Abstract

Background: Streptozotocin as diabetogenic can damage the pancreatic β cells of animals tried through the oxidative stress process to increase blood sugar levels. Giving ethanolic extract of squash fruit has hypoglycemia effect because it contains flavonoids that act as antioxidants and antihiperglichemia. This study aimed is analyze the effect of ethanol extract of squash fruit to decrease blood sugar level and the change of pancreatic β cell diameter.Subjects and Method: This study was an experimental study with post-randomized controlled group design, using male white mice (Mus musculus L.) DD Webster strains randomized into 4 groups: negative control group, positive control group, group with extract ethanol of 100 mg/ kgBB, and a group of ethanol extract of 200 mg/ kgBW of pumpkin.Results: The results showed a significant reduction in blood sugar levels if compared with the control group. The presence of changes in β pankreas cell diameter on ethanol extract of 100 mg/ kgBB and 200 mg/ kgBB.Conclusion: The conclusion of this study is the extract of ethanol fruit of 200mg/ kgBB squash significantly reduce blood sugar level of mice, the change of β pankreas cell diameter on ethanol extract of 100mg/ kgBB and 200mg/ kgBB.Keywords: streptozotocin, antihipoglikemia, flavonoidCorrespondence: Jekson Martiar Siahaan. Abstract

Background: Streptozotocin can cause hyperglycemia in guinea pig through the mechanism of oxidative stress which damages pancreatic b cells. Ethanol extract of chayote can decrease oxidative stress.  This study aimed to determine the effect of Ethanol Extract of Chayote ((EEBLS) Ekstrak Etanol Buah Labu Siam) on decreasing blood sugar content and increasing the activity of glutathione peroxide enzyme. Subjects and Method: This was an experimental study. The samples were using random sampling technique. The control group was using male white mice (Musmusculus L.) Strain DD Webster which randomized into four groups: negative control group, positive control group, group which got EEBLS of 100 mg/kgBB, and group which got EEBLS of 200 mg/kgBB.Results: The result of the research showed that there was significant decrease in blood sugar, compared with the control group. There was insignificant decrease in the activity of glutathione peroxide enzyme, compared with the control group.Conclusion: The conclusion of the research was that EEBLS of 200 mg/kgBB decreased blood sugar content of mice significantly, but there was no significant change in the activity of glutathione peroxide enzyme when EEBLS was given to the mice.   Keywords: streptozotocin, oxidative stress, antioxidant, flavonoidCorrespondence: Jekson Martiar Siahaan. Abstract

Background: Diabetes mellitus (type 2 DM) is now ranked first in health problems and its prevalence tends to increase worldwide. The Diabetes Control and Complication Trial (DCCT) study showed that lowering HbA1c rates may delay or prevent chronic complications. One of the risk factors for CHD in type 2 DM is dyslipidemia, i.e lipid metabolism disorders in the form of elevated total cholesterol, triglyceride (TG), low-density lipoprotein (LDL), and decreased levels of high-density lipoprotein (HDL). This study aimed to determine the correlation between HBA1C and blood sugar, as well as the lipid profile in patients with type 2 DM.Subject and Methods: This was an analytic observational study with a cross-sectional design. The study was conducted at the Endocrinology and Metabolic polyclinic Pirngadi Hospital, Medan, North Sumatera, from January to February 2018. A total sample of 10 patients diagnosed with type 2 diabetes mellitus with obesity was selected for this study. The independent variables were blood sugar, lipid profile including total cholesterol, high-density lipoprotein (HDL), low-density lipoprotein (LDL), and triglyceride. The dependent variable was the HbA1c level. Lipid profile was measured by blood serum examination. HbA1c level was measured by blood serum examination. The data were analyzed by Pearson correlation.Results: Hba1c was positively correlated with blood sugar levels and was statistically significant (r= 0.81; p= 0.005). HbA1c correlated weakly and was statistically non-significant with total cholesterol (r= 0.14, p= 0.702), LDL (r= 0.15; p= 0.683), HDL (r= 0.04; p= 0.922), and triglycerides (r= 0.06; p= 0.860).Conclusion: HbA1c correlates with blood sugar levels but does not show a correlation with lipid profiles.Keywords: HBA1C, lipid profile, blood sugar, type 2 diabetes mellitusCorrespondence:Endy Julianto. Abstract

