Abstract

This study aimed to screen and characterize halotolerant bacterial isolates, which could enhance plant growth performance in saline soil. Halotolerant bacteria was isolated from weeds rhizosphere grown in saline soil of coastal agricultural land located at Brondong District, Lamongan Regency, East Java Province, Indonesia. This research was conducted from June to September 2018. Seven bacterial isolates can grow in a Nutrient Agar medium containing 10% of NaCl, suggesting that these bacteria were halotolerant. Furthermore, all bacterial isolates were shown to produce indol acetic acid (IAA) and do not induce a hypersensitive response when infiltrated into tobacco leaves. These results explain that these bacteria had potency as plant growth-promoting rhizobacteria (PGPR) and were not tend to be the plant pathogen. The growth of seedlings when inoculated in cucumber seed grown in saline media were higher than those in control. This result suggests that the halotolerant bacteria can enhance the development of cucumber seedlings in saline stress conditions. Three potential halotolerant bacteria i.e., SN22, SN23, SN26 were selected and molecularly identified as Bacillus megaterium, Bacillus sp., and B. megaterium, respectively.

Abstract

The agroforestry system with its diversity of vegetation has the potential on the existence of antagonistic and entomopathogen fungi. This study was conducted to evaluate the diversity of antagonistic fungi and entomopathogen fungi in the rhizosphere and pine leaf litters on pine monoculture and intercropping pine - coffee. The research was started from plot determination, sampling, fungal identification, antagonist test and pathogenicity test. The results of isolation of fungi from pine leaf litter on monoculture pine and intercropping pine-coffee fields obtained 17 genus of fungi. In monoculture pine, there 9 genus of fungi were found, while other 4 were still unidentified. In pine-coffee intercropping land 13 genus were observed. Based on the potential and bility tests, Acremonium sp. 3 and Penicillium sp. 2 has the highest inhibiton capacity, while isolate Paecilomyces sp. 1 and Paecilomyces sp. 2 had the best level of pathogenicity and mortality. Temperature and humidity did not affect the diversity of fungi. The diversity of entompathogenic and antagonistic fungi was higher in the pine-coffee intercropping land use. The litter plots had higher fungal diversity than the rhizosphere.

Abstract

This study aims to examine several inoculation methods for pathogenisity test ofBDB on banana fruit. The Results showed that injection method and smearingsuspension bacteria using cotton bud on the base of fruit was capable in producingsymptoms of blood disease on banana fruit. The injection method is shown to bethe most effective method because it caused blood disease symptoms in thebanana fruit at the first day after inoculation. Inoculums densities of 108 and 109CFU/ml of BDB could develop disease on third and sixth day after inoculation.BDB produced unique symptoms by causing yellowing and browning on the fleshof a banana fruit as well as browning at the fruit placenta in 7 days afterinoculation, the symptom is different with caused by other pathogen like asXanthomonas oryzae, Pantoea sp dan Erwinia carotovora, dan Ralstoniasolanacearum.Keywords: banana, Blood Disease Bacterium (BDB), inoculation

Abstract

Potato (Solanum tuberosum L.) is one of the commodities of potential as a sourceof carbohydrates and important on Indonesia's economy. One of the constraints ofpotato production is soft rot disease caused by Erwinia carotovora (syn.Pectobacterium carotovorum) which is needed to be controlled. One alternativecontrol measures is the used of bacteria Bacillus subtilis and Pseudomonasfluorescens for biological control. The purpose of this research is to know thepotential of B. subtilis and P. fluorescens in single and in combination to inhibitpotato soft rot diseases caused by E. carotovora. The results showed thatbiological control agents B. subtilis and P. fluorescens in single or in combinationboth of them (T3 to T17) have potential in inhibiting the growth of pathogenic E.carotovora by showing the clear zone. Biological agents provides nearly the sameeffect if compared with the bactericide Agrept (T2). Based on the results of thestatistical analysis, the treatment 11th and treatment 14th showed a clear zonewider than the Agrept treatment (T2). The treatment 11th contain combination ofbiological agents B. subtilis (109 cfu/ml) and P. fluorescens (105cfu/ml), while the treatment 14th contain combination of biological agents B.subtilis (107 cfu/ml) and P. fluorescens (105 cfu/ml).Keywords: Erwinia carotovora, Bacillus subtilis and Pseudomonas fluorescens

