Abstract

Perbanyakan bibit melon secara secara in vitro menghasilkan bibit yang bebas patogen, seragam, true to type yang dapat diperoleh dalam jumlah yang banyak dengan cepat. Penelitian ini bertujuan menentukan kombinasi IAA dan BAP yang optimal untuk inisiasi kultur tunas melon in vitro. Penelitian ini dilaksanakan pada bulan Januari-Juli 2018, menggunakan rancangan kelompok lengkap teracak tiga faktor dengan 6 ulangan. Faktor pertama adalah tiga taraf IAA yaitu 0.0, 0.25, dan 0.5 ppm, faktor kedua adalah empat taraf BAP yaitu 0.0, 1.0, 1.5, dan 2.0 ppm, dan faktor ketiga adalah jenis eksplan yaitu pucuk dan buku. Faktor tunggal (IAA, BAP, jenis eksplan) berpengaruh nyata terhadap jumlah tunas dan daun, interaksi dua atau 3 faktor tidak berpengaruh nyata terhadap jumlah tunas dan daun, kecuali interaksi IAA dan BAP berpengaruh nyata terhadap jumlah akar. Eksplan pucuk menghasilkan tunas yang lebih banyak dibandingkan buku. Hasil analisis regresi menunjukkan bahwa pengaruh BAP cenderung membentuk pola kuadratik positif terhadap peubah jumlah tunas dan jumlah daun, namun linier negatif dengan peubah jumlah akar. Perlakuan yang optimal untuk multiplikasi tunas adalah BAP 1.0 ppm tanpa penambahan IAA. Konsentrasi IAA dan BAP yang makin tinggi menginduksi pembentukan kalus dan menekan pertumbuhan tunas. Abstract

Rainfed lowland rice cultivation is an alternative to increase national rice production. Breeding of high yielding rice varieties suitable for rainfed lowland condition can be accelerated by using doubled-haploid (DH) as genetic materials. This study aimed at obtaining information on the agronomic performance including yields in several DH rice lines and selecting DH lines suitable for rainfed lowland paddy field. The experimental design used was a Randomized Complete Block Design with 3 replications. The treatment was thirty DH lines and 4 check varieties namely RJ31 Ciherang, RJ32 Inpari 18, RJ33 Inpari 40, and RJ43 Inpari 41. The results showed that there were variability in all agronomic performances, i.e., plant height, number of tillers, days to heading and to harvest, panicle length, number of filled and empty grains, 1000-grain weight and grain yield. The DH lines, namely RJ19 DR8-43-3-1 and RJ25 DR10-14-1-1, gave the same productivity as 4 check varieties. Index selection showed that twelve DH lines with medium number of productive tillers, early maturing, and productivity of more than 4.40 tons.ha-1 were selected for further evaluation.

Abstract

Melon (Cucumis melo L.) is a fruit commodity that gets a great interest to be developed in Indonesia and has a high nutritional value. However, the fungal infection and pathogens in melon cultivation are considered significant problems that are difficult to manage. Therefore, efforts are needed to improve the productivity and quality of melon and prevent pest and disease attacks. One mineral nutrient that is assumed to enhance plant resistance and increase the quality and production of melon is silica. The purpose of the research was to evaluate silica absorption from foliar-applied silicon nanoparticles in melon under soilless culture and improve melon fruit's growth and quality by applying silica fertilizer. The experimental design used was a split-plot randomized complete block design 3 x 2 factorial pattern with four replicates. The main plot factor is silica fertilizer (Novelgro, water-soluble), consisting of three silica concentrations of 0.67; 1.33 ppm, and control. The spray volume of each treatment was 160 ml per plant with seven days' intervals and a frequency of three times. The subplot factor is melon varieties consisting of “Alisha” and “Glamour”. The findings showed that silica fertilizer significantly increased the plant height, stem diameter, internode length, total number of hermaphrodite flowers, number of hermaphrodite flowers that are swelling, while decreased the fruit moisture content and gave the best average fruit position. Instead, The “Glamour” variety gave the best response to plant growth and fruit quality. Moreover, the melon plant could absorb silica in the low category (<1% Si). The highest silica contents were found in the “Alisha” cultivar leaves and the “Glamour” cultivar's rinds treated with the silica concentration of 1.33 ppm as much as 0.34% and 0.30%, respectively.

