Abstract

The shrimp fishery has a high economic value. Data from the Ministry of Maritime Affairs and Fisheries (2011) states that the group of marine crustaceans in Indonesia has high potential with production reaching 381,288 tons / yr. From these results more than half of its cultivation. Aquaculture has the risk of which is the onset of the disease caused by viruses, fungi, bacteria, fungi and parasites that cause high levels of mortality and loss of quality of meat. One way to overcome these problems is to use feed supplementation. This study aimed to determine the effect of supplementation of extracts G. verrucosa on feed on shrimp growth (weight and length), specific growth rate, survival rate of shrimp, feed conversion ratio, and Vibrio bacterial populations. The research was conducted through laboratory experimental method to the study design completely randomized design (CRD) with 3 treatments and each treatment with three replications. The treatments tested were feed by supplementing 0.5%, 1% and 2% respectively supplementation performed 3 times repetition. Feed obtained by extraction G. verrucosa, that is by G. verrucosa put in a pot submerged Stainless steel up to boiling and boil for ± 2 hours and obtained extracts G. verrucosa, then performed using a rotary solvent evaporation process to obtain dry extracts G. verrucosa. Further supplementing the feed. Results showed that the extract of G. verrucosa able to improve and control the growth of white shrimp vibrio bacteria populations in the intestines white shrimp (L. vannamei). In this study the condition of seaweed extract supplementation G. verrucosa at a concentration of 2% were able to increase the weight of shrimp growth of 131.43% and a growth of 32.50% shrimp length than controls during 28 days of treatment, as well as being able to control populations of bacteria vibrio, vibrio that reduces the total amount of 78.18% compared to controls at the third week.

Abstract

ABSTRAK : Rajungan (Portunus pelagicus) memiliki nilai ekonomi tinggi, dimana berbanding lurus dengan penangkapan yang meningkat. Tingkat pemanfaatan yang tidak mengindahkan ukuran dan kondisi rajungan dapat mempengaruhi struktur ukuran dan stok rajungan di suatu perairan. Penelitian ini bertujuan untuk melakukan kajian morfometri, rasio jenis kelamin dan tingkat kematangan gonad rajungan yang didaratkan di Kelurahan Pacar, Kabupaten Rembang. Metode yang digunakan dalam penelitian bersifat deskriptif dan eksploratif. Pengamatan rajungan dilakukan pada salah satu pengepul di Kelurahan Pacar. Rajungan yang diteliti berdasarkan morfometri, rasio seksual dan tingkat kematangan gonad selama 30 hari. Informasi ini dapat dijadikan pedoman dalam menentukan kebijakan mengenai pengelolaan perikanan rajungan yang berkelanjutan. Hasil dari penelitian diketahui bahwa ratio kelimpahan rajungan jantan sebesar 45% (1342 ekor); rajungan betina 55% (1658 ekor) dari 3000 ekor sampel rajungan. Rasio perbandingan jantan dan betina 0,80:1 berdasarkan uji chi square. Berdasarkan distribusi ukuran lebar karapas rajungan di perairan Kelurahan Pacar adalah berkisar antara 60–179 mm dan distribusi berat sebesar 23–251 gram. Distribusi tingkat kematangan gonad pada perairan Kelurahan Pacar adalah 558 ekor pada TKG 1; 761 ekor pada TKG 2; dan 339 pada TKG 3. Data menunjukkan bahwa pola pertumbuhan rajungan yang ada di perairan Kelurahan Pacar adalah allometrik negatif yaitu pertumbuhan lebar karapas lebih cepat bertambah daripada berat tubuhnya baik rajungan jantan maupun rajungan betina.ABSTRACT : Blue swimming crab (Portunus pelagicus) has a high economic value, which is causing the crab catch to rise as well. The catch rate that does not consider the size and maturity of sea crab can affect the size structure and population stock of sea crab in waters. This research to study morphometry, sexual ratio and maturity level of the gonads landed in Pacar Village, Rembang Regency.The method used in this study descriptive and exsplorative. Observation of blue swimming crab samples was carried out in one of the collectors in the village. Blue swimming crab was observed based on morphometry and sexual ratio for 30 days. The results of the study revealed that the abundance of male blue swimming crab was 45% (1342 male blue swimming crabs) and the abundance of the female blue swimming crabs was 55%  (1658 female blue swimming crabs) of 3000 blue swimming crab samples. The ratio of male and female   0.80 ; 1 based on the chi square. Based on the size distribution of the width of the blue swimming crab carapace in the waters of Pacar Village it range from 60-179 mm and the distribution of weight 23-251 grams. The distribution of gonad maturity level in the waters Pacar Village is 558 female blue swimming crab at TKG 1; 761 female blue swimming crab at TKG 2; and 339 female blue swimming crab at TKG 3. Data is showed that the blue swimming crab growth pattern in Pacar Village waters was negative allometric both male and female blue swimming crab.

