Abstract

Introduction: Malaria is one of global health problems. Splenomegaly is one of malaria symptoms. Antimalarial drug resistance had been reported. Alternative treatment is by using traditional medicinal plants such as kelakai (Stenochlaena palustris). Kelakai contains alkaloid and flavonoid which had been reported to have antimalarial activity. The aim of this study was to discover antimalarial activity of ethanol extract of kelakai leaves to parasitemia and splenomegaly of Plasmodium berghei ANKA in infected BALB/c mice.Methods: This study was based on a modified Peter test using BALB/c mice infected with P. berghei ANKA treated with ethanol extract of kelakai leaves, with chloroquine diphosphate as a positive control. The negative control was P. berghei ANKA infected mice without any additional treatment. Administration of ethanol extract of kelakai leaves was performed for 4 days with a serial doses of 100, 10, and 1 mg/kg body weight. The positive control was given chloroquine diphosphate 20 mg/kg body weight. Parasitemia was observed daily prior to the calculation of the percentage of parasite growth and parasite growth inhibition. At the end of the test, the mice were sacrificed and spleens were isolated to measure their sizes. Probit analysis was performed to obtain ED50 to find the effect of extract in parasite killing by 50%. Spearman test was performed to analyze the correlation of doses of extract and splenomegaly.Results: Parasitemia growth inhibition was directly proportional to the dose. Higher parasitemia inhibition was obtained at higher doses and vice versa. Result of probit analysis showed an ED50 was 77.05 mg/kg body weight. Statistical analysis resulted in insignificant correlation between doses and splenomegaly p = 1.0 (significancy < 0.05).Conclusion: Ethanol extract of kelakai leaves possessed good antimalarial activity and there was no correlation between extract doses and splenomegaly in Plasmodium berghei ANKA-infected mice.

Abstract

The community service activity was carried out with the partner of Karang Taruna Bimantara, as a youth organization located in kampung Kalikepiting Jaya VI, RT 5, RW 5, Kelurahan Pacarkembang, Kecamatan Tambaksari, Surabaya City. This activity was aimed at reviving this organization which has been inactive for a long time and empowering the youths in preventing the transmission of Covid-19. The activities offered were providing enlightenment about Covid-19, assistance in producing liquid soap, hand sanitizers and non-medical cloth masks which could be developed into productive economic businesses. The implementation of these activities also involved the organization of Family Welfare Education (Pendidikan Kesejahteraan Keluarga or PKK) which was an organization of mothers in this kampung.  The enlightenment were included Covid-19 and diseases transmitted by mosquitoes, their prevention and transmission. By producing the items than can be used to prevent transmission of Covid-19 followed by obedience to the health protocols, they could participate in preventing the transmission of Covid-19. In order to increase their knowledge on productive economy, the enlightenment on productive economy, marketing of goods and financial management of business, the enlightenment was also given. Hopefully the activities which can be developed into a productive economy business can increase the income of this kampung. The enlightenment provided pre-test and post-test. Comparison of both tests resulted in an insignificant increased in knowledge on both topics. More participants and more questions are recommended to improve the more significant scores of tests. The sales of liquid soap and hand sanitizer resulted in profit can be used to reproduce. Furthermore, more assistance is needed to develop a business into a productive economy.abstrakKegiatan pengabdian masyarakat ini dilaksanakan bersama mitra Karang Taruna Bimantara yang berada di kampung Kalikepiting Jaya VI, RT 5, RW 5, Kelurahan Pacarkembang, Kecamatan Tambaksari, Kota Surabaya. Kegiatan ini bertujuan untuk menghidupkan kembali organisasi yang sudah lama tidak aktif ini dan memberdayakan para pemuda dalam rangka pencegahan penularan Covid-19. Kegiatan yang ditawarkan antara lain adalah penyuluhan tentang Covid-19, bantuan pembuatan sabun cair, hand sanitizer dan masker kain non medis yang dapat dikembangkan menjadi usaha ekonomi produktif. Pelaksanaan kegiatan ini juga melibatkan ibu-ibu yang tergabung dalam organisasi PKK (Pendidikan Kesejahteraan Keluarga) yang merupakan wadah ibu-ibu di kampung ini. Penyuluhan yang diberikan adalah mengenai Covid-19 dan penyakit yang ditularkan oleh nyamuk, pencegahan dan penularannya. Dengan memproduksi barang-barang yang dapat digunakan untuk mencegah penularan Covid-19 diikuti dengan ketaatan terhadap protokol kesehatan, mereka dapat berpartisipasi dalam mencegah penularan Covid-19. Untuk menambah pengetahuan mengenai ekonomi produktif juga diberikan penyuluhan mengenai ekonomi produktif, pemasaran barang dan cara pengelolaan keuangan hasil usaha. Diharapkan dengan adanya kegiatan yang dapat dikembangkan menjadi usaha ekonomi produktif ini dapat meningkatkan pendapatan RT setempat. Dalam penyuluhan diberikan pre-test dan post-test. Perbandingan kedua tes menghasilkan peningkatan pengetahuan yang tidak signifikan pada kedua topik tersebut. Untuk meningkatkan nilai tes yang lebih signifikan, maka diperlukan jumlah peserta dan pertanyaan yang lebih banyak. Penjualan sabun cair dan hand sanitizer menghasilkan keuntungan yang dapat dimanfaatkan untuk memproduksi lagi. Pendampingan dibutuhkan untuk mengembangkan usaha ini menjadi ekonomi produktif.

