Abstract

Kunyit Hitam (Kaempferia parviflora) adalah tanaman rimpang berwarna ungu kehitaman yang banyak manfaat sebagai bahan obat tradisional. Perbanyakan bibit kunyit hitam secara konvensional tidak memungkinkan untuk budidaya skala besar, dikarenakan membutuhkan waktu yang cukup lama dan tidak efisien. Rendahnya pasokan bahan tanaman berkualitas dan masalah perkecambahan yang rendah menjadi dasar dalam upaya pengembangan kunyit hitam secara in vitro. Perbanyakan kunyit hitam melalui multiplikasi tunas dapat mempercepat produksi bibit yang efisien. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian berbagai konsentrasi hormon BAP terhadap multiplikasi kunyit hitam. Rancangan penelitian menggunakan RAL dengan faktor konsentrasi hormon BAP yang terdiri dari 5 taraf konsentrasi antara lain BAP 0 mg/L (P0), BAP 2 mg/L (P1), BAP 4 mg/L (P2), BAP 6 mg/L (P3), dan BAP 8 mg/L (P4). Hasil penelitian menunjukkan bahwa, perlakuan BAP 2 mg/L memperlihatkan hasil terbaik dengan kedinian tunas tercepat yaitu 8,3 hari setelah tanam, jumlah tunas terbanyak yaitu 5,8 tunas, rata-rata tunas tertinggi mencapai 4,8 cm, dan jumlah daun terbanyak yaitu 8,3 helai. Perlakuan BAP 2 mg/L menghasilkan persentase tunas dan akar 100%.

Abstract

Vanilla is a plant that has many benefits, high selling value and potential to be developed. Conventional propagation of vanilla such as through cuttings has not been effective, so a more effective method of propagation is needed, one of which is propagation through tissue culture. In-vitro propagation of vanilla can be through shoot multiplication to produce fast plantlets. One of the successes of shoot multiplication is influenced by the hormone given. This study aims to determine the effect of giving a combination of 2,4-D and BAP on the multiplication of vanilla shoots. The study design used CRD with a combination factor of 2,4-D (concentration 0.5 mg/L, 1.0 mg/L, and 1.5 mg/L) and BAP (concentration 0.25 mg/L, 0.5 mg/L, and 0.75 mg/L). The explants used were vanilla stem segments. Based on the research results, the combined treatment with concentrations of 2,4-D 1 mg/L and BAP 0.5 mg/L had a very significant effect on the multiplication of vanilla shoots with the fastest emergence of shoots with an average of 6.6 wap, the highest number of shoots was 7 shoots, and the survival percentage was 100%.

Abstract

Tanaman tembakau (Nicotiana tabacum L.) merupakan tanaman musiman bernilai ekonomi tinggi, namun produksinya di Indonesia terus mengalami penurunan dalam beberapa tahun terakhir. Penggunaan varietas hibrida dapat menjadi solusi karena memiliki produktivitas dan ketahanan penyakit yang lebih baik, tetapi pembentukannya secara konvensional memerlukan waktu lama. Teknik kultur antera dapat digunakan untuk mempercepat pembentukan galur murni karena menghasilkan tanaman homozigot hanya dalam satu generasi. Penelitian ini bertujuan untuk menganalisis interaksi antara jenis media kultur (MS dan Chu’s N6) dengan penambahan beberapa konsentrasi Nanopartikel silver (AgNPs) yaitu 0,0; 2,5; 5,0; 7,5; dan 10,0 mg L⁻¹ terhadap pertumbuhan antera tembakau secara in vitro. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 10 perlakuan dan 3 ulangan, kemudian data dianalisis menggunakan ANOVA dan diuji lanjut DMRT dengan taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa media MS merupakan media terbaik dalam mempercepat dan memperbanyak induksi swelling, kalus, tunas, dan akar, sedangkan konsentrasi AgNPs terbaik diperoleh pada 2,5 mg L⁻¹ yang mampu meningkatkan respons pertumbuhan serta sedikit mengurangi tingkat kontaminasi media. Hasil penelitian ini menunjukkan bahwa kombinasi media MS dan AgNPs dosis rendah mampu meningkatkan efisiensi regenerasi antera tembakau dan berpotensi digunakan sebagai protokol standar dalam produksi galur murni secara cepat, efisien, dan steril di laboratorium kultur jaringan. ABSTRACT Tobacco (Nicotiana tabacum L.) is a seasonal crop with high economic value, but its production in Indonesia has continued to decline in recent years. The use of hybrid varieties can be a solution because they have better productivity and disease resistance, but their conventional formation requires a long time. Anther culture techniques can be used to accelerate the formation of pure lines because they produce homozygous plants in just one generation. This study aims to determine the interaction between the types of culture media (MS and Chu's N6) with the addition of several concentrations of silver nanoparticles (AgNPs), namely 0, 2.5, 5, 7.5, and 10 mg L⁻¹ on the growth of tobacco anthers in vitro. The study used a Completely Randomized Design (CRD) with 10 treatments and 3 replications, then the data were analyzed using ANOVA and further tested with DMRT at the 95% level. The results showed that MS medium was the best medium in accelerating and increasing the induction of swelling, callus, shoots, and roots, while the best concentration of AgNPs was obtained at 2.5 mg L⁻¹ which was able to increase the growth response and slightly reduce the level of media contamination. The findings of this study indicate that the combination of MS medium and low-dose AgNPs can increase the efficiency of tobacco anther regeneration and has the potential to be used as a standard protocol in the production of pure lines quickly, efficiently, and sterilely in tissue culture laboratories.

Abstract

Anther culture is an effective biotechnological approach to accelerate the production of homozygous lines in tobacco breeding. This study evaluated the interaction between culture media (Murashige and Skoog/MS and Chu’s N6) and silver nanoparticle (AgNP) concentrations on in vitro anther regeneration of tobacco (Nicotiana tabacum L.). The experiment was arranged in a completely randomized design with ten treatment combinations consisting of two basal media and five AgNP concentrations (0, 2.5, 5, 7.5, and 10 ppm), each replicated three times. Quantitative parameters included callus formation, shoot regeneration, rooting response, regeneration percentage, and contamination rate. MS medium supplemented with 2.5 ppm AgNPs (K2) produced the optimal response, showing 100% callus formation and the highest shoot regeneration percentage (89%). Root formation was observed only in treatments K2 and K4. In contrast, higher AgNP concentrations (≥7.5 ppm), particularly in Chu’s N6 medium, significantly reduced regeneration percentages (ANOVA, p<0.05). The promotive effect of low AgNP concentration is associated with suppression of ethylene activity, antimicrobial action, and regulation of reactive oxygen species (ROS), which collectively enhance cell division and organogenesis. These findings indicate that MS medium supplemented with 2.5 ppm AgNPs is optimal for tobacco anther regeneration and suitable for doubled-haploid production.
