Abstract

The common palm civet (Paradoxurus hermaphroditus) is one of the mammalian in Indonesia which have anal gland in female and fefemale that located in the lower tail. This research was aimed to describe the morphology of anal gland of common palm civet female. One female of common palm civet was used in this research which observed macroscopic and microscopic.The result showed that common palm civet have a pair of anal gland which hasshaped like ball and located in the anal sacs wall’s in the ventrolateral of anus.Each anal gland has a ostium of anal canal dorsolaterally in the cutaneous zone of anal canal . Microscopic observation showed a well developed sebaceous gland and apocrine sweat gland in the wall of anal sac. Sebaceous and apocrine glands were excreted their product into main ductus to the anal canal.

Abstract

This research aimed to compare the morphology and carbohydrate content of parotid andmandibular glands of goat, cat and pig (herbivore, carnivore and omnivore). The microscopic observationwas done using histochemical method with hematoksilin eosin (HE), alcian blue (AB) pH 2.5 andperiodic acid Schiff (PAS) staining methods.The results showed that the parotid and mandibular glandsconsist of acinar cells and ducts. The parotid gland of goat and pig were serous while mixed in the cat.The mandibular gland of goat, cat and pig were mixed type. The acinar of mandibular gland is composedof serous and mucous cells in which the mucous cell predominates. The AB and PAS stain showed weakto high concentration of acid and neutral carbohydrates found in the mucous cells of mandibular gland.However, these carbohydrates were not found in the serous cells of both glands, while acid carbohydratewas found in the parotid gland of cat. The present result suggested that the carbohydrates in the salivaof the goat, cat and pig might be produced mainly by the mucous cells.Keywords: parotid gland, mandibular gland, PAS, AB pH 2.5

Abstract

This study utilized conventional and immunohistochemical methods, to describe the morphology and distribution of glucagon and insulin cells in the pancreas of adult male babirusa (Babyrousa babyrussa). The results of research showed that the endocrine portion (Langerhans islet) spread evenly on the acinar portion of pancreas. Observation using immunohistochemical method showed that glucagon cells were distributed on the perifer, whereas insulin cells in the center of the Langerhans islet of pancreas. In general, the number of insulin cells was higher than glucagon cells. It is interesting to note, in the pancreas babirusa also found that neonatal elements similar to the Langerhans islet, but have larger sizes (ranging between 200 -700 mm). The picture is similar to the large Langerhans islet was reported in ruminant.  These cells give a negative reaction to insulin and glucagon antisera.

Abstract

The presence of glycoconjugates in caput, corpus, and cauda epididymis of the Javan muntjacs was performed using lectin histochemical method. The epididymis organs were obtained from two adult male Javan muntjacs in hard antler stage and then immediately fixed in Bouin’s solution and processed in histological manner. Six types of biotinylated lectins namely Peanut agglutinin (PNA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), and Ulex europaeus agglutinin I (UEA I) were applied to detect the specific glycoconjugates in the epididymal tissue of muntjacs. The result showed the presence of glycoconjugates were detected by five types of lectins except UEA I in caput, corpus, and cauda epididymis with specific distribution and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, N-acetyl-galactosamine, N-acetyl-glucosamine, and sialic acid were stained with various intensity in epithelial cells, basal cells, stereocilia, spermatozoa, and stroma of caput, corpus, and cauda epididymis. The presence of sialic acid on the surface of spermatozoa detected by WGA are thought to be play a role on sperm protection during their maturation and transit along epididymal lumen. In conclusion, the difference of glycoconjugates distribution pattern and lectin binding in the epididymal caput, corpus, and cauda of the Javan muntjac indicates the regional differences in epididymal function in spermatozoa maturation.

Abstract

The aim of this research was to analyze the anti-oxidant activities of ethanol extract of mahogany seeds on the liver tissues of diabetic experimental rats to overcome oxidative stress condition. This study used male Rattus norvegicus strain Sprague Dawley which were divided into 5 groups: (i) negative control group (K-), (ii) positive control group/diabetes mellitus (DM) (K+), (iii) DM group treated with 500 mg/kg BW ethanol extract of mahogany seeds (EM), (iv) DM group treated with acarbose (KO), and (v) non-DM group treated with 500 mg/kg BW ethanol extract of mahogany seed (KE). Alloxan with dose of 110 mg/kg BW was used to induce diabetes in rats. The treatments were conducted for 28 days. At the end of treatment, the liver tissues were collected and analyzed for malondialdehyde (MDA) content using thiobarbituric acid reactive substance (TBARS) method and anti-oxidant cooper zinc superoxide dismutase (Cu,Zn-SOD) content using immunohistochemical technique. The result showed that level of MDA were significantly different (P0.05) in K-; K+; EM; KO; and KE with values were 0.81±0.17; 1.19±0.26; 1.10±0.04; 0.95±0.13; and 0.92±0.0 µmol/gram, respectively. The anti-oxidant content (Cu, Zn-SOD) of rat liver tissue in K+ lower than in K-, while anti-oxidant content in EM and KE were higher compared to K+. The study concluded that ethanol mahogany seeds extract decreased blood glucose levels and increased the anti-oxidant status in the liver tissues of diabetic rats.

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 µg/ml of concentration for PNA lectins and 15µg/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis. Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak

Abstract

Indo-Pacific bottlenose dolphin (Tursiops aduncus) is a marine mammal that lives in relatively small populations. The geographic ranges of this species are susceptible to the effects of human activities, thereby necessitating conservation efforts to prevent extinction. Therefore, this study aimed to evaluate the daily quality of dolphin sperm after several days of refrigeration. The sperm of two male dolphins were stored at 4oC for 4 days, and the quality was observed daily to determine the motility, viability, membrane integrity, and sperm abnormalities. Sperm samples were divided into four groups, consisting of two centrifuged followed by the removal of seminal plasma, and two groups without centrifugation, containing 100x106 and 200x106 sperm/ml each. After liquid storage, the motility of sperm was 63-75% with no significant reduction in the first 3 days. Sperm viability following storage was 65-75% and the percentage with abnormal morphology ranged from 2-6%. Furthermore, there was no significant increase in abnormal morphology of sperm on any day of storage for 3 days. Sperm membrane integrity was 36-49%, with no significant reduction in the membrane integrity in the first 2 days. There was no significant difference in sperm quality, although centrifugation and removal of seminal plasm had a slight effect. The results of this study showed that Indo-Pacific bottlenose dolphin sperm could be stored for a short period as liquid storage while maintaining a quality that allows for future use.
