Abstract

Penduduk Nusa Tenggara Timur memanfaatkan kulit batang tumbuhan Faloak (Sterculia quadrifida R.Br.) sebagai tumbuhan obat untuk mengobati penyakit liver, gastroentritis, dan penambah stamina. Kulit batang Faloak mengandung senyawa fenol, flavonoid, dan terpenoid. Penelitian ini bertujuan menganalisis daya antibakteri dan antioksidan dari fraksi hasil pemisahan ekstrak etanol kulit Faloak. Fraksi 3 menunjukan aktivitas antibakteri yang tinggi (IC50) pada bakteri B. subtilis (90.51 µg/mL), E. coli (80.12 µg/mL), S.aureus (77,87 μg/mL), dan S.thypi (61.23 µg/mL). Uji aktivitas antioksidan menunjukan bahwa fraksi 2 mempunyai aktivitas antioksidan dan kandungan fenolik total yang paling tinggi (34,16 ± 0,76 mg GAE).

Abstract

Endophytic fungi Chaetomium sp isolated from Phyllanthus niruri Linn. Mycelium powder was extracted by using ethyl acetate. Extract was fractionated using n-hexane, dichloromethane and ethanol 96%. The antimicrobial test was carried out using disc diffusion and microdilution methods. The antioxidant activity of the fraction was determined using hydrogen peroxide free radical scavenging and reducing power capacity activities. The cytotoxicity assay of the fraction against T47D breast cancer cell was carried out using dimethylthiazol-2-yl-2,5-diphenyltetrazolium bromide method (MTT). The in silico prediction of chemical substances which are reported exist in Chaetomium sp. performed using AutoDockVina embedded in PyRx version 8.0. Dichloromethane fraction was found as the most active sample against Escherichia coli (IC50 20.76 mg/mL), Staphylococcus aureus (IC50 70.15 mg/mL), Salmonella typhi (49.13 mg/mL) and was found as the most high phenolic content with value 47.44 mg GAE/g fraction, whereas the best antioxidant activity was performed by ethanol 96% fraction (85%). Cytotoxicity assay against T47D cell line showed dichloromethane fraction have highest activity with IC50 10.76 mg/mL. The docking studies showed that compounds bearing xanthone structure were potential for maltose binding periplasmic and human aromatase associating with their potencies as antibacteria and anticancer. Endophytic fungi Chaetomium sp. was isolated from Phyllanthus niruri using n-hexane, dichloromethane and ethanol fractions was studied its various biological activities as antimicrobial, antioxidant and cytotoxic agent against breast cancer cell.

Abstract

Chemotherapeutic agent such as cisplatin used as cancer cure with side effects and resistance. Ursolic acid is a compound found in Hedyotis corymbosa L., (HCoL) while 8-hydroxydehydrodunnione is found in Sterculia quadrifida R.Br (SQuR) which are proven to have cytotoxic effect to cancer cells. This research aims to review the effect of cisplatin, ethanolic extract of HCoL and SQuR to sensitivity increase on breast cancer cells, which will be confirmed through apoptosis induction and cell cycle modulation. The cytotoxic effect was tested using MTT assay on T47D cell by using IC50 parameter. The combination was tested by determining their combination index (CI) and cell viability. The combination effect in apoptosis induction and cell cycle modulation were observed using flow cytometry method. The cytotoxic test result of the combination shows CI value of below 1 at the concentration of HCoL ethanolic extract as much as 0,21 µg/mL, SQuR ethanolic extract as much as 0,45 µg/mL, and cisplatin as much as 2,5 µM. The combination of HCoL ethanolic extract, SQuR ethanolic extract, and cisplatin results in phase S cell accumulation (27,73%) on breast cancer cell T47D and was able to induct apoptosis. The result proves that ethanolic extract of HCoL and SQuR can developed as a co-chemotherapeutic agent to increase the effectivity of breast cancer treatment.

