Abstract

Biofilm formation is a virulence factor contributing to the pathogenicity and antibiotic resistance of Klebsiella pneumoniae. This process occurs due to biofilm genes in clinical isolates, especially the mrkA, mrkD, and fimH genes. These three genes are related the quality of biofilms formed, which lead to chronic and prolonged infections. This study was conducted to determine the presence of mrkA, mrkD, and fimH genes as biofilm formers in Klebsiella pneumoniaeclinical isolates. The study used a descriptive in vitro laboratory study, with total of 24 Klebsiella pneumoniae clinical isolates from the collection of the Department of Microbiology, Faculty of Medicine, Universitas Tanjungpura. Isolates were subculture in MacConkey Agar, Gram staining, followed by DNA extraction and gene detection using PCR. The results showed the presence of the mrkA gene in 91.6% of the isolates, while the mrkD and fimH genes were each in 87.5% of the isolates. The high percentage of biofilm gene detected, should increase health workers and stakeholder awarness about the impact.

Abstract

Background: Squash orange squash is a natural drink with the ingredients consist of ice and oranges that are sold in many places such as eatery and vendor. Iced orange squash might not be safe for consumption because bad quality oranges, unclean water, unhygienic sellers, and unstandarlized environmental sanitation of the beverage processing. These could cause indigestion with the symptoms of nausea, stomachache, vomitting and diarrhea. The bacteria often used as the indicator that causes contamination in the beverage is Coliform, for example Escherichia coli. Objectives: To find out about the food safety of iced orange squash in Southeast Pontianak. Research Metodes: This research is descriptive with cross-sectional approach. This research uses bacterial quantification method with Most Probable Number (MPN) as the technique and purposive sampling as the sampling method. Results: This research indicates that 16 samples of Squeezed Orange ice exceed the maximum microbial limit that is with the value range of 20 MPN/100 ml - >1100 MPN/ 100 ml contaminated with Coliform (100%) and Escherichia coli (100%). Conclusion: The tested iced orange squash drinks are not safe to consume and unstandardized according to Permenkes 2010, National Standard of Indonesia and World Health Organization.

Abstract

Latar Belakang: Resistensi Proteus mirabilis meningkat; eksplorasi antibakteri bawang hitam dan bawang putih siung tunggal menjadi alternatif. Tujuan: Menilai serta membandingkan potensi antibakteri ekstrak etanol herbal bawang hitam siung tunggal serta herbal bawang putih siung tunggal terhadap Proteus mirabilis. Metode: Merupakan penelitian true experiment dengan melakukan observasi setelah perlakuan diberikan. Uji antibakteri menggunakan metode difusi cakram. Uji statistik memakai rumus uji Shapiro Wilk, uji alternatif menggunakan rumus Kruskal Wallis serta rumus uji Mann Whitney sebagai penutup. Hasil: Percobaan memperlihatkan ekstrak etanol bawang putih siung tunggal tidak memiliki aktivitas antibakteri apapun pada semua konsentrasi uji. Sementara itu, ekstrak etanol tanaman herbal bawang hitam siung tunggal dengan kepekatan 75% serta 100% menunjukkan adanya zona jernih (zona antibakteri) dengan ukuran masing-masing 8,2 milimeter dan 8,4 milimeter. Simpulan: Bawang hitam siung tunggal memiliki potensi dalam mencegah pertumbuhan bakteri Proteus mirabilis, dengan konsentrasi minimum efektif sebesar 75%.

Abstract

Pityriasis versicolor is a superficial fungal infection characterized by changes in skin pigment due to stratum corneum colonization by Malassezia furfur. The Increasing of antifungal resistance and high recurrence rate, requires alternative treatment of medicinal plants. Pegagan (Centella asiatica L.) is known to produce secondary metabolites that have antifungal activity produced by endophytic bacteria that live on pegagan plant tissue. The study aimed to determine the antifungal activity of pegagan endophytic bacteria isolates to Inhibit the Growth of Malassezia furfur. The endophytic bacteria isolates were re cultured on NA media with a streak plate method. Antifungal activity test used the disk diffusion method which is characterized by the formation of inhibitory zones. Total of 37 endophytic bacteria isolates were successed re cultured, and there were 15 endophytic bacterial isolates that have potentcy as antifungal agent with inhibitory zones ranged from 6.5 to 15.52 mm. The most potential isolate was similarity with genus Pseudomonas. The secondary metabolites contained of alkaloids, terpenoids and saponins. The potential Centella asiatica endophytic bacteria had antifungal activity and similarity with the genus Pseudomonas.