Abstract

This community service activity was carried out with the partner of the Muslim Metal Islamic Boarding School, Pasuruan. This activity aims to improve the habits of clean and healthy living behavior (personal hygiene. The activities offered include examination of intestinal parasitic infections for residents of the Muslim Metal Islamic Boarding School, counseling on intestinal parasitic infections and personal hygiene and improvement of PHBS facilities. The counselling given is about various kinds of parasites that cause intestinal infections, transmission of intestinal parasitic infections, and how to prevent intestinal parasitic infections by implementing personal hygiene (PHBS). By knowing about intestinal parasites and how they are transmitted, be expected that they will be able to avoid infection with intestinal parasites by practicing PHBS discipline.

Abstract

Malaria remains an important health problem in Indonesia with the highest transmission in Papua Province, an easternpart of this country. The genetic diversity of malaria parasites is the main problem in understanding several aspects ofmalaria infections and the dynamics of their transmission, which also play a role in the development of a vaccine.Plasmodium falciparum is the deadliest of the human malaria parasites. Plasmodium falciparum glutamate-rich protein(Pfglurp) is one of the many erythrocytic stages antigens currently under development for a vaccine. The Pfglurp gene hasbeen extensively used as a marker to investigate the genetic diversity, Multiplicity of Infection (MOI), the level of malariatransmission, immunity against malaria, as well as a discriminatory instrument to distinguish new from recrudescentinfections of the field parasite population. Thus, this genotyping study aimed to find out the genetic population ofP.falciparum at the Merauke District, Province of Papua, Indonesia. DNA samples were isolated from Dried Blood Spots(DBS) obtained from P.falciparum infected patients in the Regional Public Hospital of Merauke, Province of Papua, Indonesiaduring May 2019-July 2019. The isolated DNAs were then amplified for nested Polymerase Chain Reaction (PCR) prior toPfglurp genotyping. The glurp gene was identified in all 51 DBS samples of P.falciparum-infected patients, and 18 variants ofallele were found. Among them, 45.10% were found to bear multigenotype infections. The size of the dominant allele(12.5%) was 701-750 bp. The MOI was 1.58. The genetic population of P.falciparum in Merauke Hospital has contained ahigher percentage of multigenotypes compared with monogenotypes indicating the high transmission of malaria in thestudied area.

Abstract

Background: Malaria is still a universal health problem, especially in tropical countries because of high morbidity and mortality rates. Infection by Plasmodium falciparum and Plasmodium vivax could result in asymptomatic disease of malaria and be found in Trenggalek, Jawa Timur. Differences in pathogenesis among affected individuals are affected by many factors, and the immune system is one of them. Among substances involved in the malarial immunity is Tumor Necrosis Factor (TNF)-α and Interleukin (IL)-10, produced by the body's defense system as the reaction to the parasite. Therefore a study was designed to detect the level of TNF-α and IL-10 in asymptomatic malaria patients.Materials and Methods: A cross-sectional study was conducted. Thirty male asymptomatic malaria subjects, age 21 to 60 years were selected. Blood from each subject was collected and the levels of TNF-α and IL-10 were analyzed using enzyme-linked immunosorbent assay (ELISA) method. Significant values considered at p<0.05.Results: There was an increased level of TNF-α with the average of 218.760 pg/µL, and an increased level of IL-10 with an average of 257.574 pg/µL in asymptomatic malaria subjects. In normal person IL-10 level is 12.6 (8.5-16.7) pg/mL and the levels of TNF-α in normal person is 0-1.5 pg/mL because they are not produce. There was a positive correlation of TNF-α with IL-10 (r=0.332; p>0.05), and positive correlation between TNF-α and the rate of hemoglobin (r=0.002; p>0.05). IL-10 was correlated negatively with the rate of hemoglobin (r=-0.363; p<0.05).Conclusion: The results from this study conclude that TNF-α and IL-10 levels increase in asymptomatic malaria subjects.Keywords: asymptomatic malaria, TNF-α, IL-10, parasite, hemoglobin

