Abstract

A study on molecular docking-based virtual screening has been conducted to select virtual hit of compounds, reported its existence in fungal endophytes of Chaetomium sp. as cytotoxic agent of breast cancer. The ligands were docked into Human Estrogen Receptor alpha (HERa) as the protein which regulates the breast cancer growth via estradiol-estrogen receptor binding intervention. The results showed that two compounds bearing xanthone and two compounds bearing benzonaphtyridinedione scaffolds were selected as virtual hit ligands for HERa leading to the conclusion that these compounds were good to be developed as anti breast cancer.

Abstract

Good pharmacy services are expected to build the positive image of the hospital. This includes the enhancement of competitive advantage among hospitals. The expected output of this research is identifying the effect of three dimensions of pharmacy services; interpersonal relations, therapy management and general satisfaction, to the image of the Baptis hospital, Batu City. The implementation of SWOT analysis is expected to determine the effect between variables, and the hospital can improve the competitive advantage. This study used quantitative and qualitative methods. Quantitative research involved 200 respondents and the method of statistical analysis was Multiple Linear Regression to investigate the effect of pharmacy services on the hospital image. The results of the study prove that pharmacy personnel at the Pharmacy Installation of Baptist Hospital Batu has provided good service, cared for patients, showed respect, and gave their willingness to provide information, education, counseling, and good activities. While the rating is classified as low, but still in good range is the patient's response time which is in the dimensions of general patient satisfaction. Qualitative research used the SWOT analysis and involved pharmacists of the Baptis Hospital. Generally, the result of statistical analysis revealed that the dimensions of pharmacy services have a positive effect on the hospital hospital image, both partially and simultaneously. In addition, the result of SWOT analysis demonstrated that Baptis Hospital is included in the first quadrant, which means the hospital runs a strategy that leads to the growth of the hospital. The hospital is in the perfect position that allows the management to rely on the strength in order to develop rapidly.

Abstract

Karang gigi merupakan salah satu keadaan patologis dari gigi. Salah satu penyebab karang gigi adalah bakteri Streptococcus mutans. Bakteri tersebut tidak bekerja sendiri tetapi didukung oleh beberapa faktor lainnya yaitu host (gigi dan saliva), substrat (makanan), dan waktu. Salah satu cara yang dapat dilakukan dalam pencegahan tumbuhnya karang gigi adalah dengan mengontrol akumulasi plak pada permukaan gigi. Tumbuhan masoyi merupakan salah satu tumbuhan yang tumbuh di Indonesia. Dari kajian teoritis yang ada, minyak atsiri masoyi (Massoia aromatica) mengandung senyawa yang mampu menghambat pertumbuhan bakteri. Penelitian ini bertujuan untuk mengetahui efektifitas minyak atsiri masoyi (Massoia aromatica) sebagai antibakteri terhadap  Streptococcus mutans dengan melihat nilai KBM (Kadar Bunuh Minimum) dan KHM (Kadar Hambat Minimum). Penentuan nilai KBM dan KHM dilakukan metode kualitatif dan kuantitatif. Pada pengujian dengan metode kualitatif dilakukan  disc diffusion test, metode dilusi cair, dan metode dilusi padat. Sedangkan pada pengujian kuantitatif dilakukan metode mikrodilusi. Penelitian ini menggunakan 8 konsentrasi yaitu 2000, 1000, 500, 250, 125, 62,5, 31,25, dan 15,625 µg/mL. Hasil penelitian dengan metode kualitatif menunjukkan nilai KHM didapatkan pada konsentrasi 500 µg/mL dan nilai KBM didapatkan pada konsentrasi lebih dari 2000 µg/mL. Hasil penelitian dengan metode kuantitatif menunjukkan nilai IC50 85,047 µg/mL dan IC90 491,481 µg/mL. Kesimpulan dari penelitian ini adalah minyak atsiri masoyi (Massoia aromatica) terbukti dapat menghambat pertumbuhan bakteri Streptococcus mutans.