Abstract

Munculnya permasalahan lingkungan akibat plastik sintetik mendorong perlunya penelitian bahan kemasan mudah terurai (bioplastik). Salah satu bahan yang tersedia di alam dan berpotensi sebagai pembentuk bioplastik adalah alginat karena mampu membentuk gel. Bioplastik dari alginat memiliki sifat yang tidak fleksibel sehingga perlu ditambah gliserol sebagai plasticizer. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi alginat dari Sargassum sp. terhadap sifat fisik dan mekanik bioplastik dengan penambahan gliserol dan CaCl2. Materi penelitian ini adalah alginat hasil ekstraksi Sargassum sp. yang diambil dari Perairan Pantai Sundak, Gunung Kidul. Penelitian ini menggunakan metode eksperimental laboratoris yang terdiri dari empat perlakuan konsentrasi alginat (0,5%, 1%, 3% dan 5%)  dengan tiga pengulangan.  Sifat bioplastik yang diuji meliputi sifat fisik (ketebalan, ketahanan air, kehilangan berat) dan sifat mekanik (kuat tarik dan elongasi). Data yang diperoleh dianalisis menggunakan analisis sidik ragam (ANOVA). Hasil penelitian menunjukkan semakin tinggi konsentrasi alginat akan meningkatkan  ketebalan, ketahanan air dan kuat tarik bioplastik namun menurunkan elongasi dan kehilangan berat bioplastik. Konsentrasi alginat berpengaruh terhadap sifat fisik (ketebalan, ketahanan air, kehilangan berat) dan sifat mekanik (kuat tarik  kecuali elongasi), konsentrasi alginat terbaik yang memenuhi Japanese Industrial Standard dan standar SNI 7188.7:2016 adalah 0,5% The emergence of environmental problems due to synthetic plastics has led to the need to research biodegradable packaging materials (bioplastics). One of the ingredients available in nature and has the potential to form bioplastics is alginate because it is capable of forming gel. Bioplastic from alginate has inflexible properties so it needs to be added by glycerol as a plasticizer. This research aims to determine the effect of alginate concentration from Sargassum sp. on the physical and mechanical properties of bioplastics with the addition of glycerol and CaCl2. The method of this research is alginate from Sargassum sp. taken from Sundak Beach Waters, Gunung Kidul. This research used a laboratory experimental method consisting of four treatments of alginate concentration (0.5%, 1%, 3% and 5%) with three repetitions. The properties of the bioplastics tested included physical properties (thickness, water resistance, weight loss) and mechanical properties (tensile strength and elongation). Data obtained were analyzed using variance analysis (ANOVA). The results showed that the higher the concentration of alginate would increased thickness, water resistance and tensile strength of bioplastics but decreased the elongation and weight loss of bioplastics. The alginate concentration affects the physical properties (thickness, water resistance, weight loss) and mechanical properties (tensile strength and except elongation), the best alginate concentrations that meet the Japanese Industrial Standard and SNI 7188.7: 2016 are 0.5%

Abstract

Blue crab is one of the economically marine commodities and have high potential export. During this blue crab requirement derived from nature catches because in blue crab aquaculture not yet good branch out. To low survival rate of blue crab caused by bacterium vibrio disease, in addition to the content of nitrogen and phosphate can also affect the blue crab medium. This research as a purpose to test the effect of probiotic bacillus aplication to water quality and the total number of bacteria in the blue crab medium. the research was conducted with 2 stages, that is isolation and selection bacillus bacteria stages as probiotic and testing phase the effect of probiotic bacillus on water quality and the total number of bacteria. The result showed that probiotic bacillus with density 106 cfu/ml to reduce the content of ammonia (NH3), nitrite (NO2) and the total number of bacteria. While probiotics with density 105 cfu/ml to reduce the total number of bacteria in the blue crab medium, but have not been able to reduce the content of ammonia (NH3) and nitrite (NO2).