Abstract

Background: Nowadays, microbial infections remain as the leading cause of infectious diseases and human death worldwide. The use of plant-derived medicines is currently increasing in the treatment of various diseases. Papaya leaves have proteolytic enzymes and phytoconstituents with antimicrobial properties. Rwandan citizens use papaya leaves to treat hair dandruff, wounds and burns.Materials and Methods: Papaya leaves were collected and allowed to dry under the shed at room temperature for 14 days. The powdered plant materials were soaked separately in clean flask and extracted successively using maceration method with water and methanol. Qualitative phytochemical screening was conducted by using specific standard procedures. Antimicrobial activity assays of all the extracts were performed by agar well diffusion method and determined by measuring the zones of inhibition with transparent scale.Results: Phytochemical screening revealed the presence of alkaloids, carbohydrates, tannins, flavonoids, steroids and phenolic compounds. In this observation, all the extracts exhibit significant inhibitory activity against all test pathogens ranging from 2 mm to 26 mm of diameter. Methanol extracts showed the maximum activity against Candida albicans (inhibition zone: 26±0.11 and activity index: 1.23). Minimum inhibition concentration values ranges between 3.175 mg/mL and 12.5 mg/mL.Conclusion: The results indicate that Carica papaya leaves could be very potent source of antimicrobial agents and secondary metabolites that can be used by pharmaceutical industries to produce medicines.Keywords: Carica papaya, antimicrobial, agar well diffusion, phytochemical screening, zone of inhibition, activity index

Abstract

Cassia spectabilis is one of the Indonesian medicinal plants traditionally used to treat different diseases, including malaria. Quality of a drug derived from plants is also influenced by the quality of its raw materials. Thus, in order to assure the quality of products made from plants, it is necessary to standardize the raw materials and following with the antimalarial activity test. The aim of this study was to standardize the quality of C. spectabilis leaves and to evaluate its antimalarial activities of ethyl acetate extract. The fresh material of C. spectabilis leaves were observed its specific and non-specific parameters. In vitro test was done by using Plasmodium falciparum 3D7. In vivo test was done using 4-day suppressive test method against mice infected with P. berghei for four consecutive days. Heme detoxification inhibitory activity test was carried out using the modified Basilico method. The leaves of C. spectabilis meet the quality requirement for raw material of traditional medicine. The ethyl acetate extract showed in vitro antiplasmodial activity against P. falciparum 3D7 and in vivo antimalarial activity against P. berghei infection with IC50 value of 27.28 μg/mL and ED50 value of 1.74 mg/kg, respectively. The extract also showed heme detoxification inhibitory activity with IC50 value of 0.33±0.01 mg/mL. The leaves of C. spectabilis leaves meet the quality requirement and the ethyl acetate extract from standardized C. spectabilis leaves possessed a potential an antimalarial activity which deserves to be further developed.