Abstract

ABSTRACTThe objective of this research was to study the variance of hybrid ricegenotypes that showed resistance to leaf blast disease caused by Pyriculariaoryzae Cavara and got genotypes refined of the resistance . The research wasconducted at the Department of Disease Laboratory of Plant Pests and Diseases,Faculty of Agriculture, Univeristy of Brawijaya and in the P.T. DuPont’s researchfield in Ketawang Village, Bululawang, Malang since April until June 2012. Thisresearch used Randomized Block Design with 3 replications and 80 treatments.Eighty-treatments consisted of 24 restorer genotypes, 2 CMS genotypes, 24hybrid A genotypes, 24 hybrid B genotypes and 6 control genotypes (Maro,Hipa8, LP27P31, LP27P53, Ciherang and IR64). The results showed that therewere occurrence of intermediate resistance on 24 hybrid genotypes A and B.Twenty four hybrid A genotypes showed resistance reaction with moderateclassification which was equivalent to IR64 as the best control genotypes.Key word : Resistance, Leaf blas, Pyricularia oryzae Cavara

Abstract

ABSTRACTTomato (Lycopersicum esculentum) is an annual fruit plant that can be planted on high or low-land. Tomato has many beneficial vitamins and minerals to fulfillhuman body need for health. Tomato contains vitamins and minerals in amount of 30 calories, vitamin C 40 mg, vitamin A 1.500 SI, zinc, and calcium. Main cause of areal cultivation reduction are land degradation, erosions, and high intensity of bacterial wilt disease Ralstonia solanacearum that was the main consequences of perpetually tomato cultivation. Liquid fertilizer application gives some alternate way to control bacterial wilt disease Ralstonia solanacearum that also known as pathogen soil infection. Liquid fertilizer that used in this research is made from amino acid residual process. Purpose of this research is to observe the effect of liquid fertilizer giving that applied to the soil and leaves towards tomato plant growth. Result of this research shows that liquid root or foliar fertilizer applicationeffects significantly on plant height and number of leaf parameters of alltreatments and all observation time.Keywords: tomato, Ralstonia solanacearum, liquid fertilizer

Abstract

ABSTRAK Bulai merupakan penyakit penting pada tanaman jagung yang disebabkan oleh jamur patogen Peronosclerospora maydis, dengan tingkat serangan mencapai 95%. PGPR diketahui pengendali patogen hayati dengan menghasilkan berbagai senyawa atau metabolit seperti siderophore, β-1,3 glukanase, kitinase, antibiotik dan sianida serta dapat meningkatkan pertumbuhan tanaman dengan meningkatkan konsentrasi AIA, giberelin dan sitokinin.Penelitian bertujuan untuk mengetahui pengaruh PGPR terhadap penyakit bulai pada tanaman jagung. Penelitian menggunakan isolat Bacillus sp.danPseudomonas sp. serta fungisida berbahan aktif dimetomorf 50%. Perlakuan isolat Bacillus sp.(UB-ABS 4 dan 5) dapat menekan penyakit sebesar 50% sedangkan Pseudomonas sp. (UB-APF2) sebesar 40%. Kandungan khlorofil tertinggi pada Pseudomonas sp. UB-APF2 sebesar 32,03. Berat segar akar tidak berbeda nyata tetapi pada berat kering akar tertinggi pada perlakuan Pseudomonas sp. (UB-APF 5) sebesar 2,56 gram dan akar terpanjang terdapat pada Pseudomonas sp. (UB-APF2) sebesar 49,77 cm. Kata kunci: Tanaman jagung, PGPR, dan Penyakit Bulai.

