Abstract

Growing evidence suggests that microRNAs (miRNAs) play a pivotal role in viral infection. The objective of this study was to assess the association between the expression of miR-150, hsa-let-7e, and miR-146a on cytokine expression during dengue infection. Dengue virus (DENV) strain SJN-006, a serotype 2 DENV strain of the Cosmopolitan genotype, isolated in Bali, Indonesia, was used to infect peripheral blood mononuclear cells (PBMCs) isolated from healthy individuals. The relative gene expressions of miR-150, hsa-let-7e, and miR-146a as well as the gene expression of cytokines (IL-6, IL-8, IP-10, and MIP-1β) were determined using quantitative real time - polymerase chain reaction (qRT-PCR) at 6, 12 and 24 hours post infection (hpi). Correlations between the microRNAs and cytokines were analyzed by means of causality tests. Our data suggests that miR-150 and hsa-let-7e were significantly higher in infected-PBMCs after 12 hpi compared to the uninfected-PBMCs (p<0.05). The causality tests demonstrated that miR-150 and hsa-let-7e were negatively correlated with IL-8 expression, meanwhile miR-146a was the contrast. DENV infection was negatively and positively correlated with miR-150 and hsa-let-7e, respectively, after 24 hpi. In conclusion, our data demonstrates the vital role of miR-150, hsa-let-7e, and miR-146a in regulating IL-8 expression with possible different pathways.

Abstract

Inability to understand the pathogenesis of severe dengue, in particular the control mechanism of immune responses, has led to high mortality rate for patients with dengue shock syndrome (DSS). The aim of this study was to determine the control mechanism of cytokine production by mediator suppressor of cytokine signaling (SOCS), toll-like receptor 3 (TLR-3) and nuclear factor kappa B (NFκB) during DENV infection. Peripheral blood mononuclear blood cells (PBMC), isolated from healthy individuals, were infected with dengue virus (DENV)-2 strain SJN-006 Cosmopolitan genotype (isolated from Bali, Indonesia). The relative gene expression of SOCS-3, TLR-3, NFκB, and the cytokine genes (interleukin (IL)-6, IL-8, interferon inducible protein 10 (IP-10), and macrophage inflammatory protein-1 beta (MIP-1β)) were measured using qRT-PCR at 6, 12 and 24 hours post infection (hpi). Student t-test and Mann-Whitney test were used to compare the gene expressions while causal correlations were analyzed using regression test and path analyses. DENV-2 infection increased the gene expression of SOCS-3, TLR-3, and NFκB after 12 and 24 hpi. The expression of IL-6, IL-8, IP-10, and MIP-1β genes was increased and peaked at different times post-infection. NFκB and SOCS-3 genes likely have role in the upregulation of IL-8 and IL-6 gene expression, respectively. MIP-1β gene expression was significantly induced by both NFκB and SOCS-3. In conclusion, our study suggested that SOCS-3, TLR-3, and NFκB are important in regulating the production of IL-6, IL-8, IP-10, MIP-1β during early phase of DENV-2 infection. This enriches our understanding on pathogenesis pathway of DENV-associated cytokine storm.

Abstract

This study aims to determine the implementation of ice breaking on student learning motivation. The research method used is Experiment. The population of grade 5 SDN 1 Wanayasa was 32 students, sampling was carried out using the Total Sampling technique, the research sample amounted to 32 students. The research instrument used a questionnaire (questionnaire). Hypothesis testing using paired Sample T test, which is to test the normality and homogeneity of the research data Because the significance value is less than 0.0020 <0.05, it can be concluded that the research data is accepted. The results of the increase from the Pre-test and Post-test increased by 5.7%. So it can be concluded that ice breaking can increase student learning motivation. So that it can spur students to be more active in the learning process at school.

Abstract

Micro businesses play a crucial role in Indonesia's economy. Their significance in economic recovery is underscored by their substantial contribution to the labor sector. The community engagement partner, "Katbag," specializes in the production of bags and accessories, also souvenir and corporate merchandise. Partnering with Katbag was motivated by the competitiveness of its offerings and high entrepreneurial drive. However, despite their product excellence, Katbag faces challenges and limitations, particularly in business administration, HR, and marketing. This program aims to assist in enhancing the partner's capacity in terms of business planning and digital marketing so that they can develop their business further. The method involves planning, implementation (training in digital marketing and business administration), along with rigorous monitoring and evaluation. After participating on the program, the partner has gained knowledge in business administration and website management to build digital presence and reach more consumers online.