Abstract

Malaria masih menjadi masalah kesehatan masyarakat di Indonesia, di mana sekitar 80% dari malaria wilayahnya endemik, dengan 45% populasi memiliki risiko infeksi malaria. Prevalensi malaria di Indonesia masih cukup tinggi. Pemberantasan infeksi malaria masih mengalami beberapa kendala, yaitu adanya resistensi Plasmodium terhadap obat antimalaria seperti klorokuin, pirimetamin, sulfadoksin, kombinasi sulfadoksin-pirimetamin (SP) dan artemisinin-based kombinasi therapi (ACT). Penyebaran parasit Plasmodium yang resisten terhadap obat antimalaria mendorong perlunya ditemukan obat baru yang lebih efisien, antara lain pengembangan bahan alam sebagai obat herbal. Meniran merupakan salah satu tanaman yang digunakan secara tradisional oleh masyarakat untuk mengobati penyakit malaria. Pemberian ekstrak etanol meniran dapat mempengaruhi sistem imun. Penelitian ini bertujuan untuk mengetahui efek ekstrak etanol meniran dosis rendah terhadap jumlah limfosit T-CD4+ dan ekspressi interferon gamma (IFN-γ) pada mencit BALB/c yang diinfeksi P. berghei ANKA. Penelitian ini menggunakan Post Test Only Control Design. Sampel dalam penelitian ini sebanyak 25 ekor mencit, yang terdiri dari 5 kelompok. Mencit diberi Ekstrak etanol meniran dengan dosis 40 mg/kgBB dan dosis 120 mg/kgBB selama 7 hari. Dua kelompok yang diberi EEM yaitu kelompok perlakuan 1 (KP1) diberi EEM dosis 40 mg/kgBB dan kelompok Perlakuan 2 (P2) diberi EEM dosis 120 mg/kgBB dan diinfeksi P. berghei sebanyak 1x106. Kelompok kontrol positif diinfeksi Plasmodium berghei sebanyak 1x106 tanpa ekstrak etanol meniran dan kelompok negatif kontrol 1 (KN1) diberikan EEM dosis 40 mg/kgBB dan kontrol negatif 2 (KN2) diberikan EEM dosis 120 mg/kgBB tanpa diinfeksi P. berghei ANKA. Pengamatan parasitaemia dilakukan setiap hari selama 4 hari pada sediaan darah tipis yang diwarnai dengan Giemsa 10%. Jumlah sel T CD4+ dan ekspresi IFN-γ dilakukan dengan metode flocytometry. Analisis data menggunakan Oneway ANOVA. Hasil penelitian ini menunjukkan pemberian ekstrak etanol meniran dengan dosis 40 mg/kgBB dan dosis 120 mg/kgBB tidak berpengaruh terhadap jumlah sel T CD4+ pada semua kelompok perlakuan, tetapi berpengaruh secara signifikan terhadap ekspresi IFN- γ pada semua kelompok perlakuan

Abstract

ABSTRAK  Penyakit Demam Berdarah Dengue (DBD) menimbulkan sakit sehingga menghentikan produktivitas manusia, kematian, bahkan wabah (Kejadian Luar Biasa/ KLB). DBD ditularkan melalui perantaraan nyamuk. Tujuan kegiatan pengabdian masyarakat ini adalah memberikan penguatan materi kepada para guru tentang bahaya dan kewaspadaan terhadap nyamuk. Kegiatan ini dilaksanakan secara daring menggunakan zoom. Peserta kegiatan ini adalah semua guru mata pelajaran Biologi yang tergabung di dalam MGMP Biologi SMA Kabupaten Sidoarjo. Instrumen yang digunakan adalah laptop, materi presentasi format ppt, zoom, dan google form. Hasil kegiatan adalah kegiatan berjalan dengan lancar. Ada peningkatan pengetahuan para peserta dari dimana rata-rata nilai pre test = 53 dan post test = 81. Kata Kunci: Nyamuk, guru, pengetahuan  ABSTRACT  Dengue hemorrhagic fever (DHF) causes pain that stops human productivity, death, even plague. DHF is transmitted through mosquito intercession. These community service activities aim to provide material reinforcement to teachers about the dangers and vigilance against mosquitoes. This activity is carried out online using zoom. Participants of this activity are all teachers of Biology subjects who are members of MGMP Biology SMA Sidoarjo. The instruments used are laptop, ppt format presentation material, zoom, and google form. The result of the activity is that the training runs smoothly. Abstract

Soil transmitted helminth (STH) infections is kind of helminth infection which transmitted by contact with warm and moist soil especially affect the farmer. The purpose of this study was to investigate the profile of STH infection and the intensity of STH infection among farmers in Gelgel Village, Klungkung, Bali. The cross sectional study was conducted in Gelgel Village, Klungkung District, Bali and obtained stool and quitionaires from farmers by informed consent. Kato-Katz technique was used to diagnose the STH infection and to determine the intensity of infection based on the number of eggs per gram feces (EPG). Total 162 stool samples were collected from farmers aged 25-80 years, 22 farmers was positive of STH infection and 140 farmers was negative of STH infection. The result showed prevalence of STH infection was 13,5%, single infection of A. lumbricoides was 1.85%, 9.26% of T. trichiura single infection, and 0.61% of Hookworm single infection. The mixed infection was detected that were 1.23% of A. lumbricoides with T. trichiura, and 0.61% of A. lumbricoides with Hookworm. STH infection is the health problem among farmer. The farmer must be admitted antihelminthic drugs for preventing and eleminating STH infection.