Abstract

Melons are horticultural fruits with increasing demand and high economic value. In 2020, the export demand for melons was 388.98 tons, with key destinations including Hong Kong, the United Arab Emirates, Qatar, Saudi Arabia, Germany, Malaysia, East Timor, and Singapore. However, quality degradation, such as fruit cracking, can negatively impact marketability. Dorais et al. (2004) identified several factors contributing to fruit splitting, including genetics, pruning time, water management (rain, high humidity, or intensive irrigation following drought), high temperatures, light, calcium content, and the strength of fruit cell walls. Effective and efficient cultivation practices are essential to mitigate these issues. This research aimed to determine the effect of watering volume and topping treatment on the growth and quality of two melon varieties in a substrate hydroponic system. The experiment was conducted from March 2021 to May 2021 in the greenhouse of the Leuwikopo IPB University Experimental Garden, located at an altitude of 218.79 meters above sea level. Two melon varieties were used: “Alisha” (V1) and “Glamour” (V2), with four replications for each treatment. The treatments were watering volume and topping. Watering consists of consistent watering volume of 250 ml per plant until harvest, variable watering volume according to the growth phase, i.e., 200 ml per plant, 300 ml per plant, and 350 ml per plant for phase, 1, 2, and 3, respectively. The effects of the treatments on all agronomic traits and fruit quality were assessed. The results showed that topping did not significantly affect any of the agronomic traits or fruit quality. Maintaining a consistent watering volume of 250 ml per plant until harvest is beneficial for the growth and quality of melon varieties “Alisha” and “Glamour” in a substrate hydroponic system. Effective water management is crucial in melon cultivation to ensure high-quality fruit production and minimize issues such as fruit cracking. Future research could explore additional factors influencing melon quality and yield to further optimize cultivation practices.

Abstract

Maize is a significant food crop consumed in numerous countries worldwide. Increasing the yield potential of maize can be achieved through the breeding of high-yielding hybrid varieties. The objectives of this study were to assess the performance of hybrid means and parent combining ability for grain yield and other agronomic traits using a line x tester mating design. The study took place at the Gunung Gede Experimental Station of IPB University, Bogor, from June to October 2022. The experiment was conducted using a randomized complete block design with a single genotype factor and three replications. Twenty-two hybrids were utilized as genetic materials, including 20 hybrid candidates and two check varieties. Six maize lines (H1, L26, Nei, P2B, and P42) were employed as line parents, while L15 and P10 were used as tester parents in the line x tester analysis. The neighbor-joining tree analysis indicated that the female parent has a significant influence on the agronomic and yield performance of the hybrids. Correlation analysis revealed that the yield is more strongly influenced by ear diameter rather than ear length, and it is more affected by the number of kernel rows than the number of kernels per row. Three selected hybrids with favorable traits were Nei x B1, L15 x L26, and L26 x P10. The parent L26 demonstrated relatively good general combining ability for yield and may be considered for future maize breeding program.

Abstract

Breeding programs to improve rice varieties for tidal swamp areas have successfully developed a number of promisingbreeding lines. The objectives of this study were to evaluate the performance of ten advance breeding lines in multilocationyield trials and to analyze grain quality of the lines. The lines and two check rice varieties, IR42 and Batanghari, wereevaluated in replicated yield trials in six different tidal swamp environments. Grain quality of these lines were analyzed todetermine physical and chemical properties of the milled and cooked rice. Result from multilocation yield trials showedthat the breeding lines had higher yield potential compared to popular variety IR42 and their yields were comparable tothe control variety Batanghari. The lines showed different adaptability against different environments; some of the linesdemonstrated wide adaptability while the others showed specific adaptation ability. All of the lines had good grain qualitywhile they had different shape and texture which made them potential to be accepted in different regions. The result fromthis study indicated that all of the lines have potential to be commercially cultivated in tidal swamp areas. In addition, dataobtained from this study have been used in the registration of three lines as new varieties for swampy area namely IPB 1RDadahup, IPB 2R Bakumpai and Inpara 6.Keywords: adaptation, grain quality, multilocation trials

