Abstract

Diabetes merupakan penyakit menahun atau kronis berupa gangguan metabolik yang ditandai dengan kadar gula darah yang melebihi batas normal. Penelitian ini bertujuan untuk mengetahui gambaran tingkat perilaku ketidakpatuhan penggunaan obat antidiabetes pada kelompok kontrol dan kelompok intervensi sebelum dan sesudah edukasi flipchart. Hasil analisis tingkat ketidakpatuhan, didapatkan adanya penurunan dalam persen ketidakpatuhan pada pertanyaan 2, 5, 6, 7, dan 8 yang cukup tinggi perbedaannya setelah adanya intervensi. Pada pasien kelompok kontrol yang tidak diberikan perlakuan, cenderung konsisten dengan tingkat perilaku ketidakpatuhannya. Pada hasil analisis pola ketidakpatuhan menunjukan bahwa pasien memiliki alasan beragam. Beberapa pasien mengalami ketidakjelasan dalam informasi yang tertera pada label, sehingga menggunakan aturan sendiri. Kecocokan pola ini berkisar diantara 75% hingga 88%. Ada juga pasien yang mengganti obat mereka dengan jamu, serta pasien menghemat penggunaan obat agar cukup digunakan. Kecocokan pola ini berkisar antara 69% dan 94%. Hasil tersebut mengidentifikasi beberapa faktor yang memengaruhi ketidakpatuhan, pemahaman yang kurang baik tentang penggunaan obat, jadwal harian yang padat, dukungan sosial, dan kendala fisik. Meskipun edukasi flipchart berhasil meningkatkan pemahaman pasien dan mengurangi tingkat ketidakpatuhan, masih diperlukan penekanan lebih lanjut pada aspek-aspek tertentu yang menunjukkan ketidakpatuhan yang tinggi setelah intervensi.

Abstract

In recent years, the search for new anticancer drugs from local plants in Indonesia has become an interesting topic for research. One of the typical Indonesian local plants that have been used by the community as an anticancer traditional medicine (jamu) is Spatholobus littoralis Hassk (SLH) or better known as Bajakah. The native Dayak people of Kalimantan have used it for years for the treatment of breast cancer. In this study, the separation of single compounds from the ethyl acetate fraction of SLH stems was carried out, their molecular structure was analyzed, and anticancer bioactivity was tested. The isolation used the preparative HPLC method. The elucidation of the isolates from the ethyl acetat fraction used LCMS-MS, FTIR, and NMR. Anticancer bioactivity was determined using the MTT bioassay. The result is the acquisition of two isolates; it has been predicted that both are pseudobaptinin. The anticancer IC50 of the pseudobaptinin was 62.85 ppm, and Pseudobaptinin with impurities was 145.60 ppm.

Abstract

ABSTRAK: Faloak (Sterculia quadrifida R.Br) digunakan secara empiris oleh penduduk Nusa Tenggara Timur untuk mengobati hepatitis, tifus, maag, dan pemulih stamina. Informasi senyawa aktif yang terkandung didalam kulit faloak secara spesifik belum dipublikasi. Penelitian ini bertujuan untuk mengetahui senyawa aktif yang terdapat didalam kulit faloak sebagai antikanker. Ekstraksi menggunakan metode maserasi, isolasi menggunakan metode isolasi bertingkat, elusidasi menggunakan penggabungan informasi dari spektra IR, 1D-NMR, 2D-NMR dan LC-MS, dan uji aktivitas antikanker pada sel kanker payudara T47D menggunakan metode MTT. Hasil isolasi diperoleh isolat turunan senyawa naptokuinon yaitu 2,3-dihydro-6-hydroxy-2-methylenenaphtho[1,2-b]furan-4,5-dione yang aktif sebagai antikanker dengan nilai IC50 pada sel kanker payudara sebesar 9,88 µg/mL dan dengan nilai selektivitas indeks sebesar 30,23. ABSTRACT: Faloak (Sterculia quadrifida R.Br) is used empirically by residents of East Nusa Tenggara to treat hepatitis, typhoid, ulcers, and stamina restorers. The information of the active compounds contained in the faloak skin is not specifically published. This study aims to determine the active compounds contained in the bark of faloak as anticancer. The extraction was conducted with maceration method followed by a multilevel isolation method. The elucidation was carried out using information of IR spectra, 1D-NMR, 2D-NMR and LC-MS. The anticancer activity test on T47D breast cancer cells was also conducted using MTT method. Based on the results obtained, the active compound is naphthoquinone derivative compound which is 2,3-dihydro-6-hydroxy-2-methylenenaphtho [1,2-b] furan-4,5-dione that has anticancer activity on breast cancer cell (T47D) with IC50 value of 9.88 µg/mL and index selectivity value of 30.23.