Background: The prevalence of diabetes mellitus (DM) from year to year is increasing. Hyper­glycemia that occurs in DM is caused by oxidative stress that damages pancreatic ? cells. This situation can be controlled with synthetic hypoglycemic drugs, but it requires high medical costs. Therefore, alternative therapies that are easily available, relatively inexpensive, have potential anti-hypoglycemia and anti-cholesterolemia which is found in conjoined flasks (Sechium edule (Jacq.) Swartz) which contains flavonoids are needed. The purpose of this study was to analyze the effectiveness of ethanol extract and fractionation of ethyl acetate extracts of Sami (pumpkin) (Sechium edule (Jacq.) Swartz) as anti-hipoglycemia and anti-hypertriglycerides in white male wistar rats (Rattus novergius sp.) induced by STZ-NA-HFDD.Subjects and Method: A randomized controlled trial was conducted to assess the effectiveness of ethanol extract and fractionation of pumpkin fruit ethyl acetate extract. The sample included white male wistar rats (Rattus novergius sp.) Hyperglycemia induced by STZ-NA-HFD. The dependent variable was triglyceride levels. The independent variables were administration of ethanol extract and fractionation of pumpkin fruit ethyl acetate extract.Results: Ethyl acetate fraction 45 mg/kgBW better reduce triglyceride levels but statistically does not have significant differences between groups.Conclusion: The group that received Metformin and the group that was given ethanol extract 45 mg/kgBW had lower triglyceride levels compared to the other therapy and control groups.Keywords: Hipertrigliseridemia, diabetes mellitus, STZ-NA-HFDCorrespondence: Jekson Martiar Siahaan. Abstract

Background: Trichuris trichiura worm infec­tion is still a major public health problem in Indo­­nesia, especially in peripheral areas. Until now, the search for antihelmintic combinations that have a high healing ability against T. tri­chi­ura infections and in the search for truly effective worm­ing is still being done. Albendazole and Mebendazole single doses still show limited ef­fi­ca­cy in the treatment of T. trichuria worms. The combination of Albendazole with Levamisol can increase effectiveness, but this combination still needs further investigation. Based on that fact, it is necessary to study the effectiveness compa­rison of the degree of infection intensity between the treatment of Albendazol-Levamisol and Me­ben­dazol-Levamisol against T. trichiura.Subjects and Method: A double blind ran­domized control trial was conducted at the State Elementary School in Medan, North Sumatera, from April to October 2019. A sample of 60 children was selected for this study. The depen­dent variable was the number of eggs decreased, the cure rate, and side effects. The independent variables were Albendazole 400 mg-Levamisol 50 mg/ 100 mg and Mebendazole 500 mg-Levamisol 50 mg/100 mg. The stools of elementary school children were examined using the Kato-Katz method to find eggs of T. trichiura infection. Data analysis was performed using the Chi square test. Results: Samples in the Albendazol-Levamisol and Mebendazol-Levamisol groups were 30 children, respectively. Children infected with T. trichiura consisted of 58 children of mild in­tensity (96.7%) and 2 children of moderate inte­nsity (3.3%). On the results of stool examination, a significant difference in the number of eggs decreased in the two groups was obtained (p <0.001) and 96.7% was the cure rate for T. trichiura infection in the two groups of Alben­dazol-Levamisol and Mebendazol-Levamisol. In this study, there were no significant differences in side effects in the two treatment groups.Conclusion: Treatment of T. trichiura infection with single doses of Albendazol-Levamisol and Mebendazol-Levamisol showed that there were sig­nificant differences in the Number of D­ecreased Eggs. Abstract