Abstract

ABSTRAK   Bibit tanaman durian yang dibudidayakan di Desa Plangkrongan beberapa yang terserang penyakit dan mengalami kematian bibit. Jenis patogen yang menyebabkan penyakit pada bibit tanaman durian belum diketahui. Pengendalian penyakit yang ramah lingkungan diperlukan untuk menjaga keseimbangan ekosistem. Salah satu pengendalian yang ramah lingkungan dengan cara menggunakan agen pengendali hayati seperti bakteri antagonis. Penelitian ini bertujuan untuk mengetahui jenis patogen yang menyebabkan kerusakan pada bibit tanaman durian di Desa Plangkrongan, dan mengetahui potensi bakteri antagonis untuk mengendalikan patogen tersebut secara in vitro. Penelitian ini dilakukan dengan beberapa tahap, yaitu tahap pengamatan gejala dan intensitas serangan, tahap isolasi patogen, uji patogenisitas, identifikasi patogen, dan uji antagonis  bakteri Bacillus sp. (UB-ABL1), B. subtilis (UB-ABS1), dan Pseudomonas fluorencens (UB-APF1) terhadap pathogen secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL), dengan 4 perlakuan, masing-masing perlakuan diulang sebanyak lima kali. Analisis data dilakukan dengan uji F dengan tingkat kesalahan sebesar 5%. Hasil penelitian menunjukkan penyakit penting yang ditemukan pada pembibitan tanaman durian di Plangkrongan adalah hawar daun yang disebabkan oleh patogen yang berupa jamur Fusarium sp. dan bercak daun yang belum diketahui penyebabnya. Bacillus sp. (UB-ABL1), dan B. subtilis (UB-ABS1) efektif menghambat pertumbuhan jamur Fusarium sp. dalam uji antagonis secara in vitro, sehingga berpotensi dikembangkan sebagai agen hayati untuk pengendalian  Fusarium sp. penyebab penyakit hawar daun pada bibit tanaman durian. Abstract

ABSTRAKSalah satu penyakit penting yang menyerang pertanaman cabai adalah penyakit antraknosa yang disebabkan oleh jamur Colletotrichum sp. Penelitian ini bertujuan untuk mengetahui potensi konsorsium mikroba antagonis yang terdiri dari Bacillus subtillis, Pseudomonas fluorescens, serta Trichoderma harzianum sebagai agens hayati untuk mengendalikan penyakit antraknosa pada cabai merah besar yang disebabkan oleh cendawan C. capsici. Berdasarkan hasil penelitian, pengujian konsorsium mikroba antagonis secara nyata dapat menekan perkembangan jamur C. capsici. Konsorsium mikroba antagonis dengan dosis aplikasi 30 ml/liter memberikan hasil terbaik dalammenekan perkembangan penyakit antraknosa pada buah cabai besar dibandingkan dengan dosis 10 ml/liter dan 20 ml/liter serta isolat mikroba antagonis secara individu. Hasil percobaan pada buah cabai besar di laboratorium dan di lapang menunjukkan perlakuan konsorsium mikroba antagonis dengan dosis 30 ml/l lebih tinggi dalam menekan perkembangan penyakit antraknose dibandingkan perlakuan yang lain kecuali perlakuan fungisida dengan bahan aktif propineb 70%.Kata Kunci : antraknosa, antagonis, mikroba, konsorsium

Abstract

ABSTRAK Tujuan penelitian ini untuk mengetahui pengaruh pemberian kompos terhadap perkembangan penyakit busuk hati yang disebabkan oleh jamur patogen Phytophthora sp. Hasil survei ditemukan satu lokasi 45F yang banyak terserang penyakit busuk hati. Hasil analisis tanah menunjukkan kelembaban tanah di lokasi 45F lebih tinggi dibanding lahan pada lokasi yang tidak terserang penyakit busuk hati. Hasil percobaan rumah kaca menunjukkan bahwa penambahan kompos dapat memperlambat munculnya gejala penyakit busuk hati (masa inkubasi penyakit busuk hati menjadi lebih panjang). Pemberian kompos dengan dosis yang berbeda tidak berpengaruh dalam menekan perkembangan penyakit busuk hati. Terdapat hubungan erat antara populasi bakteri dengan tingkat penekanan penyakit busuk hati. Terdapat hubungan antara pH, kandungan N-total, dan kandungan C/N rasio dalam tanah dengan persentase penyakit. Semakin tinggi populasi bakteri, pH dan C/N rasio, persentase penyakit semakin kecil. Sebaliknya semakin tinggi kandungan N-total tanah, semakin tinggi persentase perkembangan penyakit busuk hati.   Kata kunci: Nanas, Kompos, Penyakit busuk hati, Phytophthora sp.