Abstract

Aktivitas antibakteri yang terdapat dalam Holothuria atra dapat dimanfaatkan sebagai makanan fungsional yang dapat mengendalikan mikroflora dalam saluran pencernaan. Tujuan dari penelitian ini adalah untuk mengetahui potensi aktivitas antibakteri dari teripang H. atra terhadap bakteri B. cereus (gram positif) dan V. alginolyticus (gram negatif) sebagai foodborne pathogens. Sampel teripang H. atra diambil dari ekosistem lamun perairan Pulau Panjang dengan metode purposive sampling method untuk mengambil teripang berukuran panjang > 16 cm sebanyak 1 kg. Sampel H. atra dibersihkan lalu dipisahkan antara daging dan jeroannya, kemudian di maserasi menggunakan pelarut etil asetat dengan perbandingan 1:3 (w:v) selama 1x24 jam. Hasil maserasi diuapkan dengan rotary evaporator pada suhu 38o C. Ekstrak daging dan jeroan yang diperoleh adalah 669,8 mg dan 625,2 mg. Uji aktivitas antibakteri dilakukan dengan metode paper disk diameter 6 mm dengan konsentrasi 500 µg/disk, 1000 µg/disk, 2000 µg/disk, dan 4000 µg/disk. Aktivitas antibakteri yang diperoleh dari perbedaan konsentrasi yang diberikan berbeda secara nyata  dengan adanya peningkatan Activity Unit (AU) di setiap kenaikan konsentrasi. Aktivitas antibakteri terbesar yang diperoleh dari ekstrak daging H. atra pada konsentrasi 4000 µg/disk terhadap bakteri V. alginolyticus  (gram negatif) dan B. cereus (gram positif) masing-masing adalah 14214,08 mm2/ml dan 10508,62 mm2/ml. Aktivitas antibakteri terbesar yang diperoleh dari ekstrak jeroan H. atra pada konsentrasi  4000 µg/disk terhadap bakteri V. alginolyticus  (gram negatif) dan B. cereus (gram positif) masing-masing adalah 7858,72 mm2/ml dan 4919,68 mm2/ml. Berdasarkan penelitian ini, daging dan jeroan teripang H. atra keduanya memiliki aktivitas antibakteri terhadap bakteri B. cereus (gram positif) dan V. alginolyticus (gram negatif). The antibacterial activity contained in the bioactive compounds of sea cucumber Holothuria atra can be used as functional foods that can control microflora in the digestive tract. The purpose of this study was to determine the potential antibacterial activity of sea cucumber H. atra against B. cereus (gram positive) and V. alginolyticus (gram negative) as foodborne pathogens. The samples of H. atra in this study were taken from the seagass ecosystem of Panjang Island waters using the purposive sampling method so that the sea cucumbers taken were > 16 cm long. The obtained sea cucumber samples were cleaned and separated between the meat and the viscera, then macerated using ethyl acetate with a ration of 1:3 (w/v) for 24 hours. The solvent was evaporated using rotary evaporator at 38o C. The obtained meat and viscera extract of H. atra in this study were 669,8 mg and 625,2 mg. The antibacterial activity test was carried out using the diffusion method with a 6 mm paper disk (Kirby-Bauer Method), and the concentrations of each extract are 500 µg/disk, 1000 µg/disk, 2000 µg/disk, and 4000 µg/disk. The antibacterial activity of extracts from different concentrations were significantly different in line with the increasing results of activity unit (AU) at each increased concentration.  The largest activity unit was obtained from meat extract at a concentration of 4000 µg/disk. The antibacterial activity unit from meat extract against V. alginolyticus (gram-negative) and B. cereus (gram-positive) are 14214,08 mm2 / ml and 10508,62 mm2 / ml. The largest antibacterial activity unit of viscera extract against V. alginolyticus and B. cereus are 7858,72 mm2 / ml and 4919,68 mm2 / ml. Based on this study, meat and viscera extracts from H. atra both have antibacterial activity against B. cereus (gram-positive) and V. alginolyticus (gram-negative).

