Abstract

Abstract. Hadianto W, Purwoko BS, Dewi IS, Suwarno WB, Hidayat P. 2023. Selection index and agronomic characters of doubled haploid rice lines from anther culture. Biodiversitas 24: 1511-1517. Development of superior rice varieties requires selection of the best lines having good agronomic characters and high yields. This study aimed to select high-yielding doubled haploid rice lines obtained from anther culture through index selection and agronomic characters. The study was conducted using a single-factor randomized complete block design (RCBD) with genotype as treatment. Sixty doubled haploid lines and 5 check varieties were evaluated along with three replications, hence, the total experimental unit was 195. Analysis of variance showed that the genotypes had a significant effect on all observed characters, except for the number of productive tillers. The agronomic characters observed had a high broad sense of heritability, except for the number of productive tillers which was in the medium category. Furthermore, index selection was carried out based on productivity, the number of productive tillers, the total number of grains, as well as the percentage of filled grains. Based on the index selection, 29 lines were selected among the 60 doubled haploid rice lines tested. These lines had the range in productivity from 2.95 to 6.77 ton ha-1, the number of productive tillers from 12.3 to 20.8 tillers, the total number of grains from 135 to 275.4 seeds, the filled grain percentage from 53.23 to 87.61%, and the selection index from -1.48 to 7.88. The selected lines will be further tested at the next experimental stage in several locations to produce high-yielding, brown planthopper-resistant, and drought-tolerant rice varieties.

Abstract

Abstract. Saputra HE, Syukur M, Suwarno WB, Sobir. 2022. Diversity and similarity of melon (Cucumis melo L.) groups and determination of distinguishing morphological characters. Biodiversitas 23: 6254-6261. Characterization is an important activity for the study of genotype diversity and similarity. The study was aiming at obtaining information about the diversity and similarity of melon groups and to determine morphological characters as differentiators between melon groups. The genotypes tested were IPB240, IPB283, IPBM21, IPBM23, IPBME5, IPBMETA9, UME20, UME38, UME39, UME90, UME91, UME98, UME99, UME100, UME101. Characterization was based on UPOV and IPGRI guidelines for qualitative and quantitative characters. Two qualitative characters showed no variation in all tested genotypes, namely sex expression (EK) and secondary skin color outside the groove (WKSDA). Qualitative characters that have high diversity were groove color (WA), maximum width between grooves (LMAA), groove color intensity (IWA), and flower stalk thickness (KTBN). The UME98 genotype had the most different color appearance from the other genotypes on the heatmap. The color and depth of fruit grooves are distinguishing characteristics of all genotypes. There were genotype differences in the inodorus and reticulatus types. Unlike the IPBMETA9 genotype, UME100, UME101, and UME38 formed the same clusters with the characteristics of the makuwa group. Melon group makuwa had different characteristics with the inodorus and reticulatus groups. The distinguishing characters for the makuwa type were groove depth, ratio length to diameter, the diameter of hermaphrodite flower stalks, fruit diameter, and fruit skin thickness. Melon groups in the inodorus and reticulatus groups had high similarities, making it difficult to find specific characters in all melon genotypes observed.