Abstract

Endophytic fungi used as a new resource to produce bioactive compounds such as antibacterial, antioxidant and anticancer. This study aimed to analyze the antibacterial, antioxidant, cytotoxic fraction of the separation ethyl acetate extract of mycelium of endophytic fungi genus Fusarium sp. Diethyl ether fraction showed high antibacterial activity in E.coli (20,75 µg/mL), S.typhi (35,08 µg/mL), and S.aureus (51,96 µg/mL). Test showed that the antioxidant activity of ethanol 96% fraction has highest antioxidant activity and total phenolic content (75,85 ± 0.87 mg GAE). Cytotoxic test on T47D breast cancer cells showed that the fraction of diethyl ether have highest activity with IC50 of 10,16 ± 0,88 µg /mL.

Abstract

Endophytic fungi can produce compound that similar with the host, so it can be  used as mass production of compound. The optimal and valid assay method is needed to obtain the proper culture condition, which one using densitometric Thin Layer Chromatography (TLC). This research used 11 samples of endophytic fungi culture extract from Phyllantus niruri within various culture condition. Optimization of mobile phase was done using 3 kinds of mobile phase mixture, chloroform: ethyl acetate: methanol (8:8:4); chloroform: ethyl acetate: methanol: NH4OH (8:8:4:0.005); chloroform:ethyl acetate:NH4OH (8:8:0.005). Validation method measured using some parameters such as linearity, accuracy, and precision. The result of optimization and validation showed that TLC densitometry method can be used for measuring alkaloid level within the endophytic fungi extract of P. niruri using chloroform: ethyl acetate: NH4OH (8:8:0.005), with R value is 0.977, percentage of recovery is between 90–110, and RSD ≤7% on each concentration series. While the proper condition of endophytic fungi culture is using sucrose as carbon source and at pH 6 condition. The content of alkaoid reached 25.9 ± 1.4 mg 100 g-1 fresh weight.

Abstract

Cisplatin is a chemotherapeutic agent with varying side effects. Berberine compounds in Tinospora crispa and ursolic acids in Hedyotis corymbosa L are known to have high cytotoxic activities. This study was aimed to examine the effects of the combination of cisplatin, the ethanolic extract of Hedyotis corymbosa L.(EEHC), and the ethanolic extract of Tinospora crispa roots (EETC) on enhanced breast cancer cell sensitivity by apoptotic induction and cell cycle modulation. Cytotoxic effects were tested using the MTT assay in breast cancer cells, 4T1. The combination test of cisplatin and the extracts yielded combination index (CI) and cell viability, and its effects on the induction of apoptosis and cell cycle modulation were observed by the flow cytometry method. The results of the cytotoxic test showed CI of less than one for 1 mg/mL EEHC, 6 mg/mL EETC, and 1.68 mg/mL cisplatin. The EEHC-EETC-cisplatin combination accumulated cells in the S phase (29.98%) and induced apoptosis in 4T1 cells. It proves that EEHC and EETC can be developed as co-agents with cisplatin to improve the effectiveness of breast cancer chemotherapeutic treatment.

Abstract

Faloak (Sterculia quadrifida R.Br) is used empirically by residents of East Nusa Tenggara for drug plant. The information of the active compound contained in the faloak bark yet specifically published. This study aims to determine the active compound contained in the faloak bark that have cytotoxic activity to T47D breast cancer cells. Extraction process using maceration method, isolation process using gradien isolation method, elusidation using combination of information of IR spectra, 1D-NMR, 2D-NMR and LC-MS, and cytotoxic activity test on T47D breast cancer cells using MTT method. The isolation result obtained that isolate of naphthoquinone derivative compound is 2,3-dihydro-6-hydroxy-2-methylenenaphtho [1,2-b] furan-4,5-dione with IC50 value on T47D breast cancer cell was 9,88 µg/mL and with an index selectivity value was 30,23.

