considered a golden period of cognitive, affective, and psychomotor development. To address this issue, the research employed a qualitative descriptive method combined with a Project-Based Learning (PjBL) approach, which allowed the teaching process to be linked with practical and contextual activities. Data were collected through direct observation, structured interviews with parents and teachers, and careful documentation of student progress. The analysis was carried out through systematic steps of data reduction, presentation, and conclusion drawing. The findings reveal significant improvements in children’s ability to recognize, read, and write hijaiyah letters, although variations remain, particularly in creativity and independence. Furthermore, Qur’anic literacy activities meaningfully contributed to strengthening the understanding of religious moderation values, such as national commitment, tolerance, anti-violence, and acceptance of local culture. In conclusion, student service activities through Qur’anic literacy not only enhanced technical skills but also functioned as an effective medium for building religious moderation foundations from an early age.

Abstract

The research discusses the perceptions of students using Google Docs as a collaborative tool to enhance their writing skills in English. The study employed an online survey using Google Forms, conducted on 50 students from the English education program at Indraprasta University PGRI in the 2021 academic year. The survey results indicated that most respondents found Google Docs to be very useful in collaborative writing practice. They also highlighted the importance of features such as spell checking and real-time revision in supporting the writing process. The study concludes that there is a need for more comprehensive usage guides and training to overcome technical barriers and maximize the potential of Google Docs in improving writing and collaboration skills among college students

Abstract

In tropical regions such as East Lombok Regency, where food production is highly dependent on rainfall, drought poses a major threat to the agricultural sector. This study aims to monitor drought patterns over time using Google Earth Engine (GEE). Vegetation indices, including the Normalized Difference Vegetation Index (NDVI), Normalized Difference Water Index (NDWI), and Normalized Difference Drought Index (NDDI), were derived from Landsat 8 satellite imagery. The analysis revealed that during the dry season, particularly in August and September, the southern region—especially Jerowaru Sub-district—experienced severe drought conditions. The western parts, including Sambelia, Pringgabaya, and Suela Sub-districts, were also significantly affected, with the most impacted areas being rain-fed rice fields, corn plantations, and mixed horticultural crops. Temporal trend analysis indicated an increasing drought intensity in the later years of observation. The resulting information can support decision-making in drought risk mitigation and sustainable water resource management. By integrating satellite-based drought assessment with agricultural planning, this approach can strengthen food security and promote adaptive agricultural practices in drought-prone regions such as East Lombok Regency.

Abstract

The advancement of information technology has driven rapid development across various fields, including traditional, digital, and fundamental research methods. This study aims to analyze the effect of brand image, promotion, and online customer reviews on consumer purchase decisions on Shopee in Bekasi City, both partially and simultaneously. Primary data were collected from 200 respondents using a questionnaire with purposive sampling, and analyzed using SPSS through validity and reliability tests, normality, multicollinearity, heteroscedasticity, multiple linear regression, t-test, F-test, and coefficient of determination (R²). The results show that brand image (t = 6.141; p < 0.001) and online customer review (t = 10.195; p < 0.001) have a significant partial effect on purchase decisions, while promotion also shows a significant partial effect (t = 5.289; p < 0.001). Simultaneously, these three independent variables significantly influence purchase decisions with F = 110.198; p < 0.001 and adjusted R² = 0.622, indicating that 62.2% of the variation in purchase decisions can be explained by these variables. These findings confirm that brand image, promotion, and customer reviews play a crucial role in enhancing consumer confidence and purchase decisions on Shopee in Bekasi City.

Abstract

Student satisfaction serves as a fundamental indicator for evaluating the effectiveness of Guidance and Counseling services in educational settings. This study aims to assess student satisfaction levels and perceptions regarding the role of guidance and counseling services in Junior High Schools in Cilegon City. Utilizing a quantitative descriptive approach, data were collected from 311 students selected via random sampling from a population of 7,583. The analysis employed descriptive statistics to measure central tendencies and data distribution. The results indicate that overall student satisfaction falls within the high category (Mean = 3.40), as does the perception of the guidance and counseling service's role (Mean = 3.33). Students responded positively to interpersonal aspects, particularly counselor empathy and the assurance of confidentiality. However, the study identified significant gaps in specific areas, notably the lack of effective academic interventions for low-achieving students and dissatisfaction with the physical comfort of counseling facilities. The findings suggest that while guidance and counseling services are generally effective and well-received, strategic improvements are necessary. To optimize service quality, schools must prioritize proactive academic support, enhance physical facilities, and ensure continuous professional development for counselors. These measures are essential for transforming guidance and counseling into a holistic instrument for student development.