Abstract

Background: During the COVID-19 pandemic, teaching and learning activities have to be conducted online. Many students have difficulty in dealing with online learning, and it may become a stress factor. Stress may cause several dermatological disorders such as acne. Objective: To determine the relationship between stress caused by online learning and acne severity during the COVID-19 period. Methods: A cross-sectional study with observational analytic research design. The population is Medical Study program students, Faculty of Medicine, Tanjungpura University. The sampling method uses the Proportionate Stratified Random Sampling technique with a minimum of 169 samples; 57 third-year students, 55 second-year students, and 57 first-year students. Stress level was measured with the MSSQ questionnaire, acne severity was measured with GAGS. Bivariate analysis used the Spearman correlation test. Results: A total of 62.7% of males and 46.8% of females experienced a moderate level of stress on online learning. A total of 66.7% of males and 56.4% of females had moderate acne. Spearman’s test for the relationship between online learning stress levels and acne severity obtained a value of 0.048 (p <0.05) and r = 0.152. Conclusion: The relationship between the stress level of online learning and the severity of acne among Medical Study program students at, the Faculty of Medicine, Tanjungpura University was very weak. Abstract

Irrational use of antibiotics is currently still a major health problem in the world. Worldwide around 50% of medicines are sold, distributed, and given to patients inappropriately. Irrational use of antibiotics can trigger antibiotic resistance. Knowledge and behavior regarding antibiotic stewardship are important in preventing antibiotic resistance. This cross-sectional research was conducted to determine the relationship between knowledge and behavior regarding antibiotic stewardship among healthcare workers. This study's 58 respondents were general practitioners/dentists, specialist doctors/dentists, and pharmacists. The instrument used in this study was a valid and reliable questionnaire. The data analysis used was univariate analysis and Spearman analysis with IBM SPSS Statistics 26 and significance set as p0.05. In this study, the results showed that the majority of respondents worked as specialist doctors/dentists (53.44%), aged in the range 26-35 years (36.20%), of the type female (58.62%), have work experience for more than 10 years (39.65%) and all of the surveyed respondents have good knowledge and behavior (100%). Spearman’s rank analysis showed no significant relationship between knowledge and antibiotic stewardship behavior (p0.05). The high level of expertise of respondents does not significantly affect the level of behavior because it can influence several factors, both internal and external. Further study is needed to identify other factors influencing antibiotic stewardship and rational use of medicine.

Abstract

 AbstrakTingginya angka kejadian jerawat dan meningkatnya resistensi terhadap antimikroba memerlukan alternatif pengobatan yang berasal dari bahan alam. Beberapa studi menunjukkan bahwa bakteri endofit tertentu memproduksi senyawa bioaktif yang memiliki efek bagi kesehatan, terutama bakteri endofit yang diisolasi dari tumbuhan obat. Pegagan (Centella asiatica L.) merupakan tumbuhan obat yang mempunyai banyak manfaat seperti untuk mengobati masalah kulit, menyembuhkan luka, serta menjadi agen antimikroba. Penelitian ini bertujuan untuk mengetahui aktivitas antimikroba isolat bakteri endofit daun pegagan terhadap Propionibacterium acnes. Pengujian aktivitas antimikroba dilakukan dengan metode difusi cakram melalui pengukuran diameter zona hambat. Bakteri endofit yang didapat dari penelitian sebelumnya dikarakterisasi berdasarkan ciri-ciri dari morfologi koloni, morfologi sel, dan aktivitas biokimia. Sembilan belas isolat bakteri endofit memiliki aktivitas antimikroba terhadap Propionibacterium acnes, ditandai dengan terbentuknya zona hambat berdiameter antara 11,46–25,34 mm. Dua isolat yang memiliki kemampuan aktivitas antimikroba yang paling besar, yaitu isolat nomor 3 dan 17 dengan hasil identifikasi termasuk ke dalam genus Aeromicrobium. Berdasarkan hasil penelitian dapat disimpulkan bahwa bakteri endofit dari daun pegagan (Centella asiatica (L.) berpotensi sebagai antimikroba terhadap bakteri P. acnes.AbstractThe high prevalence of acne cases and the increasing resistance to antimicrobials requires alternative treatments that originate from natural ingredients. Several studies have shown that certain endophytic bacteria produce bioactive compounds that benefits human health, especially those whom isolated from medicinal plants. Gotu kola or pegagan (Centella asiatica L.) is one of medicinal plant that has many benefits such as treating skin problems, healing wounds, and also known for an antibacterial agent. The aim of the study was to determine the antimicrobial activity of the endophytic bacterial isolates of pegagan against Propionibacterium acnes. The antimicrobial activity test were carried out using the disc diffusion method by measuring the diameter of the inhibition zone within isolate. Endophytic bacteria obtained from previous studies were characterized based on the characteristics of colony morphology, cell morphology and biochemical activity. 19 isolates of endophytic bacteria showed antimicrobial activity against P. acnes, indicated by the formation of inhibition zones ranging from 11.46 to 25.34 mm. Two isolates that had the greatest antibacterial activity, namely isolates number 3 and 17 were identified and belongs to the genus Aeromicrobium. Based on the results of this study, it can be concluded that endophytic bacteria from pegagan (Centella asiatica L.) leaves has potential as an antibacterial agent against Propionibacterium acnes bacteria.