Abstract

Bakteri asam laktat (BAL) sangat baik digunakan dalam pencegahan efek samping bakteri patogen. Bakteri ini termasuk dalam golongan bakteri baik dan menguntungkan yang memiliki peranan besar untuk mencegah dan mengurangi pertumbuhan bakteri patogen. Sisa air cucian beras terkandung kikisan dari karbohidrat sehingga dapat digunakan BAL untuk berkembangbiak. Fermentasi air cucian beras diidentifikasi untuk mengetahui kandungan BAL spesifik yang terdapat didalamnya menggunakan Polymerase Chain Reaction (PCR). Identifikasi ini dilakukan menggunakan tahapan-tahapan metode yang dimulai dari pencarian isolat murni, uji aktivitas antimikroba, uji katalase, ekstraksi DNA, siklus PCR dan elektroforesis gel agarosa. Hasil penelitian terbentuk pita pada kisaran 300 bp yang artinya kedua primer terdeteksi dikarenakan Lactobacillus casei berada pada 254 bp sedangkan Lactobacillus rhamnosus berada pada 159 bp pada fermentasi air cucian beras.

Abstract

Stinkvine (Paederia foetida L.) is known as a plant that has many properties that are empirically used in traditional medicine. However, information of utilization of Stinkvine leaf outside as a traditional medicine has not been discovered yet. This research aims to determine the potential larvicidal extracts and fractions of stinkvine leaf. The ethanol extract of stinkvine leaf fractionated using column chromatography with n-hexane, ethyl acetate, and methanol. Larvicidal activity test against Aedes aegypti and Anopheles III instar was done using 25 larvae for each treatment extracts and fractions in 5 series of concentration, and left exposed for 24 hours. Total mortality of larvae is calculated and analyzed using a modified Finney probit to determine the LC50 and LC90 values. The results showed larvicidal activity on larvae of Aedes aegypti mosquitoes with LC50 and LC90 value of n-hexane, ethyl acetate, and methanol fraction respectively 67,89 ; 10,92 ; 30,98 µg/mL and 114,11 ; 18,80 ; 54, 12 µg/mL. Larvicidal activity on larvae of Anopheles mosquitoes with LC50 and LC90 value of n-hexane, ethyl acetate, and methanol fraction respectively 50,76 ; 14,96 ; 60,82 µg/mL and 87,23 ; 25,65 ; 90,92 µg/mL. Ethyl acetate fraction of ethanol extract of stinkvine leaf (Paederia foetida L.) shows to have highest larvicidal activity against the mosquito larvae of Aedes aegypti and Anopheles. The results of GC-MS analysis of the ethyl acetate fraction showed there are 15 compounds, a constituent component consisting of lupeol (20,32 %), gamma-sitosterol (12,22 %), cyclohexanecarboxamide (11,82 %), campesterol (11,45 %), 3-butenol, 4- (2,6,6-trimethyl-1-sikloheksenil) (7,98 %), siklolanos-24-en-3-ol (7,87%), and there are 9 other components with a percentage amount of 0,18 to 5,86 %.

Abstract

Desa Gondowangi adalah salah satu desa yang sebagian penduduknya mengandalkan sektor pertanian. Pada umumnya, petani di Desa Gondowangi mengalami masalah bedanya hasil pertanian dari berbagai lahan yang dimiliki. Pengabdian masyarakat ini dilaksanakan dengan metode pelatihan pengukuran kualitas tanah dan pembuatan infografik kualitas tanah. Kegiatan pengabdian dilaksanaan dengan cara pelatihan secara langsung di lahan milik para petani. Pelatihan pengukuran dilaksanakan secara langsung oleh petani pemilik lahan sehingga petani terbiasa menggunakan soil meter. Berdasarkan hasil pengabdian didapatkan berbagai lahan memiliki kualitas tanah yang berbeda dari segi pH dan kandungan unsur hara.