Abstract

Abstract. Rohaeni WR, Suwarno WB, Susanto U, Trikosoemaningtyas, Ghulamahdi M, Aswidinnoor H. 2023. Genetic diversity, heritability, and accumulation patterns of Zn content in biofortified rice. Biodiversitas 24: 208-214. The study on zinc accumulation patterns in various rice organs is valuable information to support effective biofortified breeding. Research reports related to zinc accumulation have only been limited to seeds, while the pattern of zinc accumulation in each part of the plant has not been reported, especially for Indonesian rice germplasm. The study aims to provide information on the pattern of accumulation and genetic diversity of Zn content in rice plants. The experiments used a randomized complete block design (RCBD) with 3 replications and genotypes as a treatment. The genotypes were 29 lines of BC4F3 Inpari IR Nutri Zinc////Inpari 23, 5 lines of BC4F3 Inpari IR Nutrizinc////Sintanur, and 1 line of BC4F3 Inpari IR Nutrizinc////Inpari 35. A total of 6 varieties were used as checks. The Zn content of dehulled (brown rice) grains sample was measured using the Oxford Supreme8000 XRF machine with the Rice15 method that had been validated by the Inductively Coupled Plasma (ICP) method, while the analysis of the pattern of Zn content of several organs (seeds, leaves, stems, and roots) used the Inductively Coupled Plasma Optical Emission Spectrometry (ICP-IOS) method. The coefficient of genetic variation of the grain zinc content (GZn) was moderate and had high heritability criteria (H2bs = 0.83). Yield characters have a narrow coefficient of genetic variation and low heritability. There were differences in the pattern of Zn accumulation in plant organs between high GZn and low GZn genotypes. The distribution of Zn content in high GZn genotypes showed 26.4% in seeds, 13.7% in leaves, 39.3% in stems, and 20.6% in roots. The distribution of Zn content in low GZn genotypes showed 14.2% in seeds, 10.9% in leaves, 31.0% in stems, and 44.0% in roots. The results of this study indicate that a different breeding strategy is needed to develop new high-yielding rice varieties based on zinc accumulation patterns in cross-parents.

Abstract

Abstract. Humam BK, Suwarno WB, Azrai M, Makkulawu AT, Efendi R. 2023. Variability of kernel morphological and flour physicochemical properties in tropical dent corn hybrids. Biodiversitas 24: 1600-1609. Corn is an important crop worldwide for food, feed, and industry. The aims of this study were to study kernel morphological and corn flour physicochemical properties and hybrids selection according to nutritional values for food industries. Plant materials were 15 test hybrids and four check varieties, namely RK457, RK57, BISI18, and NASA29. Kernel morphological and corn flour physicochemical variations are influenced by kernel type. All dent hybrids had 40/0 mesh corn flour yielded significantly higher compared to NASA29. °Dent scores were negatively correlated to 40/0 mesh corn flour yields. The total and soluble carbohydrates of dent corn flour hybrids are significantly higher compared to semi-dent and semi-flint corn flour hybrids. According to corn flour nutritional values in polygon GT biplot analysis, dent hybrids had relatively high starch, total carbohydrate, and soluble carbohydrate contents. Semi-dent hybrids had relatively high moisture, crude protein, and total ash contents. Semi-flint hybrid had relatively high crude fat and crude fiber contents. Cluster-2 was dominated by dent hybrids according to the fuzzy clustering method and selected as potential hybrids. MSM53 was the female parent of dent hybrids in cluster-2. Potential hybrids could be evaluated further by corn flour functional properties analysis.

Abstract

Abstract. Sintia M, Ardie SW, Suwarno WB. 2023. Genetic variability of F2 foxtail millet population derived from ICERI-5 and Botok-10 cross. Biodiversitas 24: 3559-3567. Foxtail millet (Setaria italica (L.) P. Beauv.) is a carbohydrate source with good tolerance to abiotic stress. High productivity, short stature, and early flowering time are the breeding targets of this species. This study aimed to obtain genetic variability of F2 foxtail millet population derived from ICERI-5 and Botok-10 crossand to predict selection response based onweighted selection index. This experiment was conducted in Bogor and consisted of 522 F2 individuals and two parental genotypes. The results showed that the F2 population had shorter plant height and earlier flowering time compared to Botok-10, and higher grainweight per plant compared to ICERI-5. Plant height, flowering time, and grainweight per plant showed moderate to high genotypic coefficient of variation with moderate to high broad-sense heritability. Weighted selection index using the three target traits resulted in ten F2 individuals with higher selection index compared to both parents, with individual number I5B10-4-96 having the highest selection index. Selection responses based on a weighted selection index indicate an expected decrease in flowering time and an increase in grain weight per plant in the next generation.