Abstract

Background: Diabetes Mellitus is a non-contagious disease characterized by hyperglycemia. Diabetes mellitus occurs when the body cannot receive or use insulin properly. If you already have diabetes, then the patient must take medication continuously because diabetes mellitus is a lifelong disease. Because medicines are quite expensive, alternative ways to cure the disease are needed by consuming traditional medicines, one of which is bitter melon (Momordica charantia). Objectives: This research aims to predict the secondary metabolite compounds in the bitter melon plant, analyze molecular interactions, and identify compounds that can lower blood sugar levels. Material and Methods: 12 compounds from the Momordica charantia plant and six proteins (1IR3, 1RHF, 1XU7, 4PNZ, 4YVP, 2NT7) that will be docked using Pyrx and Yasara Dynamics applications. Results: From the molecular docking results, three compounds with the highest binding affinity were found in Momordica charantia: momordenol, oleanolic acid, and momordicin. Based on molecular dynamics simulations, these three compounds were stable in their interactions with the six proteins tested, namely 1IR3, 1RHF, 4PNZ, 4YVP, 1XU7, and 2NT7. Momordenol and momordicin showed the most stable interaction profiles. Furthermore, ADMET tests showed that momordenol, oleanolic acid, and momordisin have drug-like characteristics. Conclusions: The Momordica charantia plant has the potential to act as an antidiabetic agent.

Abstract

Growing evidence suggests that biofilm formation in Pseudomonas aeruginosa (PA) results from quorum sensing, reducing bacterial sensitivity to antibiotics and fostering the development of antibiotic-resistant strains. Addressing the need for a compound that can inhibit quorum sensing signals to mitigate biofilm formation in PA, this study screened 40 essential oils for their potential as quorum sensing inhibitors against PA ATCC9027. Utilizing the agar well diffusion assay, the antibacterial activity of these essential oils was compared with levofloxacin, revealing that 25 out of 40 essential oils exhibited antibacterial activity against PA ATCC9027. The highest antibacterial activity was exhibited by essential oils from Cinnamomum burmannii (Cinnamon Bark Oil), Cananga odorata (Ylang-ylang Oil), and Eucalyptus globulus (Eucalyptus Oil), confirmed by respective MIC values of 0.09765% (v/v), 0.390625% (v/v), and 1.5625% (v/v), and MBC values of 0.09765% (v/v), > 1.5625% (v/v), and > 1.5625% (v/v). Growth curve inhibition assay followed by crystal violet biofilm formation assay revealed that only Ylang-ylang Oil exhibited biofilm formation inhibition at 1/4 MIC concentration. Docking analysis of compounds from these essential oils against quorum sensing enzymes (LasR, PqsR, and QscR) revealed that Ylang-ylang Oil compounds exhibited the highest binding affinity. In conclusion, among the 40 essential oils tested, 25 show potential as antibacterials against PA ATCC9027, with Ylang-ylang Oil standing out as a promising candidate for quorum sensing inhibitor development. The use and development of quorum sensing inhibitors from Ylang-ylang Oil compounds are expected to reduce the burden of antibiotic resistance, especially in Pseudomonas aeruginosa.

Abstract

Faloak (Sterculia quadrifida R.Br) is used empirically by residents of Timor island to treat hepatitis, typhoid, ulcers, and to restore stamina. Information of active compound contained in the bark of faloak specifically unpublished. This study aims to determine the active compound contained in faloak bark that can be used as antibiofilm. The extraction was performed by soxletation method, the isolation was performed by gradien isolation method, the elusidation was performed by merging information from NMR and LC-MS spectra analysis. The biofilm inhibition activity test was performed by microdilution method formed on flat bottom flexible microplate U-bottom PVC 96 wells with staining using 1% crystal violet. The isolation process obtained 3 gallic acid derivatives, named isolate 1, isolate 2, and isolate 3. Test of inhibition of biofilm formation showed isolate 1 has IC50 of 46,87 µg/mL, isolate 2 has IC50 of 45,87 µg/mL, and isolate 3 has IC50 42,65 µg/mL.The biofilm test results showed that isolates 1-3 had high biofilm inhibition potency.