Tingkat pemanfaatan yang tidak konservatif akan mempengaruhi  ukuran dan kondisi rajungan di suatu populasi, serta stok rajungan di suatu perairan. Penelitian bertujuan untuk mengetahui kajian konservasi rajungan (Portunus pelagicus) berdasarkan morfometri dan sex ratio yang didaratkan oleh nelayan  di Perairan Desa Gegunung Wetan, Kecamatan Rembang, Kabupaten Rembang. Metode yang digunakan dalam penelitian bersifat deskriptif. Pengamatan rajungan dilakukan pada salah satu pengepul di Desa. Rajungan di amati berdasarkan morfometri dan sex rasio selama 30 hari. Hasil Informasi dapat dijadikan pedoman dalam menentukan kebijakan mengenai pengelolaan perikanan rajungan yang berkelanjutan dan bersifat konservatif. Hasil dari penelitian diketahui bahwa kelimpahan rajungan jantan sebesar 46% (1379 ekor) rajungan betina 56% (1621 ekor) dari 3000 ekor sampel rajungan. Rasio perbandingan jantan dan betina 1:1,18. Berdasarkan distribusi ukuran lebar karapas rajungan di perairan Rembang adalah berkisar anatara 72–167  mm dan distribusi berat sebesar 40–303 gram. Distribusi tingkat kematangan gonad pada perairan Rembang adalah 608 ekor pada TKG 1; 658 ekor pada TKG 2; dan 355 pada TKG 3. Data menunjukan bahwa pola pertumbuhan rajungan yang ada di perairan Rembang adalah allometrik negatif  baik rajungan jantan maupun rajungan betina. Abstract: Blue swimming crab (Portunus pelagicus) has a high economic value, directly proportional to the increased catch. The level of non-conservation utilization will affect the size and condition of the blue swimming crab in a population, as well as the blue swimming crab stock in a waters. This study is aimed to determine the blue swimming crab conservation study (Portunus pelagicus) based on morphometry and sex ratio brought by fishermen in the waters of Gegunung Wetan Village, Rembang District, Rembang Regency. The method used in this study is descriptive. Observation of blue swimming crab samples was carried out in one of the collectors in the village. Blue swimming crab was observed based on morphometry and sex ratio for 30 days. Informative results can be used as a guide in determining policies regarding sustainable and conservation management of blue swimming crab fisheries. The results of the study revealed that the abundance of male blue swimming crab was 46% (1379 male blue swimming crab) and the abundance of the female blue swimming crab was 56%  (1621 female blue swimming crab) of 3000 blue swimming crab samples. The ratio of male and female  is 1:1.18. Based on the size distribution of the width of the blue swimming crab carapace in the waters of Getanung Wetan it ranges from 72-167 mm and the distribution of weight is 40-303 grams. 13.8% (413 blue swimming crab) sized≤10 cm and 86.2% (2587 blue swimming crab) sized ≥10 cm. The distribution of gonad maturity level in the waters of Gegunung Wetan is 608 female blue swimming crab at TKG 1; 658 female blue swimming crab at TKG 2; and 355 female blue swimming crab at TKG 3. There were 355 female blue swimming crab (22%) of the total female blue swimming crab laying eggs. The data showed that the blue swimming crab growth pattern in Gegunung Wetan waters was negative allometric both male and female blue swimming crab.

