Abstract

Areca nut (Areca catechu) belongs to the monocotyledonous and palm family of plants known as betel nut or supari plants. The Center of origin areca nut comes from India. Areca nut is a cross-pollinated plant with a heterozygous heterogeneous genetic constitution. Next-generation sequencing is widely used for plant breeding activities, including developing SSR markers with an in-silico approach. This research aims to develop an SSR marker to detect genetic diversity in local areca nut from Indonesia. The genomic data used to develop SSR markers were obtained from the Whole Genome Sequencing of Pinang Reyan Hainan Cultivars obtained from NCBI. Fragments containing SSR regions were isolated using MISA software, and primers were designed with Primer3web v.4.1.0 (https://bioinfo.ut.ee/primer3/). The average total size of the sequences tested was 168770731.8, with a total number of SSRs identified as many as 1149,813. SSR motifs are used as a step in genetic diversity research because these motifs will be used in primary attachments which in turn can affect genetic diversity. There were twenty-eight primers generated from four amplicons (100-150, 150-200, 200-250, and 250-300 bp) used for multiplex primers with motifs (AAT)37, (TA)53, (TA)54, (TC)52, (TC)52, (TG)53, and (AG)54.

Abstract

One essential objective of rice breeding is to obtain high-yielding varieties. This study aimed to (1) determine the effect of genotype (G), environment (E), and genotype by environment (G×E) interaction on agronomic traits and yield of 12 lowland rice genotypes, (2) estimate variance components and repeatability (3) identify promising rice genotypes with good agronomic performance and high yield potential. The trials were conducted in three irrigated lowland locations from June to November 2020, using a randomized complete block design with three replications. The results showed that the G×E interaction effect was significant on days to flowering, days to harvest, plant height, number of tillers, and panicle length. The genotype's main effect was significant on yield. Four IPB lines (IPB189-F-13-1-1, IPB189-F-23-2-2, IPB193-F-17-2-3, and IPB193-F-30-2-1) had a higher average yield than Ciherang and Inpari 32 varieties. The IPB189-F-23-2-2 had a panicle length stability across the three test locations and a higher average yield than the checks.

Abstract

Abiu merupakan salah satu tanaman hasil introduksi dari Amazon yang potensial untuk dikembangkan di Indonesia. Buah abiu memiliki cita rasa enak dan juga kaya akan manfaat kesehatan. Salah satu permasalahan yang umum ditemukan pada buah abiu yakni tingkat kemulusan buah yang rendah. Penelitian ini bertujuan mengetahui pengaruh bahan aktif pestisida yang digunakan dengan periode aplikasi yang berbeda terhadap tingkat kemulusan buah abiu. Penelitian dilakukan di Kampung Babakan Lebak, Kelurahan Balumbang Jaya, Kecamatan Bogor Barat dan Laboratorium Pasca Panen Departemen Agronomi dan Hortikultura, Institut Pertanian Bogor. Rancangan percobaan yang digunakan adalah rancangan petak terbagi (split-plot) RKLT dengan 5 ulangan. Faktor petak utama yaitu periode aplikasi yang berbeda, yaitu: 1,2,3 minggu setelah antesis (MSA) (V1); 1,3,6 MSA (V2); dan 1,4,7 MSA (V3). Faktor anak petak yaitu bahan aktif pestisida yaitu: mankozeb 80 WP dan streptomisinsulfat 20 WP (P1); propinep 70 WP (P2); profenofos 500 EC (P3); dan kontrol (P4). Hasil penelitian menunjukkan bahwa perlakuan bahan aktif pestisida memberikan pengaruh yang nyata terhadap variable kemulusan buah abiu. Aplikasi pestisida pada periode berbeda dan interaksinya dengan perlakuan bahan aktif pestisida tidak memberikan pengaruh yang nyata terhadap kemulusan buah abiu. Perlakuan terbaik yaitu pemberian bahan aktif mankozeb dan streptomisinsulfat yang dapat mempertahankan kemulusan buah abiu sebesar 77.31%. Abstract

Foxtail millet (Setaria italica L. Beauv.) is a grain-producing crop with high nutritional benefits and adaptability to broad environmental conditions. Indonesian local foxtail millet accessions are valuable genetic resources for crop improvement of the species. This study aims to assess the biodiversity of Indonesian local foxtail millet accessions and estimate their salinity tolerance level utilizing the SiDREB2-based SNAP marker and early evaluation at the seedling stage. Our results showed that eight Indonesian local foxtail millet accessions, namely Toraja, Mauliru-2, Hambapraing, NTB-1, ICERI-5, ICERI-6, Botok-4, and Botok-10, had high variability based on agro-morphological traits. The Hambapraing and Mauliru-2 were the potential accessions with high yield estimates (2.33 ton.ha-1 and 1.93 ton.ha-1) and early harvest time (<110 DAT). ICERI-6 was indicated as a salinity-tolerant genotype based on the SiDREB2-based marker and early salinity evaluation at the seedling stage, while Toraja, Mauliru-2, NTB-1, and Botok-4 were categorized as sensitive genotypes. Germplasm variability and the estimated salinity tolerance level of Indonesian local foxtail millet accessions presented in this study serve as essential information in the foxtail millet breeding for high productivity, early harvest time, and salinity tolerant variety.