Abstract

Essential oils had been used as antibacterial, antifungal, antiviral, antioxidant and insecticidal in medicine and industry. Increasing the usage of essential oils as an alternative medication also lead more research of essential oils in vivo and in vitro. Cinnamon with cynamaldehide as a main active substance and masoyi with massoia lacton as a main active substance have antibacterial activity.This study aimed to determine the activity of masoyi and cinnamon essentials oils combinationin E.coli, S.aureus, and P.aeruginosabacteria. The research encompasses antibacterial activity screening and antibacterial evaluation. The aims of essential oil activity screening using Kirby-Bauer method is to find the optimum concentration of masoyi and cinnamon essentials oils combination. Determination of MIC50, MIC90, and MBC concentration is using microdilution method. Abstract

Uncaria gambir Roxb (URG) is one of the plants from West Kalimantan predicted to contain antioxidant and anti-breast cancer. This study aims to test the antioxidant and anti-breast cancer potential. UGR leaves were extracted by infusion method using water for 15 minutes with 4 repetitions at a temperature 70oC. A thick extract of 96.2351 grams (29.979%) was obtained from a sample weight of 321 grams of dried UGR leaves. In vitro antioxidant assay of the extract was investigated using 2,2-diphenylpicrylhydrazyl (DPPH) with positive control using quercetin and ascorbic acid, and the Ferric Reducing Antioxidant Power (FRAP) method. In silico screening showed that the biological agents in UGR had the potential as TP53 expression enhancer, antioxidant, anticarcinogenic, chemopreventive, and free radical scavenger. The antioxidant bioassay results showed IC50 values of 81.21 μg/mL, 73.39 μg/mL, and 9.17 μg/mL in DPPH for extract samples with positive control quercetin, sample extracts with positive control Vitamin C and Vitamin C with positive control quercetin, respectively. Meanwhile, the antioxidant activity of extract samples with the FRAP method showed value of 66,05 μg/mL. Anticancer bioassay result showed that UGR leaves extract with water solvent had the stron potentgial to inhibit 4T1 cells with IC50  87.72 μg/mL.

Abstract

Indonesia has a diversity medicinal plants as traditional medicine. Traditional medicine has shortcomings in the use of the main composition and components used, making it less effective. Therefore, medicinal plants need to be optimized in composition and add two or more plants to maximize treatment. Medicinal plants can be antibacterial. Indonesia has the ciplukan and areca nut plants that have antibacterial potential. There are 5 stages of work procedures in the research method. The first is plant determination. The second is the optimization composition using the simplex lattice design method. The third is extraction by the maceration method. The fourth is an antibacterial activity test. The last of data analysis uses software design experts and SPSS. The results were composition optimization with the simplex lattice design method resulting in 8 compositions. The results of the antibacterial activity test were presence of antibacterial activity in composition 7 (87.5 μg and areca nut shell 62.5 μg) with an inhibition diameter of 11.32 mm and composition number 8 (62.5 μg and areca nut shell 87.5 μg) with an inhibition diameter of 11.58 mm. The final result of the confirmation of composition 8 has the strongest antibacterial potential shown by the diameter of inhibition of 15.27 mm and the desirability value, which is 1.

Abstract

Pada luka diabetes, kadar TNFα meningkat dan menginduksi sekresi MMP9 secara berlebihan sehingga meningkatkan laju degradasi ECM. Hal ini beresiko menyebabkan luka kronis yang tak kunjung sembuh dan kemungkinan terjadinya infeksi. Oleh karena itu peningkatan proses penyembuhan luka menjadi sangat penting, salah satunya dengan menghambat aktivitas MMP9 dan TNFα. Bahan alam terutama rempah-rempah khas Indonesia dipilih karena memiliki efek samping yang minimal dan ketersediaannya melimpah. Tujuan dari penelitian ini adalah skrining aktivitas konstituen utama dalam rempah-rempah khas Indonesia terhadap MMP9 dan TNF-α dengan metode penambatan molekul secara in silico. Dari skrining terhadap 106 senyawa diperoleh hasil sebanyak 4 senyawa yang memiliki energi ikatan terendah pada kedua protein yaitu hinokinin, procyanidin, xanthorrhizol, cubebin, dan rosmarinic acid yang menunjukkan potensi untuk menghambat kedua protein tersebut sekaligus. Terlebih lagi, procyanidin memiliki energi ikatan yang lebih rendah dibandingkan ligan natif masing-masing protein (-9,2 kkal/mol terhadap MMP9 dan -9,1 kkal/mol terhadap TNFα) dengan nilai Ki sebesar 0,180 µM dan 0,213 µM secara berturut-turut. Dengan demikian dapat disimpulkan bahwa procyanidin merupakan kandidat yang paling menjanjikan sebagai agen terapeutik dalam penyembuhan luka pada pasien diabetes melitus.