Abstract

Infeksi bakteri patogen menjadi masalah kesehatan dunia. Antibiotik untuk penanggulangan infeksi menjadikan bakteri resisten karena pemakaiannya tidak sesuai dosis. Permasalahan ini dapat diatasi salah satunya dengan pencarian sumber antibiotik baru termasuk diantaranya pada rumput laut. Penelitian ini bertujuan untuk menentukan aktivitas antibakteri ekstrak metanol G. verrucosa terhadap Escherichia coli, Bacillus cereus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Vibrio anguillarum, dan Vibrio alginolyticus. Penelitian dilakukan dengan metode eksperimental laboratoris. Sampel rumput laut hasil budidaya Balai Besar Perikanan Budidaya Air Payau (BBPBAP) Jepara dikeringkan kemudian dimaserasi dengan metanol selama 2x24 jam. Aktivitas antibakteri ekstrak diuji pada konsentrasi 100 µg/disk, 50 µg/disk, 25 µg/disk, dan 10 µg/disk kemudian dilakukan uji fitokimia. Hasil uji aktivitas antibakteri menunjukkan bahwa ekstrak metanol G. verrucosa tidak memiliki aktivitas antibakteri. Uji fitokimia menunjukkan ekstrak mengandung flavonoid, saponin, steroid, dan triterpenoid. Pathogenic bacterial infection become a world health problem. Antibiotics for prevention of infection make bacteria resistant due to the incorrect dosage. One of these problems can be overcome by finding a new source of antibiotics including seaweed. This study aims to determine the antibacterial activity of methanol extract of G. verrucosa against Escherichia coli, Bacillus cereus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Vibrio anguillarum, and Vibrio alginolyticus. Laboratory experimental method was used in this study. Seaweed samples from the cultivation at Brackish Water Aquaculture Center (BBPBAP) Jepara were dried and then macerated with methanol for 2x24 hours. The antibacterial activity of the extract was tested at concentration of 100 µg / disk, 50 µg / disk, 25 µg / disk, and 10 µg / disk then phytochemical tests were carried out. The results of antibacterial activity test showed that G. verrucosa methanol extract did not have antibacterial activity. The phytochemical test showed that seaweed extract contained flavonoids, saponins, steroids, and triterpenoids

Abstract

ABSTRAK: Perairan Laut Kendal terhubung dengan Sungai Buntu, Sungai Kendal, dan Sungai Blorong yang membawa limbah dari kegiatan di darat seperti Mandi Cuci Kakus (MCK), peternakan, tambak, pertanian, dan kegiatan kapal dan Tempat Pelelangan Ikan (TPI). Hal ini akan mempengaruhi kualitas bakteriologis perairan laut Kecamatan Kendal. Penelitian ini dilakukan untuk menganalisa kualitas bakteriologis air dan sedimen perairan laut Kecamatan Kendal berbasis jumlah total bakteri dan Coliform. Sampel air laut dan sedimen diambil pada bulan Oktober Tahun 2019. Analisa bakteriologis dilakukan di Laboratorium Balai Pengujian Mutu Hasil Perikanan Semarang menggunakan metode berdasarkan ISO 6222: 1999 dan SNI 01-2332.3 – 2015, SNI ISO 9308 –1: 2010 dan SNI 01-2332.1 – 2006. Hasil penelitian menunjukkan jumlah total bakteri pada sampel air laut di stasiun 1 berkisar antara 190 Cfu - 240 Cfu, di stasiun 2 antara 340 Cfu - 380 Cfu. Jumlah total Coliform pada sampel air laut di stasiun 1 dan 2 adalah 1 Cfu/ 100 ml. Jumlah total bakteri pada sampel sedimen di stasiun 1 dan 2 yaitu antara 19.000 Cfu - 90.000 Cfu dan jumlah total coliform di kedua stasiun tersebut sebesar 3,6 MPN/g Coliform. Nilai total Coliform ini berada dibawah baku mutu air laut untuk biota laut, yang maksimum adalah 1.000 dengan toleransi <10%. Sedangkan untuk jumlah total bakteri tidak diatur dalam baku mutu air laut. Tetapi secara umum jumlah total bakteri di air laut terdapat 106 sel/ ml. Sehingga nilai hasil analisis total bakteri menunjukkan kualitas air laut yang masih baik. ABSTRACT: Kendal sea waters are connected to the Buntu River, Kendal River, and the Blorong River which carry waste from human activities such as domestic waste, fishery, agriculture, ship activities, and fish auction activities. These activities can influence the bacteriological quality of the waters of Kendal District. This research was conducted to analyze the bacteriological quality of seawater and sediments from Kendal Seawaters based on the total number of bacteria and Coliform. Seawater and sediment samples were taken on October 2019. Bacteriological analysis was conducted at the Semarang Fisheries Product Quality Testing Laboratory (BPMHP) using methods based on ISO 6222: 1999 and SNI 01-2332.3 - 2015, SNI ISO 9308-1: 2010 and SNI 01-2332.1 - 2006. The results showed the total number of bacteria in sea water samples at station 1 ranged from 190 Cfu - 240 Cfu, at station 2 between 340 Cfu-380 Cfu. The total amount of Coliform in seawater samples at station 1 and 2 are 1 Cfu / 100 ml. The total number of bacteria in sediment samples at station 1 and 2 are between 19,000 Cfu - 90,000 Cfu and the total number of coliforms at both stations are 3.6 MPN / g Coliform. The total value of this coliform is below the sea water quality standard for marine life, the maximum is 1,000 with a tolerance of <10%. Where as the total number of bacteria is not regulated in the seawater quality standard. But in general the total number of bacteria in seawater is 106 cells / ml. So the value of the total bacterial analysis shows that the quality of sea water is still good.