Background: Cholesterol imbalance occurs in Diabetes Mellitus, where high-density lipo­protein (HDL) levels are low and low-density lipoprotein (LDL) increases, where this im­balance results in the appearance of athero­sclerosis. HDL has antiarterosclerotic poten­tial so it needs to be targeted for therapy. One of the bioactive compounds that have this effect is found in chayote (Sechium edule (Jacq.) Swartz). This study aimed to examine the effects of ethanol extract, Chayote (Sechium Edule (Jacq.) Swartz) fraction and juice on the hdl level in male white miceSubjects and Method: A randomized controlled trial study was conducted to test HDL level in 54 male white rats of the Wistar strain (Rattus novergius sp.) after received ethanol extract, Chayote (Sechium Edule (Jacq.) Swartz) fraction and juice. The depen­dent variable was the HDL level. The indepen­dent variable was the variation in the dose of ethanol extract, fraction and Sechium edule (Jacq.) Swartz Juice. The data were analyzed using Anova test.Results: The treatment group induced by Streptozotocin 50 mg / kgBW + nicotinamide (120 mg/kgBW) + HFD and obtained ethanol extract of chayote fruit 150 mg / kgBW, orally had the highest average HDL levels compared to other groups (Mean = 17.31; SD = 3.14).Conclusion: The ethanol solvent found in the ethanol extract is better because it attracts biaoctive compounds so that it has a better effect than the ethyl acetate fraction and the n-hexane fraction.Conclusion: The ethanol solvent found in the ethanol extract is better because it attracts biaoctive compounds so that it has a better effect than the ethyl acetate fraction and the n-hexane fraction.Keywords: HDL, Flavonoid, Sechium edule (Jacq.) Abstract

Background: The rising diabetes mellitus (DM) population is a public health concern. Pharmaco­therapy for DM can cause side effects and is often inaccessible, necessitating alternative therapies like medicinal plants. Breadfruit leaves (Artocarpus altilis) have antidiabetic effects. This study aimed to examine the antihyperglycemic effect of ethanol extract and ethyl acetate fraction of bread­fruit leaves on alloxan-induced diabetic male white rats (Rattus norvegicus). Subjects and Method: This was a randomized controlled trial. Sample was 35 male white Wistar rats (Rattus norvegicus sp.) divided into 7 groups. The study was conducted at the Phytochemical Laboratory and Animal House, Faculty of Medicine, Indonesian Methodist University, from April to May 2023. The dependent variables were body weight, blood glucose levels, and diameter of the islets of Langerhans. The independent variables were variations in the dose of ethanol extract and ethyl acetate fraction of breadfruit leaves. Body weight was measured by scales. Blood glucose was measured using glucometer. Diameter of the islets of Langerhans was measured using computer-based image analysis system. Data were analyzed using Anova and Kruskal-Wallis tests. Results: Weight loss was better in the group given the ethyl acetate fraction at a dose of 200 mg/kgBW compared to the other groups. Administration of ethanol extract at a dose of 200 mg/kgBW showed a better reduction in blood glucose levels compared to the other groups, where P < 0.05 between K4 and K3, K5, and K7 on day 5. Ethanol extract at a dose of 200 mg/kgBW was able to improve the diameter of the islets of Langerhans induced by alloxan. Conclusion: A 200 mg/kg BW dose of ethyl acetate fraction reduces body weight. A 200 mg/kg BW dose of ethanol extract reduces blood glucose and improves pancreatic islet diameter.

Abstract

Background: Diabetes mellitus is a chronic disease in the form of metabolic disorders, which is characterized by blood glucose levels (KGD) that exceed normal limits and are accompanied by impaired metabolism of carbohydrates, lipids, and proteins, as a result of abnormalities in insulin secretion. Diabetics are known to have slow wound healing. Moringa oleifera leaves or moringa plants contain flavonoid compounds, which can decrease glucose absorption. This study aims to determine the effect of moringa leaf infusion on blood sugar levels, glucose tolerance, cholesterol levels, body weight, and wound healing speed in mouse model animals. Subject and Method: This study is experimental using a posttest only controlled group design, using 25 male mice (Mus musculus) divided into 5 groups. The study was conducted at the Phytochemistry and Animal House Laboratory, Faculty of Medicine, Methodist University Indonesia from March to May 2022. The bound variables were body weight measured using a scale, current blood glucose levels and blood glucose levels with glucose loading measured with strips and glucometers, total cholesterol levels measured using strips and cholesterol measuring devices, and the length of mice wounds measured using a ruler. The free variable is the variation in the dose of moringa leaf infusion. The data were analyzed using the Anova and Kruskal-Wallis tests. Results: The administration of moringa leaf infusion had a significant effect on the reduction of the levels of each variable, namely a dose of 10% in body weight, a dose of 30% in KGD, 20% in KGD with glucose loading, 20% in cholesterol, and 20% in wound closure, which was proven based on the results of statistical tests. Conclusion: Moringa leaf infusion can lower blood sugar levels, blood sugar levels by glucose depletion, cholesterol levels, body weight, and accelerate wound healing in sucrose-induced mice, high fat diet, and PTU.