Abstract

Background: Treating diabetic foot ulcers (DFU) sometimes takes a long time. During treatment, the patient's feet are usually given a cushion (such as a sleeping pillow or bolster) to make the wound seen. This way, some patients reported that their feet tingled because they were not positioned rightly for an extended period. So far, no unique pillow is available for this function. This pillow would help in treating DFU and should be comfortable for patients. Thus, it is essential to research designing such a product.Purpose: To develop and test a special pillow for treating DFU.Methods: This research adopts Research and Development (R&D). This research is divided into two stages: designing and developing and product testing.Results: This pillow is made of multiplex and foam, covered with Polyester fabric. It is 75mm in length and 45mm in width. It is equipped with a foot care area and waste storage tank. The test results showed that the score for comfort for the patients was 92%, while the score from the nurse was 88%.Conclusion:The pillow product helps the nurse conduct the DFU treatment and is comfortable for the patients.

Abstract

Latar Belakang: Musculoskeletal disorders (MSDs) dapat menyerang seseorang yang bekerja dalam posisi statis yang cukup lama, salah satunya yaitu para pekerja batikTujuan: Penelitian ini bertujuan untuk menganalisis faktor-faktor apa sajakah yang berhubungan dengan kejadian musculoskeletal disorders pada pekerja batik di Kecamatan Sokaraja Banyumas.Metode: Penelitian ini menggunakan metode analitik korelasi, populasi penelitian adalah para pekerja batik di Kecamatan Sokaraja Kabupaten Bayumas, Jawa Tengah, jumlah berjumlah 45 responden pekerja batik yang mengalami masalah MSDs. Variable yang diukur dalam penelitian ini adalah usia, jenis kelamin, lama kerja, jam kerja perhari, posisi kerja dan banyaknya keluhan MSDs. Instrument penelitian menggunakan Nordic Body Map dan Rapid Upper Limb Assessment (RULA). Analisis data menggunakan Pearson correlation.Hasil: Hasil penelitian menunjukan ada hubungan yang negative antara usia dengan keluhan MSDs (r: -0,327; p<0,05). Ada hubungan yang positif antara jenis kelamin dengan keluhan MSDs (r: -0,379; p<0,05). Ada hubungan yang negative antara lama kerja dengan keluhan MSDs (r: -0,301; p<0,05). Ada hubungan yang positif antara jam kerja perhari dengan keluhan MSDs (r: 0,445; p<0,01) dan ada hubungan yang positif antara posisi kerjai dengan keluhan MSDs (r: 0,642; p<0,01).Kesimpulan: Berdasarkan hasil penelitian dapat disimpulkan bahwa usia, jenis kelamin, lama kerja, jam kerja perhari dan posisi kerja berhubungan dengan keluhan MSDs.

Abstract

Background: Symptoms experienced hemorrhoid sufferers are feeling pain in the anus, especially in sitting position. It is necessary to innovate special underwear which can reduce pressure on the anus and the pain experienced by the patientObjective: the study aimed to create and analyze special underwear for hemorrhoid sufferers which increased comfort and reduce the intensity of pain.Methods: This research imitated Research and Development (R&D). It consisted of 3 stages; stage I (literature study), stage II (product development) and stage III (product experiment).Result: The literature study in stage I obtained materials used to create special underwear for hemorrhoid sufferers by Cotton Combed 30s of 100% premium cotton and foam as a cushion. In the second phase of the research, designs and shapes were obtained. The results of product experiment in the third phase of the study discovered that respondents felt more comfortable by the special underwear than ordinary underwear and it was very effective in reducing pain.Conclusion: The innovation of hemorrhoid underwear is effective to increase comfort and reduce pain intensity in hemorrhoid sufferers.

Handling stunting should not only focus on the nutrition fulfillment sector. A fundamental factor to consider as the root cause of high stunting in Indonesia is unwanted or unplanned pregnancies.

Abstract

Background: The air bubbles in the intravenous tubing can be fatal if it enters the body. It can result in an air embolism. Air embolism is a condition in which air bubbles enter the blood vessels. Several methods can be used to remove air bubbles in the intravenous tubing. One is replacing it with new intravenous tubing, but this method is considered less efficient. This article describes effective and efficient ways to remove air bubbles from intravenous tubing and tests them for effectiveness.Technique: Four techniques can be applied when the intravenous tubing is filled with air, namely the roll-up technique, flick technique, combination roll-up and flick technique, and push-pull technique.Conclusion: From the test, the push-pull technique is the most effective and efficient in removing air from intravenous tubing compared to the others.
