Abstract

Sebagai salah satu pangan pokok rakyat Indonesia, kenaikan harga beras yang terjadi belakangan ini tentunya memberikan dampak yang signifikan dan respon yang beragam. Secara lebih sederhana, penyebab kenaikan harga beras di hampir seluruh wilayah negeri dapat dilihat dari neraca supply dan demand-nya.

Abstract

Efficiency and effectiveness in controlling crossbreeding are pivotal in yielding superior hybrid varieties with desired traits such as genetic purity and high productivity. Cytoplasmic Male Sterile (CMS) is a crucial component in hybrid variety assembly. This study aims to identify the genotypes that can induce male sterile lines based on genetic diversity and the level of pollen sterility in the prospective male sterile genotypes tested, consisting of seventeen genotypes. The research findings revealed that the genetic diversity among the seventeen observed male-sterile candidate genotypes tends to vary in quantitative traits. Among the quantitative traits, JHD14 exhibited indications of being a male sterility inducer compared to the other genotypes observed, demonstrating a value of 0.00 g in terms of pollen weight. This is further substantiated by the microscopic examination of JHD14 pollen grains, which displayed 100% sterility.

Abstract

Foxtail millet hybrid development encounters challenges due to its self-pollinating nature, small florets, and tool availability for confirming true hybrid. Our research aimed to assess flowering phenology and explore artificial hybridization employing SiDREB2 based-SNAP marker. The research consisted of three experiments. The first experiment focused on the panicle phenology of foxtail millet, while the second experiment focused on floret phenology and reproductive organs. The artificial hybridization successfulness was validated with SNAP markers based on the SiDREB2 gene in the third experiment. Observations on two genotypes (ICERI-5 and ICERI-6) revealed panicle initiation at 49–69 days after planting (DAP), with anthesis occurring 6 days after panicle emergence (DAPE). Florets exhibited the formation of reproductive organs at 3 DAPE, delineating distinct stages of pollen development leading up to anthesis. Our findings emphasized the significance of implementing male-sterile induction treatment at 4 DAPE to regulate pollen development for successful hybridization by 6 DAPE. The hybrid evaluation revealed varied germination rates and confirmed hybrid percentages across different crossing series, validated by SiDREB2-based SNAP marker. ICERI-5×Botok-4 exhibited high germination but a low confirmed hybrid percentage, while Botok-4×ICERI-5 and Botok-10×ICERI-6 showed the opposite trend. Reciprocal crossings showed unilateral incompatibility in confirmed hybrid percentage.

Abstract

Bacterial leaf blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo) is a significant disease attacking rice crops worldwide. This disease attacks at various stages of plant growth and causes significant yield loss. Breeding new varieties resistant to BLB is important to support sustainable agriculture in the future. This study aimed to identify new superior green super rice (GSR) lines resistant to BLB disease. The experiment evaluated the resistance of lowland rice lines obtained from anther culture using a factorial randomized complete block design. The 1st factor was genotype, consisting of 20 lines, 2 checks of commercial varieties (Inpari 42 Agritan GSR and Inpari 18), a resistant check (Conde), and a susceptible check (Taichung Native 1). The 2nd factor was BLB pathotypes (i.e., III, IV, and VIII). Quantitative data on disease severity and severity index were analyzed using analysis of variance and t-Dunnett test at 5% level. The results showed that the interaction between genotype and pathotype affected the disease severity and severity index in both growth phases. The tested lines exhibited varying resistance, from susceptible to resistant, to BLB. Four lines (SN 11, 13, 57, and 58) showed moderate to resistant criteria for BLB disease of 3 pathotypes in both growth phases. The selected lines can be used as a source of parents for breeders and candidates for new superior varieties with BLB resistance properties to support the reduction of synthetic chemical bactericide inputs and control BLB disease. However, further field evaluations are necessary to assess their performance.

Abstract

Areca nut (Areca catechu) belongs to the monocotyledonous and palm family of plants known as betel nut or supari plants. The Center of origin areca nut comes from India. Areca nut is a cross-pollinated plant with a heterozygous heterogeneous genetic constitution. Next-generation sequencing is widely used for plant breeding activities, including developing SSR markers with an in-silico approach. This research aims to develop an SSR marker to detect genetic diversity in local areca nut from Indonesia. The genomic data used to develop SSR markers were obtained from the Whole Genome Sequencing of Pinang Reyan Hainan Cultivars obtained from NCBI. Fragments containing SSR regions were isolated using MISA software, and primers were designed with Primer3web v.4.1.0 (https://bioinfo.ut.ee/primer3/). The average total size of the sequences tested was 168770731.8, with a total number of SSRs identified as many as 1149,813. SSR motifs are used as a step in genetic diversity research because these motifs will be used in primary attachments which in turn can affect genetic diversity. There were twenty-eight primers generated from four amplicons (100-150, 150-200, 200-250, and 250-300 bp) used for multiplex primers with motifs (AAT)37, (TA)53, (TA)54, (TC)52, (TC)52, (TG)53, and (AG)54.