Abstract

Kesenjangan antara kepatuhan formal dan kecukupan substantif dalam dokumen Analisis Mengenai Dampak Lingkungan (AMDAL) menjadi isu kritis yang belum banyak dikaji, khususnya pada kegiatan pelabuhan. Belum ada studi yang secara khusus menganalisis kecukupan substantif evaluasi dampak potensial pada KA-ANDAL kegiatan pelabuhan menggunakan kerangka regulasi PP No. 22/2021 dan Kepmen LHK No. 137/2024. Penelitian ini menganalisis kesenjangan tersebut pada kolom evaluasi dampak potensial dari tiga Formulir Kerangka Acuan (KA-ANDAL) kegiatan pelabuhan kewenangan pusat tahun 2024–2025. Menggunakan metode kualitatif-deskriptif dengan pendekatan analisis isi, setiap dampak dievaluasi berdasarkan empat kriteria yang diamanatkan PP No. 22 Tahun 2021 dan Kepmen LHK No. 137 Tahun 2024: (1) besaran kegiatan dan pengelolaan yang direncanakan, (2) kondisi rona lingkungan terdampak, (3) pengaruh terhadap kegiatan sekitar, dan (4) intensitas perhatian masyarakat. Hasil menunjukkan paradoks yang signifikan: Dokumen A memenuhi kelengkapan formal 100% namun hanya 4,17% kriteria dinilai tepat secara substantif; Dokumen B menunjukkan profil heterogen dengan 95,24% lengkap dan 47,62% tepat; sementara Dokumen C mencapai 100% ketepatan namun dengan kolom rona lingkungan yang hampir seluruhnya kosong. Pola ini membuktikan bahwa kepatuhan prosedural tidak menjamin kecukupan substantif. Sumber utama variasi ini adalah ambiguitas panduan teknis dalam regulasi, khususnya tidak adanya standar minimum pengisian setiap kriteria. Penelitian ini merekomendasikan penyusunan panduan teknis sektoral yang operasional untuk kegiatan pelabuhan sebagai langkah konkret mencegah luputnya dampak penting dari proses penetapan Dampak Penting Hipotetik (DPH).   The gap between formal compliance and substantive adequacy in Environmental Impact Assessment (EIA) documents remains a critical yet underexplored issue, particularly for seaport activities. No prior study has specifically examined the substantive adequacy of potential impact evaluation in seaport EIA Terms of Reference (ToR) documents using the current regulatory framework of Government Regulation No. 22/2021 and Ministerial Decree No. 137/2024. This study analyzes this gap in the potential impact evaluation columns of three EIA ToR documents for central-government-authorized seaport projects from 2024 to 2025. Using a descriptive qualitative content analysis approach, each identified impact was assessed against four criteria mandated by these regulations: (1) activity scale and planned management, (2) baseline environmental conditions, (3) influence on surrounding activities, and (4) level of public concern. Results reveal a striking paradox: Document A achieved 100% formal completeness yet only 4.17% substantive accuracy; Document B showed a heterogeneous profile with 95.24% completeness and 47.62% accuracy; while Document C attained 100% accuracy despite having nearly empty baseline environmental data columns. This pattern demonstrates that procedural compliance does not guarantee substantive adequacy. The primary driver of this variability is the ambiguity of technical guidance within existing regulations, particularly the absence of minimum content standards for each evaluation criterion. This study recommends the development of operationally specific sectoral technical guidelines for seaport EIA to prevent the omission of significant impacts from the Significant Impact Hypothesis (DPH) determination process.