Abstract

Albuminuria adalah kondisi adanya albumin pada urin. Adanya albumin dapat menjadi indikator penyakit ginjal dan kardiovaskular. Albumin dapat dideteksi berdasarkan perubahan warna biru menjadi kecoklatan menggunakan sensor perak iodida – metilen biru. Tujuan dari pengembangan tes cepat ini adalah mendeteksi albuminuria pada urin dengan memanfaatkan reagen perak iodida (AgI) dan metilen biru. Sensor kimia yang dikembangkan merupakan sensor kolorimetri yang terimobilisasi kertas dan dideteksi dengan metode scannometri. Analisis perubahan warna sensor untuk albumin pada penelitian ini menggunakan perangkat lunak imageJ. Sensor albumin berbasis perak iodida - metilen biru yang diusulkan memiliki waktu respon 7 menit dengan rentang linier 200 hingga 500 ppm dan batas deteksi adalah 83,039 ppm batas kualifikasi. Sensor memiliki reprodusibilitas (RSD = 2,6917%) dengan waktu pakai lebih dari 5 bulan dengan nilai akurasi 100,662%. Kata kunci: albuminuria, sensor kimia, tes cepat. ABSTRACT Albuminuria is a condition of the presence of albumin in the urine. The presence of albumin can be an indicator of kidney and cardiovascular diseases. Albumin can be detected based on the change of color from blue to brownish using a silver iodide – methylene blue sensor. This rapid test was developed to detect albuminuria in the urine by utilizing silver iodide (AgI) and methylene blue reagents. The chemical sensor was a colorimetric sensor immobilized with paper and detected by scannometric methods. The analysis of the sensor color changes for albumin was carried out using ImageJ software. The proposed silver iodide - methylene blue-based albumin sensor had a response time of 7 minutes with a linear range from 200 to 500 ppm and the detection limit of 83,039 ppm qualification limit. The sensor had reproducibility (RSD = 2.6917%) with a service life of more than 5 months with an accuracy value of 100.662%. Keywords: albuminuria, chemical sensor, rapid test.

Abstract

ABSTRACTPumpkin (Cucurbita moschata Duch ex Poiret), a fruit that contains several compounds, has activities to protect the human body from free radicals. Provitamin A is recognized as an antioxidant compound. Antioxidants can protect the body’s damage caused by free radicals. The purpose of this study was to isolate beta carotene compounds and test the purity of beta carotene isolates obtained using DR-UV and infrared spectrophotometry. Pumpkin is extracted using acetone: petroleum ether with a ratio (1: 1). Identification of beta carotene from extracts was carried out using TLC with the mobile phase of ethanol: acetone: n-hexane with a ratio (2: 1: 1) and scan spectrum using a UV-Vis spectrophotometer. Beta carotene was identified as the result of TLC elution on UV 254 and 366 nm which showed 2 spots. Detection of beta carotene spectra was carried out in the range of 350 to 550 nm and obtained beta carotene peaks at wavelength x nm. The extract purification process was carried out using Preparative TLC with the mobile phase of ethanol: acetone: n-hexane with a ratio (2: 1: 1) and obtained isolates cleaned first using chloroform: methanol ratio of 1: 1. The isolates were dried and tested using DR-UV and infrared spectrophotometry. Abstract

ABSTRACTLycopene is one of the phytochemical compounds of the carotenoid group, the main producer of red pigments found in watermelons. Watermelon is a round-shaped vines containing vitamin C and vitamin A and has high antioxidant levels. This study aims to identify lycopene compounds in watermelons. Extraction of watermelon flesh by maceration method. Identification of lycopene compounds with UV-Visibel spectrophotometry instrument followed by identification with the TLC method and fractionation of extracts using preparative TLC with chloroform: ethanol (1: 1) solvents, and identification of fractionation results with Fourier Transform Infrared Spectroscopy (FT-IR). The results of the identification of watermelon fruit extract using the TLC method obtained three spots and the results of UV-Vis spectrophotometry showed three peaks namely between 440-520 nm. The identification of fractionation of watermelon extract using FT-IR obtained a constituent group of lycopene structures, but did not show the R-CH = CH-R group that is owned by lycopene, whereas the results of the test with UV-Vis spectrophotometry appearing at 206 and 245 which indicate the presence of other compounds. From the results of the study it can be concluded that the testing of known extracts of watermelon fruit contains lycopene compounds, but the testing of lycopene identification on the results of fractionation of watermelon fruit extracts was not found with certainty the existence of lycopene because the FT-IR results were only shown and contained on the results UV-Vis spectrophotometry was found to be the wavelength of other compounds.keywords: Watermelon, Lycopene, UV-Vis Spectrophotometry, IR Spectrophotometry