Abstract

Melon breeding for fruit quality improvement is important to produce new varieties that meet consumers’ demand. The aim of this study was to elucidate the fruit characteristics of melon at five different maturity stages, involving several genotypes belonging to two cultivar groups: reticulatus and inodorus. The experiments were conducted in three planting seasons: (1) January-March 2015 (13 genotypes), (2) December 2015-February 2016 (56 genotypes), (3) February-May 2016 (9 genotypes). These trials were conducted at the Tajur II experimental station of IPB, Bogor, and each trial was arranged in a single factor randomized complete block design with three blocks. Maturity stages was determined based on fruit rind color and net coverage, and their effects on fruit quality were studied using combined analyses over seasons. Maturity stage significantly affected the fruit length, fruit diameter, flesh thickness, fruit weight, and sugar content. The average weight of the fruits at maturity stage 4 (838.90 g) and 5 (931.79 g) was significantly greater than that of stage 1 (584.42 g). The sugar content increased from maturity stage 2 (5.51 oBrix) to 3 (6.13 oBrix) and to 5 (8.18 oBrix). Fruit weight significantly correlated with fruit length (r = 0.53), fruit diameter (r = 0.85), fruit rind thickness (r = 0.33), and flesh thickness (r = 0.63). Maturity stages affected the quality of melon fruits, and therefore the determination of appropriate harvesting criteria is important in melon cultivations.Keywords: cantalupensis, correlation, inodorus, fruit quality, skin colour

Abstract

ABSTRACTSelection of transgressive segregants is considered as one effective way of obtaining peanut genotypes with high yield on early segregation. The objective of the study was to estimate genetic parameters and to identify transgressive segregants of peanut family in F3 generation. The experiment was conducted at Leuwikopo, Dramaga, Bogor on August-Desember 2014. The genetic materials evaluated were 218 family F3 generation from 5 biparental population (Jerapah/GWS79A1, GWS79A1/Zebra, GWS79A1/Jerapah, Zebra/GWS79A1 and Zebra/GWS18) and 4 commercial varieties as checks (Gajah, Jerapah, Sima, and Zebra). The experiment was conducted in an augmented randomized complete block design with 4 replications for the checks. The results showed that quantitative characters had continuous distribution and they were controlled by many genes with additive, dominance, complementary epistasis, or additive epistasis gene action. Total number of pod and number of pod had high heritability and moderate genetic coefficient of variation. Selection with 10% intensity showed that 22 families had large total number of pods while six families had larger total number of pods than both parents. Family U2-39 estimated was identified as a putative transgressive segregant with high total number of pod and had smaller variance within family than Sima variety. Abstract

Hybrid maize breeding relies on the availability of good performing inbred lines with superior combining ability. The objectives of this study were to: (1) estimate genetic paramters and heritability for agronomic and yield traits, (2) estimate correlations among traits, and (3) estimate selection-balanced inbreeding depression level from S1 to S2 generations. The S1 trial was planted from June to September 2014 at two locations, followed by the S2 trial from January to May 2015 at the same locations. Each trials was arranged in an augmented design with three blocks consisting of 72 unreplicated test genotypes, 3 unreplicated inbred lines, and 5 replicated commercial hybrid varieties as checks. The results showed that the heritability estimates were medium to high for most observed traits. Best yielding genotypes in S1 and S2 generations were derived from P27xNK6326, Pertiwi3xP31, NK6326xP31, P31xNK6326, and P31xNK33 populations. Grain yield was positively correlated with plant height, ear height, ear length, and ear diameter in S1 and S2 generations. The S2 generation exhibited inbreeding depression for grain yield. The estimates of heritability, correlations, and inbreeding depression obtained from this study could be useful for selection in corn breeding programs.Keywords: correlation, heritability, inbreeding depression

Abstract

ABSTRACTHybrid rice varieties  is an alternative technology to improve  productivity of low land rice. The results of previous studies have identified and found the male sterile lines Wild Abortive type and Kalinga are resistant to bacterial leaf blight pathotype III, IV and VIII. The objectives of the research were to obtain information on agronomic characters, yield evaluation, genetic parameters, and repeatability information. The experiment was conducted in two locations namely  Muara and Indramayu experimental stations. The design used was a Randomized Complete Block Design with three replication at each site.  Each replicate consisted of 17 hybrid rice and three check varieties. The results from locations showed that genotype BI485A/BP1 (IR53942) has the highest yield of 5.8 ton ha-1. The coefficient of genetic and phenotypic diversity of six agronomic characters was low. The repeatability for seven character observed were low, except for days to flowering. The  scoring value  of bacterial leaf blight disease in Indramayu showed that nine genotypes exhibit resistance.Keywords: cytoplasmic male sterile lines, disease resistance