Abstract

Background: Diabetes Mellitus is character­rized by metabolic disturbances due to pro­longed hyperglycemia, causing oxidative stress, which destroys pancreatic β cells. Adjuvant therapy that has antihyperglycemic effective­ness and is required to improve the diameter of pancreatic beta cells, one of which comes from Sechium edule (Jacq.) Swartz has poten­tial as an antihyperglycemic, antioxidant, anti­apop­tosis, cardioprotective, insulin resistance. This study aimed to examine the effectiveness of ethanol extract, Chayote (Sechium Edule (Jacq.) Swartz) fraction, and juice on the pancreatic β-cell diameter of white male rats Wistar strain with type 2 diabetes mellitus.Subjects and Method: This was a rando­mized controlled trial. Sample was Rattus novergius sp. 54 tails. The dependent variable was the diameter of the pancreatic β cells. The independent variables were ethanol extract, Sechium edule (Jacq.) Swartz fraction and juice, at a dose of 50 mg/kg BW, 100 mg/ kg BW, 150 mg/kg BW. The data were analyzed by the ANOVA test.Results: The group of mice induced by Strep­tozotocin 50 mg/kg BW + nicotinamide (120 mg/ kg BW) + HFD and obtained ethanol extract of chayote fruit 150 mg/ kg BW, orally, had the highest pancreatic β cell diameter com­pared to the other groups (Mean= 284.03; SD= 5.15).Conclusion: Sechium edule (Jacq.) Abstract

Background: Wounds are skin disorders in the form of damage to continuity due to a pathological process, if not treated properly it can cause serious complications such as failure to heal and chronic problems due to untreated wound infections, especially post-operative infections and scar tissue. The next problem is that excessive antibiotics can cause side effects and increase the risk of antimicrobial resistance and delay wound healing. So a new strategy is needed with herbal-based therapy that has the effect of treating infections and does not cause side effects in patients. Subjects and Method: The research was conducted with a post test only randomized control group design to test macroscopic observations of the average size of the wound length, hyperemia and swelling in 30 white Wistar rats (Rattus norvegicus) after receiving ethanol extract of rosella petals (Hibiscus sabdarifa L.). The dependent variables of the study were wound closure time, hyperemia and wound swelling. The independent variable of the research was varying doses of ethanol extract of rosella flower petals (Hibiscus sabdarifa L.) 6%, 12% and 15%. The parameters assessed are the length of the wound and the macroscopic assessment of the wound. Data were analyzed using the Kruskal-Wallis test, p<0.05 was considered statistically significant. Results: Administration of Hibiscus sabdariffa L. 6%, 12% and 15% ethanol extract gel on the 12th day resulted in a significant acceleration of the wound healing process (p=0.024). There was a significant difference between negative control (Mean=3,281 mm ; SD=1,681 mm), positive control (Mean=2,813 mm ; SD=1,106 mm), 6% Rosella flower petal extract gel (Mean=2,916 mm ; SD=1,224 mm ), 12% (Mean=1,290 mm ; SD=1,098 mm), and 15% (Mean=1,035 mm ; SD=0.952 mm) (p=0.024), in the macroscopic assessment there was no significant difference in the wound healing process on day 3, 6, 9, 12 and 15 except for signs of swelling on days 6 (p=0.038) and 9 (p=0.038) which had a significant difference compared to controls. Conclusion: Administration of Hibiscus sabdariffa L. 15% ethanol extract gel had the fastest time in wound closure and healing (Mean=1.035 mm ; SD=0.952 mm) for 12 days (p=0.024), the longest time in wound closure and healing was the negative control (gel placebo) (Mean=0.976 mm ; SD=1.103 mm) namely for 15 days (p=0.158).