Abstract

Ganoderma boninense attack and its rapid spread in oil palm plantations have caused significant economic losses. The use of Ganoderma resistant plant materials can be a solution, but the allelic segregation that occurs causes not all DxP progeny obtained have the nature of resistance to Ganoderma. The role of molecular markers is needed to assist initial selection of Ganoderma resistant DxP progenies in nurseries. This study aims to identify targeted SNP markers from the Laccase-24 gene (EgLCC24), which is thought to play a role in oil palm resistance to Ganoderma. Three oil palm DxP populations were used in this study, namely populations A, B, and C. Ganoderma screening in nurseries was carried out on these three populations to obtain progeny samples with resistant and susceptible phenotypes, which were then further analyzed. The methods used include SNP analysis and the use of SNAP primers for marker development. The SNP differences obtained from this study led to amino acid changes, but did not cause stop codons. The validation results using SNAP primers on the Laccase-24 gene showed that DxP progeny from populations A, B, and C relatively have moderate allele characteristics of Ganoderma resistance.

Abstract

Climate change and reduced crop yields caused by drought stress have increased the demand for drought-tolerant varieties. The anther culture technique allows the production of improved varieties with high homozygosity in a short time. The study aimed to select the drought-tolerant double haploid lines at the seedling stage. This research was conducted in the greenhouse of the Indonesian Center for Agricultural Biotechnology and Genetic Resources Instrument Standard Testing (BBPSI Biogen), Bogor, from August to November 2023. The drought-tolerant selection of 12 doubled-haploid rice lines (AE1-AE12) and four check varieties, i.e., two commercial checks (“Inpari 18” Tadah Hujan Agritan or AE13, “Bioni63” Ciherang Agritan or AE14), one drought-tolerant check (“Salumpikit” or AE15), and one drought-sensitive check (“IR20” or AE16). The research used a randomized complete block design with three replications. The characters observed were leaf rolling, leaf drying, recovery ability, and plant fresh and dry weight. The Friedman test results showed that the lines with the lowest rankings, three lines (AE2, AE5, and AE12) with mild tolerant criteria for leaf rolling, five lines (AE1, AE2, AE5, AE8, and AE12) with mild tolerant criteria for leaf drying, and three lines (AE1, AE5, and AE12) with tolerant criteria for recovery ability. The selection index for drought tolerance at the seedling stage identified seven lines with positive values. Based on the Friedman test, selection index, and heatmap visualization, AE12, AE1, AE5, and AE8 exhibited a tolerance similar to “Salumpikit” and were deemed suitable based on drought tolerance characters.

Abstract

Current climate change has caused drought in various regions, which has decreased rice yields. Green super rice (GSR) has the characteristic of being tolerant to water limitation. GSR lines were bred through anther culture technique to obtain doubled haploid (DH) plants. This study aimed to analyze the response of DH GSR lines to drought stress at the vegetative stage and identify potential lines based on the weighted selection index. The experiment was arranged in a randomized complete block design with three replications. The materials used consisted of twenty DH lines, Inpari 42 Agritan GSR, and Inpari 18 as check varieties, Salumpikit as a drought-tolerant check, and IR 20 as a drought-sensitive check. Qualitative data were analyzed using the non-parametric Friedman test. The result showed different responses among the lines for leaf rolling, leaf drying, and recovery ability. Line SN14 exhibited moderate leaf drying and showed improvement to mild tolerance during the recovery phase. Nine lines (SN12, 14, 32, 40, 51, 57, 58, 59, and 60) were selected using a selection index based on leaf rolling, leaf drying, and recovery ability. These lines can be further tested for drought tolerance tests until the reproductive stage, and the tolerant lines could be useful for future development. Abstract

Proses pembelajaran sepanjang hayat merupakan salah satu upaya mengembangkan potensi peserta didik berbasis nilai kearifan lokal Bali sangat mendukung terwujudnya profil pelajar Pancasila. Tujuan dari kajian ini adalah untuk menganalisis nilai kearifan lokal perspektif kebudayaan Bali dalam membangun profil pelajar Pancasila sebagai bagian dari kurikulum merdeka belajar untuk mengarahkan peserta didik agar menghargai dan mencintai budayanya sendiri. Metode dalam penelitian ini menggunakan metode penelitian kualitatif dengan pendekatan studi literatur (literature review). Pendekatan studi literatur dilakukan dengan cara membaca, mencatat, dan menelaah bahan penelitian melalui berbagai sumber yang terkait dengan tujuan dan masalah penelitian melalui data pendukung yang bersumber dari buku, hasil lokakarya, prosiding, jurnal penelitian baik nasional maupun internasional dan sumber-sumber lainnya seperti internet. Hasil kajian menemukan bahwa nilai kearifan lokal perspektif kebudayaan Bali dapat dijadikan salah satu strategi untuk menciptakan paradigma baru dalam mewujudkan kompetensi global menuju profil karakter siswa Pancasila. Kearifan lokal merupakan salah satu aspek budaya yang dimiliki daerah-daerah termasuk Bali sangat banyak dan beraneka bentuknya, secara selektif dapat diangkat menjadi aset nasional dan dapat direvitalisasi sehingga dapat difungsikan sebagai wahana untuk membangun profil pelajar Pancasila.