Abstract

ABSTRACT People on Timor island use Faloak (Sterculia quadrifida R.Br.) bark as herbal remedy to cure diseases such as liver diseases, gastroenteritis and as stamina booster. This study aims to analyze the ability of anticancer fraction of faloak bark ethanol extract.  The mashed  bark of  faloak  plants extracted by maceration method using ethanol. The ethanol extract fractionated using preparative thin layer chromatography with  mixture  mobile phase which, chloroform, n-butanol, ethyl acetate with ratio (3: 4: 1,5). Chromatogram detected with visible light, UV254, UV366, and cerium sulfate reagent, then marked. Fraction obtained five fractions.  Cytotoxic  test against T47D cell and Vero cell  conducted using MTT, IC50  and selectivity sndex (SI) used as an anticancer efficacy parameter. Fraction 4  showed moderate anticancer activity with IC50 of 21.89 μg/mL in T47D cell line. Keywords: Faloak bark, anticancer activiy, T47D cell line

Abstract

INTISARI Antioksidan merupakan zat yang mampu memperlambat atau mencegah proses oksidasi. Zat ini secara nyata mampu memperlambat atau menghambat oksidasi zat yang mudah teroksidasi meskipun dalam konsentrasi rendah. Antioksidan juga sesuai didefinisikan sebagai senyawa-senyawa yang melindungi sel dari efek berbahaya radikal bebas oksigen reaktif jika berkaitan dengan penyakit, radikal bebas ini dapat berasal dari metabolisme tubuh maupun faktor  eksternal lainnya Radikal bebas adalah spesies yang tidak stabil karena memiliki elektron yang tidak berpasangan dan mencari pasangan elektron dalam makromolekul biologi. Protein lipida dan DNA dari sel manusia yang sehat merupakan sumber pasangan elektron  yang baik.Kondisi oksidasi dapat menyebabkan kerusakan protein dan DNA, kanker, penuaan, dan penyakit lainnya.Komponen kimia yang berperan sebagai antioksidan adalah senyawa golongan fenolik dan polifenolik.Senyawa-senyawa golongan tersebut banyak terdapat dialam, terutama pada tumbuh-tumbuhan, dan memiliki kemampuan untuk menangkap radikal bebas.Antioksidan yang banyak ditemukan pada bahan pangan, antara lain vitamin E, vitamin C, dan karotenoid. Kata kunci: Antioksidan, Musa Paradisiaca L, Karakteristik.  ABSTRACT  Antioxidants are substances that can slow down or oxidation process. This substance can significantly slow or overcome oxidation of oxidizable substances in low concentrations. Antioxidants also suit their forms containing reactive oxygen-free  fuel cells when they are associated with disease, these free radicals can be derived from body metabolism and other external factors Free radicals are unstable species because they have unpaired electrons and look for electron pairs in biological macromolecules. Lipid proteins and DNA from healthy human cells are good sources of electron pairs. Oxidation conditions can cause damage to proteins and DNA, cancer, aging, and other diseases. Chemical components that act as antioxidants are phenolic and polyphenolic group compounds. These compounds are widely found in the garden, especially in plants, and have the ability to fight free radicals. Abstract