Abstract

Zinc (Zn) is an essential micronutrient that is very useful for plants as well as humans. Breeding high zinc rice varieties and pigmented varieties will increase the added nutritional value of the rice consumed. Knowing the distribution of zinc content based on pigmented rice is necessary to see the opportunities for obtaining pigmented rice lines with high zinc. This study aimed to determine the potential distribution of zinc based on the pigment of brown rice. The research was carried out at the Breeding Laboratory of the Indonesian Center for Rice Research from February to July 2022. A total of 970 lines derived from various biparental crosses and 10 varieties were used as research material. The samples were brown rice from a field research plot arranged in an augmented design with 5 blocks. Analysis of zinc content used the X-ray Fluorescence method using the XRF Machine Supreme8000. The results showed that there was a significant difference in Zn content based on CPC color. White rice had the widest range of zinc content compared to brown and black rice. The maximum zinc content found in white rice was 55.82 mg kg-1 and the maximum yield potential was 138.29 g per hill. The black rice genotypes in all categories had a zinc content exceeding the Inpari IR Nutri Zinc potency. Black rice type C was the one with the widest variety. Types of black rice with high zinc content and high yield potential are Black B and Black E types. ABSTRAKZinc (Zn) merupakan mikronutrisi esensial untuk tanaman dan juga manusia. Perakitan varietas  padi zinc tinggi pada beras-beras berwarna akan meningkatkan nilai tambah gizi beras yang dikonsumsi. Keragaman kandungan zinc berdasarkan warna beras perlu diketahui untuk melihat peluang diperolehnya varietas beras berwarna dengan keunggulan zinc tinggi. Penelitian ini bertujuan untuk mengetahui potensi zinc berdasarkan warna beras pecah kulit. Penelitian dilaksanakan Laboratorium Pemuliaan BB Padi dari bulan Februari sampai Juli 2022. Sebanyak 970 galur hasil persilangan biparental dan 10 varietas digunakan sebagai materi penelitian. Sampel berupa beras pecah kulit dari plot penelitian lapangan dengan rancangan augmented 5 blok. Analisa kandungan zinc menggunakan metode X-ray Fluorescence mesin XRF Machine Supreme 8000. Hasil menunjukkan bahwa terdapat perbedaan nyata kandungan zinc berdasarkan warna BPK. Beras putih memiliki rentang kandungan zinc yang paling luas dibandingkan beras merah dan hitam. Potensi zinc maksimum terdapat pada beras putih dapat mencapai 55,82 mg kg-1 dan potensi maksimum hasil 138,29 g per rumpun. Genotip padi semua kategori tipe beras hitam memiliki rentang kandungan zinc di atas potensi Inpari IR Nutri Zinc. Tipe beras Hitam C adalah beras yang memiliki varian paling luas. Tipe beras hitam yang memiliki peluang kandungan zinc sangat tinggi dan potensi hasil tinggi adalah tipe Hitam B dan Hitam E.

Abstract

The utilization of controlled greenhouses can be an alternative for melon (Cucumis melo L.) cultivation. Growing melon in greenhouses lowers the risk of pests and infections by diverse pathogens. Low solar irradiation during the rainy season and pollution in the greenhouse roof, may cause shade stress conditions for the plants inside. This study aimed to elucidate the plant and fruit characteristics of nine genotypes of melon grown under shade conditions in a greenhouse and the variability among them. The experiment was conducted in a greenhouse of Cikabayan Experimental Station, IPB University, Bogor, from November 2016 to January 2017. The genetic materials evaluated were nine melon genotypes from the Center for Tropical Fruit Studies at IPB University. A randomized complete block design with three replicates was followed. Shade intensity in the greenhouse was approximately 25%. The results showed that the genotype effect was significant for internode length, petiole length, plant height fruit, flesh thickness, fruit rind thickness, fruit weight, and total soluble solids. Genotype means for fruit weight were small in this experiment (< 300 g), whereas the total soluble solids were moderate to high (8.7-14.3 °Brix). Fruit diameter had a positive and significant correlation with leaf length, leaf width, and fruit length, whereas fruit weight had a positive and significant correlation with fruit diameter and fruit length. Abstract