Abstract

Abstract. Maghfiroh RN, Suwarno WB, Sobir, Gunawan E. 2023. Assessment of relationship among traits and genotypes for melon (Cucumis melo) breeding. Biodiversitas 24: 4523-4531. Melon (Cucumis melo L.) is widely consumed as fresh fruit or juice, and therefore deeper analyses of fruit quality are needed for future breeding purposes. Breeders have a role in providing improved melon varieties that suit consumer ideotypes or preferences. This study aimed to (i) evaluate the performance of melon genotypes, (ii) estimate the correlation between quantitative traits, and (iii) determine the clustering among genotypes based on plant and fruit traits. This research evaluated 24 genotypes from inodorus, reticulatus, and makuwa groups that were carried out from March to June 2022 at a greenhouse in the Tajur experimental station of the Center for Tropical Horticulture Studies, Bogor, Indonesia. IPB HD 2-111 and IPB HD 2-100 had good quality based on soluble solids content, fruit shape, flesh texture, and rind color, and G30 was relatively superior on flesh thickness, fruit weight, and vitamin C. Based on the multiple regression analysis, it was known that petal width, petiole length, and days to harvest were significantly affected the proportion of fruit cavity width to the fruit diameter. The adjusted coefficient of determination (R2) was 0.569. Cluster analysis grouped 23 genotypes into three main clusters based on the neighbor-joining method. These results may be useful for selection in melon breeding programs.

Abstract

Abstract. Romadhon MR, Sobir, Suwarno WB, Matra DD. 2024. Profile microsatellite mining of whole genome sequencing and transcriptomic assembly in dwarf and tall areca nut (Areca catechu L.) in Indonesia. Biodiversitas 25: 1081-1088. Areca nut (Areca catechu L.) has high diversity in fruit shape and flowering time. Two superior varieties are commonly cultivated in Indonesia, namely Betara areca nut (tall) and Emas areca nut (dwarf). Molecular level diversity from genomic and transcriptome of areca nut data is available at NCBI, but not yet for Indonesian areca nut. The research compared the results of SSR with two method approaches to detect genetic diversity in the plants accurately. This study aimed to compare the SSR motifs of Betara areca nut and Emas areca nut varieties from whole genome sequencing and transcriptome assembly. The research was conducted at the Leuwikopo Seed Centre Laboratory of the IPB University, Bogor, Indonesia. The methods used are Whole Genome Sequencing (WGS) and transcriptome assembly. A total number of identified SSRs from WGS approach from Betara areca nut of 95 SSRs and Emas areca nut of 95 SSRs, while Betara areca nut SSRs from transcriptome assembly of 466 SSRs and Emas areca nut of 357 SSRs. The percentage of contigs and transcripts from Betara areca nut containing SSR was 7.43% and 4.22%, respectively, while Emas areca nut was 8.06% and 2.04%, respectively. AT is the dominant SSR motif in WGS, while the GA motif dominates the transcriptome assembly results.

Abstract

Abstract. Saputra HE, Syukur M, Suwarno WB, Sobir. 2025. Analysis of similarity and dissimilarity of combining ability and heterosis for melon (Cucumis melo) fruit characters using multi-environments. Biodiversitas 26: 1870-1880. Evaluating combining ability and heterosis simultaneously across different environments facilitates the identification of hybrids with comparative advantages for melon breeding. Therefore, this study aimed to evaluate the consistency of General Combining Ability (GCA), Specific Combining Ability (SCA), heterosis, and heterobeltiosis on the characteristics of melon fruit in three environments, namely Bengkulu, Cilegon, and Argamakmur. The method used a completely randomized block design with one factor for each environment. The full diallel population (30 F1+6 parents) was repeated thrice, leading to 108 experimental units in each environment. The results showed that the best combined GCA value was followed by the optimal GCA value in each environment, despite genetic × environment and GCA × environment interaction. The consistency of GCA was due to the additive effect role that was not influenced by the environment. The best combined SCA value was not followed by the optimal SCA value in each environment, showing inconsistency due to non-additive effect roles (dominance and epistasis), which were easily influenced by the environment. The results also showed that genetic distance affected the values of heterosis and heterobeltiosis. Furthermore, crossbreeding between melon groups produced better heterosis and heterobeltiosis values than crossbreeding within the same group. Combining ability (GCA and SCA), mid-parent value, heterosis, and heterobeltiosis across multiple environments helped identify hybrids with comparative advantages for melon breeding.