Abstrak Rimpang lengkuas (Alpinia galanga L. Willd) merupakan tanaman yang memiliki banyak khasiat, salah satunya sebagai antijamur. Kandungan senyawa pada rimpang lengkuas dianggapmemilikiaktivitas enghambatan terhadap jamur khususnya Candida albicans yang merupakan penyebab penyakit kandidiasis.Penelitian ini menggunakan ekstrak lengkuas sebagai alternatif obat antijamur alami pada Candida albicans serta untuk mengetahui kadar efektif ekstrak rimpang lengkuas (Alpinia galanga(L) Willd) terhadap jamur Candida albicans dalam nilai KHM50 dan KHM90.Rancangan penelitian ini menggunakan experimental murni sederhana atau posttest onlycontrol group design dengan menggunakan dua metode uji yaitu metode difusi cakram dan mikrodilusi. Kemudian dilakukan pengamatan zona hambat yang akan terlihat zona bening pada cawan petri, menghitung persen penghambatan dengan cara perhitungan absorbansi atau optical density (OD) dan dianalisis menggunakan Analisis Probit untuk melihat nilai KHM50 dan KHM90.Hasil penelitian menunjukkan bahwa ekstrak rimpang lengkuas memiliki zona hambat sebesar 1,75 mm. Sedangkan dalam perhitungan nilai KHM50 KHM90 menggunakan analisis probit didapatkan nilai KHM50 terdapat pada konsentrasi 1834.933 µg/mL dan nilai KHM90 terdapat pada konsentrasi 13374.761 µg/mL. Wild.) is one of plants which have many savors, one of them is used for anti-fungus. The structure contained in Galanga rhizome is well known for the obstructive activity to the fungus, for especially Candida albicans which is the cause of the Candidiasis disease. This study used galanga rhizome extract as an alternative to natural antifungal for Candida albicans and to find out the effectiveness amount of extract Galanga rhizome (Alpina galanga L. Wild) to the Candida albicans fungus in the term of MIC50 and MIC90. This study is plain experimental or post –test only control group design by using two test methodology. There are disk diffusion method and microdilution. Then, followed by observing obstruction zone which will be appeared on the clear zone on the petri dish, valuating obstruction presentation by using absorbency counting or optical density (OD) and analyzed using probit analysis to see MIC50 and MIC90. The result of the study showed that extract Galanga rhizome had obstructive zone at least 1,75 mm. But, on the valuating MIC50, MIC90 point using probit analysis, the MIC50 concentration value was 1834.933µg/mL and the MIC90 concentration value was 13374.761 µg/mL. Keywords: Alpinia galanga L. Abstract

AbstrakPterygota alata (Roxb) R. Br atau yang biasa dikenal dengantanaman Bipa atau Coconut Buddha adalah tanamanmultifungsi yang tersebar di Myanmar serta India danmemiliki manfaat sebagai obat penyakit kulit, diabetesmellitus, gembur- gembur dan lain sebagainya. Tujuan daripenelitian ini adalah untuk melihat aktivitas sitotoksik,pengamatan apoptosis serta siklus sel terhadap sel kankerpayudara MCF-7. Tanaman di ekstrak menggunakan metodemaserasi sehinga diperoleh ekstrak kental yang digunakandalam pengujian. Uji sitotoksik menggunakan metode MTTassay dimana akan terbaca hasil absorbansi menggunakanELISA reader dengan parameter dari pengujian ini yakni nilaidari IC50. Pengujian Apoptosis dan siklus sel menggunakanmetode flowcytometry. Hasil hasil penelitian pengujiansitotoksik didapatkan nilai IC50 sebesar 2,44µg/mL.Pengujian siklus sel didapatkan akumulasi yang terjadi padafase S sebesar (22,11%) dan dapat menginduksi apoptosis.Dari hasil penelitian maka dapat disimpulkan bahwa bijipterygota alata memiliki aktivitas sitotoksik, dapatmeningkatan induksi apoptosis dan juga siklus sel terhadap selkanker payudara MCF-7 dan dapat dikembangkan sebagaiagen kemoterapi kanker payudara.Kata Kunci: Pterygota alata, kanker payudara, MCF-7,sitotoksik, apoptosis, siklus sel.AbstractPterygota alata (Roxb) R. The purposeof this study was to look at cytotoxic activity, pregnancy cycle,and cell cycle against breast cancer MCF-7. Plants wereextracted using the maceration method so thick viscous extractwas used for testing. Cytotoxic test using the MTT assaymethod which will read the results of absorbance using ELISAreader with the parameters of this test is the value of IC50.Apoptosis and cell cycle testing uses the flow cytometrymethod. The results of the cytotoxic test results obtained IC50values of 2.44?g / mL. Cell cycle testing was obtained whichoccurred in the S phase by (22.11%) and can induce apoptosis.From the results of the study, it can be concluded that thepterygoid alata seeds have cytotoxic activity, can increaseapoptosis induction, and also the cell cycle to breast cancerMCF-7 and can be developed as a chemotherapy agent forbreast cancer.Keywords: Pterygota alata, Breast Cancer, MCF-7, cytotoxic,apoptotic, cell cycle.