Karakteristik yang diinginkan dari buah melon oleh konsumen meliputi rasa manis, ukuran buah sedang hingga besar, daging tebal dengan warna menarik dan tekstur renyah, serta masa simpan yang relatif lama. Memprediksi waktu panen menjadi hal yang penting terkait masa simpan buah dengan harapan buah melon dapat mencapai konsumen dalam keadaan dan kualitas terbaik, serta memberikan pengalaman yang memuaskan bagi konsumen. Saat ini, ketersediaan tenaga kerja pemanen dengan kemampuan yang mumpuni dalam menentukan buah melon yang akan dipanen menjadi salah satu kendala. Penggunaan robot pertanian dalam pemanenan buah melon merupakan salah satu solusi yang efektif dalam mengatasi permasalahan tersebut. Robot pertanian ini membutuhkan sistem yang mampu memprediksi stadia kematangan buah melon untuk dipanen. Penelitian ini fokus pada analisis perbandingan kinerja antara dua algoritma machine learning yaitu Support Vector Machine (SVM) dan Random Forest (RF), dengan tujuan menentukan pilihan optimal saat menerapkannya pada perangkat komputasi terbatas. SVM dan RF memiliki nilai akurasi tinggi, masing-masing 82% dan 73%. Keduanya juga memiliki waktu komputasi yang cepat, dengan rata-rata waktu inferensi masing-masing 2.14 detik dan 2.15 detik. Rata-rata penggunaan CPU pada algoritma SVM lebih tinggi dibandingkan dengan algoritma RF yaitu 17.80% sedangkan RF 15.48%. Meskipun SVM memiliki precision, recall, dan f-scored yang sedikit lebih tinggi dibandingkan dengan RF, namun setelah dilakukan independent 2-samples t-test terhadap inference time dan penggunaan CPU, didapatkan hasil bahwa tidak ada perbedaan nyata antara SVM dan RF. Keduanya sama-sama memiliki kinerja yang baik dan masuk ke dalam kategori good classification. Meninjau hal tersebut, algoritma RF menjadi algoritma yang disarankan karena memiliki tingkat akurasi yang baik, waktu komputasi cepat, dan penggunaan rata-rata sumberdaya CPU lebih rendah

Abstract

Penelitian ini bertujuan untuk menggali unsur-unsur yang berkaitan dengan asal usul sejarah munculnya ter-teran dalam rangkaian Usaba Mumu serta untuk mengetahui fungsi dan maknanya dalam rangkaian prosesi Usaba Mumu di Desa Adat Jasri. Penelitian ini menggunakan metode penelitian kualitatif deskriptif. Kemudian pengumpulan data dilakukan melalui  observasi, wawancara, dan beberapa dokumentasi kepustakaan yang ada relevansinya dengan masalah yang diteliti. Hasil penelitian menunjukkan bahwa Ter-teran merupakan suatu keharusan dalam rangkaian Ngusaba Mumu atau Ngusaba Dalem Nganggih berfungsi sebagai pelengkap pemuput Wali (upacara) pada saat Usaba Mumu yang dilaksanakan di pempatan (catuspata) dengan menggunakan sundih (bobok) yang saling dilemparkan. Upacara ini mengandung makna untuk mempertebal keyakinan dan ketaatan dalam diri setiap individu masyarakat Desa Adat Jasri sehingga tetap melaksanakan ajarannya dan mencegah hal-hal negatif yang dianggap dapat mengganggu hubungan harmonis, sehingga kesejahteraan kehidupan masyarakat Desa Adat Jasri dapat terwujud.

Abstract

Setaria italica L. or foxtail millet is known for its nutritious grains and adaptability to unfavorable environmental conditions. High productivity, early heading, medium stature, and tolerance to drought- or salinity stress are among the breeding objectives for foxtail millet. The objective of this study was to select F3 families of foxtail millet from the cross of Botok-10xICERI-6 by weighted selection index and assisted by SiDREB2-based SNAP marker. Genotyping of 178 F3 families using the SiDREB2-based SNAP marker resulted in 29 A/A genotypes, 121 A/G genotypes, and 28 G/G genotypes. Further evaluation was conducted on 48 F3 families consisting of 27 A/A genotypes and 21 A/G genotypes in an augmented randomized complete block design together with their parental genotypes (Botok-10xICERI-6) and three check genotypes (Mauliru-2, NTB-1, and Toraja). Plant height and heading time had high broad-sense heritability, whereas grain weight per plant had a moderate broad-sense heritability. Ten potential F3 families were selected based on a weighted selection index with 20% intensity, comprised of seven A/G genotypes and three A/A genotypes with a weighted selection index ranging from 0.84 to 3.76. The F3 family with pedigree numbers B10I6-15-136, B10I6-15-161, and B10I6-15-70 with A/A genotypes are considered putative transgressive segregants and could be continued to the next generation for further breeding process.

Abstract

Sheet pipe is a type of perforated pipe used for drainage designed initially for drainage but has the potential for sub-surface irrigation. The objectives of this study were to experiment and observe the performance of the sub-surface irrigation control system with sheet pipe. This investigation covered the observation of water table control and its effect on soil moisture. The detailed process of water flow during the setting of the water table was numerically modeled in 2 dimensions to observe the distribution of soil moisture, soil pressure, and flux. The results showed that the system successfully controlled the water table at the desired level in the experiment. The developed two-dimensional numerical simulation showed the distribution of soil moisture in the model center as a response to the water table increase, represented by the variable head. The soil wetting advances toward soil surface driven by the water table, which was increased gradually and reached saturation at the height of water table setpoint.
