Abstract

The purpose of the research was to know  the effect of Insulin-Like Growth Factor-I (IGFI) derived from pregnant crossbreed mare serum (PMS) on endometrium thickness of mice (Mus musculus). The subject of this research were 35 female mice. The research was arranged by Completely Randomized Design (CRD) with seven treatment and five replications. The treatment were P0 = 10 ng/ml of physiological NaCl, P11 = 10 ng/ml of IGF-I PMS, P12 = 20 ng/ml of IGF-I PMS, P13 = 40 ng/ml of IGF-I PMS, P21 = 10 ng/ml of IGF-I recombinant mouse, P22 = 20 ng/ml of IGF-I recombinant mouse, and P23 = 40 ng/ml of IGF-I recombinant mouse. Observed variables include histopatological endometrium thickness of mice. The data were analyzed by Analysis of Variance (ANOVA), followed by HSD (Honestly Significant Difference) test. The data was also be analyzed using General Linear Model Univarieted to see the comparison between IGF-I PMS and recombinant mouse. The result showed that the addition of IGF-I PMS did not significantly affect (p>0,05) on endometrium thickness of mice . It showed that did not significantly difference (p>0,05) between the effect of IGF-I PMS and IGF-I recombinant mouse against the endometrium thickness of mice.

Abstract

This study was aimed to determine the effect of urea in maturation medium on in vitro oocyte maturation rate. The medium used was TCM-199 added with Hepes, NaHCO3, Kanamycin 0.15 IU/mL, PMSG, 0.15 IU/mL hCG, and 10% FBS. Cumulus oocyte complexes (COCs) of cows derived from follicle aspiration were divided into three groups. In control group (P0), the COCs were matured in vitro in a maturation medium without urea addition, meanwhile in the P1 and P2 groups, the medium was added with urea 20 and 40 mg/dL, respectively. Each petri dish contained three drops of maturation medium (300 µl/drops) according to the groups. Microdrops were coated with mineral oil and then incubated in a 5% CO2 incubator, at 39 ËšC with maximum humidity. Aceto-orcein staining was conducted to evaluate the maturation of oocytes based on the achievement of metaphase II phase that is indicated by the presence of metaphase plate and/or first polar body. The result showed that the oocyte maturation rates of P0, P1, and P2 were 51.25, 52.43 (p >0.05), and 46.88 % (p <0.05) respectively. It could be concluded that the presence of urea at 40 mg/dL in maturation medium reduced the percentage of bovine oocyte maturation in vitro.

Abstract

 This research aimed to determine the effect of α-tocopherol on the count of spermatogenic and Leydig cells in rats (Rattus norvegicus) exposed to 2,3,7,8 tetrachlorodibenzo-p-dioxin (dioxin). Dioxin is an endocrine-disrupting agent that adversely affects reproductive health, while α-tocopherol maintains fertility. This research used 25 rats aged 10-12 weeks weighing 150-200 grams. Rats were divided into five groups (K, P0, P1, P2 and P3). The K (control group) was administered with corn oil 1 ml/day. P0 was exposed to 700 ng/kg/day dioxin. P1, P2 and P3 was exposed to dioxin at a dose of 700 ng/kg/day and administered with α-tocopherol at a dose of 77, 140 and 259 mg/kg/day respectively. Dioxin exposure, corn oil and α-tocopherol administration were conducted orally for 20 days. On day-21, all rats were sacrificed for histological slides preparation of testicles with hematoxylin-eosin staining. Data were analyzed with analysis of variance and continued with the Duncan test. The results indicated that exposure to dioxin caused a decrease in the number of spermatogenic and Leydig cells. The administration of α-tocopherol at a dose of 140 mg/kg/day eliminated the effect of reducing the number of spermatogenic and Leydig cells caused by exposure to dioxin. The conclusion was the administration of α-tocopherol at 140 mg/kg/day was effective in maintaining the number of spermatogenic and Leydig cells in rats (Rattus norvegicus) exposed to 2,3,7,8 tetrachlorodibenzo-p-dioxin.
